Hachiku;Sasa kurilensis cv. Chishimazasa, which are of different classification or of different ecological distributions, using the “Sandwich Method”, which assays the dried leaves on growth of lettuce seedlings. Only small difference of activity was found among the four bamboo species. In addition, “Protoplast Co-culture Method” for assay of allelopathy in a 50 μL liquid medium using a 96 well culture plate, was applied to the suspension cultures of the four bamboo species. Protoplasts were isolated from two-week cultured suspension cells of four bamboo species using Cellulase RS and Pectolyase Y-23 in 0.6 M mannitol. At low protoplast densities of bamboo, B. multiplex and P. bambusoides stimulated the recipient lettuce growth, i.e., non-spherically cell enlargement and cell divisions observed under an inverted microscope, while protoplasts of P. nigra and S. kurilensis were less stimulatory or inhibitory. Inhibitory effect of S. kurilensis was the strongest among four bamboo species. Furthermore, highly inhibitory effects of S. kurilensis protoplasts on yellow color accumulation of lettuce protoplasts were clearly observed by analysis of a scanned digital image of a 96-well culture plate. Differences and causes of the allelopathic activities were discussed comparing with other plant species studied using the same assay methods.展开更多

Much of these unique collections of great value to education and research are not currently accessible due to distance, form, and technical barriers. This project is to find new ways to enable users to access and exploit these significant research collections via global network. As GMNet is ending its first 5-year phase in October 2005, it has contributed substantially to the community building in digital library development by ac- commodating numerous collaborators and technical staff from various parts of the world to spend 3 to 5 months as a full-member of the GMNet team in Boston. They have come from different parts of China—such as Sichuan, Hainan, Shanghai and Xi’an; Croatia; and Hanoi, Vietnam. In addition to contribute to the overall system development and enhancement of system function- alities, they have brought valuable sample image collections of their own institutions/countries, and actually developed prototype collections as a part of GMNet. This paper describes the exciting and productive experience of the first of this visiting research group in developing the GMNet’s Version 2.0 PHP-based system under Prof. Chen’s overall supervision. It also describes both the system’s technical level structure—user/Web-based application/data, and complex functionalities with multi-collection, multi-lingual, multi-modal searching capabilities; system management capabilities; as well as provisions for user uploads and retrieval for our own projects. This Version 2.0 system is built on the Linux/Apache/PHP/MySQL platform. What is described in this paper is an actual case which has formed a base for further new development by others in the research group. It demonstrates fully the value of the synergistic collaboration among global partners for universal digital library development. More information can be found in http://www.memorynet.org/.展开更多

First, nodes of 11 different bamboo species that belong to seven major bamboo genera (Bambusa, Dendrocalamus, Phyllostachys, Tetragonocalamus, Chimonobambusa, Pleioblastus, and Sasa) were cultured using 2 mL per well of a liquid medium in a 6-well microplate to form a small-scale liquid culture environment (SLCE). The dormant lateral buds of all bamboo nodes resumed expanding and elongating within 7 days in the SLCE. The dormant and active lateral buds were sectioned longitudinally and stained with Sytox green (SG) to monitor mitotic activity and counterstained with safranin (SF) to detect the inward region of the SAM region. Further, mitotic activity was calculated using a digital imaging analysis, which showed an increase of up to 1.2- to 3.8-fold in terms of the SG/SF ratio after 7 days in the culture. Moreover, we used in vitro node cultures of two typical bamboo species, the sympodial clump-forming type (Bambusa multiplex Raeush, Bm) and the monopodial single culm-forming type (Phyllostachys meyeri McClure, Pm), and noted the following: 1) since gradual white-to-green tinge shoots were observed, we investigated the relation between color variation in the outer regions of culm and node tissues and their suitability as explants. By checking the autofluorescence property of whole shoots under LED 365 nm illumination with an RGB (red, green, and blue) digital imaging analysis using ImageJ software, we specified the color variation of explants as the relative intensity of the blue value. 2) Since the obtained shoots of a 1-month-old culture box showed growth variation, we distinguished shoot types based on plant height, i.e., short (less than 5 cm), medium (ca. 5 - 10 cm), and tall (more than 10 cm). Tall shoots that have ca. 5 nodes on average were suitable for explant. 3) Three types of node portions—the first node (the base node near a rhizome tissue), middle nodes (upper nodes near the 1st node), and the top meristem—were independently cultured in the SLCE, and it was found that the first node showed the best growth performance. 4) By culturing the first node in the SLCE system, we performed a quick survey during the 3 weeks in the culture and found that a combination of 10 μM benzyl adenine and 3 μM thidiazuron was effective for in vitro SAM development, while the addition of 2, 4-D was effective for promoting in vitro RAM development. 5) The detailed autofluorescence properties of the outer regions of culm and node tissues were also identified using an inverted fluorescent microscope under B- and U-excitation lights with RGB and HSB (hue, saturation, and brightness) digital imaging analysis.展开更多

