The development...The localization of ion channels on myelinated axon is closely related with the saltatory conduction of action potential (AP). Abnormal changes in these channels contribute to multiple mental diseases. The development of cryo-Electron Tomography (cryo-ET) has provided a promising prospect for peering into ion channels in their native environment at high resolution. Previous achievements are reviewed here on cryo-ET. Accordingly, a cryo-ET workflow is designed for understanding ion channels localization in myelinated axon, especially nodes of Ranvier, which are significant for the saltatory conduction involved in the propagation of high-speed AP. The workflow is divided into six parts: the preparation of neural cultures with myelin, antibodies and immunofluorescence staining, frozen-hydrated sample preparation, cryo-ET imaging, cryo-correlative light and electron microscopy (cryo-CLEM) imaging, three-dimensional (3D) reconstruction and refinement. The purpose is to conceive a possible solution for the problems related to ion channel compounds including localization, conformation dynamics, accessory structures of ion channel and transient regulatory factors, and thus provide insights into treating neurological diseases caused by abnormal ion channels activity.展开更多

Myelinated nerves also carry out plasticity in which microglia remove myelin debris by phagocytosis. However, it remains unknown whether microglia explore structures on nerve fibers, such as Ranvier’s node(RN) or myelin sheath, before they become debris. By double or triple staining RNs or myelin sheathes and microglia in healthy rat corpus callosum, this study unveiled direct contacts of microglia pseudopodia with RNs and with para-and inter-nodal myelin sheathes, which was then verified by electron microscopic observations. Our data indicated that microglia also explore unmyelinated nerve fibers. Furthermore, we used the animals with matured white matter;therefore, microglia may be actively involved in plasticity of matured white matter tracts as it does for synapse pruning, instead of only passively phagocytize myelin debris.展开更多

In recent years,bamboo has been widely used in a broad range of applications,a thorough understanding of the structural characteristics of bamboo nodes is essential for better processing and manufacturing of biomimeti...In recent years,bamboo has been widely used in a broad range of applications,a thorough understanding of the structural characteristics of bamboo nodes is essential for better processing and manufacturing of biomimetic materials.This study investigated the complex anatomical structure for the nodes of two bamboo species,Indocalamus latifolius(Keng)McClure and Shibataea chinensis Nakai,using a high-resolution X-ray microtomography(μCT).The results show that the vascular bundle system in the nodal region of I.latifolius and S.chinensis is a net-like structure composed of horizontal and axial vascular bundles.Furthermore,the fiber sheath surrounding metaxylem vessels tended to be shorter in the tangential direction.This structure of bamboo nodes facilitates the tangential and axial transport of moisture and nutrients.The anatomical structure of I.latifolius and S.chinensis nodes has obvious differences,especially in the arrangement of vascular bundles.Vascular bundle frequency was significantly higher in S.chinensis nodes than in I.latifolius nodes.These findings indicate thatμCT is a nondestructive three-dimensional imaging method that can used to examine the anatomical structure of bamboo nodes.展开更多

BACKGROUND Castleman's disease(CD)is a rare lymphoproliferative,emulating both benign and malignant diseases.The diagnosis of CD is formulated upon the combination of clinical and laboratory criteria and ultimatel...BACKGROUND Castleman's disease(CD)is a rare lymphoproliferative,emulating both benign and malignant diseases.The diagnosis of CD is formulated upon the combination of clinical and laboratory criteria and ultimately confirmed by histopathological assessment.Due to its rarity,CD presents a challenge in treatment selection,with available options encompassing surgery,chemotherapy,and autologous stem cell transplantation.However,studies suggest that surgical resection of the lesion is the most effective treatment modality,especially for unicentric CD(UCD).CASE SUMMARY Here,we describe the case of a 25-year-old woman who presented with painless left thigh swelling for 10 wk.She had been following a low-fat diet to lose weight and had normal laboratory results.Magnetic resonance imaging revealed a wellcircumscribed,demarcated cystic lesion located in the left inguinal region with eccentrically positioned signal void vascular structures,measuring 4.3 cm×3 cm×3.2 cm,likely of lymphoid origin.The patient underwent surgical resection,and the final histopathology showed a vascular proliferation and hyalinization of the vessel walls,along with atretic germinal centers traversed by penetrating vessels,consistent with CD.The patient was discharged home one day after the procedure in good condition,with a follow-up appointment scheduled in our outpatient clinic.CONCLUSION Although surgical resection is the mainstay for UCD,a multidisciplinary approach is needed due the lack of specific diagnostic features and treatments.展开更多

