BACKGROUND Despite significant advancements in the medical treatment of primary hepato-cellular carcinoma(PHC)in recent years,enhancing therapeutic effects and im-proving prognosis remain substantial challenges worldw...BACKGROUND Despite significant advancements in the medical treatment of primary hepato-cellular carcinoma(PHC)in recent years,enhancing therapeutic effects and im-proving prognosis remain substantial challenges worldwide.AIM To investigate the expression levels of serum vascular endothelial growth factor(VEGF)and interleukin(IL)-17 in patients with PHC and evaluate their diagnostic value while exploring their relationship with patients’clinical characteristics.METHODS The study included 50 patients with confirmed PHC who visited Wuhan Han-yang Hospital from January 2021 to January 2022,and 50 healthy individuals from the same period served as the control group.Serum VEGF and IL-17 levels in both groups were measured by Enzyme-Linked Immunosorbent Assay,and their diagnostic value was assessed using receiver operating characteristic(ROC)curves.Pearson correlation analysis was performed to examine the relationship between serum VEGF and IL-17 levels.Pathological data of the PHC patients were analyzed to determine the relationship between serum VEGF and IL-17 levels and pathological characteristics.RESULTS Serum VEGF and IL-17 levels were significantly higher in the study group com-pared to the control group(P<0.05).No significant association was observed between serum VEGF and IL-17 levels and gender,age,combined cirrhosis,tumor diameter,or degree of differentiation(P>0.05).However,there was a significant relationship between clinical TNM stage,tumor metastasis,and serum VEGF and IL-17 levels(P<0.05).Correlation analysis revealed a positive correlation between serum VEGF and IL-17(P<0.05).ROC analysis demonstrated that both serum VEGF and IL-17 had good diagnostic efficacy for PHC.CONCLUSION Serum VEGF and IL-17 levels were significantly higher in PHC patients compared to healthy individuals.Their levels were closely related to pathological features such as tumor metastasis and clinical TNM stage,and there was a significant positive correlation between VEGF and IL-17.These biomarkers may serve as valuable reference in-dicators for the early diagnosis and treatment guidance of PHC.展开更多

METHODS: Eighty-eight patients with exudative AMD and 45 sex-and age-matched healthy controls were enrolled in this study conducted in China. Serum samples was obtained from the patients with exudative AMD and from the controls. Serum sCD146 and VEGFR2 protein levels were measured using an enzyme-linked immunosorbent assay.RESULTS: We found that serum sCD146 and VEGFR2 protein levels were significantly higher in the patients with exudative AMD group than in the controls(t=3.859, P<0.001 and t=3.829, P<0.001, respectively). Serum sCD146 levels were significantly higher in patients with classic choroidal neovascularization(CNV) than in those with occult CNV(t=9.899, P<0.001). There was a significant difference in the trend for exudative AMD in the highest versus lowest quartile of circulating sCD146 levels(χ2=10.29, P=0.001). The receiver operating characteristic curve analysis showed that the area under the curve was 0.696 for s CD146(95%CI: 0.601-0.791) with an optimum diagnostic cut-off value of 157.16 ng/mL, a sensitivity of 55.7%, and a specificity of 82.2%.CONCLUSION: The serum sCD146 level increases and may be a biomarker for exudative AMD.展开更多

Methods: Enzyme linked immunosorbent assay (ELISA) was used to determine the serum VEGF concentration in 40 patients with advanced cancer [non-small cell rung cancer (NSCLC), esophageal cancer (EC) and nasopharyngeal carcinoma (NPC)] before and after chemotherapy and 10 healthy volunteers as control group. Results: The serum VEGF concentrations in 40 cases of advanced cancer patients were significantly higher than those of 10 healthy control cases [(477.07 ± 374.10 ) pg/mL vs (139.09 ± 133.41 ) pg/mL; P = 0.016]. The serum VEGF concentrations in patients with NSCLC, EC and NPC were (518.53 _± 378.99) pg/mL, (399.21 ± 393.69) pg/mL and (500.68 ± 348.48) pg/mL, respectively. The differences were all statistically significant as compared with healthy control group (P values were 0.011,0.044 and 0.019, respectively). The serum VEGF concentrations of the patients in response to chemotherapy was significantly lower than those of the same patients before they undergoing chemotherapy [(400.41 ± 332.84) pg/mL vs (777.10 ± 666.01) pg/mL; P = 0.034]. VEGF levels were examined for each serum sample using the Quantikine Human VEGF-enzyme-linked immuno-absorbent assay(ELISA),whereas tryptase serum concentrations were assessed for each serum sample by means of fluoro-enzyme immunoassay(FEIA) using the Uni-CAP100 tool.Differences between serum VEGF and tryptase values before and after TACE were evaluated using Student t test. Person's correlation was used to assess the degree of association between the two variables.RESULTS: VEGF levels and serum tryptase in HCCpatients before TACE had a mean value and standard deviation(SD) of 114.31 ± 79.58 pg/mL and 8.13± 3.61 μg/L, respectively. The mean levels and SD of VEGF levels and serum tryptase in HCC patients after TACE were 238.14 ± 109.41 pg/mL and 4.02 ±3.03 μg/L. The changes between the mean values of concentration of VEGF and tryptase before treatment and after treatment was statistically significant(P <0.000231 and P < 0.00124, by Wilcoxon-Mann-Whitney respectively). A significant correlation between VEGF levels before and after TACE and between tryptase levels before and after TACE was demonstrated(r =0.68, P = 0.003; r = 0.84, P = 0.000 respectively).CONCLUSION: Our pilot results suggest that the higher serum VEGF levels and the lower tryptase levels following TACE may be potential biomarkers changing in response to therapy.展开更多

