However, the enthalpies of most TRFsare not high, and the preparation methods are still at the laboratory scale. It remains a great challenge to use industrial spinning equipment to achieve continuous processing of TRF_(s) with excellent thermal and mechanical properties. Here, polyamide 6(PA6) based TRF_(s) with a sheath-core structure were prepared by bicomponent melt-spinning. The sheath-core TRF(TRF_(sc)) are composed of PA6 as sheath and functional PA6 as core, which are filled with the shape stable phase change materials(ssPCM),dendritic silica@polyethylene glycol(SiO_(2)@PEG). With the aid of the sheath structure, the filling content of SiO_(2)@PEG can reach 30 %, so that the enthalpy of the TRF_(s) can be as high as 21.3 J/g. The ultra-high enthalpy guarantees the temperature regulation ability during the alternating process of cooling and heating. In hot environment, the temperature regulation time is 6.59 min, and the temperature difference is 12.93℃. In addition, the mechanical strength of the prepared TRF_(sc) reaches 2.26 cN/dtex, which can fully meet its application in the field of thermal management textiles and devices to manage the temperature regulation of the human body or precision equipment, etc.展开更多
In this editorial,we comment on the article by Zhou et al.The study reveals the connection between ferroptosis and pyroptosis and the effect of silent information regulator sirtuin 1(SIRT1)activation in acute liver fa...In this editorial,we comment on the article by Zhou et al.The study reveals the connection between ferroptosis and pyroptosis and the effect of silent information regulator sirtuin 1(SIRT1)activation in acute liver failure(ALF).ALF is characterized by a sudden and severe liver injury resulting in significant hepatocyte damage,often posing a high risk of mortality.The predominant form of hepatic cell death in ALF involves apoptosis,ferroptosis,autophagy,pyroptosis,and necroptosis.Glutathione peroxidase 4(GPX4)inhibition sensitizes the cell to ferroptosis and triggers cell death,while Gasdermin D(GSDMD)is a mediator of pyroptosis.The study showed that ferroptosis and pyroptosis in ALF are regulated by blocking the p53/GPX4/GSDMD pathway,bridging the gap between the two processes.The inhibition of p53 elevates the levels of GPX4,reducing the levels of inflammatory and liver injury markers,ferroptotic events,and GSDMDN protein levels.Reduced p53 expression and increased GPX4 on deletion of GSDMD indicated ferroptosis and pyroptosis interaction.SIRT1 is a NAD-dependent deacetylase,and its activation attenuates liver injury and inflammation,accompanied by reduced ferroptosis and pyroptosis-related proteins in ALF.SIRT1 activation also inhibits the p53/GPX4/GSDMD axis by inducing p53 acetylation,attenuating LPS/D-GalN-induced ALF.展开更多

During the plugging process, the violent vibration will occur by the flow field, which can cause serious damage to the pipeline and PIPR. In this paper, we propose a dynamic regulating strategy to reduce the plugging-induced vibration by regulating the spoiler angle and plugging velocity. Firstly, the dynamic plugging simulation and experiment are performed to study the flow field changes during dynamic plugging. And the pressure difference is proposed to evaluate the degree of flow field vibration. Secondly, the mathematical models of pressure difference with plugging states and spoiler angles are established based on the extreme learning machine (ELM) optimized by improved sparrow search algorithm (ISSA). Finally, a modified Q-learning algorithm based on simulated annealing is applied to determine the optimal strategy for the spoiler angle and plugging velocity in real time. The results show that the proposed method can reduce the plugging-induced vibration by 19.9% and 32.7% on average, compared with single-regulating methods. This study can effectively ensure the stability of the plugging process.展开更多

Hippocampal neuronal loss causes cognitive dysfunction in Alzheimer’s disease.Adult hippocampal neurogenesis is reduced in patients with Alzheimer’s disease.Exercise stimulates adult hippocampal neurogenesis in rode...Hippocampal neuronal loss causes cognitive dysfunction in Alzheimer’s disease.Adult hippocampal neurogenesis is reduced in patients with Alzheimer’s disease.Exercise stimulates adult hippocampal neurogenesis in rodents and improves memory and slows cognitive decline in patients with Alzheimer’s disease.However,the molecular pathways for exercise-induced adult hippocampal neurogenesis and improved cognition in Alzheimer’s disease are poorly understood.Recently,regulator of G protein signaling 6(RGS6)was identified as the mediator of voluntary running-induced adult hippocampal neurogenesis in mice.Here,we generated novel RGS6fl/fl;APP_(SWE) mice and used retroviral approaches to examine the impact of RGS6 deletion from dentate gyrus neuronal progenitor cells on voluntary running-induced adult hippocampal neurogenesis and cognition in an amyloid-based Alzheimer’s disease mouse model.We found that voluntary running in APP_(SWE) mice restored their hippocampal cognitive impairments to that of control mice.This cognitive rescue was abolished by RGS6 deletion in dentate gyrus neuronal progenitor cells,which also abolished running-mediated increases in adult hippocampal neurogenesis.Adult hippocampal neurogenesis was reduced in sedentary APP_(SWE) mice versus control mice,with basal adult hippocampal neurogenesis reduced by RGS6 deletion in dentate gyrus neural precursor cells.RGS6 was expressed in neurons within the dentate gyrus of patients with Alzheimer’s disease with significant loss of these RGS6-expressing neurons.Thus,RGS6 mediated voluntary running-induced rescue of impaired cognition and adult hippocampal neurogenesis in APP_(SWE) mice,identifying RGS6 in dentate gyrus neural precursor cells as a possible therapeutic target in Alzheimer’s disease.展开更多

