Stigma-Specific Comparative Proteomic Analysis Reveals the Distyly Response to Self-Incompatibility in Plumbago auriculata Lam

In plants,heteromorphic self-incompatibility(HetSI)is a strategy for avoiding self-pollination and promoting outcrossing,and during this process,numerous protein-protein interaction events occur between the pistil and pollen.Previous studies in Primula and Fagopyrum that focused on HetSI systems have provided interesting insights;however,the molecular mechanism underlying HetSI remains largely unknown.In this study,we profiled the proteome of Plumbago auriculata stigmas before and after self-incompatible(SI)and self-compatible(SC)pollination.Comparative analyses were conducted by 4D-DIA(Four-dimensional data independent acquisition),a promising technology that increases the sensitivity and reduces the spectral complexity of proteomic analysis by adding a fourth dimension,ion mobility.The results revealed 33387 peptides and 5311 proteins in all samples.The pathways in which the differentially expressed proteins(DEPs)identified in the P×P(Pin style self-pollinated with pin pollen)vs.PS(Pin style)and T×T(Thrum style self-pollinated with thrum pollen)vs.TS(Thrum style)comparisons were significantly enriched were biosynthesis of secondary metabolites and pentose and glucuronate interconversions.In the P×T(Pin style cross-pollinated with thrum pollen)vs.PS and T×P(Thrum style cross-pollinated with pin pollen)vs.TS comparison,the top three pathways were biosynthesis of secondary metabolites,pentose and glucuronate interconversions,and phenylpropanoid biosynthesis.The phenylpropanoid biosynthesis,cutin,suberine and wax biosynthesis,and flavonoid biosynthesis pathways were enriched in the P×T vs.P×P comparison,and starch and sucrose metabolism,glycerophospholipid metabolism,and alpha-linolenic acid metabolism were abundant in the T×T vs.T×P comparison.The enriched pathways between PS and TS were the biosynthesis of secondary metabolites,phenylpropanoid biosynthesis,and pentose and glucuronate interconversion.Self-incompatibility protein S1(SI S1),Mitogen-activated protein kinase 3/4(MPK3/4),Mitogen-activated protein kinase kinase 2/3(M2K2/3),Exocyst complex component EXO70A1(E70A1)and Thioredoxin H1/2(TRXH1/2)were found to be HetSI-related candidates,and O-fucosyltransferase 23(OFT23),3-ketoacyl-CoA synthase 6(KCS6),Receptor-like protein kinase FERONIA(FERON),Fimbrin-5(FIMB5),Pollen-specific leucine-rich repeat extensin-like protein 4(PLRX4),Transcription initiation factor IIB-2(TF2B2)and Pectinesterase 1(AL11A),etc.,were identified as other regulatory transducers.These findings combined with our morphological and reactive oxygen species(ROS)intensity analyses indicate that P.auriculata has typical dry-stigmas and that the HetSI mechanism might differ between the pin and thrum.SI S1 might be the key factor in HetSI,and ROS are overexpressed during SC pollination to rapidly activate the mitogen-activated protein kinase(MAPK)-mediated phosphorylation of E70A1 to maintain stigma receptivity in plants with HetSI. 展开更多

A new tfiterpenoid, 1β,3β,1 1α-trihydroxy-urs-12-ene (1), together with six known compounds, androsta-l,4-diene-3,17- dione (2), isoshinznolone (3), neoechinulin A (4), harman (5), ergostadiene-3β,5α,6β-triol (6) and N-(N'-benzoyl-S-phenylalaninyl)-S- phenylalaninol (7) were isolated from the aerial parts ofP. zeylanica. [ Objectives ] This study was conducted to evaluate the action of the aqueous extract of a medicinal plant Plumbago zeylanica L. in the reversal of dime- thylnitrosamine (DMN) -induced hepatic fibrosis in rats, and to provide a scientific basis for the utilization of P. zeylanica in treatment of hepatic fibrosis. [Meth- ods ] Hepatic fibrosis in rats was induced by intraperitoneal injection of DMN. The rats were then given the aqueous extract of P. zeylanica at high, medium or low concentration by garage for five weeks. Serum level of alanine aminotransferase (ALT) was determined by lactate dehydrogenase (LDH)-release assay, and serum level of aspartate transaminase (AST) was measured by UV-malate dehydrogenase (MDH) assay. Serum levels of total bilirubin (TBIL), direct bilirubin (DBIL) and indirect bilirubin (1BIL) were measured by vanadate oxidation assay. Four indices of hepatic fibrosis (hyaluronic acid, laminin, procollagen type III and colla- gen type IV) were determined by radioimmunoassay (RIA) assay. Morphological damage of liver tissue was observed by hematoxylin-eosin staining (H&E stai- ning). Immunohistochemical staining was performed to determine the location and area of deposited collagen type I, collagen type III and ct-smeoth muscle actin (a-SMA) in liver tissue. [ Results] Compared with the negative control (rats with diseased fiver and untreated with P. zeylanica aqueous extract), the serum lev- els of ALT, AST, TBIL, DBIL and IBIL were significantly decreased by P. zeylanica aqueous extract; the levels of the four serum indices of hepatic fibrosis were also obviously reduced. H&E staining showed that hepatic fibrosis in rats was obviously inhibited or even reversed by P. zeylanica aqueous extract. Immunohisto- chemical staining proved that the aqueous extract of P. zeylanica significantly reduced the area and coloration of collagen type I, collagen type III and ct-SMA in rat liver. [ Conclusions] The aqueous extract of P. zeylanica has a definite effect in reversal of DMN-indueed hepatic fibrosis in rat by promoting the recovery of liver function, reversal of histopathological changes and reducing fibrotic collagen. 展开更多