The changes of ICAM-1, VEGF and nitric oxide (NO), von Willebrand factor (vWF), endothelin-1 (ET-1) and thrombomodulin (TM) and other vascular endothelial function indexes in four groups were compared.Results:NDR group, BDR group, PDR group, ICAM-1, VEGF, NO and vWF (except NDR group), four ET-1 TM and vascular endothelial function indexes were significantly higher than the control group, while the difference between NDR vWF group was not statistical significant;differences in ICAM-1, VEGF and NO, vWF, ET-1 and TM four vascular endothelial function index among NDR group, BDR group and PDR group were statistically significant. Compared with the two groups, four vascular endothelial function indexes of ICAM-1, VEGF and NO, vWF, ET-1 and TM in retinopathy group were significantly higher than those in control group. Conclusions:There is correlation between VEGF, ICAM-1 level and microvascular injury in senile diabetic retinopathy patients. The late progression of the disease is, the higher VEGF, ICAM-1, NO, vWF, ET-1 and TM of vascular endothelial damage indexes are.It can reflect the microcirculation the extent of injury and blood rheology, and change of coagulation mechanism. 目的探讨子痫前期孕妇血清内分泌腺源性血管内皮生长因子(EG-VEGF)、中性粒细胞淋巴细胞比值(NLR)、血小板淋巴细胞比值(PLR)表达与病情严重程度的相关性。方法回顾性分析2021年10月~2023年3月在承德市中心医院妇儿院区产检的90例孕妇...目的探讨子痫前期孕妇血清内分泌腺源性血管内皮生长因子(EG-VEGF)、中性粒细胞淋巴细胞比值(NLR)、血小板淋巴细胞比值(PLR)表达与病情严重程度的相关性。方法回顾性分析2021年10月~2023年3月在承德市中心医院妇儿院区产检的90例孕妇临床资料,根据是否患有子痫前期进行分组,将健康正常的30例孕妇纳入健康对照组,将确诊为子痫前期的60例孕妇纳入观察组,且观察组根据子痫前期严重程度分为轻度组(n=30)和重度组(n=30),比较健康对照组和观察组、轻度组和重度组的血清EG-VEGF、NLR、PLR表达水平,并应用ROC曲线分析血清EG-VEGF、NLR、PLR诊断子痫前期和评价病情严重程度的效能。结果观察组的血清EG-VEGF、NLR高于健康对照组[(53.65±17.46)pg ml vs.(28.93±9.37)pg ml、(3.15±1.02)vs.(2.12±0.67)],PLR低于健康对照组[(98.53±21.86)vs.(115.31±18.36)],差异有统计学意义(t=7.237、5.010、3.613,P<0.05)。经ROC曲线分析发现,血清EG-VEGF、NLR、PLR诊断子痫前期效能良好(P<0.05)。重度组的血清EG-VEGF、NLR高于轻度组[(57.93±13.19)pg ml vs.(49.63±15.63)pg ml、(3.40±0.95)vs.(2.93±0.71)],PLR低于轻度组[(163.72±35.41)vs.(146.70±28.63)],差异有统计学意义(t=2.223、2.171、2.047,P<0.05)。经ROC曲线分析,发现血清EG-VEGF、NLR、PLR评价子痫前期病情严重程度的效能良好(P<0.05)。结论子痫前期孕妇血清EG-VEGF、NLR呈高表达水平,PLR呈低水平表达,且病情越严重的孕妇血清EG-VEGF、NLR越高,PLR越低,且经ROC分析血清EG-VEGF、NLR、PLR在子痫前期和病情严重程度诊断评价方面效能良好。展开更多

