[Objectives]This study aimed to observe the effects of different processed products of Radix Codonopsis on intestinal flora in rats with spleen deficiency.[Methods]Rat models with spleen deficiency were established by bitter-cold purgation method with Radix et Rhizoma Rhei.Normal group,model group,Radix Codonopsis group,fried Radix Codonopsis group,rice-fried Radix Codonopsis group,honey-fried Radix Codonopsis group and bran-fired Radix Codonopsis group were designed.After subjecting to corresponding treatments,the changes in the quantity of intestinal microorganisms of the rats were detected.[Results]There was no significant change in the quantity of intestinal microorganisms of the rats in the normal group and model group.The rats in the rice-fried Radix Codonopsis group were administered after successful modeling,and the abundance of Bifidobacterium and Lactobacillus in the intestines of the rats increased,and the quantity of Escherichia coli and Staphylococci reduced.The rats in the Radix Codonopsis group,fried Radix Codonopsis groups,honey-fried Radix Codonopsis group and bran-fried Radix Codonopsis group were administered after successful modeling,and the abundance of Bifidobacterium and Lactobacillus in the intestines of the rats increased(the increases were smaller than those in the rice-fried Radix Codonopsis group),and the abundance of E.coli and Staphylococci reduced,close to normal levels.[Conclusions]Different processed products of Radix Codonopsis have obvious regulation effect on intestinal flora of rats with spleen deficiency,and the regulation effect of rice-fried Radix Codonopsis on rats with spleen deficiency is better than that of Radix Codonopsis,fried Radix Codonopsis,honey-fried Radix Codonopsis and bran-fried Radix Codonopsis. 展开更多

T-cell subsets from peripheral blood and spleen were investigated prospectively by flow cytometry in a murine intrahepatic implanted tumor model. T-cell subsets from peripheral blood and spleen were investigated prospectively by flow cytometry in a murine intrahepatic implanted tumor model. Peripheral blood OX-19+ cells (pan T-cells) and W3/25+ cells (Th cells) decreased with tumor development in tumor-bearing rats, giving significant differences when compared with healthy rats. No significant changes were found in OX-8+ cells (Ts cells). Hence, Th Ts ratio in tumor-bearing rats was lower than that in control group. Meanwhile, on day 17 following tumor implantation, in tumor-bearing rats with intrahepatic or peritoneal metastasis, there were less W3/25+ cells and more OX-8+ cells than those of hepatoma-bearing rats without metastasis (p<0.01). In the spleen, the phenotypes of lymphocyte markers showed less OX-8+ cells and more OX-12+ cells (B cells) tumor-bearing rats. It was suggested that, in tumor-bearing host, not only was T-cell immune function inhibited, but also there was abnormal distribution of T-cell subsets. It seems that there is a close relationship between tumor growth and its spread and a lower Th/Ts ratio. 展开更多

Effect of <i>Chromolaena odorata</i>Extracton Hematotoxicity and Spleen Histopathology Induced by <i>Salmonella typhi</i>in Wistar Rats

Salmonella typhi is a facultative intracellular pathogen that causes typhoid fever in humans. In the present study, the effect of Salmonella typhi infection on hematological indices and spleen histology in Wistar rats was investigated and was followed by an evaluation of the ameliorative potential of the methanol leaf extract of Chromolaena odorata (MLECO) compared with ciprofloxacin treatment. The animals were divided into six groups: group 1 was normal control, group 2 was infected with Salmonella typhi without treatment (negative control), groups 3, 4 and 5 were Salmonella typhi infected and treated with 100 mg/kg, 200 mg/kg and 400 mg/kg of the extract respectively and group 6 was also Salmonella typhi infected and treated with 500 mg/70kg of ciprofloxacin. The animals were inoculated with a single infectious dose of Salmonella typhi bacteria and thereafter, treated with the extract and ciprofloxacin for a period of seventeen days, after the animals were confirmed infected. The rats were humanely sacrificed and blood samples taken for haematological investigations, and the spleen harvested and processed for histological examinations. Chromolaena odorata administration reversed the adverse hematotoxicity and histopathological changes in the spleen induced by?Salmonella typhi?infection. 展开更多

