Methods Lymphocyte phenotypes were detected using indirect immunofluorescencetechnique. The proliferation responses were determined by MTT method. Flow cytometry and automaticimage analysis were performed to evaluate the apoptosis of lymphocytes. Results The proliferationresponses of lymphocytes in the elderly were lower than that in young adults. Decreased CD_(45) RA^+cells and increased CD_(45) RO^+ cells were found in lymphocyte population of aged people, comparedwith that of young adults. The CD_(45) RO^+ cells were prone to apoptosis. There is an inhibitoryeffect of PE on apoptosis of lymphocytes in the elderly. Conclusion It is implied that thesusceptibility of lymphocyte in the elderly to apoptosis depends on activation, so called,activation-induced cell death. Present results suggest that apoptosis of lymphocytes in aged peopleplay an important role in the pathogenesis of immunosenescence. Antiproliferative activity at different concentrations of BPE and cisplatin was determined through using MTT assay on MCF-7 and L929 cell lines. An interaction effect(synergistic, additive and antagonistic) between BPE and cisplatin was determined by Compu Syn software based on MTT assay data.Results: The EC50(50% decrement of DPPH inhibition) of BPE was 0.5 mg/m L.L. terminata BPE exhibited antiproliferative activity on both cancer and normal cell lines.The IC50(concentration of drug that was required for 50% of cell inhibition in vitro) of BPE on MCF-7 was 15 mg/m L whereas in normal cell line L929 was 26 mg/m L. The IC50 for cisplatin on MCF-7 was 20 mmol/L. The combination effect of BPE and cisplatin on MCF-7 cells showed that BPE at 15 mg/m L was able to potentiate the inhibitory effect of cisplatin at all different concentrations(2.5–20.0 mg/m L). The average of cancer cells inhibition which was potentiated by BPE was around 50%. A combination index values of less than 1 reported in the Compu Syn software further proved the synergistic effect between BPE and cisplatin, suggesting that BPE was working synergistically with cisplatin.Conclusions: Our study therefore suggested that BPE of Malaysian stingless bee,L. terminata is a potential chemopreventive agent and can be used as a supplementary treatment for chemotherapy drugs. BPE might be able to be used to potentiate the effect of chemotherapy drugs with the possibility to reduce the required dose of the drugs. Background Urinary incontinence(UI)is a debilitating and common condition that adversely affects quality of life.Prescriptive and surgical approaches for managing UI symptoms may result in undesirable risks and compli...Background Urinary incontinence(UI)is a debilitating and common condition that adversely affects quality of life.Prescriptive and surgical approaches for managing UI symptoms may result in undesirable risks and complications.This randomized,double-blind,placebo-controlled,parallel study investigated the efficacy of 2 nonsolvent flower pollen extracts on UI in healthy women.Materials and methods One-hundred and fourteen women aged 40–75 years who scored≥5 on the International Consultation on Incontinence Questionnaire-Urinary Incontinence Short Form(ICIQ-SF)were randomized to receive either Graminex®RCT Fem™UI,Graminex®PollenBerry®,or placebo for 24 weeks.The primary outcome was the change in the ICIQ-SF score between the trial and placebo groups after 24 weeks of supplementation.The secondary outcomes included changes in the frequency of nocturia(recorded in 3-day void diaries)and 24-hour leakage volume(assessed via pad weight)after 6,12,18,and 24 weeks of supplementation and changes in stress-induced urinary leakage volume(after completion of a provocative maneuver challenge)after 24 weeks of supplementation.Results All the groups demonstrated improvement in ICIQ-SF scores at week 24(p<0.001).The RCT Fem™UI group had the greatest improvement in ICIQ-SF scores(−4.07±3.4),followed by the PollenBerry®group(−3.34±2.87)and placebo group(−2.61±3.52).The RCT Fem™UI group had corresponding improvements in 24-hour leakage volume(−17.68±39.84 g)and frequency of nocturia(−0.52±1.26)(p≤0.05).PollenBerry®supplementation significantly improved stress-induced urinary leakage volume(−7.12±15.64 g)at week 24.The study products demonstrated safe hematological and chemical profiles.Conclusions RCT Fem™UI supplementation resulted in significant and clinically meaningful reductions in UI severity,with corresponding improvements in daily urinary leakage volume and frequency of nocturia.PollenBerry®significantly improved stress-induced urinary leakage volume,suggesting that it may be efficacious in women who are prone to stress UI.The study products were safe and well tolerated in this population.展开更多
