Bidens pilosa is recognized as one of the major invasive plants in China.Its invasion has been associated with significant losses in agriculture,forestry,husbandry,and biodiversity.Soil ecosystems play an important role in alien plant invasion.Microorganisms within the soil act as intermediaries between plants and soil ecological functions,playing a role in regulating soil enzyme activities and nutrient dynamics.Understanding the interactions between invasive plants,soil microorganisms,and soil ecological processes is vital for managing and mitigating the impacts of invasive species on the environment.In this study,we conducted a systematic analysis focusing on B.pilosa and Setaria viridis,a common native companion plant in the invaded area.To simulate the invasion process of B.pilosa,we constructed homogeneous plots consisting of B.pilosa and S.viridis grown separately as monocultures,as well as in mixtures.The rhizosphere and bulk soils were collected from the alien plant B.pilosa and the native plant S.viridis.In order to focus on the soil ecological functional mechanisms that contribute to the successful invasion of B.pilosa,we analyzed the effects of B.pilosa on the composition of soil microbial communities and soil ecological functions.The results showed that the biomass of B.pilosa increased by 27.51% and that of S.viridis was significantly reduced by 66.56%.The organic matter contents in the bulk and rhizosphere soils of B.pilosa were approximately 1.30 times those in the native plant soils.The TN and NO_(3)^(-)contents in the rhizosphere soil of B.pilosa were 1.30 to 2.71 times those in the native plant soils.The activities of acid phosphatase,alkaline phosphatase,and urease in the rhizosphere soil of B.pilosa were 1.98-2.25 times higher than in the native plant soils.Using high-throughput sequencing of the16S rRNA gene,we found that B.pilosa altered the composition of the soil microbial community.Specifically,many genera in Actinobacteria and Proteobacteria were enriched in B.pilosa soils.Further correlation analyses verified that these genera had significantly positive relationships with soil nutrients and enzyme activities.Plant biomass,soil p H,and the contents of organic matter,TN,NO_(3)^(-),TP,AP,TK,and AK were the main factors affecting soil microbial communities.This study showed that the invasion of B.pilosa led to significant alterations in the composition of the soil microbial communities.These changes were closely linked to modifications in plant traits as well as soil physical and chemical properties.Some microbial species related to C,N and P cycling were enriched in the soil invaded by B.pilosa.These findings provide additional support for the hypothesis of soil-microbe feedback in the successful invasion of alien plants.They also offer insights into the ecological mechanism by which soil microbes contribute to the successful invasion of B.pilosa.Overall,our research contributes to a better understanding of the complex interactions between invasive plants,soil microbial communities,and ecosystem dynamics. 展开更多

Afforestation has an important role in biodiversity conservation and ecosystem function improvement.A meta-analysis was carried out in China,which has the largest plantation area globally,to quantify the effects of plantings on soil microbial diversity.The results showed that the overall effect of afforestation on soil microbial diversity was positive across the country.Random forest algorithm suggested that soil carbon was the most important factor regulating microbial diversity and the positive response was only found with new plantings on low-carbon bare lands but not on high-carbon farmlands and grasslands.In addition,afforestation with broadleaved species increased microbial diversity,whereas planting with conifers had no effect on microbial diversity.This study clarified the effects of plantings on soil microbial diversity,which has an important implication for establishing appropriate policies and practices to improve the multiple functionalities(e.g.,biodiversity conservation and climate change mitigation)during plantation establishment. 展开更多

