Pathogenesis-related proteins (PRs) play many important roles in plant defense response against pathogen attack. To better understand the molecular mechanism of PR genes involved in wheat adult plant resistance (APR) to stripe rust, based on a differentially expressed transcribed derived fragment (TDF), a novel PR gene from wheat cv. Xingzi 9104 infected by the Puccinia striiformis Westend f. sp. tritici Erikss. pathotype CY32, which was highly similar to the maize ZmPRIO gene and designated as TaPRIO, was identified using in silico cloning and RT-PCR method. This novel TaPRIO gene was predicted to encode a 160-amino acid protein with a deduced molecular weight of 17.06 kDa and an isoelectronic point (pI) of 5.19. An amino acid sequence analysis of TaPR10 demonstrated the presence of a typical conserved domain of pathogenesis related protein Bet v I family. Multiple alignment analysis based on the amino acids encoded by 10 different PRIO genes from maize (Zea mays), rice (Oryza sativa), broomcorn (Sorghum bicolor), and wheat (Triticum aestivum) indicated that PR proteins of class 10 was conserved among the 4 plant species with about 80% similarity. DNA sequence of TaPRIO suggested the presence of one 84-bp intron with the splicing sites of GT-AT bi-nucleotide sequence between 188 and 271 bp. Using a real-time quantitative RT-PCR (qRT-PCR), expression profiles of TaPRIO revealed that at the adult-plant stage, TaPRIO transcript was up-regulated as early as 12 h post-inoculation (hpi), with the occurrence of maximum induction at 24 hpi. At the seedling stage, TaPRIO was also slightly induced 18 hpi. However, the transcript amount was relatively lower than that of the adult-plant stage. Taken together, these results suggest that TaPRIO may participate in wheat defense response of APR to stripe rust. 展开更多

The purpose of this study is to explore the influence of co-suppressing tomato ACC oxidase Ⅰ on the expression of fruit ripening-related and pathogenesis-related protein genes, and on the biosynthesis of endogenous ethylene and storage ability of fruits. Specific fragments of several fruit ripening-related and pathogenesis-related protein genes from tomato (Lycopersicon esculentum) were cloned, such as the l-aminocyclopropane-1-carboxylic acid oxidase 1 gene (LeAC01), 1- aminocyclopropane-l-carboxylic acid oxidase 3 gene (LeAC03), EIN3-binding F-box 1 gene (LeEBF1), pathogenesis-related protein 1 gene (LePR1), pathogenesis-related protein 5 gene (LePR5), and pathogenesis-related protein osmotin precursor gene (LeNP24) by PCR or RT-PCR. Then these specific DNA fragments were used as probes to hybridize with the total RNAs extracted from the wild type tomato Ailsa Craig (AC++) and the LeAC01 co-suppression tomatoes (V1187 and T4B), respectively. At the same time, ethylene production measurement and storage experiment of tomato fruits were carded out. The hybridization results indicated that the expression of fruit ripening-related genes such as LeACO3 and LeEBF1, and pathogenesis-related protein genes such as LePR1, LePR5, and LeNP24, were reduced sharply, and the ethylene production in the fruits, wounded leaves decreased and the storage time of ripening fruits was prolonged, when the expression of LeACO1 gene in the transgenic tomato was suppressed. In the co-suppression tomatoes, the expression of fruit ripening-related and pathogenesis-related protein genes were restrained at different degrees, the biosynthesis of endogenous ethylene decreased and the storage ability of tomato fruits increased. 展开更多

Soybean(Glycine max(Linn.)Merr.)annual leguminous crop is cultivated all over the world.The occurrence of diseases has a great impact on the yield and quality of soybean.In this study,based on the RNA-seq of soybean variety M18,a complete CDS(Coding sequence)GmPR1L of the pathogenesis-related protein 1 family was obtained,which has the ability to resist fungal diseases.The overexpression vector and interference expression vector were transferred into tobacco NC89,and the resistance of transgenic tobacco(Nicotiana tabacum L.)to Botrytis cinerea infection was identified.The results show that:Compared with the control,the activities of related defense enzymes SOD(Superoxide dismutase),POD(Peroxidase),PAL(L-phenylalanine ammonia-lyase)and PPO(Polyphenol oxidase)in the over-expressed transgenic tobacco OEA1 and OEA2 increased to different degrees,and increased significantly at different infection time points.The activities of defense enzymes in the interfering strains IEA1 and IEA2 were significantly lower than those in the control strains.The results of resistance level identification showed that the disease spot rate of OEA1 was significantly lower than that of the control line,and the disease spot rate of OEA2 was significantly lower than that of the control line.The plaque rate of the interfering expression line IEA1-IEA2 was significantly higher than that of the control line.It is preliminarily believed that the process related protein GmPR1L can improve the resistance of tobacco to B.cinerea. 展开更多

