[Method] The aseptic seedlings of Guisangyou 12 were used as the materials and the comparative experiment on subculture multi-plication and root induction used different kinds,concentrations and combinations of exogenous hormones. [Result] The hormone combination of 0.1 mg/L IBA+1.5 mg/L 6-BA+0.03 mg/L TDZ showed the best effects to bud multiplication, and the multiple of bud multiplication was 5.28. The effects of NAA was better than IBA and IAA on root induction. The hormone combination of 2.0 mg/L NAA+1.0 mg/L PP333 was most suitable to the root induction, in which the rooting rate arrived to 100%, the root number was 7.01 and the root length was 1.38 cm on average. [Conclusion] The results wil provide some technical reference for large-scale propagation of mul-berry seedlings in vitro.展开更多

Festival, RABI-3, and Neho strawberry genotypes’ runner tips were cultiv...A study was done to produce a rapid in vitro propagation of three strawberry genotypes and tested in the field under Bangladeshi circumstances. Festival, RABI-3, and Neho strawberry genotypes’ runner tips were cultivated in vitro to induce root induction and multiple shoot proliferation. MS (Murashige and Skoog) media that were basally containing three different concentrations at 1.0, 1.5, and 2.0 of BA (6-benzyl adenine), KIN (6-furfuryl amino purine), or GA<sub>3</sub> (gibberellic acid) at 0.5 mg/L increasing tips of the runner was attained. The culture grew on the medium provided with 1.5 mg/L 6-benzyl adenine and 0.5 mg/L 6-furfuryl amino acid to increase shoot at the best level. Micro-cuttings were rooted on MS media at half strength combined with 0.5 mg/L - 1.5 mg/L IBA (indole butyric acid) or IAA (indole acetic acid). IBA attained 4 - 9 roots and 91% - 96% rooting at 1.0 mg/L. The resulting plantlets grew into hardy plants and took root in the earth. The genotype festival had the highest response rate, followed by RABI-3 and Neho.展开更多

Five Benzylaminopurine (BAP) concentrations were utilized for shoot induction—0.0 mg/L (Control), 0.5 mg/L, 1.0 mg/L, 1.5 mg/L, and 2.0 mg/L and five Indole Buteric Acid (IBA) concentrations—0.0 mg/L(Control), 0.5 mg/L, 1.0 mg/L, 1.5 mg/L, and 2.0 mg/l—were used for the induction of the root. The highest mean amount of shoots (eight) and length of the shoot (3.40 cm) were observed at a concentration of BAP of 0.5 mg/L. Festival also had the highest mean amount of leaves (6) when we used the identical concentration, while RABI-3 and Camarosa did the least well. The IBA of 0.5 mg/L concentration of rooting performed the best across all metrics tested among the five concentrations. The longest (3.3 cm) roots and most roots (7) were likewise obtained from this concentration in Festival. However, RABI-3 and Camarosa performed worse than Festival in the same concentration. Phyllanthus hainanensis is a shrub that has been used in traditional herbal medicine.It has great pharmaceutical potential for treating diseases such as cancer and diabetes.As a prerequisite for propagation of this sp...Phyllanthus hainanensis is a shrub that has been used in traditional herbal medicine.It has great pharmaceutical potential for treating diseases such as cancer and diabetes.As a prerequisite for propagation of this species on a large scale,hairy roots in P.hainanensis were induced using Rhizobium rhizogenes and various factors affecting hairy root induction and growth evaluated.Seven factors were tested:(1)type of explant,(2)type of culture medium,(3)duration of pre-culture,(4)R.rhizogenes inoculum cell density,(5)duration of infection,(6)acetosyringone concentration in the culture medium,and(7)duration of incubation.The optimal protocol for hairy root induction and growth was:young shoots,pre-cultured in Y1 for 2 d,inoculated with R.rhizogenes broth with an OD600 of 0.6 for 20 min,and incubated for 3 d.Putative transgenic hairy roots were initially identified by morphology and then confirmed by polymerase chain reaction.Successful and optimal production of hairy roots is a critical prerequisite for industrial scale clonal propagation of P.hainanensis.Being able to cultivate the plant on a large scale will provide rapid and ready supply of the plant materials that can be used in herbal medicine and in scientific and industrial exploitation.展开更多
