In the present experiment, effects of sodium hydrosulfide (NariS), a H2S donor, on the oxidative damage, antioxidant capacity and the growth of cucumber hypocotyls and radicles were studied under 100 mmol L^-1 NaCl stress. NaCl treatment significantly induced accumulation of H2O2 and thiobarbituric acid-reactive substances (TBARS) in cucumber hypocotyls and radicles, and application of NariS dramatically reduced the accumulation of H/O2 and lipid peroxidation. However, the alleviating effects greatly depended on the concentrations of NariS, and 400 ~tmol L-1 NariS treatment showed the most significant effects. Corresponding to the change of lipid peroxidation, higher activities of antioxidant enzymes as well as the antioxidant capacity indicated as DPPH scavenging ac'tivity, chelating activity of ferrous ions and hydroxyl radical (.OH) scavenging activity were induced by Naris treatment under NaCI stress, especially by 400 Ixmol L-I Naris treatment. With the alleviating lipid peroxidation, the amylase activities in cotyledons were increased, and the length of cucumber hypocotyls and radicles were significantly promoted by NariS treatment under NaCI stress. Unlike the effects of NariS, pretreatment with other sodium salts including Na2S, NazSO4, NaHSO4, Na2SO3, NaHSO3 and NaAc did not show significant effects on the growth of cucumber hypocotyls and radicles. These salts do not release H2S. Based on above results, it can be concluded that the effects of NariS in the experiment depended on the H2S rather than other compounds derived from NariS, and the alleviating effects might related with its function in modulating antioxidant capacity and amylase activities. 展开更多

[Method] The previously isolated TLSB2-K strain was identified though morphological observation, gram staining and 16S rDNA sequence alignment. In ad- dition, the effects of temperature, pH and osmotic pressure on the growth of strain were also investigated by using shaking culture. The Cu-tolerance and Cu enrich- ment capacity of TLSB2-K strain under Cu stress were also studied. [Result] The results showed TLSB2-K belongs to Bacillus spp., and its optimum growth conditions were as follows: temperature, 27 ~C; pH, 7.0; osmotic pressure, 1.1% NaCI. When the Cu concentration ranged from 100 mg/L to 500 mg/L, the strain grew well; when the Cu concentration ranged from 100 mg/L to 400 mg/L, the Cu content in bacteria was increased with the increase of Cu concentration; when the Cu con- centration was 400 mg/L and the incubation time was 30 h, the Cu content in bac- teria reached the peak (2 250 mg/kg); the highest tolerant concentration was 700 mg/L. [Conclusion] TLSB2-K strain had relatively high Cu tolerance and Cu enrich- ment capacity. They had important theoretical research and engineering application values. 展开更多

The vacuolar proton pump ATPase(V-H^+-ATPase), which is a multi-subunit membrane protein complex, plays a major role in the activation of ion and nutrient transport and has been suggested to be involved in several physiological processes, such as cell expansion and salt tolerance. In this study, three genes encoding V-H^+-ATPase subunits B(Sc VHA-B, Gen Bank: JF826506), C(Sc VHA-C, Gen Bank: JF826507) and H(Sc VHA-H, Gen Bank: JF826508) were isolated from the halophyte Suaeda corniculata. The transcript levels of Sc VHA-B, Sc VHA-C and Sc VHA-H were increased by salt, drought and saline-alkali treatments. V-H^+-ATPase activity was also examined under salt, drought and saline-alkali stresses. The results showed that V-H^+-ATPase activity was correlated with salt, drought and saline-alkali stress. Furthermore, V-H^+-ATPase subunits B, C and H(Sc VHA-B, Sc VHA-C and Sc VHA-H) from S. corniculata were introduced separately into the alfalfa genome. The transgenic alfalfa was verified by Southern and Northern blot analysis. During salt and saline-alkali stresses, transgenic lines carrying the B, C and H subunits had higher germination rates than the wild type(WT). More free proline, higher superoxide dismutase(SOD) activity and lower malondialdehyde(MDA) levels were detected in the transgenic plants under salt and saline-alkali treatments. Moreover, the Sc VHA-B transgenic lines showed greater tolerance to salt and saline-alkali stresses than the WT. These results suggest that overexpression of Sc VHA-B, Sc VHA-C and Sc VHA-H improves tolerance to salt and saline-alkali stresses in transgenic alfalfa. 展开更多

