Objective Innate lymphoid cells(ILCs)are a class of newly discovered immunocytes.Group 1 ILCs(ILC1s)are identified in the decidua of humans and mice.High mobility group box 1(HMGB1)is predicted to be one of the target...Objective Innate lymphoid cells(ILCs)are a class of newly discovered immunocytes.Group 1 ILCs(ILC1s)are identified in the decidua of humans and mice.High mobility group box 1(HMGB1)is predicted to be one of the target genes of miR-142-3p,which is closely related to pregnancy-related diseases.Furthermore,miR-142-3p and HMGB1 are involved in regulating the NF-κB signaling pathway.This study aimed to examine the regulatory effect of miR-142-3p on ILC1s and the underlying mechanism involving HMGB1 and the NF-κB signaling pathway.Methods Mouse models of normal pregnancy and abortion were constructed,and the alterations of ILC1s,miR-142-3p,ILC1 transcription factor(T-bet),and pro-inflammatory cytokines of ILC1s(TNF-α,IFN-γand IL-2)were detected in mice from different groups.The targeting regulation of HMGB1 by miR-142-3p in ILC1s,and the expression of HMGB1 in normal pregnant mice and abortive mice were investigated.In addition,the regulatory effects of miR-142-3p and HMGB1 on ILC1s were detected in vitro by CCK-8,Annexin-V/PI,ELISA,and RT-PCR,respectively.Furthermore,changes of the NF-κB signaling pathway in ILC1s were examined in the different groups.For the in vivo studies,miR-142-3p-Agomir was injected in the uterus of abortive mice to evaluate the abortion rate and alterations of ILC1s at the maternal-fetal interface,and further detect the expression of HMGB1,pro-inflammatory cytokines,and the NF-κB signaling pathway.Results The number of ILC1s was significantly increased,the level of HMGB1 was significantly upregulated,and that of miR-142-3p was considerably downregulated in the abortive mice as compared with the normal pregnant mice(all P<0.05).In addition,miR-142-3p was found to drastically inhibit the activation of the NF-κB signaling pathway(P<0.05).The number of ILC1s and the levels of pro-inflammatory cytokines were significantly downregulated and the activation of the NF-κB signaling pathway was inhibited in the miR-142-3p Agomir group(all P<0.05).Conclusion miR-142-3p can regulate ILC1s by targeting HMGB1 via the NF-κB signaling pathway,and attenuate the inflammation at the maternal-fetal interface in abortive mice.展开更多

The study of the chlorite coatings always attracts scholars in China and other countries because the chlorite coatings play an important role in the preservation of residual primary pores in sandstone reservoirs.At pr...The study of the chlorite coatings always attracts scholars in China and other countries because the chlorite coatings play an important role in the preservation of residual primary pores in sandstone reservoirs.At present,the study of the origin and the controlling factors is relatively few.The occurrence,time of formation,genesis,controlling factors,and the mechanism of chlorite coatings inhibiting quartz overgrowths were studied in detail with thin section and SEM analysis.Samples were from the sandstone reservoirs of the T3x Group in the Baojie area,the transitional zone from the middle to the south of Sichuan Basin.The results indicate that the chlorite coatings on the walls of the pore spaces are oriented perpendicular to grain surfaces in the form of isopachous(even-thickness) grain-coating,while the chlorite coatings at the contacts between adjacent detrital grains are arranged with a preferred orientation tangential to the surface of detrital grains.The chlorite coatings were formed in the eogenetic stage.They were formed by recrystallization of Fe-rich clay films during the syndepositional period,and chlorite cements would be recrystallized after the coatings’ formation.The formation of chlorite coatings was mainly controlled by the depositional environment,provenance conditions,and diagenetic environment.The presence of chlorite coatings could result in the preservation of primary pores in deeply buried sandstone reservoirs by effectively inhibiting quartz overgrowths and the development of compaction and pressure solution.展开更多

