In this study, the combined effects of procaine and hyperthermia on the growth of hematopoietic progenitors (GM-CFU) and ieukemic progenitors (L-CFU) were examined to determine if this combination resulted in a great selective killing of leukemlc cells than that achieved by procaine or heat alone. When the cells were treated simutaneously with procaine (2 mM) and hyperthermia (42℃) for one hour, the killing of L-CFU was enhanced considerably whereas GM-CFU were not markedly affected. These data Indicate that the combined treatment with procain and hyperthermia might offer an efficient mean to selectively purge residual leukemlc cells in vitro. Procaine with hyperthermia may have a role in clinical autoiogous bone marrow transplantation for acute leukemia.展开更多

Cardiac myocytes were isolated by collage-nase using Langendorff’s perfusion apparatus, and [Ca2+], was measured using the fluorescent indicator Fain-2. The normal[Ca2+], was 101. 3 ± 21. 3 nmol/L in cardic myocytes. PBS at various postburn home could very significantly increase the[Ca2+]i (P< 0. 01 ) and, 6 h PBS had the strongest effect. However, no significant difference was found between the effects of2 h PBS and 4 h PBS (P >0. 05 ). Both calcium channel antagonist verapamil(30 umol/L) and the inhibitor of ryanodine receptoron sarcoplasmic reticulum procaine (2 mmol/L), very significantly inhibited the action of 6 h PBS, with the inhibition rate of47. 7% and 67. 6% respectively. The inhibiting rate of procaine was significantly greater than that of verapamil (P < 0. 01 ). Theresults suggested that PBS could stimulate the increase of [Ca2+], in isolated cardiac myocytes of adult rats, in which calcium release from intracellular stores might play greater roles. Anesthetic residues in fish represent a potential risk to human health.Therefore,it is important to develop a sensitive and broad-specific method for the detection of anesthetics.In this study,we developed a colloidal...Anesthetic residues in fish represent a potential risk to human health.Therefore,it is important to develop a sensitive and broad-specific method for the detection of anesthetics.In this study,we developed a colloidal gold-based quadruplex immunochromatographic(Qua-ICS)assay using four highly sensitive monoclonal antibody immunotherapy(mAbs)that simultaneously detected 11 anesthetic residues in fish within 10 min.The colorimetric and cut-off values(COVs)for procaines,eugenols,bupivacaines,and tricaine(TMS)were 0.37–1.1 and 3.3–10,11–222 and 100–2000,0.37 and 3.3,and 111 and 10,000µg/kg,respectively.Quantitative analysis was achieved with a portable strip-reader,and the detection ranges were 0.15–2.6,6.3–677,0.13–2.8,and 83–1245µg/kg for procaines,eugenols,bupivacaines,and tricaine,respectively.Our developed method was reliable and accurate according to the recovery test results and analyses of real samples.Therefore,the strip can be used as an alternative method for the rapid detection of anesthetic residues in fish.展开更多
