TPA stimulated DNA synthesis of SSV-NIH3T3 cells on the third day of culture in SFM. In SDS-PAGF of medium conditioned by TPA-treated SSV-NIH3T3 cells (in SFM+TPA), the amounts of four proteins of 31.0 Kd, 28.5 Kd, 25.5 Kd and 13.5 Kd strikingly increased over that of non-TPA-treated counterpart (in SFM). The PDGF-like activity was also detected in CM of SFM+TPA. When insulin and EGF were drown off the SFM+TPA (SFM-Ins-EGF+TPA), TPA lost its ability to stimulate DNA synthesis of SSV-NIH3T3 cells on the third day and SDS-PAGE of the conditioned medium showed that the amounts of the four proteins noted above grately reduced. However, cells in SFM-Ins-EGF+TPA were in almost the same growth condition as cells in complete SFM+TPA on the third day of culture. Results were discussed in the paper.展开更多
目的对比观察电针和艾灸对佐剂性关节炎(adjuvant arthritis,AA)大鼠足跖容积测量、关节炎指数评分及分化簇4淋巴细胞(cluster of differentiation-4^(+)T-cell,CD4^(+)T)水平的影响,评价两种干预措施治疗AA的效应机制。方法选取健康雄...目的对比观察电针和艾灸对佐剂性关节炎(adjuvant arthritis,AA)大鼠足跖容积测量、关节炎指数评分及分化簇4淋巴细胞(cluster of differentiation-4^(+)T-cell,CD4^(+)T)水平的影响,评价两种干预措施治疗AA的效应机制。方法选取健康雄性大鼠24只,随机分为空白组、模型组、电针组和艾灸组,每组6只。艾灸组予以双侧“足三里”“关元”“阿是穴”悬灸20 min;电针组左侧足三里与关元配穴,右侧足三里与同侧阿是穴配穴,干预20 min。于干预第21天进行足跖容积测量及关节炎指数评分,隔天于无菌操作台进行脾脏取材,采用流式细胞术检测脾脏CD4^(+)T淋巴细胞。结果与空白组相比,模型组的足跖容积、关节炎指数评分、CD4^(+)T淋巴细胞数量均明显增多(P<0.01);与模型组相比,电针组、艾灸组的足跖容积、关节炎指数评分、CD4^(+)T淋巴细胞数量均明显减少(P<0.05或P<0.01);与电针组相比,艾灸组的足跖容积、关节炎指数评分、CD4^(+)T淋巴细胞数量差异均无统计学意义(P>0.05)。结论电针与艾灸在改善类风湿关节炎症状及免疫方面的作用机制具有类似效应,均可抑制脾CD4^(+)T淋巴细胞的增殖,维持机体免疫稳态,改善病情。展开更多

Methods A co-culture system containing irradiated antigen presenting cells (J774A.1) and unirradiated T lymphocytes (EL-4) was established to observe the effect of J774A.1 cells exposed to both low and high doses of X-rays on the unirradiated EL-4 cells. Incorporation of 3H-TdR was used to assess the proliferation of the EL-4 cells, expression of CD80/86 and CD48 on J774A.1 cells was measured with immunohistochemistry and flow cytometry, respectively. NO release from J774A.1 cells was estimated with nitrate reduction method. Results Low dose-irradiated J774A.1 cells could stimulate the proliferation of the unirradiated EL-4 cells while the high dose-irradiated J774A.1 cells exerted an inhibitory effect on the proliferation of the unirradiated EL-4 cells. Preliminary mechanistic studies illustrated that the differential changes in CD48 expression and NO production by the irradiated J774A.1 cells after high and low dose radiation might be important factors underlying the differential bystander effect elicited by different doses of radiation. Conclusion Stimulatory bystander effect can be induced in immune cells by low dose radiation.展开更多

MSC are present in several adult tissues which milieu may influence MSC behavior particularly under inflammatory conditions. Additionally, culture conditions also can modify cell function or state of activation. Methods: To address the influence of the MSC source on its characteristics, we studied a xenofree, platelet lysate supplemented MSC from dental pulp, adipose tissue and bone marrow, co-cultured with isolated T cells and PBMC subset, and studied the effect of culture animal or human supplements immunomodulatory effect. Results: All three sources were efficient in inhibiting T cells. Among all MSC sources, as also described by others, adipose MSC was capable to significantly induce Treg phenotype and decrease T CD8+. Furthermore, comparing fetal bovine serum and platelet lysate, results demonstrate that platelet lysate alone is capable to induce immunomodulatory phenotype. Additional studies have to be made to elucidate the PL immunomodulatory effect.展开更多