Preliminary Report of PGR’s Influence to Multiple Shoots Induction and Plant Regeneration on <i>Plumbago auriculata</i> 被引量:2

With the explants of young nodals and leaves of Plumbago auriculata, we studied multiple shoots induction and plant regeneration in the medium with different PGR proportions. The results showed the nodals of Plumbago auriculata could be used as the explant to induce multiple shoots. After culturing young nodals for 30 d, optimum single shoot formation was achieved on MS medium supplemented with 0.3 mg/L 6-BA and the induction rate was 70.79%. After being transferred to the medium containing 1.0 mg/L 6-BA, 1.0 mg/L NAA and 0.2 mg/L IAA for 1 - 2 generation, single shoot was induced to form multiple shoots with the multiplication rate was 506.45%. After multiple shoots grew to 3 cm high, the multiple shoots were cut into single shoots and transferred onto half-strength MS medium supplemented with 0.5 mg/L NAA and the rooted rate was 94.33%. The results showed that a new type of cutting facility used in this study significantly improved the survival rate of P. auriculata sum- merwoed cuttings ; the best time for summerwood cutting of P. auriculata was May; various hormone treatments significantly improved the rooting rate of P. auricu- /ata summerwood cuttings; the optimal concentrations of NAA, IAA and IBA were 1 000, 1 500 and 1 500 mg/L, respectively. This study provided scientific basis for the establishment of summerwoed cutting propagation technology system of P. auriculata. 展开更多

[Objectives]A method for the determination ofβ-sitosterol in Plumbago zeylanica L.was established and the content ofβ-sitosterol in different medicinal parts,different producing areas and different harvest periods were compared.[Methods]High performance liquid chromatography(HPLC)-evaporative light scattering detector assay was used.The chromatographic column was Kromasil C_(18) column(250 mm×4.6 mm,5μm);the mobile phase was pure methanol;the flow rate was 1.0 mL/min;the column temperature was 30℃.The detection parameters of evaporative light scattering detector were as follows:drift tube temperature was 40℃,carrier gas(N_(2))pressure was 3.5 bar.[Results]There was a good linear relationship betweenβ-sitosterol(1.080-4.860μg)and the natural logarithm of peak area(r=0.9995).The average recovery rate was 99.80%.The content ofβ-sitosterol in root and stem was 0.2074 and 0.4064 mg/g,respectively,but it was not found in leaves;the content ofβ-sitosterol in P.zeylanica L.in Guangxi was generally lower than that in Yunnan,and the content ofβ-sitosterol in P.zeylanica L.in Xishuangbanna was the highest;the content ofβ-sitosterol in the stem of P.zeylanica L.was stable at a relatively high level in different harvest periods.[Conclusions]The method is simple,accurate and reproducible,and can be used as one of the methods to control the quality of P.zeylanica L. 展开更多

响应面法优化兰雪醌超声辅助提取工艺的研究

目的优化白花丹中兰雪醌超声辅助提取的最佳工艺。方法采用紫外分光光度法分析兰雪醌含量。采用单因素试验考察超声时间、提取温度和料液比对兰雪醌得率的影响,并在单因素试验的基础上使用Box-Behnken响应面法优化得到最佳工艺。结果该... 目的优化白花丹中兰雪醌超声辅助提取的最佳工艺。方法采用紫外分光光度法分析兰雪醌含量。采用单因素试验考察超声时间、提取温度和料液比对兰雪醌得率的影响,并在单因素试验的基础上使用Box-Behnken响应面法优化得到最佳工艺。结果该模型的调整决定系数(R_(Adj)^(2))为0.8394,回归模型的概率值(P<0.005)显示出较高的显著性。通过Box-Behnke响应面法得到超声提取兰雪醌的最佳工艺参数为超声时间41 min,提取温度42℃,料液比1∶23(g/mL),预测兰雪醌得率为1.823%。在最佳条件下进行了5次验证试验,得到兰雪醌平均得率为1.8941%,与预测值基本吻合。结论利用超声辅助提取方法提取白花丹中兰雪醌的工艺稳定可靠。 展开更多