Before estimating BFN, obtaining blood oxygen level dependent (BOLD) representativ...Purpose: Brain functional networks (BFNs) has become important approach for diagnosis of some neurological or psychological disorders. Before estimating BFN, obtaining blood oxygen level dependent (BOLD) representative signals from brain regions of interest (ROIs) is important. In the past decades, the common method is generally to take a ROI as a node, averaging all the voxel time series inside it to extract a representative signal. However, one node does not represent the entire information of this ROI, and averaging method often leads to signal cancellation and information loss. Inspired by this, we propose a novel model extraction method based on an assumption that a ROI can be represented by multiple nodes. Methods: In this paper, we first extract multiple nodes (the number is user-defined) from the ROI based on two traditional methods, including principal component analysis (PCA), and K-means (Clustering according to the spatial position of voxels). Then, canonical correlation analysis (CCA) was issued to construct BFNs by maximizing the correlation between the representative signals corresponding to the nodes in any two ROIs. Finally, to further verify the effectiveness of the proposed method, the estimated BFNs are applied to identify subjects with autism spectrum disorder (ASD) and mild cognitive impairment (MCI) from health controls (HCs). Results: Experimental results on two benchmark databases demonstrate that the proposed method outperforms the baseline method in the sense of classification performance. Conclusions: We propose a novel method for obtaining nodes of ROId based on the hypothesis that a ROI can be represented by multiple nodes, that is, to extract the node signals of ROIs with K-means or PCA. AIM:To develop lymph node metastasis(LNM)-associated biomarkers for colorectal cancer(CRC) using quantitative proteome analysis.METHODS:Differences in protein expression between primary CRC with LNM(LNM CRC) and witho...AIM:To develop lymph node metastasis(LNM)-associated biomarkers for colorectal cancer(CRC) using quantitative proteome analysis.METHODS:Differences in protein expression between primary CRC with LNM(LNM CRC) and without LNM(non-LNM CRC) were assessed using methyl esterification stable isotope labeling coupled with 2D liquid chromatography followed by tandem mass spectrometry(2D-LC-MS/MS).The relationship to clinicopathological parameters and prognosis of candidate biomarkers was examined using an independent sample set.RESULTS:Forty-three proteins were found to be differentially expressed by at least 2.5-fold in two types of CRC.S100A4 was significantly upregulated in LNM CRC compared with non-LNM CRC,which was confirmed by Western blotting,immunohistochemistry and real-time quantitative polymerase chain reaction.Further immunohistochemistry on another 112 CRC cases showed that overexpression of S100A4 frequently existed in LNM CRC compared with non-LNM CRC(P < 0.001).Overexpression of S100A4 was significantly associated with LNM(P < 0.001),advanced TNM stage(P < 0.001),increased 5-year recurrence rate(P < 0.001) and decreased 5-year overall survival rate(P < 0.001).Univariate and multivariate analyses indicated that S100A4 expression was an independent prognostic factor for recurrence and survival of CRC patients(P < 0.05).CONCLUSION:S100A4 might serve as a powerful biomarker for LNM and a prognostic factor in CRC.展开更多