Durning the design process of hydrostatic rotary worktable,the processing and assembly tolerance,(the offset of worktable and the gap of the oil film’s thickness)is ignored.But it will cause that the real bearing of oil pocket deviates from the initial design value,and then the performance of rotary worktable will be reduced significantly.Up to now,no effort is found toward the research of influence of the processing and assembly tolerance on the performance of the rotary worktable.So the hydrostatic oil film is assumed as the elastomer in this paper,and then the bearing capacity of the oil pocket is studied with and without the mass offset of the worktable by taking an expression between the bearing capacity and the oil film’s thickness of the oil pocket as the deform compatibility equation.The influence of the processing tolerance of the oil sealing belt’s gap on the bearing capacity of the oil pocket is analyzed.In the light of the liquid hydrostatic worktable of Gantry Moving Milling Center using on the scene,the oil pocket’s pressure of the worktable is tested using Rotary Worktable Test System under the circumstance of the mass offset of the worktable and the gap tolerance of the oil sealing belt,and then the equivalent offset of worktable,the average pressure of the oil pocket and the actual thickness of the oil film are analyzed respectively.The test results show that the bearing capacity component of the oil pocket caused by G is consistent,and the component caused by M is relative to the position of the oil pocket.When the oil sealing belt’s gap is larger than the theoretical value,the bearing capacity of the oil pocket is smaller than the others;whereas the bearing capacity of the oil pocket is larger than the others.The maximum and minimum equivalent offsets are 0.256 4 mm and 0.047 5 mm,respectively,and the average oil pocket pressure varies from 0.345 MPa to 0.460 MPa,the maximum and minimum value of the actual oil film thickness are 109.976?m(No.7 oil pocket)and 93.467?m(No.10 oil pocket),respectively.The research results can be used to detect the offset of the worktable and the actual thickness of the oil film under processing and assembly tolerance,and provides a basis way for detecting the processing and assembly tolerance of rotary worktable signing reasonably of Gantry Moving Milling Center. 展开更多

Two indica rice genotypes, viz. Milyang 46 and Zhenshan 97B differing in Cd accumulation and tolerance were used as materials in a hydroponic system consisting of four Cd levels (0, 0.1, 1.0 and 5.0 μmol/L) and three N levels (23.2, 116.0 and 232.0 mg/L) to study the effects of nitrogen status and nitrogen uptake capacity on Cd accumulation and tolerance in rice plants. N-efficient rice genotype, Zhenshan 97B, accumulated less Cd and showed higher Cd tolerance than N-inefficient rice genotype, Milyang 46. There was consistency between nitrogen uptake capacity and Cd tolerance in rice plants. Increase of N level in solution slightly increased Cd concentration in shoots but significantly increased in roots of both genotypes. Compared with the control at low N level, Cd tolerance in both rice genotypes could be significantly enhanced under normal N level, but no significant difference was observed between the Cd tolerances under normal N (116.0 mg/L) and high N (232.0 mg/L) conditions. The result proved that genotypic differences in Cd accumulation and toxicity could be, at least in part, attributed to N uptake capacity in rice plants. 展开更多

Determining the responses of candidate plants to salt stress is a prerequisite for selecting and breeding suitable plants with high salt tolerance to grow in coastal mudfl at areas with high salinity.Here,2-year cutting seedlings of Ilex purpurea Hassk.(local species)and I.integra Thunb.(introduced species)were grown in pots in a glasshouse and irrigated with a Hoagland-NaCl solution at 0,24,and 48 h.Root samples were collected at 0,1,6,24,and 72 h,and concentration of Na^(+)ion;content of proline,soluble carbohydrate,malondialdehyde(MDA),H_(2)O_(2) and ascorbate;and activity of three key antioxidative enzymes were measured.Roots of I.integra accumulated relatively less Na^(+)and had less membrane lipid peroxidation and H_(2)O_(2) during salt stress,thus indicating a relatively higher salt tolerance than roots of I.purpurea.Values for ascorbate content and antioxidant enzymatic activity suggest that the antioxidant ascorbate and antioxidative catalase may play substantial roles for scavenging reactive oxygen species in I.integra roots during salt treatment.Thus,I.integra is apparently more suitable for growing in local highly saline coastal mudfl ats. 展开更多