Rat Models of Fatty Liver due to Liver Depression and Spleen Deficiency

[Objectives] To establish animal models of fatty liver due to liver depression and spleen deficiency that were suitable for activity discovery of traditional Chinese medicine( TCM) compounds,efficacy evaluation,new drug research and development and correspondence between prescription and syndrome of TCM. [Methods] A syndrome score scale that was suitable for evaluating the rat models was established according to the evolvement rules of etiology and pathogenesis of TCM and the modern clinical pathological mechanism. At the same time,bifendate pills and Sanqi Zhigan pills were selected as drug counterevidence for the models. Rats in model groups were given different proportions of high-fat and low-protein fodder and different concentrations of alcohol every day,and intraperitoneally injected with porcine serum twice a week. Drug groups were given with bifendate pills and Sanqi Zhigan pills( 8. 1 mg/kg,and 2. 7 g/kg) respectively for 14 consecutive days.During the experiment,general status,weight,daily fodder intake and daily water intake of the animals were observed,and TCM syndromes were scored. After the experiment,the levels of alanine aminotransferase( ALT),aspartate aminotransferase( AST),γ-glutamine transpeptidase( γ-GT),total cholesterol( TC),triglycerides( TG),low density lipoprotein-cholesterol( LDL-C),and high density lipoprotein-cholesterol( HDL-C) in serum were detected,and the pathological changes in liver tissue were observed. [Results] Compared with the control group,body weight,daily fodder and water intake of rats increased slowly in the model group,and the levels of ALT,AST,γ-GT,TC,TG and LDL-C increased significantly,while the level of HDL-C dropped. Pathological examination showed that steatosis and fat granule were observed in hepatocytes of rats in the model group. Behavioral observation found that main symptoms and minor symptoms of rats in the model group conformed to the syndrome manifestation of liver depression and spleen deficiency,which suggested that the three model groups were established successfully. Among them,three rats died in model group 1,and one rat died in model group 2. The manifestation of all the above lesions can be alleviated by drug counterevidence. [Conclusions]The animal model of fatty liver due to liver depression and spleen deficiency shows an obviously lower mortality and shorter duration and can be used for research of correlation between prescription and syndrome and its mechanism. 展开更多

基于细胞铜死亡分析香砂六君子汤减少脾虚高脂血症大鼠肝脏组织脂质沉积的作用机制

目的从细胞铜死亡的角度分析中药香砂六君子汤减少脾虚高脂血症大鼠肝脏组织脂质沉积的作用机制。方法将18只健康SD大鼠随机均分为正常组、脾虚高脂血症组、中药组。脾虚高脂血症组和中药组采用游泳力竭和高脂饲料喂养的复合方法造模,... 目的从细胞铜死亡的角度分析中药香砂六君子汤减少脾虚高脂血症大鼠肝脏组织脂质沉积的作用机制。方法将18只健康SD大鼠随机均分为正常组、脾虚高脂血症组、中药组。脾虚高脂血症组和中药组采用游泳力竭和高脂饲料喂养的复合方法造模,正常组仅给予普通饲料。造模12周后,中药组进行香砂六君子汤(11.34 g·kg-1)灌胃干预,正常组与脾虚高脂血症组给予同体积0.9%氯化钠溶液灌胃,灌胃干预4周后检测各组大鼠血脂指标[总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)]、肝脏脂积变化;PCR Array技术检测与铜死亡相关基因的表达。结果与正常组比较,脾虚高脂血症组血清中TC、TG、LDL-C水平显著升高(P<0.01),HDL-C水平显著降低(P<0.01);脾虚高脂血症组大鼠肝脏脂质沉积明显。与脾虚高脂血症组比较,中药组TC、TG、LDL-C水平显著降低(P<0.01),HDL-C水平显著升高(P<0.01);中药组大鼠肝脏脂质沉积明显减少。PCR Array共检测出11个铜死亡相关基因。与正常组比较,脾虚高脂血症组肝脏铜死亡差异表达基因7个;与脾虚高脂血症组比较,中药组肝脏铜死亡差异表达基因6个;脾虚高脂血症组与中药组铜死亡共同差异表达基因5个。脾虚高脂血症组与正常组、中药组与脾虚高脂血症组差异表达均发生上调≥1倍或下调≤1倍的基因3个,2种比较方法结果取交集,共同的差异表达基因为Dld和Gls,这2个基因均能够调控线粒体铜死亡水平。qPCR验证结果显示,Dld基因表达与PCR芯片结果一致。结论肝脏铜死亡相关基因(Dld、Gls)表达异常可能是血脂代谢异常的潜在机制,也可能是香砂六君子汤防治脾虚高脂血症的重要作用靶标。 展开更多