In the Loess Plateau of China,land-use pattern is a major factor in controlling underlying biological processes.Additionally,the process of land-use pattern was accompanied by abandoned lands,potentially impacting soil microbe.However,limited researches were conducted to study the impacts of land-use patterns on the diversity and community of soil microorganisms in this area.The study aimed to investigate soil microbial community diversity and composition using high-throughput deoxyribonucleic acid(DNA)sequencing under different land-use patterns(apricot tree land,apple tree land,peach tree land,corn land,and abandoned land).The results showed a substantial difference(P<0.050)in bacterial alpha-diversity and beta-diversity between abandoned land and other land-use patterns,with the exception of Shannon index.While fungal beta-diversity was not considerably impacted by land-use patterns,fungal alpha-diversity indices varied significantly.The relative abundance of Actinobacteriota(34.90%),Proteobacteria(20.65%),and Ascomycota(77.42%)varied in soils with different land-use patterns.Soil pH exerted a dominant impact on the soil bacterial communities'composition,whereas soil available phosphorus was the main factor shaping the soil fungal communities'composition.These findings suggest that variations in land-use pattern had resulted in changes to soil properties,subsequently impacting diversity and structure of microbial community in the Loess Plateau.Given the strong interdependence between soil and its microbiota,it is imperative to reclaim abandoned lands to maintain soil fertility and sustain its function,which will have significant ecological service implications,particularly with regards to soil conservation in ecologically vulnerable areas. 展开更多

[Objective] This study was conducted to expound the fertility improvement effect in continuous-cropping sugarcane field and provide reference for establishment of rational sugarcane fertilization system and improvement of soil quality in continuous-cropping sugarcane field. [Method] The soil in the experimental region is latosolic red soil which was planted with sugarcane for 11 years continuously, and 8 treatments including sole application of chemical fertilizers, sole application of organ- ic fertilizer, and combined application of organic fertilizer and chemical fertilizers were designed according to different fertilization measures. The effects of different fertilization treatments on soil microbial biomass, soil enzyme activities and related fertility factors were determined. [Result} Different fertilization treatments all showed soil microbial biomass N, C and P and activities of soil acid phosphatase, catalase, sucrase and urease higher than the CK. Soil microbial biomass N increased by 5.56%-67.13%, soil microbial biomass C increased by 4.01%-20.40%, and soil mi- crobial biomass P increased by 6.39%-67.02%. The activity of acid phosphatase was improved by 12.96%-35.19%, the activity of catalase was improved by 18.24% -78.93%, the activity of sucrase was improved by 3.00%-42.00%, and the activity of urease was improved by 1.21%-23.43%. However, the soil nutrients of different fertilization treatments increased non-significantly (P〉0.05). Soil microbial biomass N, C and P and activities of acid phosphatase, catalase and urease were in significant (P〈0.05) or very significant correlation (P〈0.01) with contents of soil rapidly available P, rapidly available K and total N. [Conclusion] The evaluation of improvement of soil fertility in continuous-cropping sugarcane field using soil microbial biomass and enzyme activities as indexes is more comprehensive and sensitive. 展开更多

An investigation on soil organic carbon, total N and P, NO3-N, available P, microbial biomass C, N and P, basal respiration and metabolic quotients (qCO2) was conducted to compare differences in soil microbial properties and nutrients between 15-year-old pure Chinese fir (Cunninghamia lanceolata) and two mixed Chinese fir plantations (mixed plantations with Alnus crernastogyne, mixed plantations with Kalopanax septemlobus) at Huitong Experimental Station of Forest Ecology (26°45′N latitude and 109°30′E longitude), Chinese Academy of Sciences in May, 2005. Results showed that the concentrations of soil organic carbon, total N, NO3^--N, total P and available P in mixed plantations were higher than that in pure plantation. Soil microbial biomass N in two mixed plantations was averagely higher 69% and 61% than that in pure plantation at the 0-10 cm and 10-20 cm soil depth, respectively. Soil microbial biomass C, P and basal respiration in mixed plantations were higher 11%, 14% and 4% at the 0-10 cm soil depth and 6%, 3% and 3% at the 10-20 cm soil depth compared with pure plantation. However, soil microbial C: N ratio and qCO2 were averagely lower 34% and 4% in mixed plantations than pure plantation. Additionally, there was a closer relation between soil microbial biomass and soil nutrients than between basal respiration, microbial C: N ratio and qCO2 and soil nutrients. In conclusion, introduction of broad-leaved tree species into pure coniferous plantation improved soil microbial properties and soil fertility, and can be helpful to restore degraded forest soil. 展开更多

