Non-monophyly of Rhacophorus rhodopus,Theloderma and Philautus albopunctatus Inferred from Mitochondrial 16S rRNA Gene Sequences 被引量:6

Mitochondrial gene fragments of 16S rRNA gene of four species (Rhacophorus rhodopus, R. reinwardtii, Philautus albopunctatus and P. rhododiscus ) from 11 populations were sequenced in this study. Homologous sequences of R. bipunctatus, Theloderma asperum, T. corticale and Buergeria japonica were obtained by screening the GenBank database. After excluding all gaps and ambiguous positions, aligned sequences were 500 bp in length with 115 variable sites and 92 parsimony-informative sites. Using B .japonica as an outgroup, phylogenetic relationships were analyzed using Bayesian inference, maximum parsimony and maximum likelihood methods. Our results indicated that neither of R. rhodopus and P. albopunctatus were monophyletic at the species level. The population of R. rhodopus from Hainan Island was more close to R. bipunctatus than to populations of R. rhodopus from Yunnan Province. Furthermore, the populations of R. rhodopus from Yunnan Province can be divided into two main lineages. Theloderma corticale and P.rhododiscus were clustered together and T. asperum was nested in P.albopunctatus. We considered that P. albopunctatus Liu and Hu, 1962, was the synonymy of T. asperum Boulenger, 1886, and suggested removing P. rhododiscus from Philautus into the genus Theloderma. 展开更多

A new species of rhacophorid of the genus Rhacophorus is described from the Dabie Mountains of west Anhui, east China. The new species, Rhacophorus zhoukaiyae sp. nov. is distinguished from its congeners by a combination of the following characters: 1) the ventral surface and front-and-rear of the femur is paler yellowish and decorated with irregular grayish blotching, and without obvious spots on the dorsum of the hand and foot webbing; 2) the outer metatarsal tubercle is small; 3) outer fingers are half-webbed and outer toes two third webbed; 4) the skin on the dorsum is smooth and without compressed warts; 5) the throat, chest and belly are pure paler yellowish; 6) the dorsal part of the fingers and toes are grayish-white; 7) the iris is golden-yellow. In addition, the phylogenetic tree showed that all the individuals of R. zhoukaiyae sp. nov. clustered into one distinct clade which suggested the validity of this species. This results could also be used to the support of species delimitation. Currently, this species is known only from mid-elevation montane evergreen forest in the Dabie Mountains of west Anhui, China. 展开更多

In anurans, the outcomes of male-male contests and female choice are often associated with body size. In some species, males evaluate an opponent’s fighting ability and females evaluate male quality on the basis of male communication signals, which are thought to reflect information concerning male body size. Nevertheless, previous studies reveal that male call structure is not always correlated with body size. In the present study we investigated the relationships between body size and call structure in the large treefrog, Rhacophorus dennysi, as well as the relationship of its calling behavior with air temperature and humidity. The results show that both the dominant and fundamental frequencies are negatively correlated with body size, while inter-note intervals are positively correlated with body size, indicating that call characters could reflect body size in this species.Additionally, calling in this tropical species exhibits a circadian rhythm insofar as relatively high temperature and low humidity during the day is associated with less vocal behavior. Thus, individual variations in call structure are mainly dependent on body size while the temporal rhythm of calling activity is affected by environmental conditions in large treefrogs. 展开更多