[Method] The suitable concentration combination of appendices was optimized by adding different concentrations of osmosis-regulating substances including mannitol, sorbitol and organic appendices such as Gln, CH and LH, into the somatic embryogenesis in wheat. [Result] The mannitol or sorbitol lower than 40 g/L was helpful for improving somatic embryogenesis; there was no significant difference in the induction rate of somatic embryogenesis when 300-500 mg/L Gln、CH or LH was respectively added into the induced medium, while somatic embryogenesis could be enhanced dramatically in the presence of 500 mg/L Gln together with 300 mg/L CH. [Conclusion] Somatic embryogenesis could be improved to some extent by different concentrations of osmosis-regulating substances and organic appendices, which laid foundation for establishing a more perfect system of somatic embryogenesis in wheat.展开更多

With the use of the AMESIM software, the changes of the piston displacement, the oil pressure, the magnitude attenuation and the phase lag of the system under different plugging states are studied after simulation. Besides, the influences of the symmetrical and unsymmetrical plugging on the system are also compared and the characteristic table is given. The duo-neural network is put forward to achieve an online diagnosis on the plugging fault of the servo valve. The first level of network helps to make the qualitative diagnosis of the plugging position while the second level is for the quantitative diagnosis of the degree of the plugged position. The research results show that plugging at different positions exerts different influences on the performance of the system. The unsymmetrical plugging mainly affects the regulation time while the symmetrical plugging leads to great changes in the magnitude attenuation and the phase lag.展开更多

The exact function of YAP in bone homeostasis remains controversial. Here we provide evidence for YAP's function in promoting osteogenesis, suppressing adipogenesis, and thus maintaining bone homeostasis.YAP is selectively expressed in osteoblast(OB)-lineage cells. Conditionally knocking out Yap in the OB lineage in mice reduces cell proliferation and OB differentiation and increases adipocyte formation, resulting in a trabecular bone loss. Mechanistically, YAP interacts with β-catenin and is necessary for maintenance of nuclear β-catenin level and Wnt/β-catenin signaling. Expression of β-catenin in YAP-deficient BMSCs(bone marrow stromal cells) diminishes the osteogenesis deficit. These results thus identify YAP-β-catenin as an important pathway for osteogenesis during adult bone remodeling and uncover a mechanism underlying YAP regulation of bone homeostasis.展开更多

In this study, we functionally characterized Ta ARR1, a cytokinin(CK) response regulator gene in Triticum aestivum, in mediating the N starvation adaptation in plants. Ta ARR1 harbors two conserved domains specified by plant ARR family members;subcellular localization analysis indicated its target onto nucleus after endoplasmic reticulum assortment. Ta ARR1 displayed modified expression upon the N starvation stressor, showing upregulated expression in roots and leaves over a 27-h N starvation treatment and whose induced transcripts were gradually recovered along with progression of the N recovery treatment. The tobacco lines overexpressing Ta ARR1 displayed improved low-N stress tolerance, displaying enlarged phenotype, increased biomass and N accumulation, and enhanced glutamine synthetase(GS) activities compared with wild type(WT) following the N starvation treatment. Expression analysis on genes encoding the nitrate transporter(NRT) and GS proteins in Nicotiana tabacum revealed that Nt NRT2.2 and Nt GS3 are upregulated in expression in the N-deprived transgenic lines, whose expression patterns were contrasted to other above family genes that were unaltered on transcripts between the transgenic lines and WT. Transgene analysis validated the function of Nt NRT2.2 and Nt GS3 in regulating N accumulation, GS activity, growth traits, and N use efficiency in plants. These results suggested the internal connection between the Ta ARR1-mediated N starvation tolerance and the modified transcription of distinct N acquisitionand assimilation-associated genes. Our investigation together indicates that Ta ARR1 is essential in plant N starvation adaptation due to the gene function in transcriptionally regulating distinct NRT and GS genes that affect plant N uptake and assimilation under the N starvation condition.展开更多
In plants,the vesicle fusion process plays a vital role in pathogen defence.However,the importance of the vesicle fusion process in apple ring rot has not been studied.Here,we isolated and characterised the apple synt...In plants,the vesicle fusion process plays a vital role in pathogen defence.However,the importance of the vesicle fusion process in apple ring rot has not been studied.Here,we isolated and characterised the apple syntaxin gene MdSYP121.Silencing the MdSYP121 gene in transgenic apple calli increased tolerance to Botryosphaeria dothidea infection;this increased tolerance was correlated with salicylic acid(SA)synthesis-related and signalling-related gene transcription.In contrast,overexpressing MdSYP121 in apple calli resulted in the opposite phenotypes.In addition,the results of RNA sequencing(RNA-Seq)and quantitative real-time PCR(qRT-PCR)assays suggested that MdSYP121 plays an important role in responses to oxidation–reduction reactions.Silencing MdSYP121 in apple calli enhanced the expression levels of reactive oxygen species(ROS)-related genes and the activity of ROS-related enzymes.The enhanced defence response status in MdSYP121-RNAi lines suggests that syntaxins are involved in the defence response to B.dothidea.More importantly,we showed that MdSYP121 forms a soluble N-ethylmaleimide-sensitive factor attachment protein receptor(SNARE)complex with MdSNAP33,and the complex may participate in regulating resistance to B.dothidea.In conclusion,by regulating the interaction of SA pathway and oxidation–reduction process,MdSYP121 can influence the pathogen infection process in apple.展开更多

