目的:探索赤芍总苷对脑出血大鼠神经功能损伤的改善及对脑组织中调节性T细胞(Treg)/辅助性T细胞17(Th17)比例的影响。方法:使用神经功能损伤评估和脑组织苏木精-伊红(HE)染色法评估赤芍总苷对脑出血大鼠神经功能损伤的改善作用;采用酶... 目的:探索赤芍总苷对脑出血大鼠神经功能损伤的改善及对脑组织中调节性T细胞(Treg)/辅助性T细胞17(Th17)比例的影响。方法:使用神经功能损伤评估和脑组织苏木精-伊红(HE)染色法评估赤芍总苷对脑出血大鼠神经功能损伤的改善作用;采用酶联免疫吸附实验(ELISA)检测脑组织中白细胞介素-17(IL-17)、白细胞介素-10(IL-10)和转化生长因子β(TGF-β)的含量,使用流式细胞术检测脑组织中Treg/Th17细胞的变化。结果:空白组及造模后3 d、7 d赤芍总苷高剂量组和赤芍总苷中剂量组大鼠神经功能损伤评分均明显低于脑出血组,且差异均有统计学意义(P<0.05);造模后6 h、1 d和2 d赤芍总苷高剂量组和赤芍总苷中剂量组及各时间段赤芍总苷低剂量组大鼠神经功能损伤评分与脑出血组相比差异均无统计学意义(P>0.05)。与脑出血组比较,空白组、赤芍总苷高剂量组和赤芍总苷中剂量组脑组织中IL-17含量、单个脑组织细胞液中Th17比例和Th17/Treg比值明显降低,而IL-10、TGF-β含量和单个脑组织细胞液中Treg比例明显升高,且差异均有统计学意义(P<0.05);而赤芍总苷低剂量组IL-17、IL-10和TGF-β的含量及Th17比例、Treg比例及Th17/Treg比值与脑出血组相比差异均无统计学意义(P>0.05)。结论:赤芍总苷对脑出血大鼠神经功能损伤具有改善作用,也可以通过调节Th17/Treg细胞的分化和细胞因子的分泌而改善脑出血症状。 展开更多

Adoptive cell therapy using chimeric antigen receptor T (CAR-T) cells, which is a promising cancer immunotherapy strategy, has been developing very rapidly in recent years. CAR-T cells are genetically modified T cells that can specifically recognize tumor specific antigens on the surface of tumor cells, and then effectively kill tumor cells. At present, exciting results are being achieved in clinical applications of CAR-T cells for patients with hematological malignancies. The research and development of CAR-T cells for various targets and for the treatment of solid tumors have become a hot topic worldwide, so an increasing number of investigational new drug applications (INDAs) and new drug applications (NDAs) of CAR-T cell products are expected to be submitted in future. The quality control and nonclinical research of these products are of great significance in ensuring the safety and effectiveness of these products;however, they also present great challenges and difficulties. This article discusses the general principles of and key issues regarding the quality control and nonclinical research of CAR-T cell products based on their product characteristics and on relevant guidelines for gene and cell therapy products. 展开更多

Chengdu University of Technology (CDUT), located on the eastern edge of the Qinghai—Tibet Plateau, a higher education and research institution with a focus on geosciences, has paid special attention to scientific research on Q—T Plateau. In the earlier 1950s Professor Zhang Zhuoyuan, the former president of CDUT and Professor Ren Tianpei joined the geological survey in Tibet as members of the Tibetan geological working group organized by the Central Government of China. Since the 1980s, research on the Q—T Plateau has reached a golden age, and recognized this, CDUT established the Resources and Environment Research Center of Qinghai—Tibet Plateau in order to lead and coordinate the research around the Plateau continuously and smoothly. There are often more than 50 geologists involved in research during any given year. Scientists from CDUT are now conducting many kinds of research programs covering the whole area of the Plateau, from the Himalayas in the south to the Tianshan Mountains in the north, from the Longmen Mountains and the West Yunnan Plateau in the east to Ali area in the west. Scientists from CDUT have completed cooperative research programs with foreign scientists from the U. S., France, U.K., Japan, Austria, Canada, Germany and Australia. Research subjects include National Key Basic Research, State Scientific Key Projects, National Natural Scientific Foundation Projects, National Outstanding Young Scientists Foundation Projects, as well as Basic Geological Survey, Mineral Resources Evaluation, Analysis of Potential Oil and Gas Resources, Geohazard Protection.. More than 40 projects and 6 geological maps in different scales (1∶1000000, 1∶200000, 1∶50000) have been completed.. Research funding has increased to billions of yuan. Thousands relevant research papers and works have been published. More than 20 projects have won scientific achievement awards at national, ministerial and provincial levels. Meanwhile, a research group of young and middle\|aged scientists led by Prof. Wang Chengshan has formed and is determined to devote itself to the Q—T geological research. Many of them have broken established rules and have become professors or gained prizes. To strengthen Q—T research, the Ministry of Land and Resources specially established the Laboratory of Continental Dynamics of the Qianghai—Tibet Plateau at CDUT in 1999. Academicians, Li Tingdong and Liu Baojun, are the Honorary Directors of the Lab and the Academic Committee of the Lab respectively. Using long-term optical, ultraviolet(UV) and X-ray data, we present a study of a classical T Tauri star CV Cha. The V-band light curve obtained from the All Sky Automated Survey(ASAS) shows short as well as long-term variability. The short-term variability could be due to rotational modulation of CV Cha. We derive the rotational period of 3.714 ± 0.001 d for CV Cha. UV light curves obtained from Swift also show the variations. X-ray light curves from XMM-Newton and Swift do not show any significant short as well as long-term variability. However, the light curve from Chandra appears to be variable, which could be due to the emergence of flaring activities. X-ray spectra from all observations are explained well by the single temperature plasma of 0.95 keV with X-ray luminosity of 1030.4erg s-1in the 0.5–7.5 keV energy band. It appears that variability in optical and UV bands could be due to the presence of both hot and cool spots on the surface, while X-ray emission is dominated by magnetic processes. 展开更多

