Rice-crab coculture (RC) is an important eco-agricultural process in rice production in northern China. Recognizing the soil fertility in RC may help develop novel sustainable agriculture. Soil carbohydrates are important factors in determining soil fertility in different culture modes. In this study, soil carbohydrates were analyzed under three different culture modes including rice monoculture (RM), conventional rice-crab coculture (CRC) and organic rice-crab coculture (ORC). Results showed that the contents of soil organic carbon and carbohydrates were significantly higher in the ORC than those in RM. The increasing effect was greater with increased organic manure. Similar tendency was found in CRC, but the overall effect was less pronounced compared with ORC. Carbohydrates were more Sensitive to RC mode and manure amendment than soil organic carbon. Compare to RM, the (Gal+Man)/(Ara+Xyl) ratio decreased in all the RC modes, indicating a relative enrichment in plant-derived carbohydrates due to the input of crab feed and manure. While the increasing (Gal+Man)/(Ara+Xyl) ratio in ORC modes with increased organic manure suggested that crab activity and metabolism induced microbially derived carbohydrates accumulation. The lower GluN/MurA ratio in ORC indicated an enhancement of bacteria contribution to SOM turnover in a short term. The findings reveal that the ORC mode could improve the quantity and composition of soil carbohydrates, effectively, to ensure a sustainable use of paddy soil. 展开更多

The objective of this study was to establish an efficient system of producing early monozygotic twin bovine embryos in vitro using the blastomere separation and coculture technique. In this study, early eight-cell embryos were chosen to optimize the separation method, and multi-coculture tactics were applied to improve the efficiency of this production system. Bovine embryo blastomeres(groups of at least 30 at the eight-cell stage) were separated into eight segments(to regard an eight-cell embryo as a tangerine, a blastomere as one segment) and one, two and four segments(blastomeres) were cultured respectively in microwells on the bottom of the four-well dish(Nunc, Denmark) with 400 μL of culture medium under paraffin oil. Four different types of coculture tactics(cocultured with nothing, intact embryos, bovine cumulus cells(b CCs), intact embryos & b CCs) were applied to the group of four segments(blastomeres). Finally, diameter and inner cell mass(ICM):trophectoderm(TE) cell ratio was measured as a criterion to assess the quality of the twin embryos which were derived from bovine separated blastomeres. Our results showed that rate of blastocyst formation of the four segments group was significantly greater than one or two group(P〈0.05). In addition, rate of blastocyst formation was significantly increased when the four segments were cocultured with intact embryo & b CCs(P〈0.05). Although the ICM, TE and total cells of blastocysts derived from separated blastomeres was less than the control group from intact embryo(P〈0.05), more important quality indicator of the blastocyst diameter and ICM:TE cell ratio was similar between our experimental group and the control group(P〉0.05). Thus, these results suggest that combined with intact embryos & b CCs coculture system, culturing four isolated segments(blastomeres) per microwell is an efficient system of producing early monozygotic twin bovine embryos. Furthermore, our results also indicate that the quality of blastocysts derived from separated blastomere may be similar to those derived from intact eight-cell embryos. 展开更多