Inhibiting proliferation and inducing apoptosis of hepatic stel...Liver fibrosis is an important health problem that can further progress into cirrhosis or liver cancer,and result in significant morbidity and mortality. Inhibiting proliferation and inducing apoptosis of hepatic stellate cells(HSCs) may be the key point to reverse liver fibrosis. At present,anti-fibrosis drugs are rare. Kinetin is a type of plant-derived cytokinin which has been reported to control differentiation and induce apoptosis of human cells. In this study,the HSCs were incubated with different concentrations of kinetin. The proliferation of rat HSCs was measured by MTT assay,cell cycle and apoptosis were analyzed by flow cytometry,and the apoptosis was examined by TUNEL method. The expression of Bcl-2 and Bax proteins was detected by immunocytochemistry staining. It was found that kinetin could markedly inhibit proliferation of HSCs. In a concentration range of 2 to 8 μg/m L,the inhibitory effects of kinetin on proliferation of HSCs were increased with the increased concentration and the extension of time(P〈0.01). Flow cytometry indicated that kinetin could inhibit the DNA synthesis from G0/G1 to S phase in a dose-dependent manner(P〈0.01). The apoptosis rates of the HSCs treated with 8,4 and 2 μg/m L kinetin(25.62%±2.21%,15.31%±1.9% and 6.18%±1.23%,respectively) were increased significantly compared with the control group(3.81%±0.93%)(P〈0.01). All the DNA frequency histogram in kinetin-treated groups showed obvious hypodiploid peak(sub-G1 peak),and with the increase of kinetin concentrations,the apoptosis rate of HSCs also showed a trend of increase. It was also found that kinetin could down-regulate the expression of Bcl-2,and up-regulate the expression of Bax,leading to the decreased ratio of Bcl-2/Bax significantly. The kinetin-induced apoptosis of HSCs was positively correlated with the expression of Bax,and negatively with the expression of Bcl-2. It was concluded that kinetin can inhibit activation and proliferation of HSCs by interrupting the cell cycle at G1/S restriction point and inducing apoptosis of HSCs via reducing the ratio of Bcl-2/Bax.展开更多

More and more basic studies have indicated that gut microbiota can regulate brain function through the gut-brain axis, and dysbiosis of intestinal microbiota was related to anxiety. However, there is no specific evidence to support treatment of anxiety by regulating intestinal microbiota. Aims To find evidence sup porting improvement of anxiety symptoms by regulation of intestinal microbiota. Methods This systematic review of randomised controlled trials was searched based on the following databases: PubMed, EMBASE, the Cochrane Library, OVID, Web of Knowledge, China National Knowledge Infrastructure (CNKI), Wanfang Data, VIP databases and SinoMed. The retrieval time dated back to 25 July 2018. Then we screened research literatures based on established inclusion and exclusion criteria. Quality evaluation for each included study was done using the Cochrane risk of bias and the Jadad scale. Results A total of 3334 articles were retrieved and 21 studies were included which contained 1503 subjects. In the 21 studies, 15 chose probiotics as interventions to regulate intestinal microbiota and six chose non-probiotic ways such as adjusting daily diets. Probiotic supplements in seven studies contained only one kind of probiotic, two studies used a product that contained two kinds of probiotics and the supplements used in the other five studies included at least three kinds of probiotics. In the studies that used treatment as usual plus interventions regulating intestinal flora (IRIF) as interventions (five studies), only non-probiotic ways were effective (two studies), which means 40% of studies were effective;in the studies that used IRIF alone (16 studies, 11 studies used probiotic ways and 5 studies used non-probiotic ways), 56% of studies could improve anxiety symptoms, and 80% of studies that conducted the non-probiotic interventions were effective, while 45% of studies that used probiotic supplementations had positive effects on anxiety symptoms. Overall, 11 studies showed a positive effect on anxiety symptoms by regulating intestinal microbiota, which indicated 52% of the 21 studies were effective, and there were five studies that used probiotic supplements as interventions and six used non-probiotic interventions. In addition, it should be noted that six of seven studies showed that regulation of intestinal microbiota could treat anxiety symptoms, the rate of efficacy was 86%.Conclusions We find that more than half of the studies included showed it was positive to treat anxiety symptoms by regulation of intestinal microbiota. There are two kinds of interventions (probiotic and non-probiotic interventions) to regulate intestinal microbiota, and it should be highlighted that the non-probiotic interventions were more effective than the probiotic interventions. More studies are needed to clarify this conclusion since we still cannot run meta-analysis so far.展开更多
Objective:An increasing number of studies indicate that autophagy plays an important role in the pathogenesis of spinal cord injury,and that regulating autophagy can enhance recovery from spinal cord injury.However,th...Objective:An increasing number of studies indicate that autophagy plays an important role in the pathogenesis of spinal cord injury,and that regulating autophagy can enhance recovery from spinal cord injury.However,the effect of regulating autophagy and whether autophagy is detrimental or beneficial after spinal cord injury remain unclear.Therefore,in this study we evaluated the effects of autophagy regulation on spinal cord injury in rats by direct and indirect comparison,in an effort to provide a basis for further research.Data source:Relevant literature published from inception to February 1,2018 were included by searching Wanfang,CNKI,Web of Science,MEDLINE(OvidSP),PubMed and Google Scholar in English and Chinese.The keywords included"autophagy","spinal cord injury",and"rat".Data selection:The literature included in vivo experimental studies on autophagy regulation in the treatment of spinal cord injury(including intervention pre-and post-spinal cord injury).Meta-analyses were conducted at different time points to compare the therapeutic effects of promoting or inhibiting autophagy,and subgroup analyses were also conducted.Outcome measure:Basso,Beattie,and Bresnahan scores.Results:Of the 622 studies,33 studies of median quality were included in the analyses.Basso,Beattie,and Bresnahan scores were higher at 1 day(MD=1.80,95%CI:0.81-2.79,P=0.0004),3 days(MD=0.92,95%CI:0.72-1.13,P<0.00001),1 week(MD=2.39,95%CI:1.85-2.92,P<0.00001),2 weeks(MD=3.26,95%CI:2.40-4.13,P<0.00001),3 weeks(MD=3.13,95%CI:2.51-3.75,P<0.00001)and 4 weeks(MD=3.18,95%CI:2.43-3.92,P<0.00001)after spinal cord injury with upregulation of autophagy compared with the control group(drug solvent control,such as saline group).Basso,Beattie,and Bresnahan scores were higher at 1 day(MD=6.48,95%CI:5.83-7.13,P<0.00001),2 weeks(MD=2.43,95%CI:0.79-4.07,P=0.004),3 weeks(MD=2.96,95%CI:0.09-5.84,P=0.04)and 4 weeks(MD=4.41,95%CI:1.08-7.75,P=0.01)after spinal cord injury with downregulation of autophagy compared with the control group.Indirect comparison of upregulation and downregulation of autophagy showed no differences in Basso,Beattie,and Bresnahan scores at 1 day(MD=-4.68,95%CI:-5.840 to-3.496,P=0.94644),3 days(MD=-0.28,95%CI:-2.231-1.671,P=0.99448),1 week(MD=1.83,95%CI:0.0076-3.584,P=0.94588),2 weeks(MD=0.81,95%CI:-0.850-2.470,P=0.93055),3 weeks(MD=0.17,95%Cl:-2.771-3.111,P=0.99546)or 4 weeks(MD=-1.23,95%Cl:-4.647-2.187,P=0.98264)compared with the control group.Conclusion:Regulation of autophagy improves neurological function,whether it is upregulated or downregulated.There was no difference between upregulation and downregulation of autophagy in the treatment of spinal cord injury.The variability in results among the studies may be associated with differences in research methods,the lack of clearly defined autophagy characteristics after spinal cord injury,and the limited autophagy monitoring techniques.Thus,methods should be standardized,and the dynamic regulation of autophagy should be examined in future studies.展开更多

