Embryogenic callus derived from clutured young leaves was used to initiate the culture.Plant growth regulators, basal medium formula and carbohydrate levels were examined to determine their various effects on suspension culture cell growth and development. The best selection of plant growth regulator, hasal medium and carbohydrate level is 2 mg / L 2,4-D: 0.5 mg / L KT,MS and 3% sucrose respectively.展开更多

As one forest management type, ginseng cultivation under larch plantations has been developed significantly in the east of Liaoning Province. However, research on the influence of the ecological environment for this mode of production is still deficient. Based on this, our study compares the plant diversity and soil properties in the ginseng cultivation under larch plantations(LG) with larch plantations(LP) and natural secondary forests(SF). First, we randomly selected three plots for each of the three stand types which have similar stand characteristics; then, we carried out a plant diversity survey and soil sampling in each of the nine plots. The results show that no significant difference was found in plant diversity between LG and LP, but theevenness of herbs was significantly lower in LG than LP. No obvious changes in soil physical properties were found in LG, but a significant decrease in most of the soil nutrient content was presented in LG. Furthermore, we found a correlation between plant diversity(H') and soil properties in the three kinds of stand types, especially between herbaceous plant diversity and soil properties. We conclude that ginseng cultivation under larch plantations has no obvious effect on plant diversity, except the herbaceous evenness. Soil fertility can be depleted significantly in LG, but physical structures are not affected. Moreover, maintaining the diversity of herbaceous plants and controlling the density of ginseng cultivation in LG by farmers are important for the ecological environment. Based on this study and its good comprehensive benefits and with the support of policy, we think this forest management type should be promoted moderately in the region.展开更多

The demand for this herb fosters illegal poaching and over-harvesting that reduces genetic variability and population viability. Five wild populations in western North Carolina were studied to better understand the production of ginsenosides in leaf and root tissues. Total ginsenoside concentration was significantly higher in leaves than roots, though total yield was higher in roots due to greater root biomass. However, some ginsensosides (Rb2, Rd and Re) had higher or more consistent yields in leaves than roots, so might be developed into a sustainable source of these medicinally-active compounds. Additionally, we identified regional root chemotypes that differed in the production of the ginsenosides Rg1 and Re and could be developed into regional cultivars depending on the desired panel of ginsenosides.展开更多

The recombinant plasmid pBIFN was transferred into Agrobacterium tumefaciens strain LBA4404. Then the hIFN-α2b gene was introduced into ginseng callus cells via Agrobacterium-mediated transformation and the ginseng cell line carrying hIFN-α2b gene was selected on G418-containing medium. The presence of target gene in transformed cells was confirmed by PCR and RT-PCR. The results indicate that hIFN-α2b gene has been integrated into the ginseng cells' genome, with transcription products, hIFN-α2b expressed by the transgenic ginseng cells was detected by Western blot. It was shown that a specific protein band at 19000 could be observed. Cytopathic effect(CPE) inhibition assay using the W1SH-VSV system shows that the mean antiviral activity of expressed hlFN-a2α was 6.0× 10^4 IU/mL.展开更多
