A complementary DNA (cDNA) containing the complete coding sequence of CAT was cloned from the hepatopancreas using reverse-transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends. The full-length cDNA of CAT (2 649 bp) contains a 5'-untranslated region (UTR) of 78 bp, a 3'- UTR of 1 017 bp, with a poly (A) tail, and an open reading frame of 1 554 bp encoding a 517-amino-acid polypeptide with predicted molecular mass of 58.46 kDa and estimated isoelectric point of 6.64. This CAT sequence contained the proximal active site signature (60FDRERIPERWHAKGAG76), proximal heme-ligand signature sequence (350RLFSYPDTH358) and three catalytic amino acid residues (His71, Asn144 and Tyr354). Sequence comparison showed that the CAT deduced amino acid sequence of E. carinicauda shared 68%-92% of identities with those of other species. Quantitative real-time PCR analysis revealed that CAT mRNA was widely expressed in the hepatopancreas (highest), hemocyte, eyestalk, heart, gill, muscle, ovary and stomach. Under low salinity stress, CAT and GPx mRNA expression levels both in the gill and hepatopancreas increased significantly at the first 48 h and 6 h respectively, indicating a tissue- and time-dependent antioxidant response in E. carinicauda. All these results indicate that E. carinicauda CAT is a member of the CAT family and might be involved in the acute response against low salinity stress.展开更多

The relation between the level of petroleum hydrocarbon in whole tissue and the contents of glutathione (GSH), the activity of selenium-dependent glutathione peroxidase (Se-GPx) and glutathione S-transferase (GST) in digestive gland and gill were investigated. The results showed: (1) The contents of petroleum hydrocarbon in oyster collected from four stations (Lundu Port, Xinglin Bay, Tong'an Bay, Huangcuo) were 380.68, 112.34, 27.31, 20.37μg/g wet weight, respectively; (2) the activity of Se-GPx and GST in digestive gland was lower than that in gill, and the content of GSH seemed reversibly; (3) among the four stations, both Se-GPx and GST activity of digestive gland and gill in Saccostrea cuculiata sampled from the four stations showed a good correlation with whole tissue petroleum hydrocarbon, could be as biomarkers of sea oil pollution.展开更多
A new selenium source, Nano red elemental selenium (Nano-Se) was used to study the effect on the GPx activity of broiler chick kidney cells (BCKC) in vitro, Sodium selenite (Na_ 2 SeO_ 3 ) and seleno-1-methionine (S...A new selenium source, Nano red elemental selenium (Nano-Se) was used to study the effect on the GPx activity of broiler chick kidney cells (BCKC) in vitro, Sodium selenite (Na_ 2 SeO_ 3 ) and seleno-1-methionine (Se-Met) were used as the controls. The results showed that the effects of three kinds of Se forms on the GPx activity of BCKC were accordant(p>0.05) compared with each other at 0.01,0.05 and 0.10 μmol/L Se concentrations treatments. In the range of 0.00-0.10 μmol/L Se concentrations, the GPx activity increased with elevation of Se concentrations in medium. For the three kinds of Se forms, the GPx activity reached the climax at 0.10 μmol/L Se concentration. At 0.20 and 0.30 μmol/L Se concentrations, the influnces of three kinds of Se forms were not accordant with one another. For Nano-Se, the GPx activity at 0.20 and 0.30 μmol/L Se concentrations remained the same as that at 0.10 μmol/L Se concentration treatment. For Se-Met, the GPx activity at 0.20 μmol/L Se concentration treatment remained the same with 0.10 μmol/L treatment; the GPx activity at 0.30 μmol/L Se concentration treatment was declined significantly(p<0.05) compared with 0.10 or 0.20 μmol/L treatment. For Na_ 2 SeO_ 3 , the GPx activity falled gradually with Se concentration increasing from 0.10 μmol/L to 0.30 μmol/L, and at 0.30 μmol/L Se concentration treatment, the GPx activity was less than the original of BCKC.