However, extraction of the photoconductivi...An in-depth understanding of the photoconductivity and photocarrier density at the interface is of great significance for improving the performance of optoelectronic devices. However, extraction of the photoconductivity and photocarrier density at the heterojunction interface remains elusive. Herein, we have obtained the photoconductivity and photocarrier density of 173 nm Sb2Se3/Si(type-Ⅰ heterojunction) and 90 nm Sb2Se3/Si(type-Ⅱ heterojunction) utilizing terahertz(THz) time-domain spectroscopy(THz-TDS) and a theoretical Drude model. Since type-Ⅰ heterojunctions accelerate carrier recombination and type-Ⅱ heterojunctions accelerate carrier separation, the photoconductivity and photocarrier density of the type-Ⅱ heterojunction(21.8×10^(4)S·m^(-1),1.5 × 10^(15)cm^(-3)) are higher than those of the type-Ⅰ heterojunction(11.8×10^(4)S·m^(-1),0.8×10^(15)cm^(-3)). These results demonstrate that a type-Ⅱ heterojunction is superior to a type-Ⅰ heterojunction for THz wave modulation. This work highlights THz-TDS as an effective tool for studying photoconductivity and photocarrier density at the heterojunction interface. In turn, the intriguing interfacial photoconductivity effect provides a way to improve the THz wave modulation performance.展开更多

AIM:To elucidate high mobility group-box 3(HMGB3) protein expression in gastric adenocarcinoma,its potential prognostic relevance,and possible mechanism of action.METHODS:Ninety-two patients with gastric adenocarcinom...AIM:To elucidate high mobility group-box 3(HMGB3) protein expression in gastric adenocarcinoma,its potential prognostic relevance,and possible mechanism of action.METHODS:Ninety-two patients with gastric adenocarcinomas surgically removed entered the study.HMGB3 expression was determined by immunohistochemistry through a tissue microarray procedure.The clinicopathologic characteristics of all patients were recorded,and regular follow-up was made for all patients.The inter-relationship of HMGB3 expression with histological and clinical factors was analyzed using nonparametric tests.Survival analysis was carried out by Kaplan-Meier(log-rank) and multivariate Cox(Forward LR) analyses between the group with overexpression of HMGB3 and the group with low or no HMGB3 ex-pression to determine the prognosis value of HMGB3 expression on overall survival.Further,HMGB3 expression was knocked down by small hairpin RNAs(shRNAs) in the human gastric cancer cell line BGC823 to observe its influence on cell biological characteristics.The MTT method was utilized to detect gastric cancer cell proliferation changes,and cell cycle distribution was analyzed by flow cytometry.RESULTS:Among 92 patients with gastric adenocarcinomas surgically removed in this study,high HMGB3 protein expression was detected in the gastric adenocarcinoma tissues vs peritumoral tissues(P < 0.001).Further correlation analysis with patients' clinical and histology variables revealed that HMGB3 overexpression was obviously associated with extensive wall penetration(P = 0.005),a positive nodal status(P = 0.004),and advanced tumor-node-metastasis(TNM) stage(P = 0.001).But there was no correlation between HMGB3 overexpression and the age and gender of the patient,tumor localization or histologic grade.Statistical Kaplan-Meier survival analysis disclosed significant differences in overall survival between the HMGB3 overexpression group and the HMGB3 no or low expression group(P = 0.006).The expected overall survival time was 31.00 ± 3.773 mo(95%CI = 23.605-38.395) for patients with HMGB3 overexpression and 49.074 ± 3.648 mo(95%CI = 41.925-57.311) for patients with HMGB3 no and low-level expression.Additionally,older age(P = 