An animal model of allergic asthma through the challenge of Pleurotus...Allergic asthma caused by mushroom spores (Pleurotus sapidus besidiospores) is a common health problem among mushroom-cultivating workers in China. An animal model of allergic asthma through the challenge of Pleurotus sapidus besidiospores in primed rats was developed for investigating the role of cytokines in the pathogenesis of the disease. In the study a series of related cytokines and their receptors, including their activity and mRNA levels of spleen lymphocytes isolated from asthmatic rats, were measured. Determined by 3H-TdR incorporation assay and NAG microcolorimetric assay, Con A-induced spleen lymphocyte proliferation and IL-2 activity in culture supernatants of spleen lymphocytes from 7-day challengedrats with allergic asthma increased significantly by 261% and 208%, respectively, as compared with those in the control. Cytokines and their receptor expression at mRNA levels were determined by RNA/cDNA hybridization, using (α-32P-dCTP radiolabeled cDNA probes for different cytokines and their receptors in vitro. The results showed that mRNA expression of IL-4, GM-CSF, IL-6, IL-2 and IL-2R, except IL-6R, in lymphocytes of 7-day-challenged asthma-suffering rats, increased significantly by 54%, 45%, 170%, 83% and 76%, respectively. In 2-day challenged-rats, mRNA levels of IL-4, IL-6 and IL-2 increased by 37%,58% and 125%, respectively, whereas mRNA leve1s of GM-CSF, IL-2R and IL-6R remained unchanged. Thus, the experimental results suggested a significant increase in TH2type cytokines in the pathogenesis of Pleurotus sapidus besidiospore-induced allergic asthma and IL-4 may play an essential role.展开更多
Objective:To observe the intervention effects of different concentrations of dexamethasone on the general situation and pulmonary histology of chronic asthma rats,and to explore the suitable intragastric dose of dexam...Objective:To observe the intervention effects of different concentrations of dexamethasone on the general situation and pulmonary histology of chronic asthma rats,and to explore the suitable intragastric dose of dexamethasone in the rat model of chronic asthma.Methods:75 healthy male Sprague-Dawley(SD)rats were randomly divided into normal group,M group,dexamethasone low dose,middle dose and high dose group by random number table.The rat model of chronic asthma was established by sensitization and challenge of ovalbumin(OVA).From the 20th day of the experiment,dexamethasone groups were given intragastric administration of 0.0625mg/(kg·d),0.125mg/(kg·d)and 0.5mg/(kg·d),respectively.The general situation of rats was observed and recorded.The histological changes of rat lungs were observed by HE staining and Masson staining.Results:The degree of inflammatory infiltration of bronchus and surrounding tissue in the M group was significantly higher than that in the normal group(P<0.01).The thickness of airway wall and smooth muscle and the deposition of airway collagen in the M group were significantly higher than those in the normal group(P<0.01).After one week of intervention,the average daily food intake and body mass of rats in the high dose group were significantly lower than those in the M group(P<0.01),and the increase in body mass in the middle and low dose groups was significantly slower than that in the M group(P<0.01).After 2 weeks of intervention,the average daily food intake of rats in the middle and low dose groups was significantly lower than that in the M group(P<0.01).After 4 weeks of intervention,the average daily food intake in the middle and low dose groups was significantly lower than that in the low dose group(P<0.01).The inflammatory infiltration of lung tissue in the middle and low dose groups was significantly less than that in the model group,and the thickness of airway wall and smooth muscle and the deposition of airway collagen in the middle and low dose groups were significantly lower than those in the model group(P<0.01).There was no difference between the two groups(P>0.05).A large number of rats in the high dose group died after 5 times of intervention.Three rats died in the DEXM group at the 5th week of intervention,and there was no death in the other group.Conclusion:Both middle and low doses of dexamethasone can be used as the positive control dose in the experiment of chronic asthmatic rats,but the late side effects of low dose of dexamethasone are less,so the low dose of dexamethasone may be a safer and more suitable dose for the experiment of chronic asthmatic rats.展开更多

Then we detected the autoantibodies to β 2adrenergic receptors through the sera of patients with asthma could inhibit the positive chronotropic action of clenbuterol. The results showed that all the sera of the patients with asthma (16 cases) had the autoantibodies to β 2 adrenergic receptors; in contrast, none of the control subjects (20 cases) had the autoantibodies to β 2 adrenergic receptors, and that the inhibitory autoantibodies were IgG type. This study suggests that the autoantibodies to β 2 adrenergic receptors may play an important role in the pathogenesis of asthma.展开更多

