Liver tumor infiltrating lymphocytes: Comparison of hepatocellular and cholangiolar carcinoma 被引量:9

AIM: To investigate the role of tumor inf iltrating lym-phocytes (TIL) in primary hepatocellular and cholangio-lar carcinomas of the liver.METHODS: Immunohistochemical analysis was per-formed including antibodies to CD3, CD4, CD8, CD20, CD56 and TIA-1 in formalin-f ixed and paraff in-embed-ded tissue of 35 liver resection specimens of hepatocel-lular or cholangiocellular carcinomas. Semiquantitative evaluation was performed with emphasis on the area of the tumor itself and of the tumor/liver interface.RESULTS: All hepatocellular carcinomas showed in-filtration of lymphocytes predominantly around the tumor in the tumor/liver interface consisting mainly of CD3+ CD4+ T lymphocytes [164.3/10 high power f ields (HPF)] and in the tumor itself of CD8+ cells (54.9/10 HPF). Cholangiocarcinomas contained a heterogeneous amount of TIL, composed mainly of CD3+ T cells with a predominance of CD8+ cells in the tumor tissue (52.6/10 HPF) and of CD4+ cells in the interface region (223.1/10 HPF). CD56+ cells of the innate immune system were scarce. There was no significant difference between hepatocellular or cholangiolar carcinoma. No correlation with the clinicopathological data was seen. CONCLUSION: Liver TIL consists of intratumoral CD8+ T cells and peritumoral CD4+ T cells indepen-dent of histogenetic origin. Different functions of lym-phocytes in these regions seem possible. 展开更多

The occurrence of massive CD4+ T cell depletion is one of the most prominent characteristics of human immunodeficiency virus type 1 (HIV-1) infection during acute phase, resulting in unrestorable destruction to the immune system. The infected host undergoes an asymptomatic period lasting several years with low viral load and ostensibly healthy status, which is presumably due to virus-specific adaptive immune responses. In the absence of therapy, an overwhelming majority of cases develop to AIDS within 8-10 years of latent infection. In this review, we discuss the roles in AIDS pathogenesis played by massive CD4+ T lymphocytes depletion in gut-associated lymphoid tissue (GALT) during acute infection and abnormal immune activation emerging in the later part of chronic phase. 展开更多

T Lymphocytes and Th1/Th2 Cytokines in Peripheral Blood of Patients with Recurrent Genital Herpes

Objective: This study analyzed the T lymphocytes and Th1/Th2 type cytokine profile shift in the peripheral blood ofpatients with recurrent genital herpes (RGH). Methods: Immunofluorescent staining or cell surface antigenand intracellular cytokines(IL-2、IL-4、IL-12、IFN-r)inperipheral blood from 20 RGH patients and 10 controls wereanalyzed using flow cytometric techniques. Results: RGH patients had signiflcantly lower levels ofCD3^+T cells, CD4^+T cells and CD4^+ T/ CD8^+ T cells ratiocompared to control levels (P<0.001), and IL-2-producing,IFN-r-producing and IL-12-producing T cells were increasedin RGH patients (CD4^+T: P<0.001, CD8^+T: P<0.05respectively), whereas IL-4-producing T cells were increased inRGH patients compared to controls (CD4^+T: P<0.05; CD8^+T:P<0.001 respectively). Conclusions: RGH patients have T lymphocyte subsetvariations and Th1/Th2 cytokine changes. The increase in Th2cells Th1/Th2 imbalance may have important implications forRGH pathogenesis. 展开更多

目的研究不同浓度雌二醇(E2)对去卵巢小鼠脾细胞雌激素受体(ER)及辅助性T细胞1(Th1)和辅助性T细胞2(Th2)之间平衡的影响。方法将小鼠脾细胞分为7组:青年组、假手术组、去卵巢对照组及去卵巢+E2(10^-11mol/L、10^-10mol/L、10... 目的研究不同浓度雌二醇(E2)对去卵巢小鼠脾细胞雌激素受体(ER)及辅助性T细胞1(Th1)和辅助性T细胞2(Th2)之间平衡的影响。方法将小鼠脾细胞分为7组:青年组、假手术组、去卵巢对照组及去卵巢+E2(10^-11mol/L、10^-10mol/L、10^-9mol/L、10^-8mol/L)组。逆转录-聚合酶链反应(RT-PCR)方法检测ERα、ERβmRNA的表达;酶联免疫吸附法(ELISA)法检测细胞因子干扰素-γ(IFN-γ)、白细胞介素-2(IL-2)和白细胞介素-4(IL-4)水平,以了解Th1与Th2的平衡关系。结果去卵巢对照组ERα、ERβmRNA水平分别为0.68±0.15和0.70±0.17,较青年组(0.95±0.21和0.93±0.1 9)及假手术组(0.98±0.28和0.94±0.20)下降(P<0.05和P<0.01);去卵巢+E2(10^10mol/L)组ERα、ERβ的表达分别为0.95±0.19和0.90±0.29,较去卵巢对照组升高(P<0.05和P<0.01)。去卵巢对照组IFN-γ、IL-2分别为(360.36±8.64)ng/L和(1485.49±5.01)ng/L,较青年组[(93.46±4.81)ng/L和(704.37±5.32)ng/L]及假手术组[(92.47±2.95)ng/ L和(716.84±1 6.48)ng/L]升高(均为P<0.01);而去卵巢对照组IL-4水平[(258.11±6.00)ng/L]低于青年组[(269.77±8.44)ng/L]和假手术组[(270.18±10.34)ng/L](均为P<0.05)。去卵巢+ E2(10^10mol/L)组的IFN-γ、IL-2分别为(94.05±7.17)ng/L和(712.15±13.84)ng/L,低于去卵巢对照组(均为P<0.01),IL-4为(269.76±2.05)ng/L,高于去卵巢对照组(P<0.05)。结论不同浓度E_2对去卵巢小鼠脾细胞ERα、ERβ基因表达起不同的调节作用:在生理范围内为正向调节,超生理范围为负向调节。小鼠去卵巢后影响脾细胞Th1和Th2的平衡,表现为Th1漂移。生理浓度的E2能部分逆转Th1漂移,部分恢复Th1和Th2的平衡。 展开更多
