Once-daily oral administration of CV or vehicle(5% Arabic gum) was started the day before the NMDA injection and continued until the end of the study. Electroretinograms(ERGs) were recorded to evaluate the retinal function at 2 d after NMDA injection. Furthermore, a histological evaluation, Western blot analysis, and immunohistochemistry were performed for assessing the signal transduction pathway. RESULTS: HT-22 cell death was induced by the addition of glutamate and co-incubation with CV protected against it. Oral administration of CV inhibited the decrease in scotopic threshold response amplitudes induced by the intravitreal injection of NMDA and those of the thickness of the inner retinal layer in the histological evaluation. The increased phosphorylated levels of extracellular signalregulated kinase(ERK) but not cAMP response element binding protein(CREB) or Akt were observed 1 h after NMDA injection in both the vehicle-and CV-treated rats;however, p ERK activation was no more upregulated at 3 h after NMDA injection. pERK upregulation was observed in Müller cells.CONCLUSION: CV shows a protective effect against both glutamate-induced HT-22 cell death and NMDAinduced retinal damage. pERK upregulation in the Müller cells plays a key role in the protective effect of CV against glutamate-induced retinal toxicity.展开更多

This breakthrough in generating iPSCs from somatic cells has overcome the ethical issues and immune rejection involved in the use of human embryonic stem cells. Hence, iPSCs form a great potential source for developing disease models, drug toxicity screening and cell-based therapies. These cells have the potential to differentiate into desired cell types, including hepatocytes, under in vitro as well as under in vivo conditions given the proper microenvironment. iPSC-derived hepatocytes could be useful as an unlimited source, which can be utilized in disease modeling, drug toxicity testing and producing autologous cell therapies that would avoid immune rejection and enable correction of gene defects prior to cell transplantation. In this review, we discuss the induction methods, role of reprogramming factors, and characterization of iPSCs, along with hepatocyte differentiation from iPSCs and potential applications. Further, we discuss the location and detection of liver stem cells and their role in liver regeneration. Although tumor formation and genetic mutations are a cause of concern, iPSCs still form a promising source for clinical applications.展开更多

Peracetic acid treatment 96 h before challenge inoculation induced significant contents change of both total and soluble calcium in true leaves of eggplant resistant cultivar Xi’anluqie and susceptible cultivar Liaoqie4. For both cultivars, the calcium temporl contents changes in the leaves of different positions reflected the pathway of signal transduction related to induced resistance within plants. After challenge inoculation, total and soluble Ca2+ content change more greatly in the induced eggplant than in the control, derivational change of calcium contents in eggplant leaves led to the change of PAL、PPO、POD、SOD. changed activities of which built up eggplant resistance against verticillium wilt, Ca2+ content change、defensive enzyme activity and eggplant resistance level showed the positive correlation. therefore Ca2+ was presumed to participate induction of eggplant verticillium wilt defensive response through signalling, activated those defensive enzymes to improve host resistance to verticillum eggplant wilt.展开更多

In the last few years, to replace the high cost, in terms of time and money, of conventional technologies, the scientific community has directed considerable research towards the development both of new detection systems for the measurement of the contamination levels of chemicals in people's body fluids and tissue, as well as in the environment, and of new remediation strategies for the removal of such chemicals from the environment, as a means of the prevention of human diseases. New emerging biosensors for the analysis of environmental chemicals have been proposed, including VHH antibodies, that combine the antibody performance with the affinity for small molecules, genetically engineered microorganisms, aptamers and new highly stable enzymes. However, the advances in the field of chemicals monitoring are still far from producing a continuous realtime and on-line system for their detection. Better results have been obtained in the development of strategies which use organisms(microorganisms, plants and animals) or metabolic pathway-based approaches(single enzymes or more complex enzymatic solutions) for the fixation, degradation and detoxification of chemicals in the environment. Systems for enzymatic detoxification and degradation of toxic agents in wastewater from chemical and manufacturing industries, such as ligninolytic enzymes for the treatment of wastewater from the textile industry, have been proposed. Considering the high value of these research studies, in terms of the protection of human health and of the ecosystem, science must play a major role in guiding policy changes in this field.展开更多

Because of the expressed interest, a certain number of pesticides and important chemicals and their toxicity are described in this article. The studies went on the determination of the concentration, the lethal amount of the organochlorinated compounds, chemical organophosphates, carbamates and compounds. One summer recorded 3 pesticides in 5 samples of the grounds of Sikasso and Segou (Mali). Their concentration varies from 20 (atrazine) with 45 g/kg of ground. The lethal amounts of the revealed poisons variable from 338 for phtalates to 28.710 mg/kg for hexane (alkane) thus evaluate their impact on the food chain. Organophosphates and the carbamates (insecticidal) involve a reduction of 34.2% of the number of Cyprinus carpio of fresh water. The atrazine contaminates drinking water, but the diuron modifies the behavior and the reproduction of fish by deteriorating their system of olfactive perception of natural substances. Important mortalities of birds are noted around the corn fields of Bougouni treated by the carbofuran. The pesticides involve at the man a reduction in fruitfulness, an increase in the risk of miscarriage of premature birth, congenital malformations and cancers.展开更多