Results: In adrenaleetomy (ADX)-asthma acupuncture group and asthma acupuncture group, two-dimensional eleetrophoresis showed that there was an obvious fuscous point (MW=44.1kD, PI=5.1), which was 60S ribosomal protein L13, or hypothetical protein FLJ32191 analyzed by mass spectrum technique, different from other groups. In ADX-asthma acupuncture group, there were two obviously fuseous points (MW=39.8kD, PI=4.9 and MW=39.5kD, PI=5.0), which were zinc finger protein 208, and thyroid hormone receptor-associated protein analyzed by mass spectrum technique, and two special proteins (MW=17.3kD, PI〈4.5 and 16.9kD, PI〈4.5), which were Cyclophilin A and zinc finger protein 91 analyzed by mass spectrum technique, different from other groups. Conclusion: There are multi-target effects regulating the whole body in acupuncture treating asthma relating to immunoloregulation, gene expression, and protein svnthesization. A decrease in microbial infection in adolescents is implicated with an increase in the incidence of asthma and allergic diseases in adulthood,indicating that the microbiome plays a critical role in asthma.However,the ...A decrease in microbial infection in adolescents is implicated with an increase in the incidence of asthma and allergic diseases in adulthood,indicating that the microbiome plays a critical role in asthma.However,the microbial composition of the lower respiratory tract remains unclear,hindering the further exploration of the pathogenesis of asthma.This study aims to explore the microbial distribution and composition in the lungs of normal rats and rats with allergic asthma via 16S rDNA sequencing.The DNA of the pulmonary microbiome was extracted from the left lungs collected from normal control group(NC),saline control group(SC),and allergic asthma group(AA)under aseptic conditions.After the 16s rDNA V4eV5 region was amplified,the products were sequenced using Illumina high-throughput technology and subjected to operational taxonomic unit(OTU)cluster and taxonomy analysis.The OTU values of AA increased significantly compared with those of NC and SC.Microbiome structure analysis showed that the dominant phylum of the pulmonary microbiome changed from Proteobacteria in NC to Firmicutes in AA.Linear discriminant analysis indicated that the key microbiomes involved in the three groups varied.展开更多

Objective:To observe the effect of moxibustion at Feishu(BL 13)on related inflammatory cells and inflammatory factors in the bronchoalveolar lavage fluid of asthma model rats,and to explore the mechanism of moxibustio...Objective:To observe the effect of moxibustion at Feishu(BL 13)on related inflammatory cells and inflammatory factors in the bronchoalveolar lavage fluid of asthma model rats,and to explore the mechanism of moxibustion in treating asthma.Methods:A total of 48 Sprague-Dawley(SD)rats were randomly divided into a normal group,a model group,a moxibustion group and a medication group,with 12 rats in each group.Except for the normal group,the rats in the other three groups were subjected to ovalbumin sensitization to stimulate the asthma.At the same time,rats in the moxibustion group received moxibustion at bilateral Feishu(BL 13),and rats in the medication group received dexamethasone by intragastric administration.Rats in the normal and the model groups only received the same fixation and normal saline by intragastric administration.After the interventions,the inspiratory resistance,the expiratory resistance,and the pulmonary compliance were measured for rats in each group;the numbers of the inflammatory cells in the bronchoalveolar lavage fluid were counted;the levels of the involved inflammatory factors in bronchoalveolar lavage fluid were detected;the pathological morphologies of the lung tissues were observed under light microscope.Results:After modeling,compared with the normal group,the rats in the model group showed obvious asthma attack-like response,significantly increased inspiratory resistance and expiratory resistance(both P<0.01),and significantly reduced pulmonary compliance(P<0.01);thickened tracheal wall and the narrowed tracheal lumen observed under the light microscope;infiltration of inflammatory cells and increased eosinophils in and around the tracheal wall;significantly increased total number of inflammatory cells and proportion of eosinophils in the bronchoalveolar lavage fluid(all P<0.01);significantly reduced levels of interleukin(IL)-10,IL-12 and interferon(IFN)-γ(all P<0.01),and significantly increased levels of IL-4,IL-5 and tumor necrosis factor(TNF)-α(all P<0.01)in the bronchoalveolar lavage fluid.After intervention,compared with the model group,rats in the moxibustion and the medication groups showed significantly reduced asthma-like reaction,pathological morphological damage of lung tissue,inspiratory resistance and expiratory resistance(all P<0.01);significantly increased pulmonary compliance(P<0.01);significantly reduced total number of inflammatory cells,proportion of eosinophils,levels of IL-4,IL-5 and TNF-αin the bronchoalveolar lavage fluid(P<0.05 or P<0.01),while significantly increased IL-12 and IFN-γlevels(all P<0.01)in the bronchoalveolar lavage fluid;rats in the medication group also showed a significantly reduced IL-10 level in the bronchoalveolar lavage fluid(P<0.01);there was no statistically significant difference between the moxibustion and the medication groups(all P>0.05).Conclusion:Both moxibustion at Feishu(BL 13)and intragastric administration of dexamethasone can improve the asthma attack-like symptoms of ovalbumin-sensitized rats;regulating the inflammatory cell numbers and the inflammatory factor contents in the lung may be one mechanism of moxibustion in treating asthma.展开更多