Linkage analysis indicated that the α- and β-globin genes were oriented head-to-head relative to each other. To identify the regulatory elements present in the intergenic and intragenic regions of the globin complex, the intergenic region alone or together with the β-globin gene first intron was cloned into the luciferase-reporter vector pGL3-Basic respectively, and the chimeric constructs were tran- siently transfected into Vero cells and primary fish erythrocytes. The intergenic region cannot support the high-level expression of luciferase. However, the promoter activity of the intergenic region was strongly stimulated by the positive regulatory elements (PRE) located in the β-globin gene intron 1. Thus, it is proposed that the intergenic promoters and intragenic PRE were necessary for the effective expression of the linked α- and β-globin genes.展开更多

文摘Obesity and related metabolic syndromes have been recognized as important disease risks,in which the role of adipokines cannot be ignored.Adiponectin(ADP)is one of the key adipokines with various beneficial effects,including improving glucose and lipid metabolism,enhancing insulin sensitivity,reducing oxidative stress and inflammation,promoting ceramides degradation,and stimulating adipose tissue vascularity.Based on those,it can serve as a positive regulator in many metabolic syndromes,such as type 2 diabetes(T2D),cardiovascular diseases,non-alcoholic fatty liver disease(NAFLD),sarcopenia,neurodegenerative diseases,and certain cancers.Therefore,a promising therapeutic approach for treating various metabolic diseases may involve elevating ADP levels or activating ADP receptors.The modulation of ADP genes,multimerization,and secretion covers the main processes of ADP generation,providing a comprehensive orientation for the development of more appropriate therapeutic strategies.In order to have a deeper understanding of ADP,this paper will provide an all-encompassing review of ADP.

文摘Temperature regulating fibers(TRF_(s)) with high enthalpy and high form stability are the key factors for thermal management. However, the enthalpies of most TRFsare not high, and the preparation methods are still at the laboratory scale. It remains a great challenge to use industrial spinning equipment to achieve continuous processing of TRF_(s) with excellent thermal and mechanical properties. Here, polyamide 6(PA6) based TRF_(s) with a sheath-core structure were prepared by bicomponent melt-spinning. The sheath-core TRF(TRF_(sc)) are composed of PA6 as sheath and functional PA6 as core, which are filled with the shape stable phase change materials(ssPCM),dendritic silica@polyethylene glycol(SiO_(2)@PEG). With the aid of the sheath structure, the filling content of SiO_(2)@PEG can reach 30 %, so that the enthalpy of the TRF_(s) can be as high as 21.3 J/g. The ultra-high enthalpy guarantees the temperature regulation ability during the alternating process of cooling and heating. In hot environment, the temperature regulation time is 6.59 min, and the temperature difference is 12.93℃. In addition, the mechanical strength of the prepared TRF_(sc) reaches 2.26 cN/dtex, which can fully meet its application in the field of thermal management textiles and devices to manage the temperature regulation of the human body or precision equipment, etc.
文摘In this editorial,we comment on the article by Zhou et al.The study reveals the connection between ferroptosis and pyroptosis and the effect of silent information regulator sirtuin 1(SIRT1)activation in acute liver failure(ALF).ALF is characterized by a sudden and severe liver injury resulting in significant hepatocyte damage,often posing a high risk of mortality.The predominant form of hepatic cell death in ALF involves apoptosis,ferroptosis,autophagy,pyroptosis,and necroptosis.Glutathione peroxidase 4(GPX4)inhibition sensitizes the cell to ferroptosis and triggers cell death,while Gasdermin D(GSDMD)is a mediator of pyroptosis.The study showed that ferroptosis and pyroptosis in ALF are regulated by blocking the p53/GPX4/GSDMD pathway,bridging the gap between the two processes.The inhibition of p53 elevates the levels of GPX4,reducing the levels of inflammatory and liver injury markers,ferroptotic events,and GSDMDN protein levels.Reduced p53 expression and increased GPX4 on deletion of GSDMD indicated ferroptosis and pyroptosis interaction.SIRT1 is a NAD-dependent deacetylase,and its activation attenuates liver injury and inflammation,accompanied by reduced ferroptosis and pyroptosis-related proteins in ALF.SIRT1 activation also inhibits the p53/GPX4/GSDMD axis by inducing p53 acetylation,attenuating LPS/D-GalN-induced ALF.