在脊椎动物中睾丸的大小不对称是常见现象,这一现象通常可以用补偿假说来解释。该假说认为两个睾丸中较小的一个的增大是为了补偿较大一个睾丸在功能上的不足,并且方向不对称的程度与雄性质量成正相关关系。对重庆市缙云山自然保护区的... 在脊椎动物中睾丸的大小不对称是常见现象,这一现象通常可以用补偿假说来解释。该假说认为两个睾丸中较小的一个的增大是为了补偿较大一个睾丸在功能上的不足,并且方向不对称的程度与雄性质量成正相关关系。对重庆市缙云山自然保护区的斑腿泛树蛙(Rhacophorus megacephalus)雄性的年龄和睾丸不对称性进行了研究。结果发现该种群的雄性成体平均年龄为2.91±0.83岁,性成熟年龄为2岁,最大寿命为5岁,并且体长和体重都随年龄的增加而增加。该种群雄性的左右睾丸大小没有差别,左睾丸减右睾丸的差值服从正态分布,表明该物种的睾丸大小不对称性属于波动不对称;左右睾丸重与相对睾丸不对称程度都没有相关性,说明左右睾丸均不充当补偿角色;绝对睾丸不对称程度同个体体长、年龄、身体状况以及精子密度都没有相关性;相对睾丸不对称程度与个体体长、年龄和身体状况也没有相关性,与精子密度显著负相关。本研究发现在这个斑腿树蛙自然种群中没有关于补偿假说的证据。此外,还发现拥有相对较大的右睾丸的个体相对于拥有较大左睾丸的个体,其精子密度更大。 展开更多

We describe a new species of Rhacophorus based on two adult specimens collected from Hoa Binh Province, northern Vietnam. Rhacophorus hoabinhensis sp. nov. is distinguishable from its congeners on the basis of a combination of the following morphological characters: size small (SVL 31.1-32.5 mm in males); head slightly longer than wide; vomerine teeth absent; snout short (SNL/SVL 0.16); dorsal skin smooth; forearm and tarsus with dermal fringes; dermal appendage at vent present; webbing formula on fingers I12/3-12/3II1-2III1-1IV and on toes I3/4-1II1/2- IIII1/2-1IVI-1/2V; dorsal surface grey yellow with brown spots; lower jaw region dark grey, throat, chest and belly cream; anterior and posterior thighs, as well as ventral surface of tibia orange. The interspecific uncorrected genetic distances (16S rRNA gene) between the new species from Hoa Binh and other analyzed congeners varied from 9.8% to 17.4%. In the phylogenetic analyses, the new species revealed to be a representative of Rhacophorus and was nested within the R. hoanglienensis-orlovi species group. 展开更多

We conducted molecular phylogenetic analyses of Japanese Rhacophorus species, especially of R. schlegelii and R. arboreus from the mainland, based on samples encompassing their known distribution ranges, and discussed about evolutionary history of Rhacophorus species within Japan. The common ancestor of Japanese Rhacophorus, except for R. owstoni from southern Ryukyus, was estimated to have diverged from a lineage occurring mainly in China about 7 MYBP. Both R. schlegelii and R. arboreus are genetically largely divergent between regions of eastern and western Japan, and this seems to have been promoted mainly by retreat to refugia. Retreats of the two species to different refugia sometimes in the past seem to have led restricted distribution of R. schlegelii in eastern and R. arboreus in western Japan, and brought their intraspecific variation patterns in morphology and breeding habit. 展开更多

The worldwide declines in amphibian populations have largely been caused by infectious fungi and bacteria. Given that vertebrate immunity against these extracellular pathogens is primarily functioned by the major histocompatibility complex(MHC) class Ⅱ molecules, the characterization and the evolution of amphibian MHC class Ⅱ genes have attracted increasing attention. The polymorphism of MHC class Ⅱ genes was found to be correlated with susceptibility to fungal pathogens in many amphibian species, suggesting the importance of studies on MHC class Ⅱ genes for amphibians. However, such studies on MHC class Ⅱ gene evolution have rarely been conducted on amphibians in China. In this study, we chose Omei treefrog(Rhacophorus omeimontis), which lived moist environments easy for breeding bacteria, to study the polymorphism of its MHC class Ⅱ genes and the underlying evolutionary mechanisms. We amplified the entire MHC class ⅡB exon 2 sequence in the R. omeimontis using newly designed primers. We detected 102 putative alleles in 146 individuals. The number of alleles per individual ranged from one to seven, indicating that there are at least four loci containing MHC class ⅡB genes in R. omeimontis. The allelic polymorphism estimated from the 102 alleles in R. omeimontis was not high compared to that estimated in other anuran species. No significant gene recombination was detected in the 102 MHC class ⅡB exon 2 sequences. In contrast, both gene duplication and balancing selection greatly contributed to the variability in MHC class ⅡB exon 2 sequences of R. omeimontis. This study lays the groundwork for the future researches to comprehensively analyze the evolution of amphibian MHC genes and to assess the role of MHC gene polymorphisms in resistance against extracellular pathogens for amphibians in China. 展开更多