0.040),extensive wall penetration(P = 0.008),positive lymph node metastasis(P = 0.005),and advanced TNM tumor stage(P = 0.007) showed negative correlation with overall survival.Multivariate Cox regression analysis indicated that HMGB3 overexpression was an independent variable with respect to age,gender,histologic grade,extent of wall penetration,lymph nodal metastasis,and TNM stage for patients with resectable gastric adenocarcinomas with poor prognosis(hazard ratio = 2.791,95%CI = 1.233-6.319,P = 0.019).In the gene function study,after HMGB3 was knocked down in the gastric cell line BGC823 by shRNA,the cell proliferation rate was reduced at 24 h,48 h and 72 h.Compared to BGC823 shRNA-negative control(NC) cells,the cell proliferation rate in cells that had HMGB3 shRNA transfected was significantly decreased(P < 0.01).Finally,cell cycle analysis by FACS showed that BGC823 cells that had HMGB3 knocked down were blocked in G1/G0 phase.The percentage of cells in G1/G0 phase in BGC823 cells with shRNA-NC and with shRNA-HMGB3 was 46.84% ± 1.7%,and 73.03% ± 3.51% respectively(P = 0.001),whereas G2/M cells percentage decreased from 26.51% ± 0.83% to 17.8% ± 2.26%.CONCLUSION:HMGB3 is likely to be a useful prognostic marker involved in gastric cancer disease onset and progression by regulating the cell cycle.展开更多
目的 分析开放性胃肠手术患儿术后血清降钙素原(procalcitonin, PCT)、正五聚蛋白3(pentraxin 3,PTX3)、高迁移率族蛋白B1(high mobility group protein B1,HMGB-1)表达及其预测早期感染的价值。方法 2020年1月~2023年1月我院收治的开...目的 分析开放性胃肠手术患儿术后血清降钙素原(procalcitonin, PCT)、正五聚蛋白3(pentraxin 3,PTX3)、高迁移率族蛋白B1(high mobility group protein B1,HMGB-1)表达及其预测早期感染的价值。方法 2020年1月~2023年1月我院收治的开放性胃肠手术患儿206例,依据术后是否并发感染分为感染组(27例)和未感染组(179例)。比较两组一般资料及围手术期指标,比较两组术前、术后1天、3天血清PCT、PTX3、HMGB-1水平;观察术后1天、3天各血清指标单一、联合检测对开放性胃肠手术患儿术后感染的预测价值;多因素Logistic回归分析术后感染的影响因素。结果 感染组术后1天、3天血清PCT、PTX3、HMGB-1水平分别为(2.42±0.39)μg/L、(3.74±0.53)μg/L,(2.07±0.66)μg/L、(3.06±0.75)μg/L,(18.35±2.74)μg/L、(26.09±4.16)μg/L,均高于未感染组的(1.71±0.35)μg/L、(2.29±0.36)μg/L,(1.48±0.52)μg/L、(1.73±0.59)μg/L,(13.04±2.26)μg/L、(15.75±2.83)μg/L,两组比较差异有统计学意义(P<0.05);绘制受试者工作特征(receiver operating characteristic, ROC)曲线显示,术后3天血清PCT、PTX3、HMGB-1联合检测预测开放性胃肠手术患儿术后感染的曲线下面积(Area under the curve, AUC)最大,为0.989;多因素Logistic回归分析显示,年龄为患儿术后感染的独立保护因素,术中出血量、手术时间、术后1天、3天血清PCT、PTX3、HMGB-1为独立危险因素(P<0.05);感染组中重度感染患儿术后1天、3天血清PCT、PTX3、HMGB-1水平分别为(2.63±0.34)μg/L、(4.12±0.56)μg/L、(2.31±0.69)μg/L、(3.39±0.81)μg/L、(19.86±2.91)μg/L、(28.84±4.40)μg/L,均高于轻度感染患儿的(2.11±0.28)μg/L、(3.19±0.49)μg/L、(1.72±0.60)μg/L、(2.58±0.73)μg/L、(16.15±2.39)μg/L、(22.09±3.96)μg/L,差异有统计学意义(P<0.05)。结论 开放性胃肠手术患儿术后血清PCT、PTX3、HMGB-1表达显著增高,且其表达与术后早期感染及感染病情程度有关,三者联合预测价值更高。展开更多

We hypothesized that activation of phosphoin...Objective: The mechanisms by which lipopolysaccharide (LPS) pretreatment induces cardioprotection following ischaemia/reperfusion (I/R) have not been fully elucidated. We hypothesized that activation of phosphoinositide 3-kinase (PI3K)/Akt and high mobility group box 1 (HMGBxl) signaling plays an important role in LPS-induced cardioprotection. Methods: In in vivo experiments, age- and weight- matched male C57BL/10Sc wild type mice were pretreated with LPS before ligation of the left anterior descending coronary followed by reperfusion. Infarction size was examined by triphenyltetrazolium chloride (TTC) staining. Akt, phospho-Akt, and HMGBxl were assessed by immunoblotting with appropriate primary antibodies. In situ cardiac myocyte apop- tosis was examined by the TdT-mediated dUTP nick-end labeling (TUNEL) assay. In an in vitro study, rat cardiac myoblasts (H9c2) were subdivided into two groups, and only one was pretreated with LPS. After pretreatment, the cells were transferred into a hypoxic chamber under 0.5% 02. Levels of HMGBxl were assessed by immunoblot. Results: In the in vivo experiment, pretreatment with LPS reduced the at risk infarct size by 70.6% and the left ventricle infarct size by 64.93% respectively. Pretreatment with LPS also reduced cardiac myocytes apoptosis by 39.1% after ischemia and reperfusion. The mechanisms of LPS induced cardioprotection involved increasing PI3K/Akt activity and decreasing expression of HMGBxl. In the in vitro study, pretreatment with LPS reduced the level of HMGBxl in H9c2 cell cytoplasm following hypoxia. Conclusion: The results suggest that the cardioprotection following I/R induced by LPS pretreatment involves PI3K/Akt and HMGBxl pathways.展开更多

