采用SDS聚丙烯酰胺凝胶电泳方法,对锯缘青蟹肌肉中可溶性蛋白质进行分离,以过敏患者阳性血清和正常人阴性血清作对照,通过Western blot鉴定出分子量为14.3kDa的蛋白为锯缘青蟹的特异性过敏原。然后用Sephadex G75凝胶过滤层析和DEAE-52... 采用SDS聚丙烯酰胺凝胶电泳方法,对锯缘青蟹肌肉中可溶性蛋白质进行分离,以过敏患者阳性血清和正常人阴性血清作对照,通过Western blot鉴定出分子量为14.3kDa的蛋白为锯缘青蟹的特异性过敏原。然后用Sephadex G75凝胶过滤层析和DEAE-52离子交换层析纯化出该过敏原,再经MALDI-TOF/TOF-MS检测,利用Blastz在线分析。结果表明,锯缘青蟹特异性过敏原为血蓝蛋白亚基,经ClustalW分析该蛋白的多肽片段序列与可口美青蟹、邓杰内斯蟹、加州龙虾、艾氏真蟹的血蓝蛋白亚基序列有很高的同源性,经Prosite数据库检索后发现肽段1和肽段2有酪蛋白激酶Ⅱ磷酸化位点和蛋白激酶C磷酸化位点,是过敏原参与机体反应的重要活性位点。 展开更多

提高浅海滩涂开发的科技含量是当前海洋开发中的重要课题之一。日光温室被称为"具有中国特色的设施园艺",由于其具有良好的保温、节能效果,被广泛应用于北方地区的蔬菜和花卉生产。已有多位学者对其作过光、热环境及建筑结构... 提高浅海滩涂开发的科技含量是当前海洋开发中的重要课题之一。日光温室被称为"具有中国特色的设施园艺",由于其具有良好的保温、节能效果,被广泛应用于北方地区的蔬菜和花卉生产。已有多位学者对其作过光、热环境及建筑结构等的试验和理论研究,并进行了优化,将它用于水产品的越冬养殖是日光温室发展中的又一新领域。影响日光温室热环境的主要因素有:太阳辐射、风速、气温、相对湿度等。附带水池的日光温室中的水有较强的蓄热能力,其热环境有别于种植温室;另一方面日光温室具有保温性,其内的水池又有别于天然水池。根据有关试验的结果,并借鉴前人关于种植温室和天然水池的结论,本文建立了用于锯缘青蟹越冬的日光温室的热环境数学模型。经试验验证,该模型的计算值与实测值吻合较好,对水产品的越冬管理具有重要意义。 展开更多

The genetic diversity and differentiation of 6 mud crab Scylla serrata populations from southeastern China are investigated using allozyme electrophoresis. The mean number of alleles per locus population is 1.3 ± 0.1; the percentage of polymorphic loci per population is 27.3; the observed heterozygosity ranges from (0.195±0.083) to ( 0.241±0.090) and the expected heterozygosity ranges from ( 0.105±0.043 ) to ( 0.131±0.047 ). The coefficient of gene differentiation among populations is low (Fst =0.032), indicating that only 3.2 % of the total genetic diversity comes from inter-population, while the remaining 96.8 % comes from intra-population differences. The genetic distance among populations is 0.000-0.008 (the average is 0.002). Gene flow among the populations is large (Nm = 7.56).Genetic structure is very similar among 6 Scylla serrata populations. 展开更多

采用RT-PCR及RACE技术克隆锯缘青蟹(Scylla serrata)的热休克蛋白Hsp70基因并进行序列分析。克隆测序后拼接得到一条长2482 bp的cDNA序列,该序列ORF(Open reading frame,开放阅读框)为1950 bp,编码649个氨基酸,分子量约为71.06 kD,理论... 采用RT-PCR及RACE技术克隆锯缘青蟹(Scylla serrata)的热休克蛋白Hsp70基因并进行序列分析。克隆测序后拼接得到一条长2482 bp的cDNA序列,该序列ORF(Open reading frame,开放阅读框)为1950 bp,编码649个氨基酸,分子量约为71.06 kD,理论等电点为5.24。3’UTR(untranslated region,非编码区)为158 bp,5’UTR为40 bp。通过antheprot分析发现2个Hsp70家族的签名序列:IFDLGGGTFDVSIL,IVLVGGSTRIPKIQK;Dnak特征基序DLGTT-S-V;非细胞器基序:RARFEEL;核定位信号标签:KKDPSESKRALRRL;胞质Hsp70特征基序EEVD。同源性分析表明,锯缘青蟹Hsp70编码区核苷酸序列与凡纳滨对虾(Litopenaeus vannamei)、斑节对虾(Penaeus monodon)、罗氏沼虾(Macrobrachium rosenbergii)的相似性分别为84.02%、83.87%和79.60%;核苷酸序列所推导出的Hsp70氨基酸序列,与凡纳滨对虾、斑节对虾和罗氏沼虾的相似性分别为92.79%、92.17%和96.47%。本研究克隆了锯缘青蟹Hsp70基因,为进一步深入研究锯缘青蟹的抗逆机理及其遗传改良奠定了基础。 展开更多

