This present work aims to functionalize poly(amidoamine) (PAMAM) dendrimers with various reported adhesive peptides, including Arg-Gly-Asp (RGD), Tyr-lle-Gly-Ser-Arg (YIGSR), and Ile-Lys-Val-Ala-Val (IKVAV) for enhancing cell responses. The RGD, YIGSR, or IKVAV functionalized PAMAM coated substrate could promote cell adhesion of bone marrow mesenchymal stem cells (BMSCs) within 1 h after incubation. The neurite differentiation and proliferation of pheochromocytoma (PC12) cells were also significantly enhanced after culturing on the peptide functionalized PAMAM dendrimers for two and foul days. This peptide functionalized PAMAM dendrimers are considered as the potential candidates for various tissue engineering applications. 展开更多

Mesoporous silica nanoparticles (MSNs) have been extensively studied and proposed as promising candidates for numerous biomedical applications.In this study,we report the design,preparation,characterization and biosafety evaluation of peptide dendron functionalized mesoporous silica nanohybrid.This nanohybrid was prepared by surface modification of MSNs via click reaction of azido-MSNs with alkynyl peptide dendrons,and characterized by TEM,SEM,TGA,dynamic light scattering (DLS).In vitro cytotoxicity of the nanohybrid was evaluated against different cell lines by CCK-8 assay.The in vivo toxicity evaluation was measured by body weight shift,blood routine test and histological analysis,suggesting that the peptide functionalized nanohybrid possessed good biocompatibility due to the non-observed significant side effects to normal organs of healthy mice.Overall,considering our results we believe that the peptide dendron functionalized mesoporous silica nanohybrid is very promising in further biomedical applications. 展开更多

We reported a kind of inorganic-organic hybrid supramolecular hydrogel with excellent anti-biofouling capability. The hydrogel was formed via ionic interaction between the negative-charged sodium polyacrylate (SPA) entwined clay nanosheets (CNS) and positive-charged polyhedral oligomeric silsesquioxane (POSS) core-based generation one (L-Arginine) dendrimer (POSS-R). Due to their strong ionic interaction, this kind of hydrogel exhibited a rapid gelation behavior which accomplished even at a low POSS-R concentration about 1% w/v. With the increase of POSS-R concentration, these hydrogels appeared more compact structure, accompanied by remarkable enhanced mechanical strength. In addition, these hydrogels demonstrated rapid thixotropic response and shape-memory capability, as well as good hiocompatibility. More importantly, these hydrogels exhibited outstanding anti-biofouling property due to the inherent anti-biofouling capability of SPA. Overall, these findings demonstrated a novel sort of inorganic-organic hybrid supramolecular hydrogel with tunable mechanical strength and excellent anti-biofouling capability, which may have a broad application potential in tissue engineering. 展开更多