文摘AIM:To elucidate high mobility group-box 3(HMGB3) protein expression in gastric adenocarcinoma,its potential prognostic relevance,and possible mechanism of action.METHODS:Ninety-two patients with gastric adenocarcinomas surgically removed entered the study.HMGB3 expression was determined by immunohistochemistry through a tissue microarray procedure.The clinicopathologic characteristics of all patients were recorded,and regular follow-up was made for all patients.The inter-relationship of HMGB3 expression with histological and clinical factors was analyzed using nonparametric tests.Survival analysis was carried out by Kaplan-Meier(log-rank) and multivariate Cox(Forward LR) analyses between the group with overexpression of HMGB3 and the group with low or no HMGB3 ex-pression to determine the prognosis value of HMGB3 expression on overall survival.Further,HMGB3 expression was knocked down by small hairpin RNAs(shRNAs) in the human gastric cancer cell line BGC823 to observe its influence on cell biological characteristics.The MTT method was utilized to detect gastric cancer cell proliferation changes,and cell cycle distribution was analyzed by flow cytometry.RESULTS:Among 92 patients with gastric adenocarcinomas surgically removed in this study,high HMGB3 protein expression was detected in the gastric adenocarcinoma tissues vs peritumoral tissues(P < 0.001).Further correlation analysis with patients' clinical and histology variables revealed that HMGB3 overexpression was obviously associated with extensive wall penetration(P = 0.005),a positive nodal status(P = 0.004),and advanced tumor-node-metastasis(TNM) stage(P = 0.001).But there was no correlation between HMGB3 overexpression and the age and gender of the patient,tumor localization or histologic grade.Statistical Kaplan-Meier survival analysis disclosed significant differences in overall survival between the HMGB3 overexpression group and the HMGB3 no or low expression group(P = 0.006).The expected overall survival time was 31.00 ± 3.773 mo(95%CI = 23.605-38.395) for patients with HMGB3 overexpression and 49.074 ± 3.648 mo(95%CI = 41.925-57.311) for patients with HMGB3 no and low-level expression.Additionally,older age(P = 0.040),extensive wall penetration(P = 0.008),positive lymph node metastasis(P = 0.005),and advanced TNM tumor stage(P = 0.007) showed negative correlation with overall survival.Multivariate Cox regression analysis indicated that HMGB3 overexpression was an independent variable with respect to age,gender,histologic grade,extent of wall penetration,lymph nodal metastasis,and TNM stage for patients with resectable gastric adenocarcinomas with poor prognosis(hazard ratio = 2.791,95%CI = 1.233-6.319,P = 0.019).In the gene function study,after HMGB3 was knocked down in the gastric cell line BGC823 by shRNA,the cell proliferation rate was reduced at 24 h,48 h and 72 h.Compared to BGC823 shRNA-negative control(NC) cells,the cell proliferation rate in cells that had HMGB3 shRNA transfected was significantly decreased(P < 0.01).Finally,cell cycle analysis by FACS showed that BGC823 cells that had HMGB3 knocked down were blocked in G1/G0 phase.The percentage of cells in G1/G0 phase in BGC823 cells with shRNA-NC and with shRNA-HMGB3 was 46.84% ± 1.7%,and 73.03% ± 3.51% respectively(P = 0.001),whereas G2/M cells percentage decreased from 26.51% ± 0.83% to 17.8% ± 2.26%.CONCLUSION:HMGB3 is likely to be a useful prognostic marker involved in gastric cancer disease onset and progression by regulating the cell cycle.