为了探究拟穴青蟹(Scylla paramamosain,简称青蟹)在耐盐碱水稻-拟穴青蟹综合种养系统中的食物来源,研究在耐盐碱水稻田内投放密度为0.6 ind./m2的拟穴青蟹,经过为期2个月的养殖实验后通过碳、氮稳定同位素技术分析青蟹的食物来源。研... 为了探究拟穴青蟹(Scylla paramamosain,简称青蟹)在耐盐碱水稻-拟穴青蟹综合种养系统中的食物来源,研究在耐盐碱水稻田内投放密度为0.6 ind./m2的拟穴青蟹,经过为期2个月的养殖实验后通过碳、氮稳定同位素技术分析青蟹的食物来源。研究结果显示:实验期间青蟹特定生长率为(2.71±0.27)%/d,存活率为(14.26±4.85)%,青蟹收获产量为(116.30±39.59)kg/ha,水稻产量为3332.25 kg/ha。拟穴青蟹的δ^(13)C为(-21.55±0.90)‰,δ^(15)N为(9.96±0.40)‰,可能食物源的δ^(13)C为(-30.51±0.25)‰—(-13.45±0.15)‰。其中,水花生的δ^(13)C值最低(-30.51±0.25)‰,其次是水稻(-28.73±0.73)‰,而稗草的δ^(13)C值最高(-13.45±0.15)‰。可能食物源的δ^(15)N为(5.97±0.13)‰—(11.38±0.11)‰,其中配合饲料的δ^(15)N值最低(5.97±0.13)‰,飞虫的δ^(15)N值最高(11.38±0.11)‰。在食物贡献方面,配合饲料对青蟹食物贡献最高,达(61.2±3.8)%,稗草和千金子次之,分别为(8.3±5.2)%和(8.8±6.1)%,飞虫的食物贡献率占比最低,为(0.9±1.4)%。研究表明青蟹在耐盐碱水稻田内生长状况良好,配合饲料可成为稻田内青蟹的主要食物来源,同时植物性食物源对青蟹食物有重要贡献。研究可为耐盐碱水稻-青蟹综合种养系统管理和饲料开发提供理论和技术支撑。 展开更多

目的探讨左归丸含药血清对成骨-破骨细胞共培养体系中Hedgehog-GLi通路关键因子mRNA表达及骨吸收功能的影响。方法建立成骨细胞、破骨细胞、骨磨片共培养体系,实验分为7组:空白对照组、假手术组、OVX组、阳性对照组、左归丸组、激动剂... 目的探讨左归丸含药血清对成骨-破骨细胞共培养体系中Hedgehog-GLi通路关键因子mRNA表达及骨吸收功能的影响。方法建立成骨细胞、破骨细胞、骨磨片共培养体系,实验分为7组:空白对照组、假手术组、OVX组、阳性对照组、左归丸组、激动剂组和激动剂+左归丸组。选用50只雌性SD大鼠,分别制备相对应组含药血清,实验各组分别加入相对应组大鼠血清。通过实时荧光定量PCR检测成骨细胞Ihh、Ptch、Smo、GLi1、OPG及RANKL的mRNA表达;并采用甲苯胺蓝染色方法检测破骨细胞骨吸收功能。结果与假手术组相比,OVX组Ihh、Ptch、Smo、GLi1mRNA表达显著上调,同时OPGmRNA表达水平显著降低,RANKL mRNA表达水平显著增加,RANKL/OPG比值明显增高。与OVX组相比,阳性对照组与左归丸组Ihh、Ptch、Smo、GLi1mRNA表达明显下调,同时OPG mRNA表达水平显著升高,RANKL mRNA表达水平显著降低,RANKL/OPG比值明显下降。OVX组加入激动剂Shh重组蛋白后,Hedgehog-GLi信号通路关键因子的mRNA表达较OVX组明显增加。与激动剂组比较,激动剂+左归丸组Ihh、Ptch、Smo、GLi1mRNA表达以及RANKL/OPG比值均明显下降。与假手术组相比,OVX组的骨吸收陷窝面积显著增加;与OVX组相比,阳性对照组和左归丸组的骨吸收陷窝面积显著下降。结论左归丸含药血清可一定程度上抑制Hedgehog-GLi通路关键因子Ihh、Ptch、Smo、GLi1的活性,进而抑制OVX所致的骨吸收增强。 展开更多

