Preparation of Tartary Buckwheat Seed Coating Agent and Its Effect on Germination

To mitigate the wastage of seed resources and reduce the usage of pesticides and fertilizers, seed coating agentshave gained popularity. This study employs single-factor and multi-index orthogonal experimental design methodsto investigate the seed coating formula and physical properties of Tartary buckwheat. The specific effects ofeach component on Tartary buckwheat seed germination are analyzed. The findings reveal that the seed coatingagent formulated with 1.5% polyvinyl alcohol, 0.15% sodium alginate, 0.2% op-10, 0.1% polyacrylamide, 8% colorant,3% ammonium sulfate, 1% potassium dihydrogen phosphate, and 0.15% carbendazim exhibits the mosteffective coating. It demonstrates optimal physical properties and promotes seed germination efficiently. The suspensionrate of this seed coating agent reaches 91.12%, with a mere 2.13% coating shedding rate and 2.5% coatingseed rot rate. Furthermore, it achieves a germination percentage of 99.17%, which is 20.84% higher than the lowestgroup. The germination potential and index are also significantly higher than the lowest group, with anincrease of 20.84% and 26.56%, respectively. Additionally, the vitality index is 553.08, a 15.75% increase comparedto the lowest group. The application of seed coating agents helps reduce seed resource loss, increase plant numbers,and ultimately enhance agricultural yields. Soybean mosaic virus (SMV) could lead to adult-plant system diseases, and cause mottling of soybean seeds. Genetic analysis and molecular mapping were conducted using an F2 population and derived F3 families from two crosses of Dongnong 3C624 (susceptible)x Dongnong 8143 (resistant) and Dongnong 3C628 (susceptible)× Tie 6915 (resistant). Simple sequence repeat (SSR) markers with bulked segregation analysis (BSA) were used to conduct genetic mapping of the resistance to SMV1 in the segregating populations. The results indicated that resistance to SMV1 in adultplants and the resistance to seed coat mottling in Dongnong 8143 and Tie 6915 was separately controlled by one single dominant gene. The two dominant genes were identified to be linked on the MLG F by Mendel's genetics and SSR genetic mapping. The order and distance of markers DPRSMV1 and DSRSMV1 were Sat 229-6.9 cM-DSRSMV1-4.6 cM-Sat_317-3.6 cM-DPRSMV1-5.2 cM-Satt335. The order and distance of markers TPRSMV1 and TSRSMV1 was Satt160-16.1 cM-TPRSMV1-7.3 cM-Satt516-2.0 cM-TSRSMV1-4.5 cM-Sat_133. This research provides the useful information for breeders to select the two types of SMV resistance simultaneously in soybean breeding through molecular marker-assisted selection (MAS). 展开更多

Brassica napus L.(B.napus)is an important oil crop worldwide and it rapidly accumulates oil at late stage of seed maturation.However,little is known about the cellular mechanism of oil accumulation and seed color changes during the late stage of rapeseed development.Here,we analyzed the ultrastructure of seed coat,aleurone and cotyledon in embryos of B.napus from 25 to 70 days after flowering(DAF).The pigments,which were deposited on the cell wall of palisade cells in seed coat,determined dark black color of rapeseed.The chloroplasts degenerated into non-photosynthetic plastids which caused the green cotyledon to turn into yellow.The chloroplasts in aleurone and cotyledon cells respectively degenerated into remnants without inner and outer envelope membranes and ecoplasts with intact inner and outer envelope membranes.From 40 to 70 DAF,there were degraded chloroplasts without thylakoid,oil bodies contacting with plastids or protein bodies,big starch deposits of chloroplasts degrading into small particles then disappearing,and small endoplasmic reticulum(ER)in aleurone and cotyledon cells.Additionally,there were decreases of chlorophyll content and dramatic increases of oil content in rapeseed.These results suggested that the rapid oil accumulation was independent on the NADPH synthesized by photosynthesis of chloroplasts and probably utilized other sources of reductant,such as the oxidative pentose phosphate pathway during the late stage of rapeseed development.The triacylglycerol assembly presumably utilizes the enzymes in the plastid,cytosol or oil body of cotyledon and aleurone cells. 展开更多

The purpose of this study was to investigate the morphology, transmission route of photosynthetic products in the seed stalk of Caragana arborescens Lam. at various stages after flowering and evaluate the role of the seed stalk in seed maturation using scanning electron microscopy. We found that the mature seed stalk mainly consisted of vessels,sclerenchymatous cells and parenchymatous cells. Elongated cells formed a ring at the junction between the seed stalk and seed, and a large hollow was produced by two layers of sclerenchymatous cells in the interior seed, which functions to sustain seed growth. The opening in the center of the ring was closed during an early stage of seed development, and the opening enlarged as the seed matured.There were two routes for material transport, which started from the seed stalk: one from the seed coat to the embryo,and another directly entering the embryo. Vessels, which are abundant in the seed stalk, are the pathway for absorption of nutrients by the seeds. The vessels were densely packed in the seed stalk, and secondary thickening of the cell wall of the vessels was characterized by net thread thickening, while the secondary thickening of the cell wall of the vessels within the seed was characterized by screw thread thickening. The morphological characteristics of the vessels were adaptive to its functions. 展开更多