每天从锯缘青蟹育苗池采集幼体,在实验室内依平衡法测定从孵化至大眼幼体各天青蟹幼体的摄食率,并同时测定青蟹幼体每天的干重和比能值,比较研究了饵料密度对青蟹不同发育阶段幼体摄食的影响,以及幼体蜕皮周期内摄食的变化。结果表明,... 每天从锯缘青蟹育苗池采集幼体,在实验室内依平衡法测定从孵化至大眼幼体各天青蟹幼体的摄食率,并同时测定青蟹幼体每天的干重和比能值,比较研究了饵料密度对青蟹不同发育阶段幼体摄食的影响,以及幼体蜕皮周期内摄食的变化。结果表明,轮虫是青蟹蚤状幼体(Z)Ⅰ期(Z1)和Ⅱ期(Z2)适宜的饵料,卤虫无节幼虫是青蟹幼体生长发育主要的能量来源,占整个幼体发育期摄入能量的96·48%。以干重和能量表示的青蟹幼体摄食率(CDW和CJ)随幼体蜕皮发育呈指数增长[CDW=13·463e0·8283(d+1)(r2=0·927),p<0·01;CJ=0·4018e0·7516(d+1)(r2=0·956),p<0·01];青蟹大眼幼体摄食强度显著上升,摄入能量高达整个幼体发育期摄入能量的77·25%。Z1、Z2期为被动摄食,青蟹幼体主动摄食的能力随幼体发育而逐渐提高,大眼幼体为主动摄食。锯缘青蟹各期幼体蜕皮周期内摄食率的变化是:Z1→Z4期蜕皮后上升,蜕皮前稍微下降;Z5期蜕皮后和蜕皮前较高,蜕皮间较低。Z5期日粮水平最低;青蟹大眼幼体期有2个摄食峰值,变化不规则。 展开更多

The regulation of three biogenic amines over the reproductive neuroendoerine activity of the male Scylla serrata was investigated by in vivo injection and in vitro incubation. The testicular index, the ratio of the mature sections in testes, and the ratio of Type B cells in androgenic gland were taken as the quantitative indexes. The in vivo injections indicated that: 5-HT can significantly promote the testicular development and the secretion of the androgenic gland in S. serrata; DA can inhibit the testicular development, but no influence on the secretion of the androgenic gland was found; no significant difference was observed between the OA-injected group and the concurrent control group. In vitro incubations showed that: 5-HT can stimulate the secretion of the brain and the thoracic ganglia, thus accelerating that of the androgenic gland; however, neither OA nor DA showed any significant influence on the secretion of the brain and the thoracic ganglionic mass. As to the optic ganglia, the three biogenic amines hardly have any effect on its secretion. It is the first time to report the regulation of biogenic amines over the reproductive neuroendocrine of male crustaceans through vitro experiments. Results corroborate that 5-HT activates the brain and the thoracic ganglia to secret GSH first, then promote the testicular development through the activity of the androgenic gland. 展开更多

N-Acetyl-β-D-glucosaminidase(NAGase, EC 3.2.1.52), which catalyzes the cleavage of N-acetylgluco- samine polymers, plays important roles in the molting, digestion of chitinous foods in green crab. In the study, the efforts of urea on the activity of NAGase purified from the viscera of green crab(Scylla serrata) have been studied. The results show that appropriate concentrations of urea can lead to reversible inactivation of the enzyme, and the value of the inhibitor concentration leading to 50% of enzyme activity lost(IC50) is estimated to be 0.63 mol/L. The inactivation kinetics has been studied via the kinetic method of the substrate reaction. The rate constants of inactivation have been determined. The value of k+0 is larger than that of k′+0, indicating the free enzyme molecule is more fragile than the enzyme-substrate complex in urea solution. It is suggested that the presence of the substrate offers the marked protection of this enzyme against inactivation by urea. 展开更多

在实验室条件下测定了在盐度5.0,10.0,17.5,25.0,35.0胁迫0,12,24,48,72和96 h,锯缘青蟹血细胞总数(THC)、血细胞吞噬能力及其血清中溶菌酶(LZM)、超氧化物歧化酶(SOD)、碱性磷酸酶(AKP)、酸性磷酸酶(ACP)、过氧化物酶(POD)、酚氧化酶(... 在实验室条件下测定了在盐度5.0,10.0,17.5,25.0,35.0胁迫0,12,24,48,72和96 h,锯缘青蟹血细胞总数(THC)、血细胞吞噬能力及其血清中溶菌酶(LZM)、超氧化物歧化酶(SOD)、碱性磷酸酶(AKP)、酸性磷酸酶(ACP)、过氧化物酶(POD)、酚氧化酶(PO)、溶血素等免疫相关因子的活力变化。结果表明:各梯度盐度胁迫下THC明显下降,其他免疫指标均在12至24 h出现一个峰值变化,而后趋于平稳。在12 h盐度5.0和35.0组吞噬百分比(PP)、吞噬指数(PI)值高于对照组,盐度10.0和25.0组的LZM,PO活力显著高于对照组,除盐度5.0组以外的ACK,APK活力均高于对照组;POD、溶血素活力在盐度10.0和25.0组显示随时间的延续而增高,分别在72和24 h达到最大值,且极显著高于对照组;SOD的变化不显著。说明盐度突变对锯缘青蟹各相关免疫因子均有影响,低浓度短时间的盐度胁迫能引起应激反应,适当提高其免疫力,而大幅度长时间的盐度胁迫可显著影响锯缘青蟹的抗病力。 展开更多