脾虚泄泻模型大鼠肠组织结构及分泌型免疫球蛋白A含量变化研究

为研究脾虚泄泻大鼠肠道结构和肠道分泌型免疫球蛋白A的变化,试验选取24只健康SD大鼠,雌雄各半,随机分为脾虚泄泻模型组和空白对照组,在不同阶段采用“饮食失调+苦寒泻下”的方法人工复制大鼠脾虚泄泻模型,连续14 d监测大鼠的采食量、... 为研究脾虚泄泻大鼠肠道结构和肠道分泌型免疫球蛋白A的变化,试验选取24只健康SD大鼠,雌雄各半,随机分为脾虚泄泻模型组和空白对照组,在不同阶段采用“饮食失调+苦寒泻下”的方法人工复制大鼠脾虚泄泻模型,连续14 d监测大鼠的采食量、饮水量、精神状态、粪便等临床特征变化,记录第7天和第14天时大鼠的空腹体质量,采集小肠组织进行结构观察,并测定盲肠内容物中分泌型免疫球蛋白A的含量。结果表明,与空白对照组相比,模型组大鼠在试验期间出现粪便稀溏、食欲下降、消瘦、蜷缩扎堆、精神萎靡等临床症状;与空白对照组相比,模型组大鼠的体质量显著降低(P<0.05);模型组大鼠十二指肠绒毛高度,空肠隐窝深度、绒毛高度、绒毛宽度,回肠隐窝深度和绒毛宽度均显著低于空白对照组(P<0.05);模型组大鼠十二指肠隐窝深度、绒毛宽度及回肠绒毛高度均极显著低于空白对照组(P<0.01);模型组大鼠小肠不同段的绒毛高度与隐窝深度比值均低于空白对照组,但差异不显著(P>0.05);模型组大鼠盲肠内容物中sIgA含量极显著低于空白对照组(P<0.01)。结果提示,本试验成功建立了脾虚泄泻大鼠模型,脾虚泄泻可损伤大鼠小肠黏膜结构并降低盲肠中sIgA含量。 展开更多

目的:探讨参杞强精颗粒对慢性疲劳综合征(CFS)脾肾阳虚证模型大鼠的影响以及免疫调节作用。方法:将50只无特定病原体(SPF)级健康雄性斯泼累格·多雷(SD)大鼠随机分为正常对照组,模型对照组,参杞强精颗粒高剂量组、中剂量组、低剂量... 目的:探讨参杞强精颗粒对慢性疲劳综合征(CFS)脾肾阳虚证模型大鼠的影响以及免疫调节作用。方法:将50只无特定病原体(SPF)级健康雄性斯泼累格·多雷(SD)大鼠随机分为正常对照组,模型对照组,参杞强精颗粒高剂量组、中剂量组、低剂量组,采用游泳力竭实验造成大鼠体力疲劳,慢性束缚应激造成大鼠心理疲劳,同时给予番泻叶煎煮液灌胃造成脾肾阳虚证候,构建CFS脾肾阳虚证大鼠模型,造模成功后分别按照各组剂量灌胃,连续给药21 d后,测定各组大鼠的胸腺和脾脏指数,酶联免疫吸附试验(ELISA)检测血清免疫球蛋白G(IgG)、免疫球蛋白M(IgM)、免疫球蛋白A(IgA)浓度以及细胞因子白细胞介素2(IL-2)、白细胞介素6(IL-6)和γ干扰素(IFN-γ)含量,MTT法检测脾脏T、B淋巴细胞增殖能力,流式细胞法检测各组大鼠外周血T淋巴细胞亚群分布情况。结果:行为学结果显示,模型大鼠的体质量增长缓慢,精神萎靡,摄食水量减少,活动减少,便质稀软,力竭游泳时间减少,悬尾静止时间增加。药物干预后,与模型组比较,各给药组大鼠精神状态、体质量、摄食水量均显著改善,力竭游泳时间增加、悬尾静止时间减少(P<0.01或P<0.05);参杞强精颗粒各剂量组能显著提升大鼠胸腺和脾脏指数,提高IgG、Ig M、IgA浓度和IL-2、IL-6、IFN-γ的含量,显著提高脾脏T、B淋巴细胞增殖能力,也能提高CD3+、CD4+T淋巴细胞百分率和CD4+/CD8+T比值,降低CD8+T淋巴细胞百分率,差异有统计学意义(P<0.01或P<0.05)。结论:参杞强精颗粒能显著改善慢性疲劳综合征脾肾阳虚证大鼠免疫功能,缓解疲劳症状,且具有很好的免疫调节作用。 展开更多

目的观察黄芪建中汤对脾胃虚寒证胃溃疡大鼠炎症因子及HGF/c-Met信号通路的影响。方法将60只大鼠随机分为4组,即正常组、模型组、黄芪建中汤组[黄芪建中汤6.8 g/(kg·d)]、奥美拉唑组[奥美拉唑肠溶胶囊4.2 mg/(kg·d)],每组15... 目的观察黄芪建中汤对脾胃虚寒证胃溃疡大鼠炎症因子及HGF/c-Met信号通路的影响。方法将60只大鼠随机分为4组,即正常组、模型组、黄芪建中汤组[黄芪建中汤6.8 g/(kg·d)]、奥美拉唑组[奥美拉唑肠溶胶囊4.2 mg/(kg·d)],每组15只大鼠。正常组除外,其他组采用耗气破气法结合饥饱失常法进行大鼠脾胃虚寒证模型造模,再采用冰醋酸法建立大鼠胃溃疡模型。比较各组大鼠体质量、溃疡指数;HE染色观察大鼠胃组织病理学变化;采用酶联免疫吸附法检测血清中白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)含量;免疫组化法检测胃组织中肝细胞生长因子(HGF)、肝细胞生长因子受体(c-Met)表达;荧光定量PCR检测胃组织HGF、c-Met m RNA水平。结果与正常组相比,模型组大鼠体质量降低,溃疡指数升高,IL-6、IL-1β、TNF-α含量升高,HGF升高,HGF、c-Met m RNA升高(P<0.05);与模型组相比,黄芪建中汤组和奥美拉唑组大鼠体质量升高,溃疡指数降低,IL-6、IL-1β、TNF-α降低,HGF升高,HGF、c-Met mRNA升高(P<0.05)。结论黄芪建中汤可以通过抑制炎症因子、调节HGF/c-Met通路的表达促进胃黏膜修复。 展开更多