An esophageal tum...Castleman's disease(CD) is an uncommon benign lymphoproliferative disorder of unknown etiology. A rare case of cervical CD diagnosed at lymph node dissection for esophageal carcinoma is reported. An esophageal tumor was identified in a 67-year-old man during a follow-up examination after surgery for oral carcinoma. Esophagoscopy revealed a type 1 tumor in the cervical esophagus. Histology of esophagoscopic biopsies indicated squamous cell carcinoma. Contrastenhanced computed tomography revealed swollen lymph nodes of the right cervical region. No distant metastasis was detected. Esophageal carcinoma, T2N2M0, Stage ⅢA was diagnosed. Neoadjuvant chemotherapy was recommended, but the patient rejected the chemotherapy. The patient underwent laparoscopic-assisted transhiatal esophagectomy. The histopathological diagnosis was moderately differentiated squamous cell carcinoma with pT1bN0M0, Stage ⅠA. On histology, the swollen lymph nodes of the right cervical region revealed CD. The patient's postoperative course was relatively good.展开更多
BACKGROUND Colorectal cancer(CRC)is one of the most common malignant tumors of the digestive tract.Lymphatic metastases of this tumor are mostly confined to the regional lymph nodes,and distant supraclavicular lymph n...BACKGROUND Colorectal cancer(CRC)is one of the most common malignant tumors of the digestive tract.Lymphatic metastases of this tumor are mostly confined to the regional lymph nodes,and distant supraclavicular lymph node metastases are very rare.CASE SUMMARY In this report,we describe a patient with sigmoid carcinoma and isolated synchronous supraclavicular lymph node metastases.A 56-year-old male presented with a left cervical mass that was confirmed as a lymph node metastasis from sigmoid cancer by several auxiliary examinations.After 6 cycles of chemotherapy with the 5-fluorouracil,leucovorin and oxaliplatin+cetuximab regimen,the sigmoid colon tumor and Virchow’s lymph node metastasis were significantly smaller than before treatment,and no new metastatic sites were observed.Considering the effects of chemotherapy on quality of life,resection of the primary tumor was performed followed by 4 cycles of chemotherapy with the original chemotherapy regimen.Virchow’s lymph node dissection was selected by mutual consultation between the patient and us.After the second surgery,the patient received capecitabine and cetuximab chemotherapy and did not experience recurrence or metastasis during follow-up.CONCLUSION In conclusion,supraclavicular lymph node metastasis without any other solid organ metastasis is a potential metastatic pathway for CRC.In addition,after resection of the primary lesion,postoperative chemotherapy combined with supraclavicular lymph node dissection is feasible for the treatment of patients with CRC and isolated synchronous Virchow’s lymph node metastases.展开更多

The orbital lifetime of lunar orbits depends significantly on the initial conditions of the orbit. Right ascension of ascending node (Ω) is one of the important orbital parameter affecting the orbital lifetime. In the present work we have analyzed the effect of Ω on the variation of lifetime with altitude for circular lunar orbits. It is found that at a particular initial altitude, a small increase in the altitude results in substantial increase in the orbital lifetime due to effect of the long periodic terms of Earth’s gravity on eccentricity and this transition altitude is different for different Ω. Further, it is observed that the variation of transition altitude with Ω follows a definite, but different trend for orbits with different inclinations. The transition altitude for polar orbits is found to be higher without the effect of Sun and Earth gravity. Variation of transition altitude with orbital inclination is also analyzed. Lifetimes of high altitude circular lunar orbits are analyzed and it is observed that at high altitudes lifetime decreases with altitude.展开更多
Wireless sensor Mobile ad hoc networks have excellent potential in moving and monitoring disaster area networks on real-time basis.The recent challenges faced in Mobile Ad Hoc Networks(MANETs)include scalability,local...Wireless sensor Mobile ad hoc networks have excellent potential in moving and monitoring disaster area networks on real-time basis.The recent challenges faced in Mobile Ad Hoc Networks(MANETs)include scalability,localization,heterogeneous network,self-organization,and self-sufficient operation.In this background,the current study focuses on specially-designed communication link establishment for high connection stability of wireless mobile sensor networks,especially in disaster area network.Existing protocols focus on location-dependent communications and use networks based on typically-used Internet Protocol(IP)architecture.However,IP-based communications have a few limitations such as inefficient bandwidth utilization,high processing,less transfer speeds,and excessive memory intake.To overcome these challenges,the number of neighbors(Node Density)is minimized and high Mobility Nodes(Node Speed)are avoided.The proposed Geographic Drone Based Route Optimization(GDRO)method reduces the entire overhead to a considerable level in an efficient manner and significantly improves the overall performance by identifying the disaster region.This drone communicates with anchor node periodically and shares the information to it so as to introduce a drone-based disaster network in an area.Geographic routing is a promising approach to enhance the routing efficiency in MANET.This algorithm helps in reaching the anchor(target)node with the help of Geographical Graph-Based Mapping(GGM).Global Positioning System(GPS)is enabled on mobile network of the anchor node which regularly broadcasts its location information that helps in finding the location.In first step,the node searches for local and remote anticipated Expected Transmission Count(ETX),thereby calculating the estimated distance.Received Signal Strength Indicator(RSSI)results are stored in the local memory of the node.Then,the node calculates the least remote anticipated ETX,Link Loss Rate,and information to the new location.Freeway Heuristic algorithm improves the data speed,efficiency and determines the path and optimization problem.In comparison with other models,the proposed method yielded an efficient communication,increased the throughput,and reduced the end-to-end delay,energy consumption and packet loss performance in disaster area networks.展开更多