一株卵形鲳鲹肠道源粪肠球菌的分离鉴定与生物学特性研究

【目的】近年来鱼源益生菌在水产养殖中的应用越来越广泛,然而卵形鲳鲹(Trachinotus ovatus)源的益生菌鲜有报道。从卵形鲳鲹中分离潜在益生菌并对其生物学特性进行分析,为该菌株在卵形鲳鲹中的应用提供理论基础。【方法】无菌采集健康... 【目的】近年来鱼源益生菌在水产养殖中的应用越来越广泛,然而卵形鲳鲹(Trachinotus ovatus)源的益生菌鲜有报道。从卵形鲳鲹中分离潜在益生菌并对其生物学特性进行分析,为该菌株在卵形鲳鲹中的应用提供理论基础。【方法】无菌采集健康卵形鲳鲹肠道内容物,进行细菌分离与鉴定;对典型菌落做形态观察、革兰染色、生化鉴定、16S rRNA基因序列分析、药敏试验、耐盐能力试验、产酸能力及生长曲线测定和毒力基因鉴定。【结果】根据分离菌株的形态特征、革兰染色、生化特性和16S rRNA基因测序结果比对,分离菌株与粪肠球菌(Enterococcus faecalis)相似度达100%,确定其为粪肠球菌。该菌株菌落呈淡灰白色,为革兰氏阳性球菌,七叶苷、纤维二塘、麦芽糖、甘露醇、水杨苷等生化检测结果均显示阳性。药敏试验结果表明,分离菌株对哌拉西林、多粘菌素B、青霉素、复方新诺明、苯唑西林等药物呈现出不同程度的耐药性;对环丙沙星、麦迪霉素、头孢曲松呈中等敏感,对万古霉素、头孢哌酮、氧氟沙星等11种药物敏感。分离菌株可在0%~8%NaCl浓度下生长,具有一定耐盐性。产酸能力和生长曲线测定结果表明,随着菌株大量繁殖,菌液pH值快速下降,其产酸能力增强。毒力基因鉴定结果表明,分离菌株包含efaA、agg、as、fsrA 4种毒力基因。【结论】该研究成功从卵形鲳鲹肠道内分离到一株潜在益生菌菌株,鉴定为粪肠球菌。该菌株具有良好的耐盐和产酸能力,并含有与肠道黏附相关的毒力基因,可为该菌株的后续应用奠定基础。 展开更多

【目的】分离鉴定一株耐盐好氧反硝化细菌,以开发高效脱氮菌种,加强海水及咸淡水养殖尾水脱氮技术研究。【方法】采集花鲈(Lateolabrax maculatus)池塘原位底泥,采用稀释涂布法初步分离一株脱氮功能最佳的菌株HL4,通过16s rDNA测序以及... 【目的】分离鉴定一株耐盐好氧反硝化细菌,以开发高效脱氮菌种,加强海水及咸淡水养殖尾水脱氮技术研究。【方法】采集花鲈(Lateolabrax maculatus)池塘原位底泥,采用稀释涂布法初步分离一株脱氮功能最佳的菌株HL4,通过16s rDNA测序以及生理生化实验鉴定菌株,通过设置不同碳源、碳氮质量比(m_(C)/m_(N))、pH、温度、盐度等条件探明其最适培养条件及脱氮性能。通过攻毒试验[以健康斑马鱼(Danio rerio)为受试生物],检验菌株的环境安全性。【结果】经鉴定,菌株HL4为黏质沙雷氏菌(Serratia marcescens),命名为S.marcescens HL4。以葡萄糖或琥珀酸钠为碳源,菌株HL4在m_(C)/m_(N)=20、pH=6.0~7.0、温度30℃的条件下具有较强脱氮能力,48 h TN去除率为81.83%~87.17%;该菌株在0~30的盐度环境下均可生长,在0~10盐度范围内72 h的总氮(TN)去除率为68.76%~76.59%,在20~30盐度范围内72 h的TN去除率为23.84%~53.94%;在NH_(4)^(+)-N质量浓度为0~100 mg·L^(-1)时,72 h NH_(4)^(+)-N去除率皆在90%以上,在NH_(4)^(+)-N质量浓度为500 mg·L^(-1)时,72 h NH_(4)^(+)-N去除率高达73.52%。健康斑马鱼在含1×10^(6)CFU·mL^(-1)菌株HL4的水体中15 d后,成活率100%。【结论】菌株HL4生物及环境安全性较高,在处理中高浓度铵废水以及治理咸淡水、低盐度海水养殖尾水等领域具有潜在的应用价值。 展开更多

Sylvester方程在数学和控制理论中经常使用,而零化神经网络(ZNN)在求解此类时变方程方面非常有效。对ZNN的收敛性能进行了研究,基于一个预定义时间稳定性定理设计了一个新的激活函数,进而得到了一个用于求解时变Sylvester方程的新的ZNN模型。该模型被称为预定义时间收敛零化神经网络(PTZNN)模型。与之前的ZNN模型相比,所提出的模型提高了收敛速度使其达到了预定义时间收敛并进一步提高了容噪声能力。最后通过仿真模拟验证了该模型在求解时变Sylvester方程方面优于其他已知模型。 展开更多