贵州地质高背景区稻鱼共生系统汞的风险评估

为了解高地质背景区稻鱼共生系统中汞的潜在环境和健康风险,本研究选取贵州省喀斯特地质高背景区黔东南丹寨县、麻江县和非喀斯特地区从江县、榕江县和锦屏县为研究对象,采集稻鱼共生系统中稻田土壤、稻米和稻花鱼样品,测定总汞(THg)和... 为了解高地质背景区稻鱼共生系统中汞的潜在环境和健康风险,本研究选取贵州省喀斯特地质高背景区黔东南丹寨县、麻江县和非喀斯特地区从江县、榕江县和锦屏县为研究对象,采集稻鱼共生系统中稻田土壤、稻米和稻花鱼样品,测定总汞(THg)和甲基汞(MeHg)含量。采用地累积指数法和潜在生态风险指数法评估土壤环境风险;通过稻米和稻花鱼MeHg含量及不同年龄段居民膳食摄入情况,评估了居民摄入稻米和稻花鱼的汞暴露风险。结果表明:喀斯特地区土壤THg和MeHg含量均高于非喀斯特地区,但其甲基化率却弱于后者;喀斯特区域分别有28%和37%的稻田土壤样品THg含量超过我国水稻生产土壤汞阈值标准(0.65 mg·kg^(-1))和农用地水田土壤污染风险管控标准(0.50 mg·kg^(-1)),而非喀斯特区域土壤均未超标。土壤生态风险评估结果显示:喀斯特地区土壤的汞污染水平和生态风险更高。喀斯特地区稻米THg和稻花鱼MeHg含量分别为(5.15±4.19)ng·g^(-1)和(32.80±23.05)ng·g^(-1),高于非喀斯特区域分别为(3.43±0.87)ng·g^(-1)和(11.77±7.08)ng·g^(-1),但稻米汞的富集系数更低,鱼米汞含量均低于我国食品安全标准限值规定的THg 0.02 mg·kg^(-1)和MeHg 0.50 mg·kg^(-1)。居民膳食风险评估表明:“稻米+稻花鱼”食用模式的居民风险值HQ较高,且随居民年龄增加而降低,女性高于男性,但总体HQ<1,对居民没有汞暴露风险。以上结果表明,地质高背景区的稻鱼共生系统整体呈现低风险水平。 展开更多

利用从野樱桃发酵底泥中筛选的一株葡萄汁有孢汉逊酵母(Hanseniaspora uvarum)YT-35与商业酿酒酵母(Sacharomyces cerevisiae)共培养发酵野樱桃饮品,以2株菌的单菌株发酵为对照,对不同发酵阶段中的微生物数量、还原糖、乙醇等动态变化分... 利用从野樱桃发酵底泥中筛选的一株葡萄汁有孢汉逊酵母(Hanseniaspora uvarum)YT-35与商业酿酒酵母(Sacharomyces cerevisiae)共培养发酵野樱桃饮品,以2株菌的单菌株发酵为对照,对不同发酵阶段中的微生物数量、还原糖、乙醇等动态变化分析,利用高效液相色谱(HPLC)对有机酸进行动态检测,采用顶空固相微萃取/气相色谱-质谱(HS-SPME/GC-MS)对发酵饮品的挥发性风味物质进行测定。结果表明,H.uvarum YT-35在共培养发酵前期占据主导优势;与单一S.cerevisiae发酵相比,共培养发酵可以获得低乙醇含量(3.51 g/L)饮品;HPLC结果表明,共培养发酵可降低柠檬酸、苹果酸和奎宁酸的产量,增加冰醋酸的产量;HS-SPME/GC-MS结果表明,共培养发酵可产生更多的酯类风味物质,如己酸乙酯、苯甲酸甲酯、辛酸异戊酯、肉桂酸甲酯、反式肉桂酸乙酯、十八酸乙酯、苯甲酸苄酯和邻苯二甲酸二异丁酯,同时可增强如月桂酸乙酯、辛酸乙酯、苯乙醇、苯甲醇、辛酸和月桂酸等风味物质的含量;聚类分析表明,H.uvarum YT-35与S.cerevisiae共培养发酵野樱桃对丰富饮品中挥发性物质种类及含量提高有重要贡献。本研究旨在为具有不同代谢潜能的混合菌株在提升果汁发酵饮品品质中的研究提供一定科学依据和理论指导。 展开更多