目的:基于“肝病实脾”理论探讨逍遥散对肝纤维化大鼠肝星状细胞铁死亡的影响。方法:采用腹腔注射四氯化碳的方法建立肝纤维化大鼠模型,随机分为模型组、逍遥散组、水飞蓟宾葡甲胺组,每组12只,另选12只SD大鼠腹腔注射等剂量玉米油设为... 目的:基于“肝病实脾”理论探讨逍遥散对肝纤维化大鼠肝星状细胞铁死亡的影响。方法:采用腹腔注射四氯化碳的方法建立肝纤维化大鼠模型,随机分为模型组、逍遥散组、水飞蓟宾葡甲胺组,每组12只,另选12只SD大鼠腹腔注射等剂量玉米油设为对照组,以逍遥散、水飞蓟宾葡甲胺分组处理后检测各组大鼠肝功能指标天冬氨酸氨基转移酶(AST)和丙氨酸氨基转移酶(ALT);检测各组大鼠肝指数并以Masson染色检测各组大鼠肝纤维化,比较其肝胶原容积分数(CVF);以免疫组织化学染色检测各组大鼠肝星状细胞活化,比较其肝组织α-SMA阳性表达;通过16SrRNA基因测序检测各组大鼠肠道菌群改变。分离培养上述各组大鼠原代肝星状细胞(HSC),以试剂盒测定各组大鼠HSC铁死亡指标:铁含量、谷胱甘肽(GSH)及丙二醛(MDA)水平;以免疫印迹法检测各组大鼠HSC铁死亡标志蛋白[长链脂酰辅酶A合成酶4(ACSL4)、环加氧酶2(PTGS2)、谷胱甘肽过氧化酶4(GPX4)]表达。结果:与对照组相比,模型组大鼠拟杆菌目丰度、HSC铁含量、MDA水平、ACSL4与PTGS2蛋白表达均明显降低(P<0.05),血清AST与ALT水平、肝指数、肝CVF、肝组织α-SMA阳性比、ACE指数、梭菌目丰度、HSC中GSH水平与GPX4蛋白表达均明显升高(P<0.05);与模型组相比,逍遥散组、水飞蓟宾葡甲胺组大鼠拟杆菌目丰度、HSC铁含量、MDA水平、ACSL4与PTGS2蛋白表达均升高(P<0.05),血清AST与ALT水平、肝指数、肝CVF、肝组织α-SMA阳性比、ACE指数、梭菌目丰度、HSC中GSH水平与GPX4蛋白表达均降低(P<0.05);与逍遥散组相比,水飞蓟宾葡甲胺组大鼠各指标差异无统计学意义(P>0.05)。结论:逍遥散可通过诱导肝星状细胞铁死亡而抑制其活性,进而减轻肝纤维化大鼠肝组织肝纤维化,减轻其肝功能损伤,最终对大鼠发挥明显肝保护作用。 展开更多

Dimethyl curcumin,a synthetic derivative of curcumin,has good anti-cancer,anti-arthritis and anti-inflammatory properties,but underline mechanism was unclear.Activation,differentiation,proliferation and apoptosis of lymphocytes have important functions during arthritis development and immune homeostasis.Over-activation of lymphocyte causes inflammatory cytokines storm,leading to an immune imbalance and tissue damages.The aim of this study was to investigate the anti-inflammation mechanism of liposome encapsulated dimethyl curcumin(Lipo-DiMC)on over-activated rat spleen lymphocytes.In this study,Lipo-DiMC was produced to increase the solubility;the primary rat spleen lymphocytes were extracted and the Concanavalin A induced cell activation model was established to study the cellular responses to Lipo-DiMC treatment in-vitro.The results showed that Lipo-DiMC suppressed Concanavalin A-induced cell proliferation,inhibited cells entering G2/M phase,and reduced the ratio of necrosis/apoptosis by manipulating intracellular p53/p21/PCNA/JNK-1 pathways.Furthermore,Lipo-DiMC increased the accumulations of intracellular matrix metalloproteinase-9 and reduced matrix metalloproteinase-9 secretion of rat spleen lymphocytes.Also,Lipo-DiMC increased intracellular caspase-3 expression and reduced Cat-C activity in activated rat spleen lymphocytes,involving in intracellular proteolysis.Our findings suggest that dimethyl curcumin effectively alleviates Concanavalin A induced over-activation of rat spleen lymphocytes,thereby inhibiting inflammatory cytokines storm and restoring cell homeostasis. 展开更多