As one of the main methods of microbial community functional diversity measurement, biolog method was favored by many researchers for its simple oper- ation, high sensitivity, strong resolution and rich data. But the preprocessing meth- ods reported in the literatures were not the same. In order to screen the best pre- processing method, this paper took three typical treatments to explore the effect of different preprocessing methods on soil microbial community functional diversity. The results showed that, method B's overall trend of AWCD values was better than A and C's. Method B's microbial utilization of six carbon sources was higher, and the result was relatively stable. The Simpson index, Shannon richness index and Car- bon source utilization richness index of the two treatments were B〉C〉A, while the Mclntosh index and Shannon evenness were not very stable, but the difference of variance analysis was not significant, and the method B was always with a smallest variance. Method B's principal component analysis was better than A and C's. In a word, the method using 250 r/min shaking for 30 minutes and cultivating at 28 ℃ was the best one, because it was simple, convenient, and with good repeatability. 展开更多

To investigate the effects of different vegetable growing regions and planting modes on soil quality,soils in high,medium and low altitude areas of Guizhou were respectively sampled under different vegetable efficient planting modes,and the variations of soil microbial flora and enzyme activities were analyzed. The soil microbial count and total bacteria of the vegetable efficient cultivation mode were significantly higher than that of the control (traditional planting mode) in each planting area,and the microbial diversity index was also improved to varying de- grees.The soil phosphatase,catalase and urease activities of the vegetable efficient planting mode were higher than that of the control.The soil catalase and urease activities were higher than that of the control by 1.37-1.44 and 1.51-2.80 times. Application of vegetable efficient planting mode in different regions will help to im- prove the soil quality in a given period. 展开更多

Impact of Vanadium on Enzyme Activity and Microbial Biomass in Soils

[Objective] This study aimed to investigate the impact of vanadium at dif- ferent concentration on enzyme activity and microbial biomass in soils. [Method] Us- ing pot experiments in the growth cabinet, we would like to investigate the changes of the soil enzyme activity and microbial biomass at different growing stages of rape (Brassica juncea L.) at different soil vanadium concentrations (soil background value was 147 mg/kg, spiked with 0, 50, 100, 150, 250 and 500 mg/kg of exogenous vanadium). [Result] Among all enzymes examined, polyphenol oxidase was most sensitive to soil vanadium. Addition of 50 mg/kg vanadium decreased its activity up to 56% of the control probably due to the vanadium toxicity. In comparison, the ac- tivities of sucrase, urease and catalase was less affected by soil vanadium. Surpris- ingly, the activity of sucrase, urease and catalase at the rape seedling stage differed significantly from at the maturity stage, highlighting the potential impact of plant growth on the vanadium-soil enzyme interaction. Different soil vanadium concentra- tions led to increases of microbial biomass to different extents. However, the corre- lation between soil microbial biomass carbon and phosphorus with vanadium con- centrations was insignificant. This revealed that the presence of additional factors (eg. plant) affected soil microbial biomass carbon and phosphorus aside from soil vanadium. [Conclusion] Polyphenol oxidase may be considered as an indicator of soil vanadium contamination. Due to the highly complicated interaction between vanadium and soil biological activities during plant growth, more investigations are required to reveal the mechanisms beyond our findings here. 展开更多

Using a scheme of agricultural fields with progressively less intensive management (deintensification), different manage- ment practices in six agroecosystems located near Goldsboro, NC, USA were tested in a large-scale experiment, including two cash-grain cropping systems employing either tillage (CT) or no-tillage (NT), an organic farming system (OR), an integrated cropping system with animals (IN), a successional field (SU), and a plantation woodlot (WO). Microbial phos- pholipid fatty acid (PLFA) profiles and substrate utilization patterns (BIOLOG ECO plates) were measured to examine the effects of deintensification on the structure and diversity of soil microbial communities. Principle component analyses of PLFA and BIOLOG data showed that the microbial community structure diverged among the soils of the six systems. Lower microbial diversity was found in lowly managed ecosystem than that in intensive and moderately managed agro- ecosystems, and both fungal contribution to the total identified PLFAs and the ratio of microbial biomass C/N increased along with agricultural deintensification. Significantly higher ratios of C/N (P < 0.05) were found in the WO and SU systems, and for fungal/bacterial PLFAs in the WO system (P < 0.05). There were also significant decreases (P < 0.05) along with agricultural deintensification for contributions of total bacterial and gram positive (G+) bacterial PLFAs. Agricultural deintensification could facilitate the development of microbial communities that favor soil fungi over bacteria. 展开更多