长期秸秆还田对稻虾共作系统稻季甲烷排放的影响

稻田甲烷排放是农业甲烷排放的主要来源。稻虾共作是稻田生态种养应用面积最大的一种模式,秸秆还田是稻虾共作模式一种重要农事操作。为研究长期稻虾共作和秸秆还田对稻虾共作系统稻季甲烷排放影响及其机理,本试验于2015年设稻虾共作秸... 稻田甲烷排放是农业甲烷排放的主要来源。稻虾共作是稻田生态种养应用面积最大的一种模式,秸秆还田是稻虾共作模式一种重要农事操作。为研究长期稻虾共作和秸秆还田对稻虾共作系统稻季甲烷排放影响及其机理,本试验于2015年设稻虾共作秸秆还田(RC-S)、稻虾共作秸秆不还田(RC-NS)、水稻单作秸秆还田(RM-S)、水稻单作秸秆不还田(RM-NS)共4种处理,对7年后稻田稻季甲烷排放、土壤碳库指标及功能微生物(mcrA和pmoA)进行测定与分析。研究结果表明:稻虾共作模式显著降低甲烷排放,比水稻单作稻季甲烷累计排放降低1.42%~26.44%;而秸秆还田稻季甲烷排放显著增加15.18%~54.36%。4个处理甲烷累计排放量依次为RM-S>RC-S>RM-NS>RC-NS。通径分析结果显示:总有机碳(TOC)、微生物量碳(MBC)、可溶性有机碳(DOC)、产甲烷菌(mcrA)对甲烷排放的直接作用是促进的,而易氧化有机碳(ROC)、甲烷氧化菌(pmoA)对甲烷排放的直接作用是抑制的。研究结果表明,稻虾共作甲烷减排机理在于土壤ROC含量增加和产甲烷菌功能基因mcrA丰度的下降,秸秆还田甲烷排放增加主要是因为土壤ROC含量的降低以及土壤DOC含量和产甲烷菌mcrA基因丰度的增加。可见,稻虾共作缓解了秸秆还田所引起的甲烷增排效应。 展开更多

为探究稻蟹共作对土壤理化性质及土壤酶活性的影响,采用水泥池模拟稻田环境的方法,对比研究了稻蟹共作与水稻单作模式7个土壤理化指标和4个土壤酶活性的差异。结果表明:与稻田单作相比,稻蟹共作对土壤容重、土壤含水率和土壤还原性物质... 为探究稻蟹共作对土壤理化性质及土壤酶活性的影响,采用水泥池模拟稻田环境的方法,对比研究了稻蟹共作与水稻单作模式7个土壤理化指标和4个土壤酶活性的差异。结果表明:与稻田单作相比,稻蟹共作对土壤容重、土壤含水率和土壤还原性物质总量均有显著影响(P<0.05);稻蟹组的总有机碳略高于稻田组,但总氮略低于稻田组;稻蟹组与稻田组的Eh存在极显著差异,两组之间差异达20.95%;稻蟹组和稻田组土壤粒径机械组成均为粉粒>粘粒>粗细砂粒,粉粒占比均超过80%,且两组之间存在显著差异(P<0.05);随着土壤深度的增加,稻田组的四种酶活性均呈显著下降趋势;稻蟹组的脲酶、纤维素酶和蔗糖酶活性变化与稻田组一致,呈显著下降趋势,但其酸性磷酸酶没有变化。可见,稻蟹共作系统由于河蟹的引入,降低了土壤容重,提高了土壤含水率,改良了土壤,其水稻根系分布较多的10~15 cm底层土壤显示了较强的土壤酶活性,促进了水稻对稻蟹共作系统中碳、氮等元素的高效利用。因此,稻蟹共作系统既有利于水稻生长,又有利于河蟹养殖,可以作为一种生态种养模式在江苏地区推广应用。 展开更多