紫花丹全草药用功能广泛,但其自然授粉率、种子萌发率和种苗存活率均非常低,自然种群分布越来越少。为保护紫花丹资源及满足未来的种苗需求,以紫花丹茎段为外植体,对其外植体消毒时间、芽增殖和壮苗生根培养基、移栽方式及基质进行研究... 紫花丹全草药用功能广泛,但其自然授粉率、种子萌发率和种苗存活率均非常低,自然种群分布越来越少。为保护紫花丹资源及满足未来的种苗需求,以紫花丹茎段为外植体,对其外植体消毒时间、芽增殖和壮苗生根培养基、移栽方式及基质进行研究。结果表明:利用0.1%氯化汞对紫花丹新生枝条消毒12 min效果较好;MS+6-BA 0.2 mg/L+IBA 0.1 mg/L可诱导茎段出芽,出芽后用MS+6-BA 2 mg/L+NAA 0.2 mg/L诱导芽增殖,增殖系数可达32.52;最佳生根培养基为1/2 MS+IBA 0.1 mg/L,15 d生根率可达61.57%;最佳移栽基质为泥炭土∶珍珠岩1∶1(V∶V),其生根率为76.67%,黑茎率最低(4.00%),新叶萌发率最高(78.00%);移栽方式以盖膜培养最佳,其植株新根及新叶萌发率最高,分别为76.67%和78%。本研究初步建立了紫花丹的离体快繁体系,为其工厂化育苗提供技术参考。 展开更多

[目的]探讨白雪花诱导舌癌细胞凋亡及对端粒酶活性的影响。[方法]用流式细胞术检测白雪花的石油醚提取物诱导体外培养的人舌鳞癌细胞凋亡作用;以Tca-8113细胞株作为靶细胞,采用原位TRAP法检测端粒酶活性。[结果]浓度8 mg/ml白雪花的石油醚提取物作用人舌鳞癌Tca细胞24和48 h后,凋亡细胞明显增加;白雪花的石油醚提取物对舌癌细胞作用72 h后,端粒酶活性明显降低,且随药物浓度增大,抑制端粒酶活性的作用增强,呈剂量依赖性。[结论]白雪花的石油醚提取物对人舌鳞癌Tca细胞的增殖具有明显抑制作用;其抗癌作用机制可能是通过抑制端粒酶活性,诱导癌细胞凋亡。 展开更多

目的研究白花丹醌对瘦素(leptin)刺激人肝星状细胞(HSC-LX2)增殖与α-平滑肌动蛋白(α-SMA)表达的影响。方法体外培养HSC-LX2,瘦素刺激24h,各药物组与细胞共孵育24h后,采用MTT法检测白花丹醌对HSC-LX2增殖的影响;采用荧光定量PCR检测各... 目的研究白花丹醌对瘦素(leptin)刺激人肝星状细胞(HSC-LX2)增殖与α-平滑肌动蛋白(α-SMA)表达的影响。方法体外培养HSC-LX2,瘦素刺激24h,各药物组与细胞共孵育24h后,采用MTT法检测白花丹醌对HSC-LX2增殖的影响;采用荧光定量PCR检测各组α-SMAmRNA的表达和免疫组织化学方法检测α-SMA蛋白表达情况。结果白花丹醌呈剂量依赖性抑制瘦素诱导HSC-LX2增殖,以16μmol·L-1浓度时最为明显,与leptin对照组比较差异有显著性(P<0.01);白花丹醌各剂量组作用24h后,HSC-LX2细胞中α-SMAmRNA的水平降低,尤以中、高剂量组明显,与leptin组比较(P<0.01),且作用呈剂量依赖性。免疫组化结果显示白花丹醌各剂量组对HSC-LX2细胞α-SMA蛋白表达有一定的抑制作用。结论白花丹醌抗肝纤维化作用机制之一是从mRNA水平和蛋白水平抑制α-SMA表达,从而抑制HSCECM的合成,发挥抗肝纤维化作用。 展开更多

采用电感耦合等离子体发射光谱法(ICP-AES)分析广西中药白花丹根、茎、叶3个部位的20种常量及微量元素。结果显示,白花丹中除了含人体必需的常量元素K,Ca,Na,Mg外,还有Zn,Fe,Mn,Cr,Co等必需的微量元素和其他元素Mo,Sb,Bi,Cd,Sr,Pb,Cd,A... 采用电感耦合等离子体发射光谱法(ICP-AES)分析广西中药白花丹根、茎、叶3个部位的20种常量及微量元素。结果显示,白花丹中除了含人体必需的常量元素K,Ca,Na,Mg外,还有Zn,Fe,Mn,Cr,Co等必需的微量元素和其他元素Mo,Sb,Bi,Cd,Sr,Pb,Cd,As等。各元素的含量在白花丹的根,茎,叶中分布不同,在叶和根中的含量比较丰富,与白花丹的抗菌,抗肿瘤及抗氧化活性的药用有效部位在根部和叶部的结果一致,如Na,K,Ca,Zn,Fe,Mn,Sr,Cu,Co等含量在叶中含量最高,根次之。在许多抗癌中药中,Zn,Mn,Fe的含量通常比较高,Cr,Sr,Cu的含量也至关重要,作为传统抗癌中药的白花丹中就含有丰富的Zn,Mn,Fe和一定量的Sr,Cr,Cu元素,测定结果为探讨中草药中常量、微量元素与生物活性之间的相互协同关系提供了有用的数据,也为更好地开发和利用白花丹药用资源提供理论依据。 展开更多