目的探讨壮骨强肌方对去卵巢大鼠骨密度及IL-17的影响。方法30只SPF级6月龄雌性SD大鼠按随机数字表分为Sham组(假手术组,n=9)和模型组(n=21),模型组予以双侧输卵管结扎并卵巢切除,Sham组切除卵巢周围等体积脂肪组织,12周后两组各随机抽... 目的探讨壮骨强肌方对去卵巢大鼠骨密度及IL-17的影响。方法30只SPF级6月龄雌性SD大鼠按随机数字表分为Sham组(假手术组,n=9)和模型组(n=21),模型组予以双侧输卵管结扎并卵巢切除,Sham组切除卵巢周围等体积脂肪组织,12周后两组各随机抽取3只大鼠检测腰椎骨密度验证造模。造模成功后,将模型组大鼠按照随机数字表分为OVX组(模型对照组)、ZGQJ组(壮骨强肌方组)和Alen组(阿仑膦酸钠组),每组6只。ZGQJ组予以壮骨强肌方,Alen组予以阿仑膦酸钠,OVX组和Sham组大鼠予以等体积生理盐水,均灌胃给药,连续给药12周后取材。DEXA检测腰椎骨密度、Micro-CT检测胫骨骨小梁微结构、股骨HE染色、血清IL-17检测、胫骨IL-17免疫荧光染色,脾脏HE及IL-17免疫组化染色。结果与OVX组比较,ZGQJ组与Alen组腰椎BMD增高(P<0.05),ZGQJ组与Alen组胫骨BV/TV、Tb.N及Tb.Th增高(P<0.05)且Tb.Sp降低(P<0.05),ZGQJ组与Alen组股骨HE染色骨小梁面积增多(P<0.05),ZGQJ组与Alen组血清Elisa检测IL-17水平降低(P<0.05),ZGQJ组胫骨免疫荧光染色IL-17表达降低(P<0.05),ZGQJ组脾脏HE染色组织病理结构改善,ZGQJ组脾脏指数提高(P<0.05),ZGQJ组脾脏免疫组化染色IL-17表达降低(P<0.05)。结论中药壮骨强肌方具有提高去卵巢大鼠腰椎骨密度、改善胫骨及股骨骨小梁结构、降低IL-17表达及改善脾脏免疫功能的作用。 展开更多

目的基于转录组学技术分析藿香正气滴丸干预湿困脾胃证的潜在作用机制。方法将SD大鼠随机分为3组:正常组、模型组、藿香正气滴丸组(1 g/kg)。采用综合物理造模方法建立湿困脾胃大鼠模型,共造模20 d,造模第11天灌胃给药。测量大鼠体质量... 目的基于转录组学技术分析藿香正气滴丸干预湿困脾胃证的潜在作用机制。方法将SD大鼠随机分为3组:正常组、模型组、藿香正气滴丸组(1 g/kg)。采用综合物理造模方法建立湿困脾胃大鼠模型,共造模20 d,造模第11天灌胃给药。测量大鼠体质量变化、观察大鼠脏器系数、结肠组织病理变化并用ELISA法检测大鼠血清免疫球蛋白G(IgG)水平。运用转录组学技术检测结肠组织中差异基因表达情况。利用RT-qPCR验证藿香正气滴丸对关键差异基因mRNA表达水平的影响。结果与正常组比较,模型组大鼠脾脏、胸腺指数及IgG水平显著降低,结肠组织变化明显。与模型组比较,藿香正气滴丸能显著回调大鼠脾脏、胸腺指数及IgG水平,能改善结肠病理变化。转录组学分析结果显示,与正常组比较,湿困脾胃证模型组差异表达基因共525个,其中有192个基因显著上调,333个基因显著下调。藿香正气滴丸共回调了113个差异表达基因。GO功能富集结果表明差异表达基因主要涉及ATP的合成、调节干扰素系统等;KEGG分析结果表明差异表达基因主要参与氧化磷酸化代谢通路、生热作用、RIG-I样受体信号通路、NOD样受体信号通路等相关信号通路;RT-qPCR验证显示,藿香正气滴丸能明显回调差异表达基因。结论藿香正气滴丸干预湿困脾胃证的作用机制可能与其调节机体能量代谢、炎症反应和改善机体免疫系统有关。 展开更多

