O-GlcNAcylation is an important post-translational modification and has been implicated in many fundamental cellular processes. Recent studies showed that O-linked N-acetylglucosamine (GlcNAc) transferase (OGT) mediated O-GlcNAcylation of histone H2B Ser 112 (H2B S 112 GlcNAcylation) plays an important role in gene transcription. However, the role of this histone modification in DNA damage response has not been studied yet. In this study, we found that OGT and OGT mediated H2B S112 GlcNAcylation are involved in DNA damage response for maintaining genomic stability and are required for resistance to many DNA-damaging and replication stress- inducing agents. OGT mediated H2B Sl12 GlcNAcylation increased locally upon the induction of double-strand breaks (DSBs), and depletion of OGT or overexpression of H2B S 112A mutant impaired homologous recombination (HR) and nonhomologous end-joining (NHEJ). Mechanistically, H2B Sl12 GlcNAcylation could bind Nijmegen breakage syndrome 1 (NBS1) and regulate NBS1 foci for- mation. Taken together, our results demonstrate a new function of histone O-GlcNAcylation in DNA damage response (DDR). 展开更多

S112是经诱变获得的抗丝氨酸结构类似物和分解能力缺陷的突变株,丝氨酸产率是1 6g/l。摇瓶试验初糖50g/l,发酵37h时补糖40g/l,可有效提高产酸率50%左右。分批培养补氮试验表明补充氮源同时补糖与只补糖相比,产酸率差异不明显,说明丝氨酸产生菌产丝氨酸过程对氮源的需求不高。经发酵培养基氮源的筛选,认为10%鱼粉水解液是最优的。在7升发酵罐试验,没有补糖的情况下46h开始产丝氨酸,46、54、58、62、66和72h产酸率大致相同,为1 8g/l。 展开更多

Non-uniform step-size distribution is implemented for split-step based nonlinear compensation in singlechannel 112-Gb/s 16 quadrature amplitude modulation (QAM) transmission. Numerical simulations of the system including a 20 × 80 km uncompensated link are performed using logarithmic step size distribution to compensate signal distortions. 50% of reduction in number of steps with respect to using constant step sizes is observed. The performance is further improved by optimizing nonlinear calculating position (NLCP) in case of using constant step sizes while NLCP optimization becomes unnecessary when using logarithmic step sizes, which reduces the computational effort due to uniformly distributed nonlinear phase for all successive steps. 展开更多
