Described in this paper is the research and development of the X70 UOE SAWL pipes by the company. Recent advances in cellular therapy have demonstrated that this platform has the potential to give patients with certain cancers a second chance at life. Unlike ... Cancer is one of the leading causes of death worldwide. Recent advances in cellular therapy have demonstrated that this platform has the potential to give patients with certain cancers a second chance at life. Unlike chemical compounds and proteins, cells are living, self-replicating drugs that can be engineered to possess exquisite specificity. For example, T cells can be genetically modified to express chimeric antigen receptors (CARs), endowing them with the capacity to recognize and kill tumor cells and form a memory pool that is ready to strike back against persisting malignant cells. Anti-CD19 chimeric antigen receptor T cells (CART19s) have demonstrated a remarkable degree of clinical efficacy for certain malignancies. The process of developing CART19 essentially follows the conventional “one gene, one drug, one disease” paradigm derived from Paul Ehrlich’s “magic bullet” concept. With major players within the pharmaceutical industry joining forces to commercialize this new category of “living drugs,” it is useful to use CART19 as an example to examine the similarities and differences in its development, compared with that of a conventional drug. In this way, we can assimilate existing knowledge and identify the most effective approach for advancing similar strategies. This article reviews the use of biomarker-based assays to guide the optimization of CAR constructs, preclinical studies, and the evaluation of clinical efficacy;adverse effects (AEs);and CART19 cellular kinetics. Advanced technologies and computational tools that enable the discovery of optimal targets, novel CAR binding domains, and biomarkers predicting clinical response and AEs are also discussed. We believe that the success of CART19 will lead to the development of other engineered T cell therapies in the same manner that the discovery of arsphenamine initiated the era of synthetic pharmaceuticals. 展开更多

Concerning this system with a concise interface, plentiful data input/output interfaces, it is featured with high integration, simple and convenient operations of applications. For a long time spleen has been an important field of study for researchers.However,details about changes of microscopic structure in spleen during the fetal period are far less available in literature.Our goals were to determine distribution of white pulp as well as its relationship with blood vessels and to observe development and distribution of blood vessels and arrangement of CD4+and CD8+lymphocytes.The study was carried out on 16 specimens of fetal spleens.After standard histological hematoxylin and eosin(H&E)staining,the specimens were stained with immunohistochemical methods.Antibodies for CD4+,CD8+and vascular endothelial cells were used.Qualitative,semi-quantitative and quantitative analyses were performed.The results were statistically analyzed and graphically presented.By the 19th week of gestation lymphocytes can be found in spleen.The 24th week of gestation is the period of the formation of a white pulp of the spleen with centrally placed blood vessels in the follicles.Also,during this week the arterioles were completely surrounded by the CD4+lymphocytes.The number of arterioles in spleen increases during fetal development.With the increase of gestational age of the fetus,lymphatic tissue in the parenchyma of the spleen increases,as well as the number of the arterioles,while the number of capillary blood vessels decreases. 展开更多

目前的工艺技术下,转子T型叶根槽车削工艺较为复杂。利用NX二次开发技术,研究了T型叶根槽的模块化编程方法,开发了叶根槽加工程序平台。根据基于拓扑学的曲线编码规则,对叶根槽结构进行了曲线编码,实现了T型叶根槽结构的自动识别。按工... 目前的工艺技术下,转子T型叶根槽车削工艺较为复杂。利用NX二次开发技术,研究了T型叶根槽的模块化编程方法,开发了叶根槽加工程序平台。根据基于拓扑学的曲线编码规则,对叶根槽结构进行了曲线编码,实现了T型叶根槽结构的自动识别。按工步自动导入定制化的加工模板后,批量生成了车削刀轨。通过工步顺序对车削刀轨进行排序,最终实现了叶根槽加工程序的自动生成。该平台可以减少复杂的人工重复操作,提升数控程序编制效率。研究成果可为类似典型结构专用程序的开发提供参考。 展开更多

The floral-organ-number mutant fon(t) was firstly discovered in the progeny of a cross between a diploid (Chunjiang 683) and a haploid (SARⅣ/-620-A) rice cultivar. The fon(t) mutant showed normal vegetative development and produced normal inflorescence structures. Difference between the mutant and the wild type was observed when the stamen primordia began to form. The mature flowers offon(t) mutant showed open-hull phenotypes, which resulted in the exposure of stamens and stigmas. Normally, a single fon(t) floret consisted of six to nine stamens and one or two pistils. In addition, stamen/pistil-like structures and bulged tissues near ovaries were also observed in a few fon(t) florets. But homeotic transformation of lodicules into palea/lemma-like organs was observed almost in all the open-hull florets. The phenotypes offon(t) flowers also suggested thatfon(t) gene might affect flower organ identity in the inner whorls. Genetic analysis showed that thefon(t) mutant was controlled by a single recessive gene. 展开更多

本文采用放射免疫分析法,对包头市160名10~16岁男学生的血清促卵泡成熟徽素(FSH)、睾酮(T)含量进行了测定,并分析了FSH、T与青春期生长发育及性发育的关系。结果表明:11岁以后T水平开始急剧增加,直到15岁接近成熟时趋于平稳。... 本文采用放射免疫分析法,对包头市160名10~16岁男学生的血清促卵泡成熟徽素(FSH)、睾酮(T)含量进行了测定,并分析了FSH、T与青春期生长发育及性发育的关系。结果表明:11岁以后T水平开始急剧增加,直到15岁接近成熟时趋于平稳。此时生长发育进入突增阶段,T与身高、体重、LBM是明显的正相关(P<0.001),表明T对生长发育有促进作用,特别是对LBM的增加有促进作用。T含量与性征发育存在明显的正相关(P<0.001),并随着性征发育而明显增加.直至成熟,表明T对性发育也有促进作用。FSH在13岁时明显增加,15岁达峰值,以后开始下降,同时还得出首次遗精年龄为14.01±0.86岁,FSH与首次道精年龄存在明显的正相关(P<0.001),说明FSH与首次道格的发生有关。另外,FSH与生长发育和性发育存在明显的正相关(P<0,001),与T的相关系数为0.5203(P<0.001),表明FSH对生长发育及性发育的作用与T分泌增多有关。 展开更多