文摘Granulomatous mastitis(GM)is a benign granulomatous condition,and its pathogenesis may be related to autoimmune disorders.Cellular immunity,humoral immunity,immunoglobulins,and complement could all play a role in the disease process,showing certain clinical patterns.Corticosteroids can quickly control disease progression,and immunosuppressants can be used for complex and refractory GM cases.In traditional Chinese medicine(TCM),“healthy qi”is similar to immune system function.For GM with deficient healthy qi,TCM treatments such as internal and external herbal applications can help regulate immune function and shorten disease duration by staged and TCM treatment,regulating viscera,reinforcing healthy qi,and eliminating pathogenic factors.

文摘Photocatalytic reduction of CO_(2) into fuel represents a promising approach for achieving carbon neutrality,while realizing high selectivity in this process is challenging due to uncontrollable reaction intermediate and retarded desorption of target products.Engineering the interface microenvironment of catalysts has been proposed as a strategy to exert a significant influence on reaction outcomes,yet it remains a significant challenge.In this study,amino alkylation was successfully integrated into the melem unit of polymeric carbon nitrides(PCN),which could efficiently drive the photocatalytic CO_(2) reduction.Experimental characterization and theoretical calculations revealed that the introduction of amino alkylation lowers the energy barrier for CO_(2) reduction into^(*)COOH intermediate,transforming the adsorption of^(*)COOH intermediate from the endothermic to an exothermic process.Notably,the as-prepared materials demonstrated outstanding performance in photocatalytic CO_(2) reduction,yielding CO_(2)at a rate of 152.8μmol h^(-1) with a high selectivity of 95.4%and a quantum efficiency of 6.6%.

文摘Pipeline isolation plugging robot (PIPR) is an important tool in pipeline maintenance operation. During the plugging process, the violent vibration will occur by the flow field, which can cause serious damage to the pipeline and PIPR. In this paper, we propose a dynamic regulating strategy to reduce the plugging-induced vibration by regulating the spoiler angle and plugging velocity. Firstly, the dynamic plugging simulation and experiment are performed to study the flow field changes during dynamic plugging. And the pressure difference is proposed to evaluate the degree of flow field vibration. Secondly, the mathematical models of pressure difference with plugging states and spoiler angles are established based on the extreme learning machine (ELM) optimized by improved sparrow search algorithm (ISSA). Finally, a modified Q-learning algorithm based on simulated annealing is applied to determine the optimal strategy for the spoiler angle and plugging velocity in real time. The results show that the proposed method can reduce the plugging-induced vibration by 19.9% and 32.7% on average, compared with single-regulating methods. This study can effectively ensure the stability of the plugging process.

文摘Neurons are highly polarized,morphologically asymmetric,and functionally compartmentalized cells that contain long axons extending from the cell body.For this reason,their maintenance relies on spatiotemporal regulation of organelle distribution between the somatodendritic and axonal domains.Although some organelles,such as mitochondria and smooth endoplasmic reticulum,are widely distributed throughout the neuron,others are segregated to either the somatodendritic or axonal compartment.For example,Golgi outposts and acidified lysosomes are predominantly present in the somatodendritic domain and rarely distributed along the axon,whereas newly formed autophagosomes and synaptic vesicles are mainly distributed in the distal axon(Britt et al.,2016).

文摘Regulated cell death is a form of cell death that is actively controlled by biomolecules.Several studies have shown that regulated cell death plays a key role after spinal cord injury.Pyroptosis and ferroptosis are newly discovered types of regulated cell deaths that have been shown to exacerbate inflammation and lead to cell death in damaged spinal cords.Autophagy,a complex form of cell death that is interconnected with various regulated cell death mechanisms,has garnered significant attention in the study of spinal cord injury.This injury triggers not only cell death but also cellular survival responses.Multiple signaling pathways play pivotal roles in influencing the processes of both deterioration and repair in spinal cord injury by regulating pyroptosis,ferroptosis,and autophagy.Therefore,this review aims to comprehensively examine the mechanisms underlying regulated cell deaths,the signaling pathways that modulate these mechanisms,and the potential therapeutic targets for spinal cord injury.Our analysis suggests that targeting the common regulatory signaling pathways of different regulated cell deaths could be a promising strategy to promote cell survival and enhance the repair of spinal cord injury.Moreover,a holistic approach that incorporates multiple regulated cell deaths and their regulatory pathways presents a promising multi-target therapeutic strategy for the management of spinal cord injury.