A previous study from our group showed that Jiawei Wendan decoction inhibits protein expression of interleukin-1β, 2, and 6, as well as plasma neuropeptide Y, P substance and somatostatin in the hippocampus of depression rat models. The present study analyzed the influence of Jiawei Wendan decoction on the mitogen-activated protein kinase signal transduction pathway in the hippocampus. Results demonstrated that Jiawei Wendan decoction effectively upregulated expression of small molecular G proteins, extracellular regulated kinase 1/2, and activated ribosomal S6 kinase protein in the rat hippocampus. In addition, Jiawei Wendan decoction exhibits antidepressant effects similar to fluoxetine. The underlying mechanisms were shown to be dependent on increased mitogen-activated protein kinase signal transduction pathway activity. 展开更多

BACKGROUND: The effect of pituitary adenylate cyclase activating polypeptide (PACAP) during traumatic brain injury (TBI) and whether it can modulate secondary injury has not been reported previously. The present study evaluated the potential protective effects of ventricular infusion of PACAP in a rat model of TBI.METHODS: Male Sprague Dawley rats were randomly divided into 3 treatment groups (n=6, each): sham-operated, vehicle (normal saline)+TBI, and PACAP+TBI. Normal saline or PACAP (1 μg/5 μL) was administered intracerebroventricularly 20 minutes before TBI. Right parietal cortical contusion was produced via a weight-dropping method. Brains were extracted 24 hours after trauma. Histological changes in brains were examined by HE staining. The numbers of CD4+ and CD8+ T cells in blood and the spleen were detected via flow cytometry.RESULTS: In injured brain regions, edema, hemorrhage, inflammatory cell infiltration, and swollen and degenerated neurons were observed under a light microscope, and the neurons were disorderly arrayed in the hippocampi. Compared to the sham group, average CD4+ CD8+ lymphocyte counts in blood and the spleen were significantly decreased in rats that received TBl+vehicle, and CD4- CD8+ were increased. In rats administered PACAP prior to TBI, damage was attenuated as evidenced by significantly increased CD4+, and decreased CD8+, T lymphocytes in blood and the spleen.CONCLUSION: Pretreatment with PACAP may protect against TBI by influencing periphery T cellular immune function. 展开更多

目的 探讨金卷升板胶囊对模型大鼠血小板减少的改善作用。方法 将36只雄性SD大鼠随机分为空白对照组(A组,等体积生理盐水),模型组(B组,等体积生理盐水),咖啡酸组(C组,9 mg/kg),以及金卷升板胶囊高、中、低剂量组(D_(1)组、D_(2)组、D_(3... 目的 探讨金卷升板胶囊对模型大鼠血小板减少的改善作用。方法 将36只雄性SD大鼠随机分为空白对照组(A组,等体积生理盐水),模型组(B组,等体积生理盐水),咖啡酸组(C组,9 mg/kg),以及金卷升板胶囊高、中、低剂量组(D_(1)组、D_(2)组、D_(3)组,1.00,0.50,0.25 g/kg),各6只。腹腔注射环磷酰胺溶液(100 mg/kg),每日1次,连续3 d,以复制血小板减少大鼠模型,同时A组大鼠腹腔注射等体积生理盐水。在建模开始后第5天,各组大鼠予相应药物或生理盐水灌胃,每日1次,连续14 d。检测血小板计数(PLT)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(Fib);采用酶联免疫吸附法检测白细胞介素10(IL-10)、肿瘤坏死因子-α(TNF-α)的水平;称定脾脏质量并计算脾脏系数;采用苏木素-伊红染色,并在显微镜下观察脾脏组织病理形态。结果 与A组比较,B组大鼠PLT显著减少,PT,APTT,TT均显著延长,Fib和TNF-α水平均显著升高,IL-10水平显著降低,脾脏系数显著降低(P<0.01)。与B组比较,C组、D_(1)组、D_(2)组大鼠PLT显著增加,PT,APTT,TT均显著缩短,Fib及TNF-α水平均显著降低;C组、D_(1)组、D_(2)组、D_(3)组大鼠IL-10水平均显著升高,脾脏系数显著升高(P<0.01);D_(1)组、D_(2)组、D_(3)组大鼠脾脏组织的B淋巴细胞数量均增加,细胞核内的嗜碱性物质着色明显,细胞核完整度较好。结论 金卷升板胶囊可改善模型大鼠的血小板减少情况,其机制可能与升高IL-10水平及降低TNF-α水平和改善大鼠的免疫有关。 展开更多