Soil health is important for the sustainable development of terrestrial ecosystem. In this paper, we studied the relationship between soil quality and soil microbial properties such as soil microbial biomass and soil enzyme activities in order to illustrate the function of soil microbial properties as bio-indicators of soil health. In this study, microbial biomass C and N contents (Cmic & Nmic), soil enzyme activities, and soil fertility with different fertilizer regimes were carried out based on a 15-year long-term fertilizer experiment in Drab Fluvo-aquic soil in Changping County, Beijing, China. At this site, 7 different treatments were established in 1991. They were in a wheat-maize rotation receiving either no fertilizer (CK), mineral fertilizers (NPK), mineral fertilizers with wheat straw incorporated (NPKW), mineral fertilizers with incremental wheat straw incorporated (NPKW+), mineral fertilizers plus swine manure (NPKM), mineral fertilizers plus incremental swine manure (NPKM+) or mineral fertilizers with maize straw incorporated (NPKS). In different fertilization treatments Cmic changed from 96.49 to 500.12 mg kg^-1, and Nmic changed from 35.89 to 101.82 mg kg^-1. Compared with CK, the other treatments increased Cmic & Nmic, Cmic/Corg (organic C) ratios, Cmic/Nmic, urease activity, soil organic matter (SOM), soil total nitrogen (STN), and soil total phosphorus (STP). All these properties in treatment with fertilizers input NPKM+ were the highest. Meantime, long-term combined application of mineral fertilizers with organic manure or crop straw could significantly decrease the soil pH in Fluvo-aquic soil (the pH around 8.00 in this experimental soil). Some of soil microbial properties (Cmic/Nmic, urease activity) were positively correlated with soil nutrients. Cmic/Nmic was significantly correlated with SOM and STN contents. The correlation between catalase activity and soil nutrients was not significant. In addition, except of catalase activity, the soil pH in this experiment was negatively correlated with soil microbial properties. In conclusion, soil microbial properties reflect changes of soil quality and thus can be used as bio-indicators of soil health. 展开更多

The contents of the soil microbial biomass nitrogen (SMBN) in the soils sampled from the Loess Plateau of China were determined using chloroform fumigation aerobic incubation method (CFAIM), chloroform fumigation anaerobic incubation method (CFANIM) and chloroform fumigation-extraction method (CFEM).The N taken up by ryegrass on the soils was determined after a glasshouse pot experiment. The flushes of nitrogen (FN) of the soils obtained by the CFAIM and CFANIM were higher than that by the CFEM, and there were significantly positive correlations between the FN obtained by the 3 methods. The N extracted from the fumigated soils by the CFAIM, CFANIM and CFEM were significantly positively correlated with the N uptake by ryegrass. The FN obtained by the 3 methods was also closely positively correlated with the plant N uptake. The contributions of the SMBN and mineral N and mineralized N during the incubation period to plant N uptake were evaluated with the multiple regression method. The results showed that the N contained in the soil microbial biomass might play a noticeable role in the N supply of the soils to the plant. 展开更多

The microbial biomass, basal respiration and substrate utilization pattern in copper mining wasteland of red soil area, southern China, were investigated. The results indicated that soil microflora were obviously different compared with that of the non-mine soil. Microbial biomass and basal respiration were negatively affected by the elevated heavy metal levels. Two important microbial ecophysiological parameters, namely, the ratio of microbial biomass C( Cmic )/organic C( Corg ) and metabolic quotient(qCO2 ) were closely correlated to heavy metal stress. There was a significant decrease in the Cmic/Corg ratio and an increase in the metabolic quotient with increasing metal concentration. Multivariate analysis of Biolog data for sole carbon source utilization pattern demonstrated that heavy metal pollution had a significant impact on microbial community structure and functional diversity. All the results showed that soil microbiological parameters had great potential to become the early sensitive, effective and liable indicators of the stresses or perturbations in soils of mining ecosystems. 展开更多