Proteomics techniques were used to study the molecular mechanisms involved in the defense of tomato against late blight (Phytophthora infestans). Proteins were extracted from resistant access BGH-2127 and susceptible cultivar “Santa Clara”. Leaves of the inoculated and non-inoculated (control) genotypes were collected at 0, 2, and 48 h after inoculation and analyzed by two-dimensional electrophoresis (2-DE), followed by identification with mass spectrometry (MALDI TOF-TOF). A total of 56 differentially abundant proteins were identified, of which 39 were resistant genotypes and 17 were susceptible. These proteins were categorized into functional groups of energy and metabolism, photosynthesis, stress and defense, transcription, other proteins, and as un-characterized ones. For access BGH-2127, oxidative stress proteins (2-cis peroxiedoxin BAS1 and 2-cis peroxiredoxin) and thaumatin-like protein showed increase in the relative abundance at 0 and 48 h of inoculation, respectively, and were therefore considered important for the defense mechanism of this genotype. The expression standards evaluated by real-time PCR differed from the results of the proteomic analysis. The protein-protein interaction networks provided important information on the cellular activities involved in the resistance of BGH-2127 late blight. 展开更多

The interactions of pyrogallol red (PR) with proteins and a derivative method for the determination of proteins were proposed. In the pH range 2 56-4 10, PR interacts with proteins, including bovine serum albumin, human serum albumin, lysozyme, chymotrypsin, resulting in enhanced resonance light scattering(RLS) with the maximal peak at 360 0 nm. With the enhanced RLS, proteins in the range 0-5 0(μg/mL can be determined with the determination limits between 10 3 and 32 2 ng/mL depending on different protein. The effects of foreign substances were tested. The method has been applied to the determination of protein in the synthetic samples and human urine with satisfaction results. 展开更多

目的 通过分析错配修复(mismatch repair, MMR)蛋白与ER、PR及p53在子宫内膜癌中的表达情况,探讨其临床病理意义及其对预后的影响。方法 收集2017年1月至2020年10月在十堰市太和医院确诊的226例子宫内膜癌患者的临床病理资料,采用免疫... 目的 通过分析错配修复(mismatch repair, MMR)蛋白与ER、PR及p53在子宫内膜癌中的表达情况,探讨其临床病理意义及其对预后的影响。方法 收集2017年1月至2020年10月在十堰市太和医院确诊的226例子宫内膜癌患者的临床病理资料,采用免疫组织化学法对肿瘤组织进行MMR蛋白、ER、PR及p53检测,分析其表达与临床病理的关系及其对患者预后的影响。结果 226例子宫内膜癌组织中MMR蛋白的缺失率为18.6%(42/226),ER及PR的阴性表达率分别为6.6%(15/226)、11.9%(27/226),p53突变型表达率为59.7%(135/226)。MMR蛋白缺失在肿瘤分级、肌层浸润深度、淋巴脉管浸润(LVSI)及术后辅助治疗中差异具有统计学意义(P<0.05)。p53突变型表达多为非子宫内膜样腺癌,且多为高级别子宫内膜癌(P<0.05)。ER与PR的表达呈正相关,ER阴性表达与组织学类型、肿瘤分级、术后辅助治疗相关(P<0.01),而PR阴性表达还与国际妇产科联盟(FIGO)分期、LVSI及淋巴结转移有关(P<0.05)。p53突变型在ER阴性且PR阴性的子宫内膜癌中表达率高(P<0.05)。结论 MMR、ER、PR和p53蛋白的表达与子宫内膜癌的病变程度相关,在ER和PR阴性表达时,MMR蛋白缺失和p53突变型表达率高提示预后不良,可以通过监测免疫组化结果进行术前手术范围的选择、术后预后的判断并予以辅助治疗。 展开更多