In vitro studies show that: (1) the AG in Stage Ⅲ can significantly accelerate the development of seminiferous tubules and spermic matu- ration; (2) the brain, thoracic ganglia and optic ganglia have no direct influence over the testicular devel- opment; (3) the brain and thoracic ganglia can significantly promote the growth of the AG cells and highly significantly boost the ratio of Type B cells, while the optic ganglia have no such effects. It is the first time for in vitro investigations to confirm that the brain and thoracic ganglia can regulate testicular development through AG in male crustaceans. 展开更多

Antioxidation and ATPase activity in the gill of mud crab Scylla serrata under cold stress

An experiment was designed to study the effect of cold stress on S. serrata. After a one-week adaptation at 28oC, the temperature is suddenly reduced to 4oC. The crabs were sampled every 2 h for 10 h and dissected immediately to measure the enzyme activity. The crabs at room temperature (28oC) were used as the control group. The activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX), the content of malondialdehyde (MDA) and the activity of 4 ATPases (Na+, K+-ATPase; Mg2+-ATPase; Ca2+-ATPase; Ca2+, Mg2+-ATPase) were measured biochemically. In contrast to the control group, the SOD activity increased significantly from 2 to 6 h after the cold stress, and then decreased. The CAT and GPX activities increased in 2 h, and then decreased gradually. The content of MDA increased gradually in 4 h. The activity of Na+, K+-ATPase decreased in 2 h, increased up to the top value at Hour 6, then decreased again. The activities of Mg2+-ATPase, Ca2+-ATPase and Ca2+, Mg2+-ATPase increased significantly in 6 h, insignificantly in any other hours. Under cold stress, the activity of antioxidative enzymes in S. serrata was reduced at first then stabilized, ROS-scavenging weakened, and MDA accumulated gradually in the gill after 6 h. The activity of the 4 ATPases in the crab decreased after 6 h, suggesting that the ability to regulate ion concentration has been paralyzed. Therefore, the maximum period to sustain healthy meat in the crab under cold stress is 6 hours. 展开更多

Acute-phase response is documented to be a significant mechanism of innate immunity in vertebrates and invertebrates.In this study,proteomic methodologies were applied for different protein expressions in hemolymph of Scylla serrata challenged by Vibrio parahaemolyticus after immunization,and in muscles of the crabs separately chal-lenged by V.parahaemolyticus,V.anguillarum and Aeromonas hydrophila.Up-regulated cryptocyanin is documented in the hemolymph and up-regulated calexcitin,wingless(fragment)and tachykinin-related peptide in the muscle as acute-phase proteins.All the four altered proteins were responsible for bacterial stress,but cryptocyanin seemed to be a memory response protein against the chal-lenge by a live bacterium after immunization of the live cells.These up-regulated proteins can be indicative of an understanding of immunity of a crab. 展开更多

A value chain analysis is a detailed description of all the activities and services needed to turn a raw product into a marketable good for delivery to final consumers.The mud crab business is considered by most marketing operators to be a profitable and viable business due to the high demand in the international market.Despite the importance of mud crab fishery,there is no structured marketing facilities in Bangladesh.This research aim to analyze the socioeconomic condition of actors,the profitability of farming,mapping the institutional and non-institutional barriers,and provide a better understanding of the possibility of technological interventions in the value chain.Qualitative and quantitative data were collected through participatory rural appraisal tools that include 200 individual interviews,16 focus group discussions,and 24 key informant interviews conducted in south-west Bangladesh from July to December of 2019.The mud crab value chain analysis revealed that the chain started with crab collection(cultivated and wild harvest),before sale to local vendors,and ended with exporters through several intermediaries.Results noted that most(±95%)exportable crab came from natural sources,with rest(±5%)from aquaculture sector.The average price(USD/kg)ranged from$10.5914.12 and$12.9419.42 for male crabs(grade:XXL),followed by$10.0011.77 and$11.7717.06 for female(grade:FF1)in domestic and international markets,respectively.Despite the high profitability of these sectors,the livelihood of these stakeholders has remained relatively hard.Multi-pronged approaches,like a nursery or increasing farming and backyard seed production,and possible value addition at the farm level may help farmers. 展开更多