The US Payor Landscape: Results from a Survey of Medical Directors and Pharmacy Directors on Comparative-Effectiveness Research

The paper is to determine the types of approaches preferred by medical and pharmacy directors of US payer organizations to enhance the Pharmacy & Therapeutic decision-making process and how medications accepted onto the formulary should be covered. An online interactive survey of US medical and pharmacy directors was conducted in 2012. In addition to a 10-point Likert scale (10 = agree completely, 1 = disagree completely), qualitative responses and interpretive analysis were used to explore beliefs about certain statements. The results showed that the 30 respondents (20 medical directors and 10 pharmacy directors) rated current progress in obtaining usable CER (comparative-effectiveness research) at only an average of 4.17 on the 10-point scale. They hoped to regularly utilize CER information in formulary decision making by 2015 (average rating, 6.03). The rating of evidence-based medicine use in coverage decision making today was somewhat higher, at an average of 7.08 (medical directors, 7.38; pharmacy directors, 6.40). The survey participants believe that emerging CER results will greatly affect the following areas: optimization/improvement of clinical guidelines, medical/pharmacy benefit management, assessments of the value and appropriateness of interventions, and pharmaceutical research and development. Therefore, payers expect CER to play an increasing role in helping them determine the value of new therapies. 展开更多

Changshou Shatian You infected with Agrobacterium rhizogenes were carried out in darkness or in light. The optimizing culture system of Ri T-DNA transformed roots for C. grandis cv. Changshou Shatian You was constructed as follows: After the ventral wounded striations on the single activation cotyledon were inoculated by A. rhizogenes A4 (logarithmic period), they were cocultured at (25 ±2)℃ in darkness for 25-30 days; some transformed roots were generated from wounded striations of most cotyledons. The genetically transformed ratio is (83 ± 11)%. Axenic Ri T-DNA transformed roots (hairy roots) were harvested after five subcultures. Explants were activated on MT medium. The MS medium was used for subculture of transformed roots. Mass Ri T-DNA transformed roots in which the hormone was produced independently were harvested from this optimizing culture system. White, fresh Ri T-DNA transformed roots were (1.14 ±0.07) cm long, (0.73 ±0.04) mm wide, and the growth direction of transformed roots was negative geotropism. 展开更多

目的:以时机理论为基础探讨嵌合抗原受体T细胞免疫治疗病人疾病的照顾需求,为嵌合抗原受体T细胞免疫病人阶段性、针对性、延续性护理实施提供依据。方法:采用目的抽样法选取44例处于不同治疗阶段的嵌合抗原受体T细胞免疫病人进行半结构... 目的:以时机理论为基础探讨嵌合抗原受体T细胞免疫治疗病人疾病的照顾需求,为嵌合抗原受体T细胞免疫病人阶段性、针对性、延续性护理实施提供依据。方法:采用目的抽样法选取44例处于不同治疗阶段的嵌合抗原受体T细胞免疫病人进行半结构化访谈,并用Colaizzi 7步分析法对所收集的资料进行整理、分析、总结、归纳提炼出主题。结果:嵌合抗原受体T细胞免疫治疗病人在事件/诊断阶段存在信息获取、身心准备、经济及家庭照顾需求;稳定准备阶段存在健康信息知晓、生理照护、心理支持需求;治疗实施阶段存在症状管理、知识指导、稳定的照顾者、同伴教育支持需求;出院准备阶段存在家庭照顾者支持、重返家园需求;社会适应阶段存在自我管理知识、持续专业医疗支持、重返工作及家庭角色需求。结论:嵌合抗原受体T细胞免疫治疗病人不同治疗阶段照顾的需求各有不同,医护人员需要根据不同阶段的照顾需求特点给予不同的支持,对病人实施全程护理管理。 展开更多

技术政治时代背景下,国际竞争焦点围绕技术权力的争夺、布局和秩序构建展开,国际科技合作的驱动结构发生深刻变化,主要国家战略转向态势明显。本文以技术政治时代的重要特征作为变革动因,选取中国和美国国际科技合作战略作为比较研究对... 技术政治时代背景下,国际竞争焦点围绕技术权力的争夺、布局和秩序构建展开,国际科技合作的驱动结构发生深刻变化,主要国家战略转向态势明显。本文以技术政治时代的重要特征作为变革动因,选取中国和美国国际科技合作战略作为比较研究对象,采用“目标-理念-路径”分析框架对两国在战略目标、战略理念、实施路径上的转变方向进行对比,接着使用文献计量分析法从总体发展态势、主导地位变迁、合作网络演化3个维度进一步开展量化验证和具体分析。战略转向趋势的多维度对比结果表明:①技术政治时代下中美战略布局呈现差异化走向,美国以排他性多边主义框架为核心的“技术联盟”战略逐步形成,中国则以“开放创新”为核心理念,由参与全球创新转向引领全球创新;②中国在时代变局中所受影响更为明显,析出的合作需求还未得到充分转移和补充;③中国在国际科技合作能级和利用国外科研资金方面尚有提升空间;④在共同关注的全球性挑战议题上,中美仍有持续合作空间。在此基础上,从战略层、战术层、操作层提出启示建议。 展开更多

基于智能建造的装配式T梁生产过程稳定性控制研究