The SMBC contents in the soils ranged from 75.9 to 301.0 μg Cg-1 with an average of 206.1 μg C g-1, accounting for 1.36%~6.24% of the total soil organic C with an average of 3.07%, and the SMBN contents from 0.51 to 68.40 μg N g-1 with an average of 29.4 μg N g-1, accounting for 0.20%~5.65% of the total N in the soils with an average of 3.36%. A close relationship was found between SMBC and SMBN, and they both were positively correlated with total organic C, total N, NaOH hydrolizable N and mineralizable N. These results confirmed that soil microbial biomass had a comparative role in nutrient cycles of soils. 展开更多

Greenhouse vegetable production has been characterized by high agricultural inputs, high temperatures, and high cropping indexes. As an intensive form of agriculture, nutrient cycling induced by microbial activities in the greenhouses is relatively different from open fields in the same region. However, the responses of soil microbial biomass carbon (MBC) and nitrogen (MBN), enzyme activities, microbial community composition, and yield to organic amendment are not well understood. Therefore, a 5-year greenhouse tomato (Solanum lycopersicum Mill.)-cucumber (Cucumis sativus L.) rotation experiment was conducted. The field experiment comprised 5 treatments: 4/4CN (CN, nitrogen in chemical fertilizer), 3/4CN+1/4MN (MN, nitrogen in pig manure), 2/4CN+2/4MN, 2/4CN+1/4 MN+1/4 SN (SN, nitrogen in corn straw) and 2/4CN+2/4SN. The amounts of nitrogen (N), phosphorus (P2O5), and potassium (K2O) were equal in the five treatments. Starting with the fourth growing season, the optimal yield was obtained from soil treated with straw. MBC, MBN, phospholipid fatty acid (PLFA) profiles, and enzyme activities were significantly changed by 5 years of substitution with organic amendments. Redundancy analysis showed that MBC accounts for 89.5 and 52.3% of the total enzyme activity and total community variability, respectively. The activities of phosphomonoesterase, N-acetyl-glucosaminidase, and urease, and the relative abundances of fungi, actinomycetes, and Gram-negative bacteria were significantly and positively related to vegetable yields. Considering the effects of organic amendments on soil microbial characteristics and vegetable yield, 2/4CN+1/4MN+1/4SN can improve soil quality and maintain sustainable high yield in greenhouse vegetable production. 展开更多

Different management practices in six agroecosystems located near Goldsboro, NC, USA were conducted including a successional field (SU), a plantation woodlot (WO), an integrated cropping system with animals (IN), an organic farming system (OR), and two cash-grain cropping systems employing either tillage (CT) or no-tillage (NT) to examine if and how microbial biomass and activity differ in response to alterations in disturbance intensity from six land management strategies. Results showed that soil microbial biomass and activity differed, with microbial activity in intermediately disturbed ecosystems (NT, OR, IN) being significantly higher (P < 0.01) than systems with either high or low disturbance intensities. There was also a significant and a highly significant ecosystem effect from the treatments on microbial biomass C (MBC) (P < 0.05) and on microbial activity (respiration) (P < 0.01), respectively. Multiple comparisons of mean respiration rates distinctly separated the six ecosystem types into three groups: CT < NT, SU and WO < OR and IN.Thus, for detecting microbial response to disturbance changes these results indicated that the active component of the soil microbial community was a better indicator than total biomass. 展开更多

