慢性自发性荨麻疹患者治疗前后外周血TPM4和CFL1的变化及意义

目的 探讨慢性自发性荨麻疹(CSU)患者治疗前后外周血原肌球蛋白-4(TPM4)和丝切蛋白-1(CFL1)mRNA表达及抗体水平的变化及其临床意义。方法 收集上海市浦东新区人民医院2023年1~6月诊治的60例7日荨麻疹活动评分(UAS7)≥28分的重度CSU患者... 目的 探讨慢性自发性荨麻疹(CSU)患者治疗前后外周血原肌球蛋白-4(TPM4)和丝切蛋白-1(CFL1)mRNA表达及抗体水平的变化及其临床意义。方法 收集上海市浦东新区人民医院2023年1~6月诊治的60例7日荨麻疹活动评分(UAS7)≥28分的重度CSU患者作为研究对象(以下称之为CSU组),予抗组胺药(咪唑斯汀缓释片)治疗12周后,UAS7<7分为有效组(共41例),UAS7≥7分为无效组(共19例)。另选同期30例健康体检者为对照组。实时荧光定量聚合酶链反应(RT-qPCR)检测外周血TPM4和CFL1 mRNA的表达,酶联免疫吸附测定(ELISA)法检测外周血TPM4和CFL1的抗体水平。比较4组中TPM4和CFL1 mRNA的表达和抗体水平。结果 重度CSU患者外周血中TPM4 mRNA表达及抗体水平均高于对照组,但CFL1均低于对照组。抗组胺药物治疗12周后有效组的TPM4 mRNA表达及抗体水平均下降,差异均有统计学意义(P<0.05),治疗前后外周血中CFL1 mRNA表达及抗体水平差异无统计学意义(P>0.05)。结论 TPM4和CFL1可作为检测重度CSU患者的生物学标记物,TPM4亦可作为评价抗组胺药治疗疗效的指标之一。 展开更多

Effect of Sanwu Huangqin Decoction on IFN-γ and IL-17A levels in mice with Behcet's disease

Objective:To investigate the effect of Sanwu Huangqin Decoction on IFN-γand IL-17A levels in mice with Behcet's disease;Methods:Clinical samples of patients with Behcet's disease were collected and mononuclear cells were isolated.High throughput RNA-seq was used to identify the different genes between Behcet's disease and healthy subjects.Mouse model was constructed by shrimp myosin sensitization and treated with Sanwu Huangqin decoction.The levels of IFN-γand IL-17A in peripheral blood and mononuclear cells were detected by ELISA and flow cytometry.The expression level of TBX21 gene in spleen was detected by RT-PCR;Results:Compared with healthy subjects,31 differential genes were identified in BD patients,of which 19 genes were up-regulated and 12 genes were down regulated.PPI analysis showed that the top 5 genes were TBX21,STAT3,IL15,PRF1 and IFNG.The results of animal experiments showed that the levels of IFN-γand IL-17A in the peripheral blood and PBMC of the model group were significantly higher compared with the control group(P<0.01).Sanwu Huangqin decoction could significantly reduce the levels of IFN-γand IL-17A in the peripheral blood and PBMC of mice(P<0.05 or P<0.01).RT-PCR results showed that the expression of TBX21 mRNA in spleen tissue of model group was significantly higher than that of the control group(P<0.01).Sanwu Huangqin decoction could significantly down-regulate the expression of TBX21 mRNA in spleen(P<0.05 or P<0.01);Conclusion:Sanwu Huangqin decoction reduce the levels of IFN-γand IL-17A by inhibiting the expression of TBX21 mRNA,so as to achieve the effect of treating BD. 展开更多

目的:探讨温胆汤对环磷腺苷效应元件结合蛋白(CREB)mRNA沉默海马细胞活性、凋亡及海马脑源性神经营养因子/原肌球蛋白相关受体激酶/CREB(BDNF/TrkB/CREB)信号通路作用的影响。方法:制备温胆汤含药血清,将SD雄性大鼠随机分为温胆汤高、... 目的:探讨温胆汤对环磷腺苷效应元件结合蛋白(CREB)mRNA沉默海马细胞活性、凋亡及海马脑源性神经营养因子/原肌球蛋白相关受体激酶/CREB(BDNF/TrkB/CREB)信号通路作用的影响。方法:制备温胆汤含药血清,将SD雄性大鼠随机分为温胆汤高、中、低剂量组、氯氮平组、正常生理盐水组,每组10只,其中每组15只。正常组予20 mL·kg-1生理盐水灌胃,氯氮平组给予0.02 g·kg-1氯氮平原药灌胃,温胆汤高、中、低剂量组分别予质量浓度为2,1,0.5 g·mL-1的等量温胆汤浓缩生药灌胃,1次/d,连续灌胃8 d后,股动脉取血,5 000 r·min-1离心15 min,倾取上清液,灭活,-80℃保存,备用。通过脂质体转染至海马细胞中,构建CREB mRNA沉默海马神经元细胞系,实时荧光定量聚合酶链式反应(Real-time PCR)验证小分子干扰核糖核酸(siRNA)转录后效果。检测海马细胞周期和凋亡,正常海马细胞和CREB基因沉默海马细胞内BDNF,TrkB,CREB,钙/钙调蛋白依赖性蛋白激酶Ⅱ(CaMKⅡ)mRNA表达水平。结果:对于细胞凋亡,与正常大鼠血清组比较,各组别在24,48 h时,细胞凋亡显著降低(P<0.01);与正常大鼠血清-siRNA组比较,各给药组细胞凋亡显著降低(P<0.01)。对于BDNF,TrkB,CREB,CaMKⅡmRNA表达,与正常大鼠血清组比较,基因沉默正常组CREB,BDNF mRNA表达均显著下降(P<0.01);与正常大鼠血清-siRNA组比较,各给药组可显著提高BDNF mRNA表达(P<0.01),其中TrkB,CaMKⅡ各组间差异无明显统计学意义。结论:温胆汤含药血清可提高BDNF mRNA表达,通过调控BDNF/TrkB/CREB信号通路,以保护海马神经元,达到防治精神分裂症认知障碍的目的。 展开更多