In order to investigate the oxygen tolerance capacity of upflow anaerobic solid-state(UASS)with anaerobic filter(AF) system, the effect of microaeration on thermophilic anaerobic digestion of maize straw was investigated under batch conditions and in the UASS with AF system. Aeration intensities of 0–431 m L O2/gvswere conducted as pretreatment under batch conditions. Aeration pretreatment obviously enhanced anaerobic digestion and an aeration intensity of 431 m L O2/gvsincreased the methane yield by 82.2%. Aeration intensities of 0–355 m L O2/gvswere conducted in the process liquor circulation of the UASS with AF system. Dissolved oxygen(DO) of UASS and AF reactors kept around 1.39 ±0.27 and 0.99 ± 0.38 mg/L, respectively. p H was relatively stable around 7.11 ± 0.04. Volatile fatty acids and soluble chemical oxygen demand concentration in UASS reactor were higher than those in AF reactor. Methane yield of the whole system was almost stable at 85 ± 7 m L/gvs as aeration intensity increased step by step. The UASS with AF system showed good oxygen tolerance capacity. 展开更多

为了量化斜拉索在安装过程中抵抗施工误差干扰的能力,从而提高斜拉桥工业化建造可施工性,提出了一种挖掘装配容差性能的区间反演框架,采用可靠度区间下界描述多源不确定性的极端干扰,将反演过程作为一个可靠性约束的多目标优化问题求解... 为了量化斜拉索在安装过程中抵抗施工误差干扰的能力,从而提高斜拉桥工业化建造可施工性,提出了一种挖掘装配容差性能的区间反演框架,采用可靠度区间下界描述多源不确定性的极端干扰,将反演过程作为一个可靠性约束的多目标优化问题求解,开发了基于机器学习的解耦式预测算法,通过集成有限元响应预测、可靠度边界计算与代理模型构建,实现了最不利可靠度指标的直接映射。通过算例对斜拉桥的斜拉索装配过程进行了容差规划,结果表明:将不确定性合理量化后,目标斜拉索能容许的最大施工张拉力误差可达到217.4~317.3 kN,其抗误差干扰率为7.29%~12.98%。区间反演方法可以显著避免多重嵌套寻优产生的巨量算力消耗;在保证结构可靠性与设计最优性前提下,有效提升了现场张拉过程的兼容性和可控性。 展开更多

为了从柿子醋醪中获得适合植物基质发酵的具有产酸能力强、耐受性能好和功能性强的乳酸菌。以自然发酵的柿子醋醪为目标乳酸菌供体,采用MRS液体培养基培养、溶钙圈法初步筛选,分离得到5株乳酸菌菌株。通过对菌株的产酸能力、耐受性、抗... 为了从柿子醋醪中获得适合植物基质发酵的具有产酸能力强、耐受性能好和功能性强的乳酸菌。以自然发酵的柿子醋醪为目标乳酸菌供体,采用MRS液体培养基培养、溶钙圈法初步筛选,分离得到5株乳酸菌菌株。通过对菌株的产酸能力、耐受性、抗氧化能力以及产β-葡萄糖苷酶水平进行测定,对综合性能较强的两株乳酸菌菌株进行形态学及16S rDNA分析。结果表明:经筛选鉴定得到的两株优良乳酸菌为嗜酸乳杆菌(Lactobacillus acidophilus)和植物乳杆菌(Lactobacillus plantarum)。两株乳酸菌菌株产酸能力分别为2.00、2.24 g/100 mL;最低耐受p H均为1.5;最高耐受乙醇浓度均为10%;胆盐耐受性均为1%、最高耐受NaCl浓度均为7%;对ABTS^(+)、DPPH、O^(2-)自由基清除能力分别为65.12%和62.26%,77.42%和68.89%,93.76%和87.89%;产β-葡萄糖苷酶能力分别为26.37,30.27 IU/L。本文从柿子醋醪中获得用于植物基质发酵的具有较强生物学性能的两株乳酸菌菌株,该菌株为益生菌的开发提供了菌种资源。 展开更多