Members of the Pathogenesis Related(PR)10 protein family have been identified in a variety of plant species and a wide range of functions ranging from defense to growth and development has been attributed to them.PR10 protein possesses ribonuclease(RNase)activity,interacts with phytohormones,involved in hormone-mediated signalling,afforded protection against various phytopathogenic fungi,bacteria,and viruses particularly in response to biotic and abiotic stresses.The resistance mechanism of PR10 protein may include activation of defense signalling pathways through possible interacting proteins involved in mediating responses to pathogens,degradation of RNA of the invading pathogens.Moreover,several morphological changes have been shown to accompany the enhanced abiotic stress tolerance.In this review,the possible mechanism of action of PR10 protein against biotic and abiotic stress has been discussed.Furthermore,our findings also confirmed that the in vivo Nitric oxide(NO)is essential for most of environmental abiotic stresses and disease resistance against pathogen infection.The proper level of NO may be necessary and beneficial,not only in plant response to the environmental abiotic stress,but also to biotic stress.The updated information on this interesting group of proteins will be useful in future research to develop multiple stress tolerance in plants. 展开更多

Results:The positive rate of PS2 protein was 50.48% (53/105) in105 cases. The positive rate of PS, in the patients whosurvived five years or more was 56.96% (45/79), whichwas higher than that of those who lived less than fiveyears (30.77%, 8/26). In the ER, PR (+) patients, thepositive rate of PS, was higher (76.74%, 33/34), thanthat of those with ER, PR (-) (22.5%, 9/40). Conclusions:Our results suggest that the expression of PS, proteinwas positively correlated with the 5-year-survival andthat of ER and PR in breast cancer. It is considered thatPS, may be as a prognostic predictor, and detection ofPS, protein expression was useful for a guidingtreatment of breast cancer. 展开更多

目的探讨子宫内膜癌中Maspin蛋白、u PA蛋白及雌激素受体(ER)、孕激素受体(PR)间表达的相关性及临床意义。方法对2004年6月至2014年6月病理科存档的蜡块,子宫内膜癌50例、不典型增生的子宫内膜50例及正常增殖期子宫内膜30例,采用免疫组... 目的探讨子宫内膜癌中Maspin蛋白、u PA蛋白及雌激素受体(ER)、孕激素受体(PR)间表达的相关性及临床意义。方法对2004年6月至2014年6月病理科存档的蜡块,子宫内膜癌50例、不典型增生的子宫内膜50例及正常增殖期子宫内膜30例,采用免疫组化SP法,检测Maspin蛋白、u PA蛋白、ER、PR的表达情况,结合临床病理特征分析4种蛋白间表达的相关性。结果 Maspin蛋白、ER、PR在增殖期子宫内膜,不典型增生的子宫内膜,子宫内膜癌中阳性表达率逐渐降低,且组间比较有统计学意义(P<0.05),而u PA的阳性表达率逐渐升高(P<0.05)。Maspin、u PA、ER、PR的阳性表达率在手术病理分期、肌层浸润深度及有无淋巴结转移组中的差异均有统计学意义(P<0.05)。Maspin和ER、PR的表达呈一致性,Maspin与u PA呈负相关(r=-0.341,P<0.05),u PA和ER呈负相关(r=-0.293,P<0.05)。结论 ER、PR的减少,Maspin蛋白表达的缺失,伴随u PA蛋白表达上调,四者均参与了子宫内膜癌的发生、发展、浸润等过程,有可能成为判断子宫内膜癌治疗及预后的指标。 展开更多