By detecting the seed quality, maize seedling indexes, which include root activity, chlorophyll content and malondialdehyde (MDA) content, and through the results we found that this seed coating agent could improve the photosynthetic capacity and enhance seedling resistance. Laboratory bacteriostasis test and study showed that the antibacterial rate of this seed coating agent has reached more than 88%. Fruit, seed morphology and seed coat sculpturing were recorded for 10 species belongs to 9 genera and five tribes of Brassicaceae in eastern region of Saudi Arabia by using light microscope (LM) and scanning electron microscope (SEM). Four seed patterns were recognized based on surface sculpturing pattern;Reticulate, with four subtypes, undulate, verrucate and ocellate. The results of numerical analysis showed that species were grouped into two major clusters and each cluster divided into two groups. The cluster analysis indicates that the characters of fruit and seed are very important characters for identification and taxonomical classification of the tribes. The results indicate that the most tribes of Brassicaceae are polyphyletic, and some are related to each other’s as Lepidieae, Alyssieae and Sisymbrieae. 展开更多

Effects of this soybean seed coating agent on soybean yield were tested through the laboratory and farm experiments. The tested results showed that the soybean yield using this seed coating agent was increased by 15.46%, and its cost was decreased by 27.8%, compared with the conventional soybean seed coating agent. Objective:To investigate the role and mechanism of tamarind seed coat extract(TSCE) on normal human skin fibroblast CCD-l064Sk cells under normal and oxidative stress conditions induced by hydrogen peroxide(H_2O_2).Methods:Tamarind seed coats were extracted with boiling water and then partitioned with ethyl acetate before the cell analysis.Effect of TSCE on intracellular reactive oxygen species(ROS),glutathione(GSH) level,antioxidant enzymes such as superoxide dismulase(SOU),glutathione peroxidase(CPx) and catalase activity including antioxidant protein expression was investigated.Results:TSCE significantly attenuated intracellular ROS in the absence and presence of H_1O_2by increasing GSH level In the absence of H_2O_2,TSCE significantly enhanced SOD and catalase activity but did not affected on CPx.Meanwhile,TSCE significantly increased the protein expression of SOD and CPx in H_2O_2-treated cells.Conclusions:TSCE exhibited antioxidant activities by scavenging ROS.attenuating GSll level that could protect human skin fibroblast cells from oxidative stress.Our results highlight the antioxidant mechanism of tamarind seed coat through an antioxidant enzyme system,the exlracl potentially benefits for health food and cosmeceutical application of tamarind seed coat. 展开更多

Merr.) in the study of genetic, molecular and biochemical processes, due to their easy recognizability. The genetic control of several soybean natural variants has not been studied. The standard phenotype of R gene is black hilum on black seed. The genetic type T16 is the only occurrence with brown hilum on black seed coat and its genetic control was not described until now. The aim of this study is to understand the genetic control of seed coat and hilum color in the genetic types T16 and in the natural variants of Bragg, BR6 and BR13. T16 was combinined with Bragg P and BR13P (black seed color) and BR6M (brown seed color) and T236 (r-m). It was found that the genetic control of the brown hilum trait in black seed coat of the T16 genotype was controlled by two loci segregating independently and controlling the expression of the color of the hilum and the seed coat color. The expression of the brown hilum trait in black seed coat is dependent on locus T_, which controls pubescence color;therefore it occurs only in genotypes with tawny brown pubescence (T_), which caracterizes the pleiotropic effect of this locus on the trait hilum brown trait in black seed coat. The color of the hilum and the seed coat color belong to the same allelic sequence. No maternal effect was found in the expression of hilum brown trait in black seed coat. 展开更多

Seed coat color affects the appearance and commodity quality of mung beans(Vigna radiata L.).The substances that affect mung bean seed coat color are mainly flavonoids,which have important medicinal value.Mapping the seed coat color gene in mung beans would facilitate the development of new varieties and improve their value.In this study,an F2 mapping population consisting of 546 plants was constructed using Jilv9(black seed coat)and BIS9805(green seed coat).Using bulk segregated analysis(BSA)sequencing and kompetitive allele-specific PCR(KASP)markers,the candidate region related to seed coat color was finally narrowed to 0.66 Mb on chromosome(Chr.)4 and included eight candidate genes.Combined transcriptome and metabolome analyses showed that three of the eight candidate genes(LOC106758748,LOC106758747,and LOC106759075)were differentially expressed,which may have caused the differences in flavonoid metabolite content between Jilv9 and BIS9805.These findings can provide a research basis for cloning the genes related to seed coat color and accelerate molecular markerassisted selection breeding in mung beans. 展开更多