文摘Hippocampal neuronal loss causes cognitive dysfunction in Alzheimer’s disease.Adult hippocampal neurogenesis is reduced in patients with Alzheimer’s disease.Exercise stimulates adult hippocampal neurogenesis in rodents and improves memory and slows cognitive decline in patients with Alzheimer’s disease.However,the molecular pathways for exercise-induced adult hippocampal neurogenesis and improved cognition in Alzheimer’s disease are poorly understood.Recently,regulator of G protein signaling 6(RGS6)was identified as the mediator of voluntary running-induced adult hippocampal neurogenesis in mice.Here,we generated novel RGS6fl/fl;APP_(SWE) mice and used retroviral approaches to examine the impact of RGS6 deletion from dentate gyrus neuronal progenitor cells on voluntary running-induced adult hippocampal neurogenesis and cognition in an amyloid-based Alzheimer’s disease mouse model.We found that voluntary running in APP_(SWE) mice restored their hippocampal cognitive impairments to that of control mice.This cognitive rescue was abolished by RGS6 deletion in dentate gyrus neuronal progenitor cells,which also abolished running-mediated increases in adult hippocampal neurogenesis.Adult hippocampal neurogenesis was reduced in sedentary APP_(SWE) mice versus control mice,with basal adult hippocampal neurogenesis reduced by RGS6 deletion in dentate gyrus neural precursor cells.RGS6 was expressed in neurons within the dentate gyrus of patients with Alzheimer’s disease with significant loss of these RGS6-expressing neurons.Thus,RGS6 mediated voluntary running-induced rescue of impaired cognition and adult hippocampal neurogenesis in APP_(SWE) mice,identifying RGS6 in dentate gyrus neural precursor cells as a possible therapeutic target in Alzheimer’s disease.

文摘[Objective]The aim was to study the response mechanism of drought stress of wheat varieties in different drought-resistance species,and protect the effect of exogenous NO on oxidative damage and photosynthetic apparatus of wheat leaves under drought stress.[Method]Using low-resistance Yumai 949 and high-resistance Xiamai 5 as test materials,drought stress was carried out to seedlings in five-leaf stage with 15% PEG-6000,and then NO(0.75 mmol/L SNP,sodium nitroprusside,exogenous NO donor) was used for regulation in drought condition,and antioxidant and photosynthetic activities was determined.Three treatments were set in the experiment.[Result]SOD,CAT and APX activities of high resistance Xiamai 5 were much higher than low resistance Yumai 949,so were MDA and chlorophyll content.And the change range of these physiological indexes of high resistance species was smaller than high-yielding and low resistance species under drought stress.NO increased the adaptation to drought stress of these physiological indexes significantly.[Conclusion]Exogenous NO could increase the activity of antioxidant enzymes of wheat leaves under drought stress,and enhance the drought resistance of wheat.

文摘The effects of exogenous growth regulating substance on the fruit weight of peach were reviewed. Paclobutrazol could increase the single fruit weight in dif- ferent degree, and cytokinins could also increase the single fruit weight. GA3 and B9 had no effect on fruit size or on reducing single fruit weight. Chlorflurenol-methy, ABT and salicylic acid had no effect on fruit size. Suggestions on the future re- search were put forward.
文摘[Objective] The study aimed to reveal the effect of osmosis-regulating substances and organic appendices on somatic embryogenesis in wheat. [Method] The suitable concentration combination of appendices was optimized by adding different concentrations of osmosis-regulating substances including mannitol, sorbitol and organic appendices such as Gln, CH and LH, into the somatic embryogenesis in wheat. [Result] The mannitol or sorbitol lower than 40 g/L was helpful for improving somatic embryogenesis; there was no significant difference in the induction rate of somatic embryogenesis when 300-500 mg/L Gln、CH or LH was respectively added into the induced medium, while somatic embryogenesis could be enhanced dramatically in the presence of 500 mg/L Gln together with 300 mg/L CH. [Conclusion] Somatic embryogenesis could be improved to some extent by different concentrations of osmosis-regulating substances and organic appendices, which laid foundation for establishing a more perfect system of somatic embryogenesis in wheat.

文摘Through the study on the output signals of the electro-hydraulic regulating system in the thermal power plant, a novel method for online diagnosis of the plugging fault in the servo valve is presented. With the use of the AMESIM software, the changes of the piston displacement, the oil pressure, the magnitude attenuation and the phase lag of the system under different plugging states are studied after simulation. Besides, the influences of the symmetrical and unsymmetrical plugging on the system are also compared and the characteristic table is given. The duo-neural network is put forward to achieve an online diagnosis on the plugging fault of the servo valve. The first level of network helps to make the qualitative diagnosis of the plugging position while the second level is for the quantitative diagnosis of the degree of the plugged position. The research results show that plugging at different positions exerts different influences on the performance of the system. The unsymmetrical plugging mainly affects the regulation time while the symmetrical plugging leads to great changes in the magnitude attenuation and the phase lag.

文摘YAP(yes-associated protein) is a transcriptional factor that is negatively regulated by Hippo pathway, a conserved pathway for the development and size control of multiple organs. The exact function of YAP in bone homeostasis remains controversial. Here we provide evidence for YAP's function in promoting osteogenesis, suppressing adipogenesis, and thus maintaining bone homeostasis.YAP is selectively expressed in osteoblast(OB)-lineage cells. Conditionally knocking out Yap in the OB lineage in mice reduces cell proliferation and OB differentiation and increases adipocyte formation, resulting in a trabecular bone loss. Mechanistically, YAP interacts with β-catenin and is necessary for maintenance of nuclear β-catenin level and Wnt/β-catenin signaling. Expression of β-catenin in YAP-deficient BMSCs(bone marrow stromal cells) diminishes the osteogenesis deficit. These results thus identify YAP-β-catenin as an important pathway for osteogenesis during adult bone remodeling and uncover a mechanism underlying YAP regulation of bone homeostasis.

