Micro-tubers are important propagules in potato breeding and potato production, and they are also dormant and easily transported and therefore good targets for mutation induction in potato mutation breeding. A prerequisite for mutation breeding is to determine optimal mutation treatments. Therefore, radio-sensitivity tests of a tetraploid and a diploid potato to gamma irradiation were undertaken. Effects of different gamma sources on radio-activity were also studied. In vitro potato cuttings were gamma irradiated using a wide dose range (0, 3, 6, 9, 12, 15 and 20 Gy). The irradiated cuttings were then cultured to induce micro-tubers directly in vitro. Micro-tuber morphotypes were assessed after irradiation of cuttings using three gamma sources with emission activities of 1.8, 7.07 and 139 Gy/min. The diploid species (Solanum verrucosum) was more radio-sensitive than the tetraploid cultivar Desirée (Solanum tuberosum). Gamma dose rates had significant influences on subsequent micro-tuber production at various mutant generations. Effects included reductions in the number, size and weight of micro-tubers produced. Gamma dose was more lethal for the diploid potato genotype and micro-tubers produced were small compared to those produced by the tetraploid genotype after irradiation. Different treatments are recommended for diploid and tetraploid potato irradiation in producing large mutant micro-tuber populations. The mutant micro-tuber populations may then be screened for interesting mutations/trait for both genetics and plant breeding purposes. 展开更多

We propose a new two-/three-stage dose-finding design called Target Toxicity(TT)for phase Ⅰ clinical trials,where we link the decision rules in the dose-finding process with the conclusions from a hypothesis test.The power to detect excessive toxicity is also given.This solves the problem of why the minimal number of patients is needed for the selected dose level.Our method provides a statistical explanation of traditional‘3+3’design using frequentist framework.The proposed method is very flexible and it incorporates other interval-based decision rules through different parameter settings.We provide the decision tables to guide investigators when to decrease,increase or repeat a dose for next cohort of subjects.Simulation experiments were conducted to compare the performance of the proposed method with other dose-finding designs.A free open source R package tsdf is available on CRAN.It is dedicated to deriving two-/three-stage design decision tables and perform dose-finding simulations. 展开更多

Phosphorus(P)limitation in soil is a major concern for crop productivity.However,the use of chemical fertilizer is hazardous to the environment and costly.Therefore,the use of phosphate-solubilizing bacteria(PSB)is an eco-friendly approach for a sustainable agricultural system.In the present study,a field trial was conducted for two consecutive years to study the effects of three PSB strains isolated,Bacillus licheniformis,Pantoea dispersa,and Staphylococcus sp.,with different P fertilizer rates on P uptake,P use efficiency(PUE),and grain yield of rice.The activities of soil enzymes were also studied in relation to PSB treatments.Comparative analysis of the yield and biochemical parameters revealed that inoculation of PSB consortium could reduce almost 50%of the recommended P dose in rice cultivation.Three PSB strains in combination with 50%P dose was most effective and showed the highest increases in P uptake and PUE as compared to the uninoculated control.Moreover,the PSB consortium combined with 50%P dose contributed to 50.58%and35.64%yield increases compared to the uninoculated control for 2018 and 2019,respectively.Significant increases in the activities of soil dehydrogenase,alkaline phosphatase,and acid phosphatase were also recorded under PSB treatment. 展开更多