The seed coat is important for embryo protection, seed hydration, and dispersal. Seed coat composition is also of interest to the agricultural sector, since it impacts the nutritional value for humans and livestock alike. Although some seed coat genes have been identified, the developmental pathways controlling seed coat development are not completely elucidated, and a global genetic program associated with seed coat development has not been reported. This study uses a combination of genetic and genomic approaches in Arabidopsis thaliana to begin to address these knowledge gaps. Seed coat development is a complex process whereby the integuments of the ovule differentiate into specialized cell types. In Arabidopsis, the outermost layer of cells secretes mucilage into the apoplast and develops a secondary cell wall known as a columella. The layer beneath the epidermis, the palisade, synthesizes a secondary cell wall on its inner tangential side. The innermost layer (the pigmented layer or endothelium) produces proanthocyanidins that condense into tannins and oxidize, giving a brown color to mature seeds. Genetic separation of these cell layers was achieved using the ap2-7 and tt16-1 mutants, where the epidermis/palisade and the endothelium do not develop respectively. This genetic ablation was exploited to examine the developmental programs of these cell types by isolating and collecting seed coats at key tran- sitions during development and performing global gene expression analysis. The data indicate that the developmental programs of the epidermis and the pigmented layer proceed relatively independently. Global expression datasets that can be used for identification of new gene candidates for seed coat development were generated. These dataset provide a comprehensive expression profile for developing seed coats in Arabidopsis, and should provide a useful resource and reference for other seed systems. 展开更多

Coating seeds with water absorbent materials can improve their survival, especially for those planted indrought or barren areas. In this study, effects of five kinds of super absorbent polymers (SAPs) on seed germinationand seedling growth of Caragana korshinskii under drought conditions were investigated. Our results showed that SAPcoatings could significantly improve the percentage and energy of seed germination, as well as reduce the relativeelectrical conductivity (REC), proline, malondialdehyde (MDA), H202 content, and peroxidase (POD) activity duringgermination. These results implied that seeds could uptake moisture from SAP coatings to alleviate drought-inducedoxidative stress and membrane damage, thus exhibiting a better vigor and germination performance. After coating C.korshinskii seeds with SAPs, more seedlings emerged and grew better. Under the combined influence of the waterabsorption capacity of SAP and other factors, the efficiencies of five SAP coatings are in the sequence D〉E〉B〉A〉C.The function of the SAP coating on promoting seedling survival was confirmed in Mu Us Sandy Land in Ordos, InnerMongolia Autonomous Region, China. The average seedling number of SAP D-coated seeds increased twofold on thatof naked seeds. Our results are expected to be helpful in understanding and utilizing SAP seed coatings in improvingplant survival under drought conditions. 展开更多

To better understand the effect and mechanism of cold tolerant seed-coating agents on the cold tolerance of rice seedlings, the physiological and biochemical effects of four cold tolerant seed-coating agents (HET, YKJ, YKZYJ, and the ABA seed coating agents) on two early indica rice varieties were studied under chilling stress. The results showed that the rice seedlings treated with cold tolerant seed-coating agents under chilling stress maintained dramatically higher root vigor, POD, CAT and SOD activities, and chlorophyll content, had lower MDA content and electrolyte leakage, and accumulated more soluble sugar and free proline, when compared with the control without the treatment, and finally showed lower plant injury rate. It was indicated that the cold tolerant seed coating agent improved the ability of rice seedlings in resisting to chilling stress. YKZYJ was ranked the first in terms of the efficiency in cold tolerance among the four cold tolerant seed-coating agents tested. 展开更多

巢式关联作图(Nested Association Mapping,NAM)群体在作物学遗传与育种研究中具有广泛的应用。本研究在前期大豆种质资源评价基础上,利用35份不同地区来源的代表性种质与中豆41(公共母本)杂交,构建了一套大豆NAM群体。PCA和聚类分析发... 巢式关联作图(Nested Association Mapping,NAM)群体在作物学遗传与育种研究中具有广泛的应用。本研究在前期大豆种质资源评价基础上,利用35份不同地区来源的代表性种质与中豆41(公共母本)杂交,构建了一套大豆NAM群体。PCA和聚类分析发现,不同亲本组合的RIL群体基本聚在一起,显示出清晰的遗传结构。利用该NAM群体亲本间花色和种皮色具有显著差异的RIL群体进行全基因组关联分析,定位到1个主要位点qFC13-1与花色显著关联,该位点与W1位点重合;定位到12个位点与种皮色显著相关,其中9个位点为3种以上方法共定位,3个位点为2种方法共定位,包括4个已知位点和8个新位点。研究结果表明,构建的NAM群体适于进行大豆相关性状遗传分析,为大豆复杂性状的遗传解析和育种实践提供了良好的基础材料。 展开更多