共培养条件下肌卫星细胞C2C12对软骨细胞ATDC5活性的影响

目的:观察共培养条件下肌卫星细胞C2C12对软骨细胞ATDC5活性的影响。方法:①将肌卫星细胞C2C12分为对照组和地塞米松组,对照组常规培养,地塞米松组采用地塞米松干预,通过CCK8法和BCA法测定地塞米松对C2C12细胞增殖和蛋白合成的影响,以... 目的:观察共培养条件下肌卫星细胞C2C12对软骨细胞ATDC5活性的影响。方法:①将肌卫星细胞C2C12分为对照组和地塞米松组,对照组常规培养,地塞米松组采用地塞米松干预,通过CCK8法和BCA法测定地塞米松对C2C12细胞增殖和蛋白合成的影响,以划痕实验和Transwell小室观察地塞米松对C2C12细胞修复和迁移能力的影响。②采用Transwell小室将正常肌卫星细胞C2C12(共培养组)和经地塞米松预处理的肌卫星细胞C2C12(预处理组)分别与软骨细胞ATDC5共培养,对照组Transwell下层小室内接种ATDC5细胞、上层小室内为无细胞的常规培养基,采用CCK8法和二氯二氢荧光素-乙酰乙酸酯荧光探针检测ATDC5细胞增殖率和细胞内活性氧含量。结果:①C2C12细胞增殖率和蛋白含量测定结果。地塞米松组C2C12细胞增殖率和蛋白含量均低于对照组[(78.402±5.401)%,(100.000±3.096)%,t=8.498,P=0.000;(5080.367±296.657)μg·mL-1,(5775.577±150.476)μg·mL-1,t=3.620,P=0.022]。②C2C12细胞伤口修复率测定结果。地塞米松组C2C12细胞伤口修复率低于对照组[(53.173±1.800)%,(79.979±10.176)%,t=4.493,P=0.011]。③C2C12细胞迁移数量测定结果。培养24 h和48 h时,地塞米松组C2C12细胞迁移数量均少于对照组[24 h:(24.200±5.630)个,(57.000±2.449)个,t=11.945,P=0.000;48 h:(57.600±8.820)个,(91.000±4.743)个,t=7.457,P=0.000]。④ATDC5细胞增殖率测定结果。3组ATDC5细胞增殖率比较,差异有统计学意义[(100.000±1.663)%,(116.894±7.917)%,(89.130±2.980)%,F=23.700,P=0.001]。对照组和共培养组ATDC5细胞增殖率均高于预处理组(P=0.037,P=0.006),共培养组ATDC5细胞增殖率高于对照组(P=0.001)。⑤ATDC5细胞内活性氧含量测定结果。3组ATDC5细胞内活性氧含量比较,差异有统计学意义(20.148±5.636,13.959±4.110,40.691±3.146,F=30.096,P=0.001)。预处理组ATDC5细胞内活性氧含量高于对照组和共培养组(P=0.001,P=0.000),对照组和共培养组ATDC5细胞内活性氧含量的差异无统计学意义(P=0.137)。结论:地塞米松可抑制肌卫星细胞C2C12增殖,应用地塞米松干预肌卫星细胞C2C12可体外模拟肌肉萎缩条件下肌卫星细胞的生长状态。正常状态下,肌卫星细胞C2C12的代谢产物可促进软骨细胞ATDC5增殖;肌卫星细胞C2C12活力降低,可抑制软骨细胞ATDC5增殖,同时会增加软骨细胞ATDC5氧化性损伤。 展开更多

【目的】脂肪酸类化合物2E,4E-癸二烯酸(2E,4E-decadienoic acid,DDA)为枯草芽孢杆菌(Bacillussubtilis)Tpb55与棘孢木霉(Trichodermaasperellum)HG1共培养发酵产物,对疫霉具有特异性抑制作用。论文旨在评价DDA防治烟草疫霉(Phytophthor... 【目的】脂肪酸类化合物2E,4E-癸二烯酸(2E,4E-decadienoic acid,DDA)为枯草芽孢杆菌(Bacillussubtilis)Tpb55与棘孢木霉(Trichodermaasperellum)HG1共培养发酵产物,对疫霉具有特异性抑制作用。论文旨在评价DDA防治烟草疫霉(Phytophthoranicotianae)引起的烟草黑胫病的效果及其对根际土壤微生物群落的影响,为该类天然产物在植物疫霉病害防治上的应用提供依据。【方法】通过温室盆栽试验测定DDA对烟草黑胫病的防治效果,采用实时荧光定量PCR技术检测处理后根际土壤中烟草疫霉DNA拷贝数,利用Illumina Hiseq高通量测序分析烟草根际土壤细菌、真菌和卵菌群落结构,基于冗余分析和Mantel检验研究土壤养分因子与微生物群落结构的相关性。【结果】与对照(0.5%DMSO)处理相比,1.25 g·L-1的DDA溶液灌根处理组烟草黑胫病病情指数和烟草根际烟草疫霉DNA拷贝数分别降低77.06%和84.35%,土壤pH由6.72增至6.82,电导率增加102.11%,有机质含量则降低13.05%。DDA灌根处理对烟草根际土壤真菌和细菌多样性和丰富度无显著影响,但卵菌的OTU水平、香农指数、Chao1指数和ACE指数显著增加,辛普森指数显著降低。主坐标分析结果显示,DDA处理显著影响烟草根际卵菌群落结构(r=0.667,P=0.028),对真菌(r=0.259,P=0.305)和细菌(r=0.593,P=0.098)则无显著影响。DDA处理组烟草根际土壤Phycicoccus、地杆菌属等革兰氏阳性细菌菌属的相对丰度显著增加,德沃斯氏菌属、鲍特氏菌属、假黄单胞菌属等革兰氏阴性细菌菌属丰度显著降低,曲霉属、Albifimbria、Arcopilus、树粉孢属等植物有益真菌菌属得到显著富集,疫霉属相对丰度显著下降、腐霉属丰度显著上升。Mantel检验结果表明,施用DDA引起的土壤理化性质变化对根际微生物群落无显著影响。【结论】2E,4E-癸二烯酸(DDA)作为一种新型天然脂肪酸化合物,能够有效防治烟草黑胫病、降低烟草根际黑胫病菌数量,并特异性调控烟草根际卵菌群落结构,具有防治作物卵菌病害的应用潜力。 展开更多