病程相关蛋白基因PR-1、PR-2和PR-5是植物抗病基因介导的抗病反应和系统获得抗性(systemic acquired resistance,SAR)中的标志基因。为了研究病程相关蛋白基因和水杨酸在小麦(TriticumaestivumL.)抗白粉病反应中所起的作用,以抗白粉病... 病程相关蛋白基因PR-1、PR-2和PR-5是植物抗病基因介导的抗病反应和系统获得抗性(systemic acquired resistance,SAR)中的标志基因。为了研究病程相关蛋白基因和水杨酸在小麦(TriticumaestivumL.)抗白粉病反应中所起的作用,以抗白粉病和感白粉病小麦品系为材料,在白粉病菌(Blumeria graminisf.sp.tritici,Bgt)诱导不同时间,或水杨酸处理不同时间后,用半定量RT-PCR技术检测了小麦叶片中PR-1、PR-2和PR-5基因的表达变化。结果表明,白粉病菌的侵染激活和显著增强了病程相关蛋白基因PR-1、PR-2和PR-5在周麦18中的转录。与感病品系相比,PR-1和PR-5在抗病品系中转录激活得更快、更强。水杨酸处理显著激活了PR-1和PR-5的转录,但对PR-2的转录影响不大。白粉病菌和水杨酸均能显著激活和增强PR-1和PR-5的表达,但白粉病菌的作用更强,说明PR-1和PR-5基因在小麦抗白粉病反应中起重要作用。PR-1和PR-5可以作为小麦SAR的标志基因。水杨酸处理后周麦18和中国春的白粉病抗性得到提高,提示水杨酸在小麦抗白粉病信号传导途径中起一定作用。 展开更多

目的:探讨分析玉米须总皂苷对高脂饮食喂养的肥胖大鼠棕色脂肪组织中解偶联蛋白-1(UCP1)和PR结构域蛋白16(PRDM16)表达的影响。方法:雄性健康清洁级SD大鼠共40只,在适应性喂养1周后随机分为正常对照组、模型对照组以及玉米须总皂... 目的:探讨分析玉米须总皂苷对高脂饮食喂养的肥胖大鼠棕色脂肪组织中解偶联蛋白-1(UCP1)和PR结构域蛋白16(PRDM16)表达的影响。方法:雄性健康清洁级SD大鼠共40只,在适应性喂养1周后随机分为正常对照组、模型对照组以及玉米须总皂苷低剂量和高剂量干预组,每组10只,给予高脂饮食12周后构建肥胖大鼠模型,治疗时玉米须总皂苷低和高剂量组分别给予玉米须总皂苷100、200 mg·kg^-1·d^-1灌胃,正常对照组和模型对照组给予1 m L·kg^-1·d^-1生理盐水灌胃,治疗12周后称重、处死大鼠,收集血清和棕色脂肪组织,检测血清血脂指标,HE染色检测棕色脂肪组织病理学,Real-Time PCR和Western Blot检测棕色脂肪组织中UCP1和PRDM16的m RNA和蛋白表达水平。结果:与正常对照组相比,模型对照组大鼠体重明显升高(P〈0.05),棕色脂肪组织重量和棕色脂肪组织重量/大鼠体质量明显降低(P〈0.05),与模型组相比,玉米须总皂苷治疗后,大鼠体质量明显降低(P〈0.05),棕色脂肪组织重量和棕色脂肪组织重量/大鼠体质量明显升高(P〈0.05);与正常对照组相比,模型对照组血清葡萄糖、甘油三酯、总胆固醇和HDL-C明显升高(P〈0.05),LDL-C明显降低(P〈0.05);与模型对照组相比,玉米须总皂苷治疗后,血清葡萄糖、甘油三酯水平、总胆固醇和HDL-C降低(P〈0.05),LDL-C明显升高(P〈0.05);与正常对照组相比,模型对照组棕色脂肪细胞分布明显减少,脂肪空泡数量明显增加,而玉米须总皂苷治疗后,脂肪空泡数量明显降低;与正常对照组相比,模型对照组大鼠棕色脂肪组织中UCP1和PRDM16的m RNA和蛋白表达明显降低(P〈0.05),玉米须总皂苷治疗后表达明显升高(P〈0.05)。结论:玉米须总皂苷治疗12周可以明显降低肥胖大鼠体质量和棕色脂肪组织中的相对含量,这可能与其可以降低棕色脂肪组织中UCP1和PRDM16的表达有关。 展开更多

