[Objectives]This study aimed to study the effects of Shenge Yifei capsule on the TGF-β1/Smad signaling pathway in rats with chronic obstructive pulmonary disease(COPD).[Methods]Ten rats were randomly selected as the ...[Objectives]This study aimed to study the effects of Shenge Yifei capsule on the TGF-β1/Smad signaling pathway in rats with chronic obstructive pulmonary disease(COPD).[Methods]Ten rats were randomly selected as the control group,and the other 40 rats were selected for modeling by fumigation combined with Klebsiella pneumoniae infection.A total of 38 rats were successfully modeled.They were randomly divided into model group(8 rats),low-dose Shenge Yifei capsule group(10 rats),high-dose Shenge Yifei capsule group(10 rats)and theophylline group(10 rats)in accordance with the principle of half male and half female.The rats in the model and control groups were given with distilled water by gavage,and the rats in the drug administration groups were given with corresponding drugs.The TGF-β1 level in the serum,and the expression levels of TGF-β1,Smad2,Smad3 and Smad7 and TGF-β1,Smad3 and Smad7 in airway tissues were detected.[Results]After 12 weeks,the serum TGF-β1 levels of the theophylline group and high-dose Shenge Yifei capsule group were lower than that of the low-dose Shenge Yifei capsule group(P<0.05).The expression levels of TGF-β1 and Smad3 in the theophylline group and high-dose Shenge Yifei capsule group were lower than that in the low-dose Shenge Yifei capsule group(P<0.05).The expression levels of TGF-β1 and Smad3 in the high-dose Shenge Yifei capsule group were lower than those in the low-dose Shenge Yifei capsule group and theophylline group(P<0.05).The expression levels of Smad7 and the proteins in the model group were lower than those in the other groups(P<0.05).The expression levels of Smad7 in the theophylline group and high-dose Shenge Yifei capsule group were higher than that in the low-dose Shenge Yifei capsule group(P<0.05).After 18 weeks,no significant difference was found in serum TGF-β1 level among the theophylline group and low and high-dose Shenge Yifei capsule groups(P>0.05).The expression levels of Smad7 and the proteins in the model group were lower than those in the other groups.The expression level of Smad7 in the high-dose Shenge Yifei capsule group was lower than that in the theophylline group(P<0.05).[Conclusions]Shenge Yifei capsule can regulate the TGF-β1/Smads signaling pathway.They can down-regulate the expression of TGF-β1,Smad2 and Smad3 and up-regulate the expression of Smad7,reducing the degree of airway modeling,delaying the development of COPD disease.Conventional high-dose Shenge Yifei capsule is more effective in inhibiting the expression of Smad2.展开更多

Effects of SGYF on body weight and lung index were evaluated. For histological evaluation of lungs injury and fibrosis, lung specimens were stained with hematoxylin and eosin(HE) and Masson trichrome.Body weight and lung index from various groups were measured, and the concentrations of monoamine(MAO) and transforming growth factor-β1(TGF-β1) were also detected in lung homogenates.Results: SGYF attenuated the bleomycin-induced weight loss and the lung index. Histologica levidence showed the ability of SGYF to decrease bleomycin-induced lung fibrosis and consolidation. SGYF reduced MAO and TGF-β1 activity in lung tissues.Conclusions: These findings suggest that SGYF may be a promising candidate to prevent bleomycininduced lung damage or other interstitial lesions.展开更多

METHODS: Cardiac hypertrophy was modeled in H9c2 cells treated with isoproterenol(ISO) to assess the effects of Shenge San( 参 蛤 散, SGS) on cell viability and mitochondrial membrane potential. We assessed mitochondrial complex m RNA levels and mitochondrial oxidative phosphorylation factor m RNA and protein levels. RESULTS: Compared with the 100 μM ISO group, cell size was significantly decreased in the 0.3 mg/m L SGS and 20 μM ZLN005(PGC-1α activator) groups(P < 0.01). Compared with the SGS(0.3) +ISO group, we observed lower phosphorylated adenosine monophosphateactivated kinase(AMPK) protein levels in the ISO and ZLN005+SGS+ISO groups(P < 0.01). Compared with the compound C group, SGS significantly increased PGC-1α expression in ISO-induced cardiac hypertrophy cells(P < 0.01), and this was inhibited by compound C pretreatment(P < 0.05). Compared with the ISO group, the mitochondrial red-green fluorescence ratio increased in the 0.3 mg/m L SGS group(P < 0.05). m RNA levels of cytochrome c oxidase subunit 1(CO1) in the ISO and compound C groups were lower than those in control group(P < 0.01), and the m RNA levels of CO1 and ATP8 were significantly lower in the ISO and compound C groups versus control(P < 0.01). Compared with the SGS(0.3) +ISO group, ATP synthetase subunit 8(ATP8) m RNA was significantly decreased in the ISO group(P < 0.01) and compound C+SGS+ISO group(P < 0.05). Compared with the SGS(0.3) +ISO group, NRF1 m RNA levels were significantly decreased(P < 0.05) in the ISO and compound C+SGS+ISO groups. CONCLUSIONS: SGS can attenuate ISO-induced cardiomyocyte hypertrophy, restore the decrease in mitochondrial membrane potential, and upregulate PGC-1α/NRF1 levels. Notably, these effects can be blocked by AMPK inhibitor-compound C.展开更多

Objectives:To discuss the mechanism of Sheng Yang San Huo decoction on diabetic peripheral neuropathy using the network pharmacology method.Methods:The BATMAN-TCM database,TCM-ID database,Chinese Natural Product Chemi...Objectives:To discuss the mechanism of Sheng Yang San Huo decoction on diabetic peripheral neuropathy using the network pharmacology method.Methods:The BATMAN-TCM database,TCM-ID database,Chinese Natural Product Chemical Composition Database,and TCMIP database were employed to screen the chemical active ingredients of each herb in Sheng Yang San Huo decoction based on the“Libinsky Drug Rules”.SwissTargetPrediction was used to screen effective action targets for each herb in the prescription.Additionally,Cytoscape 3.7.0 was utilized to construct a“drug-target”network.GeneCards,OMIM,and MaLaCards databases were utilized to gather targets related to diabetic peripheral neuropathy.VENNY 2.1 online platform was employed to match drug and disease targets,draw a Venn diagram,and construct a“drug-active compounds-common target”network using Cytoscape 3.7.0.gene ontology biological process analysis and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis for the targets were conducted using the DAVID 6.8 database.Enrichment analysis results were visualized using the OmicShareTool online platform.Molecular docking was performed using CB-Dock2.Results:Following screening,a total of 217 active compounds and 132 potential targets were identified in Sheng Yang San Huo decoction.The effects are primarily enriched in pathways such as Lipid and Atherosclerosis,AGE-RAGE signaling pathway in diabetic complications,and the IL-17 signaling pathway.The binding energy of the key active ingredients to the core protein targets of DPN was favorable.Conclusion:The study reveals the characteristics of multiple targets and pathways of Sheng Yang San Huo decoction,providing new insights for the clinical application of this prescription.展开更多