The traditional rice-fish farming system is selected as a "globally important agricultural heritage system" (GIAHS) by the Food and Agriculture Organization (FAO),United Nations Development Programme (UNDP),and Global Environment Facility (GEF),etc.In Zhejiang Province of China,where the pilot site for this GIAHS farming system is located,we compared the use of traditional rice varieties in rice-fish co-culture and rice monoculture.Further,we determined how traditional rice varieties were performed in this rice-fish system.Only 19% of the farmers who practiced rice monoculture planted traditional varieties while 52% of farmers who practiced rice-fish co-culture planted traditional varieties.Traditional varieties represented 13% of the total land cultivated under rice in the rice-fish system but only 2% in the rice monoculture system.In the rice-fish system,yield was lower for traditional rice varieties than hybrid varieties but application of fertilizers and pesticides was also lower.In a field experiment in the rice-fish system without pesticides,rice planthopper numbers and sheath blight incidence were lower from three traditional varieties than one hybrid variety;yields were 8 to 32% lower from the traditional varieties than the hybrid.Our results showed that traditional rice varieties can be preserved through conserving GIAHS rice-fish co-culture.Our study also indicated that traditional rice varieties can survive in the rice-fish system because these varieties are helpful to the whole system and beneficial to the farmers. 展开更多

By using single rice cultivation as a control, the effect of rice-fish culture on growth dynamic, plant type and yield formation of rice was studied. The results showed as follows: rice-fish culture improved the physical-chemical properties of arable layer soil of paddy field, extended growth period of rice, increased dry matter and LAI of different growth stages, improved three top leaves area, deterred the degeneration of leaves function, increased the diameter of stem, promoted the growth of roots and the formation of roots in the extended stem. At the same time, rice-fish culture extended the length of basal internodes, increased the number of internodes, uplifted the gravity of plant, and depressed the root vigor. For the grain yield and yield structure office, rice-fish culture decreased ear/tillering ratio, spikelet/panicle and seed set percentage, increased grain weight. If variety choice and cultivation technology were controlled appropriately, rice-fish culture could increase the effective panicles and improve grain yield of rice. 展开更多

Rice-fish co-culture has gained increasing attention to remediate the negative environmental impacts induced by intensive aquaculture. However, the effect of rice-fish co-culture on oxygen depletion has rarely been investigated. We constructed a rice-fish co-culture system in yellow catfish(Pelteobagrus fulvidraco) and freshwater shrimp(Macrobrachium nipponense) ponds using a new high-stalk rice variety, and conducted a field experiment to investigate the effect of rice-fish co-culture on water parameters and oxygen consumption. The results showed that rice-fish co-culture reduced the nutrients(total nitrogen, ammonia-N, total phosphorous and potassium) and the dissolved oxygen content in fish and shrimp ponds. However, they showed similar seasonal change of dissolved oxygen in the water of fish and shrimp ponds. Rice-fish co-culture reduced the total amount of oxygen consumption and optimized the oxygen consumption structure in pond. The respiration rates in water and sediment were significantly reduced by 66.1% and 31.7% in the catfish pond, and 64.4% and 38.7% in the shrimp pond, respectively, by additional rice cultivation. Rice-fish co-culture decreased the proportions of respiration in sediment and water, and increased the proportion of fish respiration. These results suggest that rice-fish co-culture is an efficient way to reduce hypoxia in intensive culture pond. 展开更多