目的比较衢枳壳不同加工饮片对脾虚型大鼠胃肠功能的影响。方法SD大鼠随机分为空白组、模型组、阳性对照组(四君子颗粒组),衢枳壳“一体化”饮片低、中、高剂量组,衢枳壳传统饮片低、中、高剂量组,每组各10只(雌雄各半)。脾虚大鼠模型... 目的比较衢枳壳不同加工饮片对脾虚型大鼠胃肠功能的影响。方法SD大鼠随机分为空白组、模型组、阳性对照组(四君子颗粒组),衢枳壳“一体化”饮片低、中、高剂量组,衢枳壳传统饮片低、中、高剂量组,每组各10只(雌雄各半)。脾虚大鼠模型采用苦寒泻下加饥饱失常法复制,连续灌胃给药7 d,给药期间观察大鼠精神状态并记录;末次给药后禁食18 h,用灌胃半固体糊的方法检测各组大鼠的胃排空率以及小肠推进率;用ELISA试剂盒检测大鼠血清中胃泌素(GAS)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)的含量;血浆中乙酰胆碱(ACh)、血管活性肠肽(VIP)、P物质(SP)、胃动素(MTL)的含量。结果与空白组比较,模型组除SP指标变化无显著性,其余各个指标均具有显著性,其中MTL、GAS、Ach、TNF-α和IL-6含量显著降低(P<0.01),VIP含量显著升高(P<0.01)。各组给药7 d后,与模型组比较,阳性给药组除SP指标无显著性差异,其余指标变化均具有显著性;传统饮片与“一体化”饮片中、高剂量组除SP指标无显著性差异,其余指标变化均具有显著性差异;传统饮片与“一体化”饮片低剂量组,除MTL、GAS、SP无显著性差异,其余指标均具有显著性差异。结论衢枳壳传统饮片与“一体化”饮片对脾虚型大鼠的胃肠功能障碍具有治疗作用,且治疗效果等效。 展开更多

目的探究软坚消瘿颗粒对肝郁脾虚型桥本甲状腺炎(Hashimoto's thyroiditis,HT)大鼠甲状腺组织SOCS1\3\JAK-STATS信号通路表达的影响。方法SD大鼠40只,随机取10只作为正常对照组,剩余大鼠采用高碘饮水与皮下注射甲状腺球蛋白联合方... 目的探究软坚消瘿颗粒对肝郁脾虚型桥本甲状腺炎(Hashimoto's thyroiditis,HT)大鼠甲状腺组织SOCS1\3\JAK-STATS信号通路表达的影响。方法SD大鼠40只,随机取10只作为正常对照组,剩余大鼠采用高碘饮水与皮下注射甲状腺球蛋白联合方案建立桥本甲状腺炎大鼠模型,通过慢性束缚应激、过度疲劳、饮食失节复合方法建立肝郁脾虚大鼠模型。模型复制成功后,随机分为模型对照组(n=10)、雷公藤多苷片组(n=10)以及软坚消瘿颗粒组(n=10)。正常对照组给予常规饲养,模型对照组予以蒸馏水2 mL,雷公藤多苷片组给予雷公藤多苷片9.45 mg/kg,软坚消瘿颗粒组给予软坚消瘿颗粒16.17 g/kg,各组干预时间为8周。干预结束后比较各组大鼠一般情况、HE染色情况、甲状腺功能指标、SOCS1\3\JAK-STATS相关蛋白表达情况。结果实验过程中无大鼠死亡,与模型组相比,软坚消瘿颗粒组大鼠血清TPOAb、TGAb、FT3、FT4水平较低,TSH水平较高(P<0.05);与雷公藤多苷片组相比,软坚消瘿颗粒组大鼠血清TPOAb、TGAb、FT3、FT4水平较低(P<0.05),TSH水平较高(P<0.05)。与模型组相比,软坚消瘿颗粒组大鼠SOCS1、SOCS2、p-JAK2、p-STAT1蛋白表达水平较高(P<0.05);与雷公藤多苷片组相比,软坚消瘿颗粒组大鼠SOCS1、SOCS2、p-JAK2、p-STAT1蛋白表达水平较高(P<0.05)。结论软坚消瘿颗粒能够有效改善肝郁脾虚型HT大鼠的甲状腺功能,提高组织SOCS1、SOCS3表达水平,抑制炎性物质传导,推测与JAK-STST信号通路异常激活有关。 展开更多