The Effect of Attendance Patterns on the Lek Mating System in the Omei Treefrog(Rhacophorus omeimontis)

Attendance patterns are a key factor affecting sexual selection and mating systems in lekbreeding species.However,there are few systematic studies on attendance patterns in lek-breeding anurans.In this study,we monitored a population of Omei treefrogs(Rhacophorus omeimontis) for four consecutive years to reveal in detail the effects of attendance patterns on mating systems.We found that(1) female nightly attendance was affected by male nightly attendance,and operational sex ratios(determined by male and female nightly attendance) ultimately determined the extent of female polyandry;(2) males surviving more than one breeding season maintained high chorus attendance and mating success,suggesting that male chorus attendance is a good predictor of male genetic quality;(3) the distributions of male mating success were not significantly different from a Poisson distribution,indicating a random mating pattern;and male mating success was not related to body size(size-assortative mating was not evident) but was significantly correlated with chorus attendance.Since the chorus attendance is a predictor of male genetic quality,females performing random mating were more likely to mate with more frequently present males and could therefore gain indirect benefits from mating with higher-than-average quality males,thereby producing good offspring without incurring the significant costs involved in prolonged searching for,and assessing males.Our systematic survey on the relationship between attendance patterns and the lek mating system in R.omeimontis could help us better understand in-depth anuran lek mating systems. 展开更多

敌敌畏和丁草胺是我国农田中使用最普遍的杀虫剂和除草剂,这些农田化学物质对当地的水生动物及种群造成很大威胁。本文以广泛分布于我国南方农田区域的斑腿树蛙蝌蚪为研究对象,用碱性单细胞电泳方法(或慧星试验)对暴露在不同浓度的敌敌... 敌敌畏和丁草胺是我国农田中使用最普遍的杀虫剂和除草剂,这些农田化学物质对当地的水生动物及种群造成很大威胁。本文以广泛分布于我国南方农田区域的斑腿树蛙蝌蚪为研究对象,用碱性单细胞电泳方法(或慧星试验)对暴露在不同浓度的敌敌畏(2·08 , 4·16 , 6·24 , 8·32 , 10·40 mg/L)和丁草胺(0·1025 ,0·205 , 0·410 , 0·820 , 1·230 mg/L)溶液中的蝌蚪进行了遗传毒性的检测。结果表明:在实验室条件下,随着农药浓度的增加,蝌蚪的DNA损伤(DNA尾长与尾宽比)随之增加;敌敌畏浓度为2·08 mg/L和丁草胺浓度为0·41 mg/L时,对蝌蚪造成显著的损伤,而且农药的剂量与蝌蚪的DNA损伤(DNA尾长与尾宽比)呈显著的线性关系:敌敌畏,y=1·136±0·0083x, r=0·957 ,P<0·01 ;丁草胺,y=0·968±0·0093x, r=0·964 ,P<0·01。本研究表明这两种农药对我国的两栖动物具有遗传毒性作用;同时也表明,在检测环境污染物对蝌蚪的基因毒性方面,碱性单细胞电泳分析是一种合适的方法[动物学报51 (3) : 447 -454 , 2005]。 展开更多

