The results showed that the mRNA expression levels of IL-17, IL-23p19, IL-23p40 and IL-6 in psoriasis leision were significantly higher than those of non-leisions (1.231±0.843 vs 1.003±0.044, 1.166±0.142 vs 0.765±0.133, 1.125±0.104 vs 0.730±0.103, 1.186±0.222 vs 0.976±0.122, respectively, all P<0.05). Meanwhile, The expression levels of IL-17 mRNA, IL-23p19 mRNA, IL-23p40 mRNA and IL-6 mRNA were higher in non-leisions than those in normal skin tissues (1.003±0.044 vs 0.620±0.104, 0.765±0.133 vs 0.584±0.078, 0.730±0.103 vs 0.000 ±0.000, 0.976±0.122 vs 0.656±0.121, respectively, all P<0.05). The overexpression of Th17 cy- tokines in the skin lesions of patients with psoriasis may indicate that Th17 cytokines play a very important role in the immunopathogenesis of psoriasis.展开更多

Psoriasis is characterized by abnormal proliferation of keratinocytes,as well as infiltration of immune cells into the dermis and epidermis,causing itchy,scaly and erythematous plaques of skin.The understanding of thi...Psoriasis is characterized by abnormal proliferation of keratinocytes,as well as infiltration of immune cells into the dermis and epidermis,causing itchy,scaly and erythematous plaques of skin.The understanding of this chronic inflammatory skin disease remains unclear and all available treatments have their limitations currently.Here,we showed that IMMH002,a novel orally active S1 P1 modulator,desensitized peripheral pathogenic lymphocytes to egress signal from secondary lymphoid organs and thymus.Using different psoriasis animal models,we demonstrated that IMMH002 could significantly relieve skin damage as revealed by PAS I score and pathological injure evaluation.Mechanistically,IMMH002 regulated CD3+T lymphocytes re-distribution by inducing lymphocytes’homing,thus decreased T lymphocytes allocation in the peripheral blood and skin but increased in the thymus.Our results suggest that the novel SIP1 agonist,IMMH002,exert extraordinary capacity to rapidly modulate T lymphocytes distribution,representing a promising drug candidate for psoriasis treatment.展开更多

The expression of p21 protein was detected by immunohischemistry in 38 psoriatic tissues and 16 uninvolved samples from psoriasis patients, and 3 normal samples from healthy control. The differences among the three groups were analyzed by Chi-square test. Results: We found that there was no significant difference in the frequency of p21 gene promoter methylation among the three groups (P=0.204), howerver the p21 protein expression were different among the three groups (P=0.007). Conclusion: This methylation marker maybe not an indicator for the detection and monitoring of psoriasis.展开更多
Psoriasis is a chronic papulosquamous skin disease occurring worldwide caused by varieties of factors.The recipe of Mongolian medicine Sendeng Decoction was recorded in“The Pleasure of the Viewer”compiled by Jigwood...Psoriasis is a chronic papulosquamous skin disease occurring worldwide caused by varieties of factors.The recipe of Mongolian medicine Sendeng Decoction was recorded in“The Pleasure of the Viewer”compiled by Jigwood Danjinzhamusu.Mongolian medicine clinical experience proved that Sendeng Decoction had good antipyretic,analgesic,anti-inflammatory and anti-rheumatism effect.However,the mechanism of this drug in psoriasis is still unclear.In this study,imiquimod cream(IMQ)was used to construct psoriasis model in 8-week-old male BALB/c mice,and Sendeng Decoction was used to treat the mice with psoriasis.The results showed that PASI score and spleen index decreased after Sendeng Decoction treatment.Meanwhile,HE staining showed that the pathological structure was improved,flow cytometry results showed that the proportion of Treg cells increased and Th17 cells decreased,and results of Western blot showed that the level of p38 MAPK,p-p38 MAPK,p-STAT3 and p-JAK1 decreased observably.More importantly,the above results were most significant in Sendeng's M group,which was similar to MTX group.Our results indicated that Mongolian medicine Sendeng Decoction can effectively relieve the symptoms of psoriasis mice induced by IMQ,mainly by regulating the ratio of Th17/Treg cells through the JAK/STAT and p38 MAPK pathways to reduce the inflammatory state in mice,thereby improving the symptoms of dermatitis.展开更多