为探究中国沙棘对土壤镉(Cd)胁迫的性别响应差异,该研究以中国沙棘2年生幼苗为材料,利用盆栽试验研究在不同浓度Cd处理下(0(CK)、25、50、100和200 mg·kg^(-1))雌、雄株幼苗的生长、叶片生理特性以及Cd富集特征的差异。结果表明:(1... 为探究中国沙棘对土壤镉(Cd)胁迫的性别响应差异,该研究以中国沙棘2年生幼苗为材料,利用盆栽试验研究在不同浓度Cd处理下(0(CK)、25、50、100和200 mg·kg^(-1))雌、雄株幼苗的生长、叶片生理特性以及Cd富集特征的差异。结果表明:(1)Cd处理下中国沙棘幼苗雌、雄株的株高和基径生长以及各器官生物量均表现出低浓度(<50 mg·kg^(-1))促进,高浓度(> 100 mg·kg^(-1))抑制的现象;低浓度Cd处理下雌株的株高、基径增长率和生物量的增幅均高于雄株;高浓度Cd处理下(200 mg·kg^(-1))雄株株高增长率、叶生物量和总生物量分别较CK显著降低,而雌株均未显著下降。(2)随着Cd浓度升高,雌、雄株叶片光合色素含量和抗氧化酶活性呈先升后降的变化趋势,丙二醛(MDA)和渗透调节物质含量呈上升趋势;Cd浓度为50~200 mg·kg^(-1)时,雌株叶片的光合色素含量、抗氧化酶活性和渗透调节物质含量均高于雄株,而MDA含量始终低于雄株。(3)随着Cd浓度升高,雌、雄株各器官Cd含量均逐渐上升且表现为根>茎>叶,雌株各器官Cd含量均高于雄株;雌、雄株地上和地下部分富集系数呈先增后减的变化趋势且均大于1,转移系数逐渐降低且均小于1;雌株地上和地下部分Cd富集系数以及转移系数均高于雄株,且地上部分Cd富集系数在Cd浓度为25~100 mg·kg^(-1)时显著高于雄株。研究发现,中国沙棘幼苗在土壤Cd浓度小于50 mg·kg^(-1)时表现出一定的Cd耐受性,当土壤Cd浓度大于100 mg·kg^(-1)时其Cd耐受性减弱,雌株对土壤Cd的生长适应性、生理耐性和富集转运能力较雄株强。 展开更多

Rolling element bearing is the most common machine element in rotating machinery.An extended life is among the foremost imperative standards in the optimal design of rolling element bearings,which confide on the fatigue failure,wear,and thermal conditions of bearings.To fill the gap,in the current work,all three objectives of a tapered roller bearing have been innovatively considered respectively,which are the dynamic capacity,elasto-hydrodynamic lubrication(EHL)minimum film⁃thickness,and maximum bearing temperature.These objective function formulations are presented,associated design variables are identified,and constraints are discussed.To solve complex non⁃linear constrained optimization formulations,a best⁃practice design procedure was investigated using the Artificial Bee Colony(ABC)algorithms.A sensitivity analysis of several geometric design variables was conducted to observe the difference in all three objectives.An excellent enhancement was found in the bearing designs that have been optimized as compared with bearing standards and previously published works.The present study will definitely add to the present experience based design followed in bearing industries to save time and obtain assessment of bearing performance before manufacturing.To verify the improvement,an experimental investigation is worthwhile conducting. 展开更多

采用气体密封舱熏气法模拟居室装修后的甲醛污染环境,对16种常见室内观赏植物进行甲醛熏气处理,以熏气12h后植物单位叶面积吸收甲醛的量作为指标评价了不同植物对甲醛气体的净化能力,同时测定了熏气后植物的外观形态变化和叶片的SPAD值... 采用气体密封舱熏气法模拟居室装修后的甲醛污染环境,对16种常见室内观赏植物进行甲醛熏气处理,以熏气12h后植物单位叶面积吸收甲醛的量作为指标评价了不同植物对甲醛气体的净化能力,同时测定了熏气后植物的外观形态变化和叶片的SPAD值、相对电导率、丙二醛含量及POD活性等指标。结果显示,被测植物在一定程度上均可以有效吸收甲醛,单位面积吸收量从大到小依次为香石竹(Dianthus caryophyllus)、瑞典常春藤(Lsodon amethystoides)、蚊净香草(Saivia spp.)、比利时杜鹃(Rhododendron hybrida)、柠檬(Citrus limon)、袖珍椰子(Chamaedorea elegans)、龟背竹(Monstera deliciosa)、冷水花(Pilea cadierei)、长寿花(Kalanchoe blossfeldiana)、‘粉冠军’(Anthurium spp.)、巢蕨(Neottopteris nidus)、吊竹梅(Zebrina pendula)、栀子花(Gardenia jasminoides var.grandiflora)、百合竹(Dracaena reflex)、口红花(Aeschynanthus radicans)、孔雀竹芋(Calathea makoyana)。不同植物在甲醛熏气后表现出不同程度的受害反应和生理生化指标的变化,综合分析结果表明香石竹、瑞典常春藤、蚊净香草、袖珍椰子、冷水花、巢蕨等对甲醛的综合净化能力和抗性较好。 展开更多