文摘Adverse drug reactions(ADRs)are major clinical problems, particularly in special populations such as pediatric patients.Indeed,ADRs may be caused by a plethora of different drugs leading,in some cases,to hospitalization, disability or even death. In addition, pediatric patients may respond differently to drugs with respect to adults and may be prone to developing different kinds of ADRs,leading,in some cases,to more severe consequences. To improve the comprehension, and thus the prevention,of ADRs,the set-up of sensitive and personalized assays is urgently needed.Important progress is represented by the possibility of setting up groundbreaking patient-specific assays. This goal has been powerfully achieved using induced pluripotent stem cells(iPSCs).Due to their genetic and physiological species-specific differences and their ability to be differentiated ideally into all tissues of the human body, this model may be accurate in predicting drug toxicity, especially when this toxicity is related to individual genetic differences. This review is an up-to-date summary of the employment of iPSCs as a model to study ADRs,with particular attention to drugs used in the pediatric field. We especially focused on the intestinal, hepatic, pancreatic, renal, cardiac, and neuronal levels, also discussing progress in organoids creation. The latter are three-dimensional in vitro culture systems derived from pluripotent or adult stem cells simulating the architecture and functionality of native organs such as the intestine, liver, pancreas, kidney, heart, and brain.Based on the existing knowledge, these models are powerful and promising tools in multiple clinical applications including toxicity screening, disease modeling, personalized and regenerative medicine.

文摘The discovery of induced pluripotent stem cells (iPSCs) unraveled a mystery in stem cell research, after identification of four re-programming factors for generating pluripotent stem cells without the need of embryos. Although tumor formation and genetic mutations are a cause of concern, iPSCs still form a promising source for clinical applications.
文摘The incidence of inflammatory bowel diseases(IBD)is rising worldwide. The therapeutic options for IBD are expanding, and the number of drugs with new targets will rapidly increase in coming years.A rapid step-up approach with close monitoring of intestinal inflammation is extensively used. The fear of side effects represents one the most limiting factor of their use.Despite a widespread use for years, drug induced liver injury(DILI)management remains a challenging situation with Azathioprine and Methotrexate.DILI seems less frequent with anti-tumor necrosis factor agents and new biologic therapies. The aim of this review is to report incidence, physiopathology and practical guidelines in case of DILI occurrence with the armamentarium of old and new drugs in the field of IBD.

文摘Peracetic acid has proved to be an effective inducer against verticillium eggplant wilt, which could achieve a 60%~90% control effect and keep over 20 days. After challenge inoculation, total and soluble Ca2+ content change more greatly in the induced eggplant than in the control, derivational change of calcium contents in eggplant leaves led to the change of PAL、PPO、POD、SOD. changed activities of which built up eggplant resistance against verticillium wilt, Ca2+ content change、defensive enzyme activity and eggplant resistance level showed the positive correlation. therefore Ca2+ was presumed to participate induction of eggplant verticillium wilt defensive response through signalling, activated those defensive enzymes to improve host resistance to verticillum eggplant wilt.

文摘Laser surgery provides clean, fast and accurate modeling of tissue. However, the inability to determine what kind of tissue is being ablated at the bottom of the cut may lead to the iatrogenic damage of structures that were meant to be preserved. In this context, nerve preservation is one of the key challenges in any surgical procedure. One example is the treatment of parotid gland pathologies, where the facial nerve(N.VII) and its main branches run through and fan out inside the glands parenchyma.A feedback system that automatically stops the ablation to prevent nerve-tissue damage could greatly increase the applicability and safety of surgical laser systems. In the present study,Laser Induced Breakdown Spectroscopy(LIBS) is used to differentiate between nerve and gland tissue of an ex-vivo pig animal model. The LIBS results obtained in this preliminary experiment suggest that the measured spectra, containing atomic and molecular emissions, can be used to differentiate between the two tissue types. The measurements and differentiation were performed in open air and under normal stray light conditions.

文摘Anabolic androgenic steroids(AASs)are a group of molecules including endogenous testosterone and synthetic derivatives that have both androgenic and anabolic effects. These properties make them therapeutically beneficial in medical conditions such as hypogonadism. However, they are commonly bought illegally and misused for their anabolic, skeletal muscle building, and performanceenhancing effects.Supraphysiologic and long-term use of AASs affects all organs, leading to cardiovascular, neurological, endocrine, gastrointestinal, renal, and hematologic disorders.Hepatotoxicity is one of the major concerns regarding AASs treatment and abuse.Testosterone and its derivatives have been most often shown to induce a specific form of cholestasis, peliosis hepatis, and hepatic benign and malignant tumors. It is currently believed that mechanisms of pathogenesis of these disorders include disturbance of antioxidative factors, upregulation of bile acid synthesis, and induction of hepatocyte hyperplasia. Most toxicity cases are treated with supportive measures and liver function normalizes with discontinuation of AAS.However, some long-term consequences are irreversible.AAS-induced liver injury should be taken in consideration in patients with liver disorders, especially with the increasing unintentional ingestion of supplements containing AAS.In this paper,we review the most current knowledge about AAS-associated adverse effects on the liver, and their clinical presentations, prevalence, and pathophysiological mechanisms.
文摘Addressing the problems related to the widespread presence of an increasing number of chemicals released into the environment by human activities represents one of the most important challenges of this century. Considering the high value of these research studies, in terms of the protection of human health and of the ecosystem, science must play a major role in guiding policy changes in this field.
文摘The contaminants of the ground are potentially harmful agents and when they are released in this medium, their persistence becomes an important concern. The pesticides involve at the man a reduction in fruitfulness, an increase in the risk of miscarriage of premature birth, congenital malformations and cancers.