METHODS:The tumor tissues of 115 patients with gastric cancer were immunohistochemically evaluated using monoclonal antibodies against NF-kB RelA. Preoperative serum levels of vascular endothelial growth factor (VEGF), interleukin-6 (IL-6) were assessed via enzyme-linked immuno-sorbent assay. C-reactive protein (CRP) and serum amyloid A (SAA) were measured via immunotrubidimetry. RESULTS:Positive rate of NF-kB RelA was 42.6%. NF-kB RelA expression in tumor tissues was also related to serum levels of IL-6 (P = 0.044) and CRP (P = 0.010). IL-6, SAA, CRP were related to depth of invasion, VEGF and SAA were correlated with lymph node metastasis. IL-6, VEGF, SAA and CRP were related to the stage. Univariate analysis demonstrated that immunostaining of NF-kB RelA, levels of IL-6, VEGF, SAA were significantly related with both disease free survival and over-all survival (OS). Multivariate analysis verified that NF-kB RelA [hazard ratio (HR): 3.40, P = 0.024] and SAA (HR: 3.39, P = 0.045) were independently associated with OS. CONCLUSION: Increased expression of NF-kB RelA and high levels of serum SAA were associated with poor OS in gastric cancer patients.展开更多

文摘目的探讨子痫前期孕妇血清内分泌腺源性血管内皮生长因子(EG-VEGF)、中性粒细胞淋巴细胞比值(NLR)、血小板淋巴细胞比值(PLR)表达与病情严重程度的相关性。方法回顾性分析2021年10月~2023年3月在承德市中心医院妇儿院区产检的90例孕妇临床资料,根据是否患有子痫前期进行分组,将健康正常的30例孕妇纳入健康对照组,将确诊为子痫前期的60例孕妇纳入观察组,且观察组根据子痫前期严重程度分为轻度组(n=30)和重度组(n=30),比较健康对照组和观察组、轻度组和重度组的血清EG-VEGF、NLR、PLR表达水平,并应用ROC曲线分析血清EG-VEGF、NLR、PLR诊断子痫前期和评价病情严重程度的效能。结果观察组的血清EG-VEGF、NLR高于健康对照组[(53.65±17.46)pg ml vs.(28.93±9.37)pg ml、(3.15±1.02)vs.(2.12±0.67)],PLR低于健康对照组[(98.53±21.86)vs.(115.31±18.36)],差异有统计学意义(t=7.237、5.010、3.613,P<0.05)。经ROC曲线分析发现,血清EG-VEGF、NLR、PLR诊断子痫前期效能良好(P<0.05)。重度组的血清EG-VEGF、NLR高于轻度组[(57.93±13.19)pg ml vs.(49.63±15.63)pg ml、(3.40±0.95)vs.(2.93±0.71)],PLR低于轻度组[(163.72±35.41)vs.(146.70±28.63)],差异有统计学意义(t=2.223、2.171、2.047,P<0.05)。经ROC曲线分析,发现血清EG-VEGF、NLR、PLR评价子痫前期病情严重程度的效能良好(P<0.05)。结论子痫前期孕妇血清EG-VEGF、NLR呈高表达水平,PLR呈低水平表达,且病情越严重的孕妇血清EG-VEGF、NLR越高,PLR越低,且经ROC分析血清EG-VEGF、NLR、PLR在子痫前期和病情严重程度诊断评价方面效能良好。

文摘AIM:To assess the prognostic significance of nuclear factor-kB (NF-kB) and its target genes in gastric cancer. METHODS:The tumor tissues of 115 patients with gastric cancer were immunohistochemically evaluated using monoclonal antibodies against NF-kB RelA. Preoperative serum levels of vascular endothelial growth factor (VEGF), interleukin-6 (IL-6) were assessed via enzyme-linked immuno-sorbent assay. C-reactive protein (CRP) and serum amyloid A (SAA) were measured via immunotrubidimetry. RESULTS:Positive rate of NF-kB RelA was 42.6%. NF-kB RelA expression in tumor tissues was also related to serum levels of IL-6 (P = 0.044) and CRP (P = 0.010). IL-6, SAA, CRP were related to depth of invasion, VEGF and SAA were correlated with lymph node metastasis. IL-6, VEGF, SAA and CRP were related to the stage. Univariate analysis demonstrated that immunostaining of NF-kB RelA, levels of IL-6, VEGF, SAA were significantly related with both disease free survival and over-all survival (OS). Multivariate analysis verified that NF-kB RelA [hazard ratio (HR): 3.40, P = 0.024] and SAA (HR: 3.39, P = 0.045) were independently associated with OS. CONCLUSION: Increased expression of NF-kB RelA and high levels of serum SAA were associated with poor OS in gastric cancer patients.