文摘AIM:To develop lymph node metastasis(LNM)-associated biomarkers for colorectal cancer(CRC) using quantitative proteome analysis.METHODS:Differences in protein expression between primary CRC with LNM(LNM CRC) and without LNM(non-LNM CRC) were assessed using methyl esterification stable isotope labeling coupled with 2D liquid chromatography followed by tandem mass spectrometry(2D-LC-MS/MS).The relationship to clinicopathological parameters and prognosis of candidate biomarkers was examined using an independent sample set.RESULTS:Forty-three proteins were found to be differentially expressed by at least 2.5-fold in two types of CRC.S100A4 was significantly upregulated in LNM CRC compared with non-LNM CRC,which was confirmed by Western blotting,immunohistochemistry and real-time quantitative polymerase chain reaction.Further immunohistochemistry on another 112 CRC cases showed that overexpression of S100A4 frequently existed in LNM CRC compared with non-LNM CRC(P < 0.001).Overexpression of S100A4 was significantly associated with LNM(P < 0.001),advanced TNM stage(P < 0.001),increased 5-year recurrence rate(P < 0.001) and decreased 5-year overall survival rate(P < 0.001).Univariate and multivariate analyses indicated that S100A4 expression was an independent prognostic factor for recurrence and survival of CRC patients(P < 0.05).CONCLUSION:S100A4 might serve as a powerful biomarker for LNM and a prognostic factor in CRC.

文摘Castleman's disease(CD) is an uncommon benign lymphoproliferative disorder of unknown etiology. A rare case of cervical CD diagnosed at lymph node dissection for esophageal carcinoma is reported. An esophageal tumor was identified in a 67-year-old man during a follow-up examination after surgery for oral carcinoma. Esophagoscopy revealed a type 1 tumor in the cervical esophagus. Histology of esophagoscopic biopsies indicated squamous cell carcinoma. Contrastenhanced computed tomography revealed swollen lymph nodes of the right cervical region. No distant metastasis was detected. Esophageal carcinoma, T2N2M0, Stage ⅢA was diagnosed. Neoadjuvant chemotherapy was recommended, but the patient rejected the chemotherapy. The patient underwent laparoscopic-assisted transhiatal esophagectomy. The histopathological diagnosis was moderately differentiated squamous cell carcinoma with pT1bN0M0, Stage ⅠA. On histology, the swollen lymph nodes of the right cervical region revealed CD. The patient's postoperative course was relatively good.