文摘Plant N starvation response is closely associated with the N signaling components that involve transduction of the low-N cues. In this study, we functionally characterized Ta ARR1, a cytokinin(CK) response regulator gene in Triticum aestivum, in mediating the N starvation adaptation in plants. Ta ARR1 harbors two conserved domains specified by plant ARR family members;subcellular localization analysis indicated its target onto nucleus after endoplasmic reticulum assortment. Ta ARR1 displayed modified expression upon the N starvation stressor, showing upregulated expression in roots and leaves over a 27-h N starvation treatment and whose induced transcripts were gradually recovered along with progression of the N recovery treatment. The tobacco lines overexpressing Ta ARR1 displayed improved low-N stress tolerance, displaying enlarged phenotype, increased biomass and N accumulation, and enhanced glutamine synthetase(GS) activities compared with wild type(WT) following the N starvation treatment. Expression analysis on genes encoding the nitrate transporter(NRT) and GS proteins in Nicotiana tabacum revealed that Nt NRT2.2 and Nt GS3 are upregulated in expression in the N-deprived transgenic lines, whose expression patterns were contrasted to other above family genes that were unaltered on transcripts between the transgenic lines and WT. Transgene analysis validated the function of Nt NRT2.2 and Nt GS3 in regulating N accumulation, GS activity, growth traits, and N use efficiency in plants. These results suggested the internal connection between the Ta ARR1-mediated N starvation tolerance and the modified transcription of distinct N acquisitionand assimilation-associated genes. Our investigation together indicates that Ta ARR1 is essential in plant N starvation adaptation due to the gene function in transcriptionally regulating distinct NRT and GS genes that affect plant N uptake and assimilation under the N starvation condition.

文摘In plants,the vesicle fusion process plays a vital role in pathogen defence.However,the importance of the vesicle fusion process in apple ring rot has not been studied.Here,we isolated and characterised the apple syntaxin gene MdSYP121.Silencing the MdSYP121 gene in transgenic apple calli increased tolerance to Botryosphaeria dothidea infection;this increased tolerance was correlated with salicylic acid(SA)synthesis-related and signalling-related gene transcription.In contrast,overexpressing MdSYP121 in apple calli resulted in the opposite phenotypes.In addition,the results of RNA sequencing(RNA-Seq)and quantitative real-time PCR(qRT-PCR)assays suggested that MdSYP121 plays an important role in responses to oxidation–reduction reactions.Silencing MdSYP121 in apple calli enhanced the expression levels of reactive oxygen species(ROS)-related genes and the activity of ROS-related enzymes.The enhanced defence response status in MdSYP121-RNAi lines suggests that syntaxins are involved in the defence response to B.dothidea.More importantly,we showed that MdSYP121 forms a soluble N-ethylmaleimide-sensitive factor attachment protein receptor(SNARE)complex with MdSNAP33,and the complex may participate in regulating resistance to B.dothidea.In conclusion,by regulating the interaction of SA pathway and oxidation–reduction process,MdSYP121 can influence the pathogen infection process in apple.

文摘Liver fibrosis is an important health problem that can further progress into cirrhosis or liver cancer,and result in significant morbidity and mortality. Inhibiting proliferation and inducing apoptosis of hepatic stellate cells(HSCs) may be the key point to reverse liver fibrosis. At present,anti-fibrosis drugs are rare. Kinetin is a type of plant-derived cytokinin which has been reported to control differentiation and induce apoptosis of human cells. In this study,the HSCs were incubated with different concentrations of kinetin. The proliferation of rat HSCs was measured by MTT assay,cell cycle and apoptosis were analyzed by flow cytometry,and the apoptosis was examined by TUNEL method. The expression of Bcl-2 and Bax proteins was detected by immunocytochemistry staining. It was found that kinetin could markedly inhibit proliferation of HSCs. In a concentration range of 2 to 8 μg/m L,the inhibitory effects of kinetin on proliferation of HSCs were increased with the increased concentration and the extension of time(P〈0.01). Flow cytometry indicated that kinetin could inhibit the DNA synthesis from G0/G1 to S phase in a dose-dependent manner(P〈0.01). The apoptosis rates of the HSCs treated with 8,4 and 2 μg/m L kinetin(25.62%±2.21%,15.31%±1.9% and 6.18%±1.23%,respectively) were increased significantly compared with the control group(3.81%±0.93%)(P〈0.01). All the DNA frequency histogram in kinetin-treated groups showed obvious hypodiploid peak(sub-G1 peak),and with the increase of kinetin concentrations,the apoptosis rate of HSCs also showed a trend of increase. It was also found that kinetin could down-regulate the expression of Bcl-2,and up-regulate the expression of Bax,leading to the decreased ratio of Bcl-2/Bax significantly. The kinetin-induced apoptosis of HSCs was positively correlated with the expression of Bax,and negatively with the expression of Bcl-2. It was concluded that kinetin can inhibit activation and proliferation of HSCs by interrupting the cell cycle at G1/S restriction point and inducing apoptosis of HSCs via reducing the ratio of Bcl-2/Bax.