文摘目的 分析开放性胃肠手术患儿术后血清降钙素原(procalcitonin, PCT)、正五聚蛋白3(pentraxin 3,PTX3)、高迁移率族蛋白B1(high mobility group protein B1,HMGB-1)表达及其预测早期感染的价值。方法 2020年1月~2023年1月我院收治的开放性胃肠手术患儿206例,依据术后是否并发感染分为感染组(27例)和未感染组(179例)。比较两组一般资料及围手术期指标,比较两组术前、术后1天、3天血清PCT、PTX3、HMGB-1水平;观察术后1天、3天各血清指标单一、联合检测对开放性胃肠手术患儿术后感染的预测价值;多因素Logistic回归分析术后感染的影响因素。结果 感染组术后1天、3天血清PCT、PTX3、HMGB-1水平分别为(2.42±0.39)μg/L、(3.74±0.53)μg/L,(2.07±0.66)μg/L、(3.06±0.75)μg/L,(18.35±2.74)μg/L、(26.09±4.16)μg/L,均高于未感染组的(1.71±0.35)μg/L、(2.29±0.36)μg/L,(1.48±0.52)μg/L、(1.73±0.59)μg/L,(13.04±2.26)μg/L、(15.75±2.83)μg/L,两组比较差异有统计学意义(P<0.05);绘制受试者工作特征(receiver operating characteristic, ROC)曲线显示,术后3天血清PCT、PTX3、HMGB-1联合检测预测开放性胃肠手术患儿术后感染的曲线下面积(Area under the curve, AUC)最大,为0.989;多因素Logistic回归分析显示,年龄为患儿术后感染的独立保护因素,术中出血量、手术时间、术后1天、3天血清PCT、PTX3、HMGB-1为独立危险因素(P<0.05);感染组中重度感染患儿术后1天、3天血清PCT、PTX3、HMGB-1水平分别为(2.63±0.34)μg/L、(4.12±0.56)μg/L、(2.31±0.69)μg/L、(3.39±0.81)μg/L、(19.86±2.91)μg/L、(28.84±4.40)μg/L,均高于轻度感染患儿的(2.11±0.28)μg/L、(3.19±0.49)μg/L、(1.72±0.60)μg/L、(2.58±0.73)μg/L、(16.15±2.39)μg/L、(22.09±3.96)μg/L,差异有统计学意义(P<0.05)。结论 开放性胃肠手术患儿术后血清PCT、PTX3、HMGB-1表达显著增高,且其表达与术后早期感染及感染病情程度有关,三者联合预测价值更高。
文摘As a thin film solar cell absorber material, antimony selenide (Sb<sub>2</sub>Se<sub>3</sub>) has become a potential candidate recently because of its unique optical and electrical properties and easy fabrication method. X-ray photoelectron spectroscopy (XPS) was used to determine the stoichiometry and composition of electroless Sb<sub>2</sub>Se<sub>3</sub> thin films using depth profile studies. The surface layers were analyzed nearly stoichiometric. But the abundant amount of antimony makes the inner layer electrically more conductive.
文摘Objective: The mechanisms by which lipopolysaccharide (LPS) pretreatment induces cardioprotection following ischaemia/reperfusion (I/R) have not been fully elucidated. We hypothesized that activation of phosphoinositide 3-kinase (PI3K)/Akt and high mobility group box 1 (HMGBxl) signaling plays an important role in LPS-induced cardioprotection. Methods: In in vivo experiments, age- and weight- matched male C57BL/10Sc wild type mice were pretreated with LPS before ligation of the left anterior descending coronary followed by reperfusion. Infarction size was examined by triphenyltetrazolium chloride (TTC) staining. Akt, phospho-Akt, and HMGBxl were assessed by immunoblotting with appropriate primary antibodies. In situ cardiac myocyte apop- tosis was examined by the TdT-mediated dUTP nick-end labeling (TUNEL) assay. In an in vitro study, rat cardiac myoblasts (H9c2) were subdivided into two groups, and only one was pretreated with LPS. After pretreatment, the cells were transferred into a hypoxic chamber under 0.5% 02. Levels of HMGBxl were assessed by immunoblot. Results: In the in vivo experiment, pretreatment with LPS reduced the at risk infarct size by 70.6% and the left ventricle infarct size by 64.93% respectively. Pretreatment with LPS also reduced cardiac myocytes apoptosis by 39.1% after ischemia and reperfusion. The mechanisms of LPS induced cardioprotection involved increasing PI3K/Akt activity and decreasing expression of HMGBxl. In the in vitro study, pretreatment with LPS reduced the level of HMGBxl in H9c2 cell cytoplasm following hypoxia. Conclusion: The results suggest that the cardioprotection following I/R induced by LPS pretreatment involves PI3K/Akt and HMGBxl pathways.
文摘A unit cell geometrical structure was found with the use of symmetry operations corresponding to the point group C3. Based on the symmetry of space group R3, a 3D braided geometrical structure was obtained by transforming the unit-cell. The features corresponding to this braided structure were studied. The fiber volume percentage and variational tendencies of the material were predicted by establishing a geometric model.