METHODS: The histological, biochemical (cholesterol protein, zinc, copper, alkaline and acid phosphatase aryl sulphatase, lactic dehydrogenase and leucine amino peptidase) and hormonal (FSH, LH and testosterone) changes of caput and cauda epididymis in ADX rats were observed. RESULTS: Organ wet weight, histological studies and morphometric measurements indicated a cellular degeneration in caput and cauda epididymis of ADX rats. Serum testosterone level was significantly lower in ADX than in sham-operated rats, while the serum FSH and LH were below the detection limit of 1 mIU/mL. The enzymatic activity was higher in ADX than in sham-operated rats. Epididymal zinc level increased whereas copper level decreased in ADX rats compared to the sham-operated. CONCLUSION: Adrenalectomy leads to degeneration of caput and cauda epididymidis epithelial cells as a result of decreased supply of testosterone.展开更多

文摘Objective:To observe the effect of moxibustion at Feishu(BL 13)on related inflammatory cells and inflammatory factors in the bronchoalveolar lavage fluid of asthma model rats,and to explore the mechanism of moxibustion in treating asthma.Methods:A total of 48 Sprague-Dawley(SD)rats were randomly divided into a normal group,a model group,a moxibustion group and a medication group,with 12 rats in each group.Except for the normal group,the rats in the other three groups were subjected to ovalbumin sensitization to stimulate the asthma.At the same time,rats in the moxibustion group received moxibustion at bilateral Feishu(BL 13),and rats in the medication group received dexamethasone by intragastric administration.Rats in the normal and the model groups only received the same fixation and normal saline by intragastric administration.After the interventions,the inspiratory resistance,the expiratory resistance,and the pulmonary compliance were measured for rats in each group;the numbers of the inflammatory cells in the bronchoalveolar lavage fluid were counted;the levels of the involved inflammatory factors in bronchoalveolar lavage fluid were detected;the pathological morphologies of the lung tissues were observed under light microscope.Results:After modeling,compared with the normal group,the rats in the model group showed obvious asthma attack-like response,significantly increased inspiratory resistance and expiratory resistance(both P<0.01),and significantly reduced pulmonary compliance(P<0.01);thickened tracheal wall and the narrowed tracheal lumen observed under the light microscope;infiltration of inflammatory cells and increased eosinophils in and around the tracheal wall;significantly increased total number of inflammatory cells and proportion of eosinophils in the bronchoalveolar lavage fluid(all P<0.01);significantly reduced levels of interleukin(IL)-10,IL-12 and interferon(IFN)-γ(all P<0.01),and significantly increased levels of IL-4,IL-5 and tumor necrosis factor(TNF)-α(all P<0.01)in the bronchoalveolar lavage fluid.After intervention,compared with the model group,rats in the moxibustion and the medication groups showed significantly reduced asthma-like reaction,pathological morphological damage of lung tissue,inspiratory resistance and expiratory resistance(all P<0.01);significantly increased pulmonary compliance(P<0.01);significantly reduced total number of inflammatory cells,proportion of eosinophils,levels of IL-4,IL-5 and TNF-αin the bronchoalveolar lavage fluid(P<0.05 or P<0.01),while significantly increased IL-12 and IFN-γlevels(all P<0.01)in the bronchoalveolar lavage fluid;rats in the medication group also showed a significantly reduced IL-10 level in the bronchoalveolar lavage fluid(P<0.01);there was no statistically significant difference between the moxibustion and the medication groups(all P>0.05).Conclusion:Both moxibustion at Feishu(BL 13)and intragastric administration of dexamethasone can improve the asthma attack-like symptoms of ovalbumin-sensitized rats;regulating the inflammatory cell numbers and the inflammatory factor contents in the lung may be one mechanism of moxibustion in treating asthma.
文摘AIM: To investigate the effect of adrenalectomy (ADX) on the epididymidis of Sprague-Dawley rats. METHODS: The histological, biochemical (cholesterol protein, zinc, copper, alkaline and acid phosphatase aryl sulphatase, lactic dehydrogenase and leucine amino peptidase) and hormonal (FSH, LH and testosterone) changes of caput and cauda epididymis in ADX rats were observed. RESULTS: Organ wet weight, histological studies and morphometric measurements indicated a cellular degeneration in caput and cauda epididymis of ADX rats. Serum testosterone level was significantly lower in ADX than in sham-operated rats, while the serum FSH and LH were below the detection limit of 1 mIU/mL. The enzymatic activity was higher in ADX than in sham-operated rats. Epididymal zinc level increased whereas copper level decreased in ADX rats compared to the sham-operated. CONCLUSION: Adrenalectomy leads to degeneration of caput and cauda epididymidis epithelial cells as a result of decreased supply of testosterone.