Soil microbial biomass is an important indicator to measure the dynamic changes of soil carbon pool.It is of great signifi cance to understand the dynamics of soil microbial biomass in plantation for rational management and cultivation of plantation.In order to explore the temporal dynamics and infl uencing factors of soil microbial biomass of Keteleeria fortunei var.cyclolepis at diff erent stand ages,the plantation of diff erent ages(young forest,5 years;middle-aged forest,22 years;mature forest,40 years)at the Guangxi Daguishan forest station of China were studied to examine the seasonal variation of their microbial biomass carbon(MBC)and microbial biomass nitrogen(MBN)by chloroform fumigation extraction method.It was found that among the forests of diff erent age,MBC and MBN diff ered signifi cantly in the 0–10 cm soil layer,and MBN diff ered signifi cantly in the 10–20 cm soil layer,but there was no signifi cant diff erence in MBC for the 10–20 cm soil layer or in either MBC or MBN for the 20–40 cm soil layer.With increasing maturity of the forest,MBC gradually decreased in the 0–10 cm soil layer and increased fi rstly and then decreased in the 10–20 cm and 20–40 cm soil layers,and MBN increased fi rstly and then decreased in all three soil layers.As the soil depth increased,both MBC and MBN gradually decreased for all three forests.The MBC and MBN basically had the same seasonal variation in all three soil layers of all three forests,i.e.,high in the summer and low in the winter.Correlation analysis showed that MBC was signifi cantly positively correlated with soil organic matter,total nitrogen,and soil moisture,whereas MBN was signifi cantly positively correlated with soil total nitrogen.It showed that soil moisture content was the main factor determining the variation of soil microbial biomass by Redundancy analysis.The results showed that the soil properties changed continuously as the young forest grew into the middle-aged forest,which increased soil microbial biomass and enriched the soil nutrients.However,the soil microbial biomass declined as the middle-age forest continued to grow,and the soil nutrients were reduced in the mature forest. 展开更多

The mineral N, Pand K fertilizers (as urea, calcium superphosphate and KCl respectively) were incorporated at 100,25, and 100 kg ha^(-1), respectively, and the various pesticides were applied at the recommendedrates. The results of the experiment demonstrated a decline in the microbial abundance and soilmicrobial biomass phospholipid contents with the advancement of crop growth, and significant changesin substrate utilization pattern of soil microbial population studied were observed with differentmanagement practices and at different growth stages. The principal component analysis (PCA) usingall 95-carbon sources (BIOLOG plates) gave good differentiation among the treatments, indicatingthat they have different patterns of carbon utilization under different habitats. The data showedthat diversity in microbial community continuously changed with the progression in crop stage,particularly at physiological maturity (PM) stage that was evident from the utilization of differentcarbon sources at various crop stages. 展开更多

Accordingly, vegetation restoration has been implemented in this area over the past two decades to remedy the soil degradation problem. Understanding the microbial community structure is essential for the sustainability of ecosystems and for the reclamation of degraded arable land. This study aimed to determine the effect of different vegetation types on microbial processes and community structure in rhizosphere soils in the Loess Plateau. The six vegetation types were as follows:two natural grassland (Artemisia capillaries and Heteropappus altaicus), two artificial grassland (Astragalus adsurgens and Panicum virgatum), and two artificial shrubland (Caragana korshinskii and Hippophae rhamnoides) species. The microbial community structure and functional diversity were examined by analyzing the phospholipid fatty acids (PLFAs) and community-level physiological profiles. The results showed that rhizosphere soil sampled from the H. altaicus and A. capillaries plots had the highest values of microbial biomass C, average well color development of carbon resources, Gram-negative (G-) bacterial PLFA, bacterial PLFA, total PLFA, Shannon richness, and Shannon evenness, as well as the lowest metabolic quotient. Soil sampled from the H. rhamnoides plots had the highest metabolic quotient and Gram-positive (G+) bacterial PLFA, and soil sampled from the A. adsurgens and A. capillaries plots had the highest fungal PLFA and fungal:bacterial PLFA ratio. Correlation analysis indicated a signiifcant positive relationship among the microbial biomass C, G- bacterial PLFA, bacterial PLFA, and total PLFA. In conclusion, plant species under arid climatic conditions signiifcantly affected the microbial community structure in rhizosphere soil. Among the studied plants, natural grassland species generated the most favorable microbial conditions. 展开更多