As new media theorists have argued, one of the defining features of the digital arts is the break-down of divisions between art forms, and between art and society (for example, Manovich 2001, 2005). This paper outlines how digital processes intersect with aesthetic and conceptual forms. Relevant frameworks, such as materiality, embodiment, hybridity, interactivity, and narrativity, form the origins of the genre. Digital artworks, like digital media, are interactive, participatory, dynamic, and customizable, incorporating shifting data flows and real-time user inputs (Paul 2003, 67). The customization of content and technology, as well as the recontextualization of information, characterize projects of digital art.展开更多

Health issues now merge with those in education, the environment, energy, culture, gender, security, and the economy. They offer enormous potential in mobilizing for political action. But they also have a dark side. Outgoing U.S. President Barack Obama issued a warning that social media in particular poses an existential threat to democracy. Their pervasive influence during the 2016 American presidential election highlighted the potential for the spread of fake news and alternative facts. These realities must be recognized and addressed if meaningful policies on climate change and future sustainability are to be achieved. The challenges ahead require a re-examination of overarching trends in mass media that date back to the middle of the last century. Warnings from scientists, scholars, and community activists indicate that, on the issue of climate change, time is running out. In his Encyclical on Climate Change and Inequality, Pope Francis amplified the urgent call to action. But collectively, we need to rethink the profit driven ways in which our proliferating digital technologies mitigate potential steps toward a sustainable future. In a myriad of forms that go well beyond the seductive and addicting trends of endless hours spent online, new and proliferating digital toys and gadgets are neither carbon neutral, nor as some proponents would have us believe, do they hold the sole promise of innovation and job creation. Additionally, their potential must be harnessed to maintain rather than disrupt social stability.展开更多

文摘Global Memory Net (GMNet) is intended to be an effective gateway to the world cultural, historical, and heritage image collections from selected academic educational and research partners in the world. More information can be found in http://www.memorynet.org/.
文摘We focus on controlling morphological and histochemical responses of the shoot apical meristem (SAM) and root apical meristem (RAM) of bamboo node by using a simple and versatile liquid culture system. 5) The detailed autofluorescence properties of the outer regions of culm and node tissues were also identified using an inverted fluorescent microscope under B- and U-excitation lights with RGB and HSB (hue, saturation, and brightness) digital imaging analysis.
文摘Digital technology has significantly facilitated the development of the cultural industry, and the cultural industry has also become one of the most important fields to which digital technology is applied.Conducting an analysis of how digital technology empowers consumers, creators, producers, social interaction and cultural communication can help us understand how it has improved the efficiency of the cultural industry, propelling digital contents into becoming the mainstay of the cultural industry and making digital communication the principal channel of cultural transmission.Digital technology has reshaped the structure of the cultural industry, leading to marked changes in the structure of cultural consumption, cultural production, the cultural market, and the proportion of domestic to foreign culture. However, whether this powerful digital technology will result in impressive technological results but lackluster cultural content remains a question. In fact, creative people's pursuit of cultural content and consumers'desire for such content will constantly give rise to outstanding cultural works with rich connotations and lasting influence in the digital era.The integration of digitization and technology can not only foster an open cultural plain filled with diverse and vibrant cultures, but can also create cultural peaks that will exhibit profound thought and superb artistic expression.
文摘In the following work of research, we shall present the results of a study on forms of digital photographic socialization as carried out by the so-called "digital natives" in order to explore their competence in managing digital photographic output and cultural practices. This shall be done under the following categories of analysis: snapshot, organization, socialization, reception, privacy, and copyrights in digital photography. The study explores the forms of digital photography production, technical problems of photographic management and the final uses that digital photographers give to their photos in terms of web tools 2.0, mobile telephones and social networks. Digital photography technology, compact cameras, mobile phones with cameras, specialized websites in photography and the lnternet generate new forms of production and emerging styles of digital photographic socialization. The study is carried out using a defined sample of university students, those digital natives who create and use digital photography.
文摘This paper explores the key vocabularies, themes, ideas, artistic movements, and technological innovations contributing to the development of the digital arts over time. The customization of content and technology, as well as the recontextualization of information, characterize projects of digital art.
文摘Digital technologies offer unprecedented opportunities in addressing climate change as public policy requirements increasingly pervade national boundaries. Additionally, their potential must be harnessed to maintain rather than disrupt social stability.