文摘BACKGROUND Colorectal cancer(CRC)is one of the most common malignant tumors of the digestive tract.Lymphatic metastases of this tumor are mostly confined to the regional lymph nodes,and distant supraclavicular lymph node metastases are very rare.CASE SUMMARY In this report,we describe a patient with sigmoid carcinoma and isolated synchronous supraclavicular lymph node metastases.A 56-year-old male presented with a left cervical mass that was confirmed as a lymph node metastasis from sigmoid cancer by several auxiliary examinations.After 6 cycles of chemotherapy with the 5-fluorouracil,leucovorin and oxaliplatin+cetuximab regimen,the sigmoid colon tumor and Virchow’s lymph node metastasis were significantly smaller than before treatment,and no new metastatic sites were observed.Considering the effects of chemotherapy on quality of life,resection of the primary tumor was performed followed by 4 cycles of chemotherapy with the original chemotherapy regimen.Virchow’s lymph node dissection was selected by mutual consultation between the patient and us.After the second surgery,the patient received capecitabine and cetuximab chemotherapy and did not experience recurrence or metastasis during follow-up.CONCLUSION In conclusion,supraclavicular lymph node metastasis without any other solid organ metastasis is a potential metastatic pathway for CRC.In addition,after resection of the primary lesion,postoperative chemotherapy combined with supraclavicular lymph node dissection is feasible for the treatment of patients with CRC and isolated synchronous Virchow’s lymph node metastases.
文摘The lifetime of a lunar satellite orbit is constrained by the non-spherical nature of the Moon’s gravity field. The orbital lifetime of lunar orbits depends significantly on the initial conditions of the orbit. Right ascension of ascending node (Ω) is one of the important orbital parameter affecting the orbital lifetime. In the present work we have analyzed the effect of Ω on the variation of lifetime with altitude for circular lunar orbits. It is found that at a particular initial altitude, a small increase in the altitude results in substantial increase in the orbital lifetime due to effect of the long periodic terms of Earth’s gravity on eccentricity and this transition altitude is different for different Ω. Further, it is observed that the variation of transition altitude with Ω follows a definite, but different trend for orbits with different inclinations. The transition altitude for polar orbits is found to be higher without the effect of Sun and Earth gravity. Variation of transition altitude with orbital inclination is also analyzed. Lifetimes of high altitude circular lunar orbits are analyzed and it is observed that at high altitudes lifetime decreases with altitude.
文摘Wireless sensor Mobile ad hoc networks have excellent potential in moving and monitoring disaster area networks on real-time basis.The recent challenges faced in Mobile Ad Hoc Networks(MANETs)include scalability,localization,heterogeneous network,self-organization,and self-sufficient operation.In this background,the current study focuses on specially-designed communication link establishment for high connection stability of wireless mobile sensor networks,especially in disaster area network.Existing protocols focus on location-dependent communications and use networks based on typically-used Internet Protocol(IP)architecture.However,IP-based communications have a few limitations such as inefficient bandwidth utilization,high processing,less transfer speeds,and excessive memory intake.To overcome these challenges,the number of neighbors(Node Density)is minimized and high Mobility Nodes(Node Speed)are avoided.The proposed Geographic Drone Based Route Optimization(GDRO)method reduces the entire overhead to a considerable level in an efficient manner and significantly improves the overall performance by identifying the disaster region.This drone communicates with anchor node periodically and shares the information to it so as to introduce a drone-based disaster network in an area.Geographic routing is a promising approach to enhance the routing efficiency in MANET.This algorithm helps in reaching the anchor(target)node with the help of Geographical Graph-Based Mapping(GGM).Global Positioning System(GPS)is enabled on mobile network of the anchor node which regularly broadcasts its location information that helps in finding the location.In first step,the node searches for local and remote anticipated Expected Transmission Count(ETX),thereby calculating the estimated distance.Received Signal Strength Indicator(RSSI)results are stored in the local memory of the node.Then,the node calculates the least remote anticipated ETX,Link Loss Rate,and information to the new location.Freeway Heuristic algorithm improves the data speed,efficiency and determines the path and optimization problem.In comparison with other models,the proposed method yielded an efficient communication,increased the throughput,and reduced the end-to-end delay,energy consumption and packet loss performance in disaster area networks.
