Low-molecular-weight(LMW)compounds are ubiquitous in living organisms and play essential roles in biological processes.The direct analysis of LMW compounds in biological tissues by matrix-assisted laser desorption/ionization mass spectrometry imaging(MALDI-MSI)could provide a more comprehensive understanding of their essential functions.Here,we evaluated 4-nitrocatechol(4-NC)as a novel positive-ion matrix for enhancing in situ detection and imaging of LMW compounds from the rat liver,brain,and germinating Chinese-yew seed by MALDI-MS.Our results showed that the 4-NC possessed remarkable features,including strong ultraviolet absorption,uniform matrix crystal,excellent chemical stability,and fewer matrix-related background peaks.The use of 4-NC led to the successful detection of 232,218,and193 LMW compounds from the three abovementioned tissue sections,respectively.Also,the use of 4-NC improved the imaging quality of LMW compounds in tissue sections through MALDI-MSI and has the potential as a matrix for MALDI tissue imaging of LMW compounds. 展开更多

The Solar Close Observations and Proximity Experiments(SCOPE)mission,which has been proposed by the Yunnan Observatories,Chinese Academy of Sciences,aiming to operate at a distance of 5 to 10 solar radii from the Sun,plans to complete the in situ detection of the solar eruption process and observation of the magnetic field structure response.The solar flux received by the satellite ranges from 10^(3) to 10^(6) Wm^(-2),which poses challenges for thermal management of the solar arrays.In this work,the solar array cooling system of the Parker Solar Probe is discussed,the developments of the fluid loop technique are reviewed,and a research plan for a next-generation solar array cooling system is proposed.This paper provides a valuable reference for novel thermal control systems in spacecraft for solar observation. 展开更多

A multifunctional Cu2O/Ag micro-nanocomposite, which has the characteristics of high cat- alytic activities under the visible light and high surface-enhanced Raman scattering (SERS) activity, was fabricated via a facile method and employed for the in situ SERS monitoring of the photocatalytic degradation reaction of crystal violet. Through the variation of the AgNO3 concentration, Ag content on the Cu2O template can be controllably tuned, which has great influence on the SERS effect. The results indicate that Ag nanopartieles form on the Cu2O nanoframes to obtain the Cu2O/Ag nanoeomposite, which can act as an excellent bifunetional platform for in situ monitoring of photocatalytic degradation of organic pollutions by SERS. 展开更多

Endogenous metabolites play key functions in many important physiological and biochemical processes.The comprehensive in situ detection and direct imaging of metabolites in bio-tissues by matrix-assisted laser desorption/ionization mass spectrometry imaging(MALDI-MSI)is very important for understanding complex and diverse biological processes and has become an essential aspect of spatial omics.In this work,4-aminoazobenzene(AAB)was successfully screened and optimized as a new negative ion(-)MALDI matrix to enhance the in situ detection and imaging of metabolites in tissues using MALDIMSI.Obviously,AAB exhibited superior properties in terms of ultraviolet absorption,background ion interference,matrix morphology,and metabolite ionization efficiency.AAB was used for in situ detection and imaging of metabolites in rat brain and germinating Chinese yew seed tissue sections,where 264and 339 metabolite ion signals were successfully detected and imaged using(-)MALDI-MS,respectively.In addition,high-resolution imaging of mouse eyeball section using MALDI-tims TOF MSI with spatial resolution of up to 10μm was successfully carried out,showing that AAB is an efficient(-)MALDI matrix for capturing high-resolution images of metabolites in biological tissue sections. 展开更多

Nucleic acid, as a type of tumor biomarker, contains vital genetic information, which is of great significance for the occurrence and development of cancer. Currently, living cell nucleic acid probes, which enable the in situ imaging and dynamic monitoring of nucleic acids, have become a rapidly developing field. This review focuses on living cell nucleic acid probes that can be used for the early diagnosis of tumors. We describe the fundamental design of the probe in terms of three units and focus on the roles of different nanomaterials in probe delivery. 展开更多

An in situ hybridization technique with 35S labelled proto-oncogene probes (c-myc & c-fes) was used to detect their expression in bone marrow cells of 22 cases of leukemia of various types and immature granulocytes and erythroblasts of 16 nomal myelograms as controls. Both c-myc and c-fes were detectable in leukemic cells as well as in immature granulocytes and erythroblasts of normal bone marrow, but the expression extent varied in different cases. The levels of c-myc expression in leukemic cells were higher than those in controls (P<0.001). There was no difference of c-fes expression in two groups of bone marrow cells (P>0.05). This technique provides us a new method in studying variations of proto-oncogene expression in leukemic cells. 展开更多

To monitor and investigate chemical reactions in real time and in situ is a long-standing,challenging goal in chemistry.Herein,an electric potential-promoted oxidative coupling reaction of organoboron compounds without the addition of base is reported,and the transmetallation process involved is monitored in real time and in situ with the scanning tunneling microscopy break single-molecule junctions(STMBJ)technique.We found that the electric potential applied determined the transmetallation.At low-bias voltage,the first-step transmetallation process occurred and afforded Au─C-bonded aryl gold intermediates.The electronic properties of organoboron compounds have a strong influence on the transmetallation process,and electron-rich compounds facilitate this transformation.At high-bias voltage,the second-step transmetallation process took place,and the corresponding intermediate(highly reactive diaryl metal complex)was detected with the assistance of Pd(OAc)_(2).Our work demonstrates the applications of STMBJ on in situ monitoring and catalyzing of chemical reactions and provides a new methodology to fabricate singlemolecule devices. 展开更多

Cytomegalovirus (CMV) genes were detected by in situ hybridization in 25 Chinese patients with viral myocarditis (VMC). The positive hybridization signals werre found in cardiomyocytes (6 cases, 24%), capillary endothelial cells (4 cases, 16%) and interstitial cells (7 cases, 28%). The difference between VMC and control group (16 cases died of brain trauma and 10 cases of congenital heart diseases was statistically significant. There was no definite pathomorphological relationship between the detection of CMV genes and myocardial lesions. The results suggest that CMV infection may be one of the causes of myocarditis and chronic stimulation of the immune system induced by CMV may be a possible pathogenesis of this disease. 展开更多

With ultrafast laser systems reaching presently 10 PW peak power or operating at high repetition rates,research towards ensuring the long-term,trouble-free performance of all laser-exposed optical components is critical.Our work is focused on providing insight into the optical material behavior at fluences below the standardized laser-induced damage threshold(LIDT)value by implementing a simultaneous dual analysis of surface emitted particles using a Langmuir probe(LP)and the target current(TC).HfO_(2) and ZrO_(2) thin films deposited on fused silica substrates by pulsed laser deposition at various O_(2) pressures for defect and stoichiometry control were irradiated by Gaussian,ultrashort laser pulses(800 nm,10 Hz,70 fs)in a wide range of fluences.Both TC and LP collected signals were in good agreement with the existing theoretical description of laser–matter interaction at an ultrashort time scale.Our approach for an in situ LIDT monitoring system provides measurable signals for below-threshold irradiation conditions that indicate the endurance limit of the optical surfaces in the single-shot energy scanning mode.The LIDT value extracted from the LP-TC system is in line with the multipulse statistical analysis done with ISO 21254-2:2011(E).The implementation of the LP and TC as on-shot diagnostic tools for optical components will have a significant impact on the reliability of next-generation ultrafast and high-power laser systems. 展开更多