文摘Background Anxiety symptoms are common in mental diseases and a variety of physical disorders, especially in disorders related to stress. More and more basic studies have indicated that gut microbiota can regulate brain function through the gut-brain axis, and dysbiosis of intestinal microbiota was related to anxiety. However, there is no specific evidence to support treatment of anxiety by regulating intestinal microbiota. Aims To find evidence sup porting improvement of anxiety symptoms by regulation of intestinal microbiota. Methods This systematic review of randomised controlled trials was searched based on the following databases: PubMed, EMBASE, the Cochrane Library, OVID, Web of Knowledge, China National Knowledge Infrastructure (CNKI), Wanfang Data, VIP databases and SinoMed. The retrieval time dated back to 25 July 2018. Then we screened research literatures based on established inclusion and exclusion criteria. Quality evaluation for each included study was done using the Cochrane risk of bias and the Jadad scale. Results A total of 3334 articles were retrieved and 21 studies were included which contained 1503 subjects. In the 21 studies, 15 chose probiotics as interventions to regulate intestinal microbiota and six chose non-probiotic ways such as adjusting daily diets. Probiotic supplements in seven studies contained only one kind of probiotic, two studies used a product that contained two kinds of probiotics and the supplements used in the other five studies included at least three kinds of probiotics. In the studies that used treatment as usual plus interventions regulating intestinal flora (IRIF) as interventions (five studies), only non-probiotic ways were effective (two studies), which means 40% of studies were effective;in the studies that used IRIF alone (16 studies, 11 studies used probiotic ways and 5 studies used non-probiotic ways), 56% of studies could improve anxiety symptoms, and 80% of studies that conducted the non-probiotic interventions were effective, while 45% of studies that used probiotic supplementations had positive effects on anxiety symptoms. Overall, 11 studies showed a positive effect on anxiety symptoms by regulating intestinal microbiota, which indicated 52% of the 21 studies were effective, and there were five studies that used probiotic supplements as interventions and six used non-probiotic interventions. In addition, it should be noted that six of seven studies showed that regulation of intestinal microbiota could treat anxiety symptoms, the rate of efficacy was 86%.Conclusions We find that more than half of the studies included showed it was positive to treat anxiety symptoms by regulation of intestinal microbiota. There are two kinds of interventions (probiotic and non-probiotic interventions) to regulate intestinal microbiota, and it should be highlighted that the non-probiotic interventions were more effective than the probiotic interventions. More studies are needed to clarify this conclusion since we still cannot run meta-analysis so far.

文摘Objective:An increasing number of studies indicate that autophagy plays an important role in the pathogenesis of spinal cord injury,and that regulating autophagy can enhance recovery from spinal cord injury.However,the effect of regulating autophagy and whether autophagy is detrimental or beneficial after spinal cord injury remain unclear.Therefore,in this study we evaluated the effects of autophagy regulation on spinal cord injury in rats by direct and indirect comparison,in an effort to provide a basis for further research.Data source:Relevant literature published from inception to February 1,2018 were included by searching Wanfang,CNKI,Web of Science,MEDLINE(OvidSP),PubMed and Google Scholar in English and Chinese.The keywords included"autophagy","spinal cord injury",and"rat".Data selection:The literature included in vivo experimental studies on autophagy regulation in the treatment of spinal cord injury(including intervention pre-and post-spinal cord injury).Meta-analyses were conducted at different time points to compare the therapeutic effects of promoting or inhibiting autophagy,and subgroup analyses were also conducted.Outcome measure:Basso,Beattie,and Bresnahan scores.Results:Of the 622 studies,33 studies of median quality were included in the analyses.Basso,Beattie,and Bresnahan scores were higher at 1 day(MD=1.80,95%CI:0.81-2.79,P=0.0004),3 days(MD=0.92,95%CI:0.72-1.13,P<0.00001),1 week(MD=2.39,95%CI:1.85-2.92,P<0.00001),2 weeks(MD=3.26,95%CI:2.40-4.13,P<0.00001),3 weeks(MD=3.13,95%CI:2.51-3.75,P<0.00001)and 4 weeks(MD=3.18,95%CI:2.43-3.92,P<0.00001)after spinal cord injury with upregulation of autophagy compared with the control group(drug solvent control,such as saline group).Basso,Beattie,and Bresnahan scores were higher at 1 day(MD=6.48,95%CI:5.83-7.13,P<0.00001),2 weeks(MD=2.43,95%CI:0.79-4.07,P=0.004),3 weeks(MD=2.96,95%CI:0.09-5.84,P=0.04)and 4 weeks(MD=4.41,95%CI:1.08-7.75,P=0.01)after spinal cord injury with downregulation of autophagy compared with the control group.Indirect comparison of upregulation and downregulation of autophagy showed no differences in Basso,Beattie,and Bresnahan scores at 1 day(MD=-4.68,95%CI:-5.840 to-3.496,P=0.94644),3 days(MD=-0.28,95%CI:-2.231-1.671,P=0.99448),1 week(MD=1.83,95%CI:0.0076-3.584,P=0.94588),2 weeks(MD=0.81,95%CI:-0.850-2.470,P=0.93055),3 weeks(MD=0.17,95%Cl:-2.771-3.111,P=0.99546)or 4 weeks(MD=-1.23,95%Cl:-4.647-2.187,P=0.98264)compared with the control group.Conclusion:Regulation of autophagy improves neurological function,whether it is upregulated or downregulated.There was no difference between upregulation and downregulation of autophagy in the treatment of spinal cord injury.The variability in results among the studies may be associated with differences in research methods,the lack of clearly defined autophagy characteristics after spinal cord injury,and the limited autophagy monitoring techniques.Thus,methods should be standardized,and the dynamic regulation of autophagy should be examined in future studies.

文摘One adult α-globin gene and one β-globin gene have been cloned from the large yellow croaker Pseudosciaena crocea. Linkage analysis indicated that the α- and β-globin genes were oriented head-to-head relative to each other. To identify the regulatory elements present in the intergenic and intragenic regions of the globin complex, the intergenic region alone or together with the β-globin gene first intron was cloned into the luciferase-reporter vector pGL3-Basic respectively, and the chimeric constructs were tran- siently transfected into Vero cells and primary fish erythrocytes. The intergenic region cannot support the high-level expression of luciferase. However, the promoter activity of the intergenic region was strongly stimulated by the positive regulatory elements (PRE) located in the β-globin gene intron 1. Thus, it is proposed that the intergenic promoters and intragenic PRE were necessary for the effective expression of the linked α- and β-globin genes.