目的:探讨隔药饼灸对克罗恩病(CD)模型大鼠的作用机制,为隔药饼灸的临床应用提供科学依据。方法:将18只雄性SD大鼠随机分为正常对照组、模型对照组和隔药饼灸组,每组6只。模型采用2,4,6-三硝基苯磺酸(TNBS)灌肠法建立,隔药饼灸组用天枢(... 目的:探讨隔药饼灸对克罗恩病(CD)模型大鼠的作用机制,为隔药饼灸的临床应用提供科学依据。方法:将18只雄性SD大鼠随机分为正常对照组、模型对照组和隔药饼灸组,每组6只。模型采用2,4,6-三硝基苯磺酸(TNBS)灌肠法建立,隔药饼灸组用天枢(双)和气海穴隔药饼灸治疗14 d。采用结直肠扩张刺激(CRD)检测内脏敏感性。采用逆转录PCR和蛋白质印迹法分别检测磷脂酰肌醇3激酶(PI3K)、蛋白激酶B(AKT)和哺乳动物雷帕霉素靶蛋白(mTOR)以及下游自噬相关16样蛋白1(Atg16L1)、免疫相关鸟苷三磷酸酶M蛋白(IRGM)的mRNA和蛋白表达水平。结果:与正常对照组比较,模型对照组和隔药饼灸组的内脏疼痛阈值均显著下降(P<0.001)。干预后,与模型对照组比较,隔药饼灸组内脏疼痛阈值均显著上升(P<0.01),与正常对照组比较,隔药饼灸组内脏疼痛阈值差异无统计学意义(P>0.05)。与正常对照组比较,模型对照组的PI3K、AKT、mTOR、Atg16L1、IRGM蛋白表达量及mRNA表达量均明显升高,差异有统计学意义(均P<0.01);与模型对照组比较,隔药饼灸组的PI3K、AKT、mTOR、Atg16L1、IRGM蛋白表达及mRNA表达均明显降低,差异有统计学意义(均P<0.001)。结论:隔药饼灸可通过P13K/AKT/mTOR信号通路调节CD模型大鼠结肠组织自噬,提高内脏疼痛阈值,促进大鼠结肠溃疡组织恢复正常。 展开更多

TMAO对脾虚高脂血症大鼠脂代谢的影响及香砂六君子汤的干预作用

目的 本研究拟探讨肠道菌群代谢物TMAO对脾虚高脂血症大鼠肝脂质代谢的影响,并进一步探讨香砂六君子汤干预肝脂质代谢紊乱的可能机制。方法 SD大鼠分为:空白对照组(C组);空白对照组+DMB(C+D组),DMB为TMAO抑制剂;脾虚高脂血症组(PG组);... 目的 本研究拟探讨肠道菌群代谢物TMAO对脾虚高脂血症大鼠肝脂质代谢的影响,并进一步探讨香砂六君子汤干预肝脂质代谢紊乱的可能机制。方法 SD大鼠分为:空白对照组(C组);空白对照组+DMB(C+D组),DMB为TMAO抑制剂;脾虚高脂血症组(PG组);脾虚高脂血症+DMB组(PG+D组);脾虚高脂血症+香砂六君子汤组(PG+XS组)。除C组、C+D组外各组构建以劳倦过度和高脂饲料相结合建立脾虚高脂血症模型(造模12周),模型建立后C+D组、PG+D组每天饮用水中给予1%DMB,PG+XS组每天灌胃香砂六君子汤(每天11.34 g生药/kg),其余给予等量生理盐水,4周后进行取材检测。全自动生化仪检测大鼠血脂含量;HE染色、油红O染色观察大鼠肝变化及脂质沉积情况;ELISA法检测大鼠肝FFA变化;试剂盒法检测各组大鼠TG、TC含量;LC-MS法检测大鼠血浆TMAO含量;qRT-PCR法检测大鼠肝PERK、FOXO1、SREBP-2、ABCA1、miR-33 mRNA相对表达水平;Western Blot检测大鼠肝SREBP-2、ABCA1蛋白表达。结果 (1)PG组大鼠血清中TC、TG和LDL-C含量较C组升高明显,HDL-C含量较C组下降显著;PG大鼠肝组织中FFA、TG、TC含量较C组增加显著;肝脂质沉积较C组加深,脂肪空泡数量大幅度增加;以上指标中C+D组较C组比较无显著性差异;PG+D组、PG+XS组与PG组相比可显著降低大鼠血清中TC、TG和LDL-C含量,升高HDL-C含量,降低肝组织中FFA、TG、TC含量,缓解肝组织脂质沉积现象,减少肝空泡;PG+D组与PG+XS组之间比较无显著性差异。(2)与C组比较,PG组大鼠血浆中TMAO含量增加显著,肝PERK和FOXO1、miR-33a mRNA相对含量明显增加,肝SREBP-2、ABCA1 mRNA和蛋白表达均降低显著;以上指标中C+D组较C组比较无显著性差异;PG+D组及PG+XS组与PG组相比可显著降低大鼠血浆中TMAO含量,降低肝PERK和FOXO1、miR-33a mRNA表达,升高肝SREBP-2、ABCA1基因mRNA和蛋白表达;PG+D组与PG+XS组之间无显著性差异。结论 TMAO可能通过PERK/FOXO1轴调控SREBP-2/miR-33a/ABCA1信号通路引起大鼠肝脂代谢紊乱,香砂六君子汤可能通过抑制TMAO含量达到抑制肝脂代谢紊乱。 展开更多