本底资源是保护区建设与发展的基础,也是生物多样性深入研究与保护重要的参考资料。湖南小溪国家级自然保护区是我国中南地区具有代表性的原始次生林区,生物资源相对丰富。自2014年以来,对区内的两栖爬行动物资源、多样性及区系进行了... 本底资源是保护区建设与发展的基础,也是生物多样性深入研究与保护重要的参考资料。湖南小溪国家级自然保护区是我国中南地区具有代表性的原始次生林区,生物资源相对丰富。自2014年以来,对区内的两栖爬行动物资源、多样性及区系进行了较为系统的调查与研究。结果表明:区内现有两栖爬行动物2目16科40属63种,其中两栖动物1目7科15属33种,爬行动物1目9科25属30种;无声囊泛树蛙Polypedates mutus和老山树蛙Rhacophorus laoshan是湖南省两栖动物新记录;核心区的物种多样性指数和均匀度指数均高于缓冲区和实验区;动物区系成分缺乏古北界种类,以东洋界种类为主(48种,占76.2%),广布种有15种(23.8%);平均动物区系相似性比较发现,其与湖南省内其他6个保护区(森林公园)属于周缘关系。鉴于保护区两栖爬行动物资源的丰富性、典型性与代表性,应加强研究和保护。 展开更多

通过对黑点树蛙、双团棘胸蛙的染色体核型和Ag-NORs的研究,其结果黑点树蛙2n=26=22m+4sm(5+8),NF=52,No.8、10染色体臂上各有一次缢痕,Ag-NORs位于No.10短臂;双团棘胸蛙2n=64,NF=64,全为端部着丝粒染色体,有一次缢痕,位于No.10,且异型。

敌敌畏是我国农田中使用最普遍的有机磷杀虫剂,它通过干扰对神经传导起重要作用的胆碱酯酶的活性,起到接触和摄入毒杀昆虫的作用,也对当地水生动物及种群造成很大威胁。本文以广泛分布于我国南方农田区域的斑腿树蛙蝌蚪为研究对象,用碱... 敌敌畏是我国农田中使用最普遍的有机磷杀虫剂,它通过干扰对神经传导起重要作用的胆碱酯酶的活性,起到接触和摄入毒杀昆虫的作用,也对当地水生动物及种群造成很大威胁。本文以广泛分布于我国南方农田区域的斑腿树蛙蝌蚪为研究对象,用碱性单细胞电泳方法比较了暴露于不同浓度(2.08,4.16,6.24,8.32,10.40mg/L)的敌敌畏溶液中三个发育阶段的遗传毒性的差异。结果表明:敌敌畏溶液暴露24h,对早期和中期阶段的蝌蚪造成极显著的DNA损伤(P<0.01),甚至在敌敌畏浓度低达2.08mg/L的溶液中也产生显著的损伤(P<0.05);统计分析表明,DNA长宽比率与敌敌畏剂量之间呈显著的线性关系,早期阶段和中期阶段的相关系数分别为0.950(P<0.01)和0.954(P<0.01)。对于后期阶段的蝌蚪,所有浓度的敌敌畏溶液都未导致明显的DNA损伤,作者认为进入变态阶段的蝌蚪对敌敌畏的敏感性明显下降。结果同时也表明,早期和中期阶段的斑腿树蛙蝌蝌是一种监测环境遗传毒性的合适指示生物。 展开更多

[目的]研究无尾两栖类的消化道组织。[方法]采用解剖学与组织学方法研究斑腿树蛙的消化道。[结果]结果表明,斑腿树蛙消化道分为口咽腔、食道、胃、十二指肠、回肠和大肠。消化道管壁由黏膜层、黏膜下层、肌层和浆膜层构成。食道为复层柱状纤毛上皮,胃和肠都为单层柱状上皮。胃和十二指肠的黏膜皱褶最丰富。食道腺为复泡状腺,胃腺属于单管状腺。具有十二指肠腺,回肠和大肠无多细胞腺体。杯状细胞存在于食道、十二指肠、回肠和大肠中,但食道、十二指肠中较少,回肠和大肠中较为丰富。[结论]斑腿树蛙肌层均为平滑肌,内层环肌较厚,外侧纵肌较薄。 展开更多