目的:探讨乳腺癌中Her-2基因及其蛋白表达状况,并研究其与ER、PR的关系及临床意义。方法:采用荧光标记原位杂交(FISH)和免疫组化法(IHC)检测45例乳腺癌组织中Her-2基因及其蛋白表达并检测ER、PR表达。实验数据采用SPSS统计软件处理。结... 目的:探讨乳腺癌中Her-2基因及其蛋白表达状况,并研究其与ER、PR的关系及临床意义。方法:采用荧光标记原位杂交(FISH)和免疫组化法(IHC)检测45例乳腺癌组织中Her-2基因及其蛋白表达并检测ER、PR表达。实验数据采用SPSS统计软件处理。结果:在45例乳腺癌中有17例出现Her-2基因扩增,阳性率为37.7%。Her-2蛋白总阳性率为66.7%。Her-2基因阳性率在ER阳性组为32.3%,在ER阴性组为50.0%,Her-2基因阳性率在PR阳性组为27.5%,在PR阴性组为56.2%。FISH法检测Her-2基因扩增与Her-2蛋白过表达之间相关(P<0.05),而与淋巴结转移、组织学分级及ER、PR的表达无关(P>0.05)。结论:FISH检测Her-2基因扩增和免疫组化检测Her-2蛋白一致性较好,ER、PR和Her-2三者在乳腺癌的发生发展中起重要作用。 展开更多

目的:观察电针联合补肾活血方对多囊卵巢综合征(PCOS)大鼠围着床期子宫内膜形态学及ER、PR蛋白的影响。方法:将70只大鼠分为正常组(G)、模型组(M)、克罗米芬组(CC)、克罗米芬+补佳乐组(CC+PVG)、克罗米芬+电针组(CC+A)、克罗米芬+补肾... 目的:观察电针联合补肾活血方对多囊卵巢综合征(PCOS)大鼠围着床期子宫内膜形态学及ER、PR蛋白的影响。方法:将70只大鼠分为正常组(G)、模型组(M)、克罗米芬组(CC)、克罗米芬+补佳乐组(CC+PVG)、克罗米芬+电针组(CC+A)、克罗米芬+补肾活血方组(CC+M)和克罗米芬+针药结合组(CC+M+A),每组10只。均于80日龄起进行干预,连续治疗5天。治疗结束后,各组于下午6:00与雄鼠按2:1比例合笼,次日早8:00查阴道涂片,以发现精虫者记为妊娠第1天(D1),于妊娠D4、D5下午4:00称重。电镜下观察饱饮突,免疫组化检测ER、PR蛋白。结果:子宫内膜组织形态学上,CC+M+A组子宫内膜发育分化程度最好;子宫内膜成熟度及胞饮突情况比较:正常组与CC+M+A组之间无明显差异(P>0.05),两组与CC+A、CC+M组比较明显升高,有统计意义(P<0.05),与CC+PVG、模型组、CC组比较有显著差异(P<0.01)。各组ER、PR免疫组化比较:正常组、CC+M+A组都有阳性表达,正常组与CC+M+A组之间无明显差异(P>0.05),两组与CC+A、CC+M、CC+PVG组比较明显升高,有统计意义(P<0.05),与模型组、CC组比较有显著差异(P<0.01)。结论:在围着床期,电针、补肾活血方结合克罗米芬治疗多囊卵综合征与正常子宫内膜发育成熟度最好,这与该治疗方法提高子宫内膜上ER、PR蛋白表达有关。 展开更多

PRDM家族蛋白是具有一个PR结构域和数量不等锌指结构(除了PRDM11),广泛存在于灵长类、啮齿动物、鸟类和两栖动物中能够调控表观遗传的蛋白。部分PRDM蛋白家族成员的PR结构域具有组蛋白赖氨酸转移酶活性(HMT),影响表观遗传,PRDM蛋白锌指结构具有DNA、RNA、蛋白识别结合能力;另一部分PRDM家族成员虽然具有PR结构域,但不具有HMT活性,在癌症发生、原始生殖细胞特化、神经系统发育、造血、血管发展、胚胎发育中发挥重要作用。PRDM家族蛋白通过甲基转移或募集染色体结构重塑复合物,调控基因表达和修改染色体结构,在细胞特化和疾病调控中起到重要作用。论文综述了PRDM家族蛋白结构与功能最新研究进展。 展开更多