Rice-duck (RD) and rice-fish (RF) ecological systems are major complex planting and breeding models of rice paddy fields in southern China. Studying the methane (CH4) and nitrous oxide (N2O) emissions and their economic value from these two ecosystems can provide theoretical and practical basis for further development and utilization of these classical agricultural techniques. CH4 and N2O emissions from RD and RF ecological systems were measured in situ by using static chambers technique. Using global warming potentials (GWPs), we assessed the greenhouse effect of CH4 and N2O and their economic value. Results showed that the peaks of CH4 emission fluxes from RD and RF appeared at full tillering stage and at heading stage, and the average emission fluxes were significantly (P〈 0.05) lower than that from CK. N2O fluxes remained low when the field is flooded and high after draining the water. Compared with CK, the total amount of N2O emissions was significantly (P〈0.05) higher and slightly lower than those from RD and RF, respectively. In 2006 and 2007, the total greenhouse effect of CH4 and N20 from RD and RF were 4 728.3 and 4 611 kg CO2 ha^-1, 4 545 and 4 754.3 kg CO2 ha^-1, respectively. The costs of greenhouse effect were 970.89 and 946.81 RMB yuan ha^-1, and 933.25 and 976.23 RMB yuan ha^-1, respectively, which were significant lower than those from CK (5 997.6 and 5 391.5 RMB yuan ha^-1). Except for the environment cost of CH4 and N2O, the economic benefits from RD and RF were 2 210.64 and 4 881.92 RMB yuan ha^-1; 3 798.37 and 5 310.64 RMB yuan ha^-1, respectively, higher than those from CK. Therefore, RD and RF complex ecological planting and breeding models can effectively decrease and control CH4 and N2O emissions, and they are two of the effective strategies to reduce greenhouse gases from rice paddy fields and contribute in alleviating global warming. Data were collected from 184 respon... We evaluated the factors influencing the adoption of rice-fish farming in the Tavalesh region near the Caspian Sea in northern Iran. We conducted a survey with open-ended questions. Data were collected from 184 respondents (61 adopters and 123 non-adopters) randomly sampled from selected villages and analyzed using logistic regression and multi- response analysis. Family size, number of contacts with an extension agent, participation in extension-education activities, membership in social institutions and the presence of farm workers were the most important socio-economic factors for the adoption of rice-fish farming system. In addition, economic problems were the most common issue reported by adopters. Other issues such as lack of access to appropriate fish food, losses of fish, lack of access to high quality fish fingerlings and dehydration and poor water quality were also important to a number of farmers. 展开更多

【目的】研究稻渔综合种养对成都平原水稻产量、外观和食味品质的影响,为成都平原稻田绿色综合种养和优质稻米生产提供理论依据。【方法】于2020-2021年在成都市崇州现代都市农业功能区选择4个不同稻渔综合种养示范点,以临近的常规水稻... 【目的】研究稻渔综合种养对成都平原水稻产量、外观和食味品质的影响,为成都平原稻田绿色综合种养和优质稻米生产提供理论依据。【方法】于2020-2021年在成都市崇州现代都市农业功能区选择4个不同稻渔综合种养示范点,以临近的常规水稻田为对照,测定稻米产量和垩白粒率、垩白度、胶稠度、直链淀粉、蛋白质等品质指标。【结果】稻渔综合种养田水稻产量2020年A3、A4样点显著高于CK,其他样点差异不显著。从产量构成因素看,产量差异主要体现在有效穗数、穗粒数和结实率。与常规稻田对照相比,稻渔综合种养田稻米的垩白粒率和垩白度显著降低。部分样点稻渔综合种养田显著降低了稻米的胶稠度,提高了直链淀粉含量,大部分样点稻米蛋白质含量有升高趋势。稻渔综合种养模式下,不同稻米品种的蛋白质含量表现出较大差异。其中2021年样点A1水稻品种川康优丝苗的蛋白质含量较CK显著降低,表现出品质改善的趋势。但是样点A3的水稻品种锦香优丝苗和A4的水稻品种宜香优2115的蛋白质含量,较CK显著增高。【结论】成都平原稻渔综合种养水稻产量较常规稻田显著提高或基本持平,显著提高了稻米的外观品质,未明显提高稻米食味品质。选择搭配适宜的水稻品种,并优化肥水管理可能是提升稻渔综合种养下稻米食味品质的有效途径。 展开更多

为了分析施入生物有机肥的稻蟹共作稻田(HXTCS)的水稻根际土壤微生物的代谢功能,以生物有机肥稻田(YJTCS)为对照,利用Illumina Novaseq测序平台对两种稻田成熟期的根际土壤进行宏基因组测序以分析微生物群落的组成及代谢功能。NR注释结... 为了分析施入生物有机肥的稻蟹共作稻田(HXTCS)的水稻根际土壤微生物的代谢功能,以生物有机肥稻田(YJTCS)为对照,利用Illumina Novaseq测序平台对两种稻田成熟期的根际土壤进行宏基因组测序以分析微生物群落的组成及代谢功能。NR注释结果显示,样品中微生物以细菌为主,约占98.09%,其次为古菌约占1.76%,真菌及其他约占0.15%。LEfSe分析结果表明,河蟹引入生物有机肥稻田后富集更多的细菌和古菌群落的显著差异分类群。COG和CAZy功能基因注释结果显示,HXTCS主要相关功能基因的相对丰度均高于对照组。碳代谢途径基因注释分析结果表明,HXTCS在16个碳代谢途径的基因注释的相对丰度均高于对照组,其中丙酮酸代谢和原核微生物的碳固定途径的相对丰度在2种稻田中差异显著(P<0.05)。氮代谢功能分析结果表明,HXTCS中的反硝化作用、硝酸盐还原作用和谷氨酰胺合成作用的关键酶基因的相对丰度均高于对照组。研究结果说明:河蟹引入生物有机肥稻田后,增加了根际土壤碳水化合物活性酶和主要代谢途径基因的相对丰度,有助于碳代谢及促进根际土壤氮循环中的反硝化作用、硝酸盐还原作用及谷氨酸的合成作用。 展开更多

该文通过优化接种比例、接种时序和碳氮比构建了小球藻和胶球藻最佳共培养体系,并探究了小球藻和胶球藻最佳共培养对生物量和油脂产率的影响.结果显示,小球藻和胶球藻种子液接种体积比为3∶7,生长起始同步接入碳氮比为32的培养基构建的... 该文通过优化接种比例、接种时序和碳氮比构建了小球藻和胶球藻最佳共培养体系,并探究了小球藻和胶球藻最佳共培养对生物量和油脂产率的影响.结果显示,小球藻和胶球藻种子液接种体积比为3∶7,生长起始同步接入碳氮比为32的培养基构建的培养体系为最佳,此培养条件下小球藻和胶球藻获得的生物量和油脂产率最大值分别为1.13 g/(L·d)和1.22 g/(L·d),是单一小球藻和胶球藻培养获得生物量和油脂产率的1.75~2.46倍和3.21~4.52倍,且生长周期较单一微藻培养均缩短72 h.共培养小球藻和胶球藻胞内淀粉和蛋白质含量较单一培养藻株显著性下调1.19~1.47倍和1.59~1.71倍,推测其可能是由于小球藻和胶球藻共培养效应驱动更多的碳流由淀粉和蛋白质合成方向转而流向脂质路径.进一步流加补料发酵,共培养小球藻和胶球藻获得的生物量和油脂产率较分批共培养小球藻和胶球藻分别提高2.54倍和1.61倍.研究结果证实了小球藻和胶球藻共培养可有效促进生物量和脂质积累,为微藻脂质工业化扩大生产奠定了基础. 展开更多
