However, other factors like the large number of species received at screening centers from different locations, rescued after being hit by vehicles, taken by the public or confiscated from illegal trade by the authorities, have increased the risk of spreading, emergence or reemergence of zoonosis. Besides the notorious importance of the procedure improvement for managing wildlife, the access to as much as possible information about the occurrence of potential infections on each particular species can be a tool of great value for mitigating the disease risk. In the present paper, it was showed the evolution of processes for wildlife translocations mostly related to mammals, we also discussed some aspects related to sylvatic animals as reservoir host of zoonosis and finally were presented several tables recording numerous mammals hosts and their respective parasitic protozoa.展开更多

Conventional lymphocyte karyotype analyses highlighted dif- ferent reciprocal balanced translocations: t(12; 13), t(4;9), t(X;8), t(8; 10) and t(3; 16). Semen analysis was performed by light and TEM. Fluorescence in situ hybridization was performed directly on sperm nuclei using centromeric probes for chromosomes 18, X and Y. The carriers of the balanced reciprocal translocations considered in the present study showed a very similar pattern of sperm pathologies: diffused presence of apoptosis and immaturity. All patients showed meiotic segregation derangements, highlighted by the presence of sperm diploidies and sex chromosome disomies particularly related to the failure of the first meiotic division. However, an increased incidence of chromosome 18 aneuploidy was detected in spermatozoa from t(X;8) and t(8;10) carriers. We have also reported values from sex chromosomes such as t(X;8), although the X chromosome was involved in translocation. Since patients with reciprocal translocations and spermatogenetic impairment are candidates for intracytoplasmic sperm injection cycles, the study of sperm parameters, and particularly of the level of aneuploidy rates, would provide better information for couples at risk and would contribute to the data in the literature for a better understanding of the effects of chromosomal rearrangement on the whole meiotic process and, in particular, on chromosomes not involved in translocation.展开更多

Conventional cytogenetic analysis plays an important role in the diagnosis...Acute lymphoblastic leukemia (ALL) is the most common malignancy in children, with the majority of cases being of precursor B-cell phenoltype. Conventional cytogenetic analysis plays an important role in the diagnosis of B-cell ALL, identifying characteristic chromosomal abnormalities associated with a given prognosis therein facilitating optimized treatment. The more recent introduction of microarray technology to the analysis of B-cell ALL has afforded both higher resolution for the detection of known abnormalities and an ability to identify novel copy number abnormalities (CNAs) with potential clinical relevance. In the current study, microarray analysis was performed on 20 cytogenetically abnormal B-cell ALL cases (10 pediatric and 10 adult), while a novel microarray-based balanced-translocation detection methodology (translocation CGH or tCGH) was applied to that subset of cases with a known or suspected recurrent balanced translocation. Standard microarray analysis identified that CNAs was not detected by previous conventional cytogenetics in 75% (15/20) cases. tCGH identified 9/9 (100%) balanced translocations defining BCR/ABL1 (x4), ETV6/RUNX1 (x3), and MLL/AFF1 (x2) breakpoints with high resolution. The results illustrate the improved molecular detail afforded by these technologies and a comparison of translocation breakpoints, CNAs and patient age offers new insights into tumor biology with potential prognostic significance.展开更多
It has been shown clinically that continuous removal of ischemia/reperfusion-induced reactive oxygen species is not conducive to the recovery of late stroke.Indeed,previous studies have shown that excessive increases ...It has been shown clinically that continuous removal of ischemia/reperfusion-induced reactive oxygen species is not conducive to the recovery of late stroke.Indeed,previous studies have shown that excessive increases in hypochlorous acid after stroke can cause severe damage to brain tissue.Our previous studies have found that a small amount of hypochlorous acid still exists in the later stage of stroke,but its specific role and mechanism are currently unclear.To simulate stroke in vivo,a middle cerebral artery occlusion rat model was established,with an oxygen-glucose deprivation/reoxygenation model established in vitro to mimic stroke.We found that in the early stage(within 24 hours)of ischemic stroke,neutrophils produced a large amount of hypochlorous acid,while in the recovery phase(10 days after stroke),microglia were activated and produced a small amount of hypochlorous acid.Further,in acute stroke in rats,hypochlorous acid production was prevented using a hypochlorous acid scavenger,taurine,or myeloperoxidase inhibitor,4-aminobenzoic acid hydrazide.Our results showed that high levels of hypochlorous acid(200μM)induced neuronal apoptosis after oxygen/glucose deprivation/reoxygenation.However,in the recovery phase of the middle cerebral artery occlusion model,a moderate level of hypochlorous acid promoted the proliferation and differentiation of neural stem cells into neurons and astrocytes.This suggests that hypochlorous acid plays different roles at different phases of cerebral ischemia/reperfusion injury.Lower levels of hypochlorous acid(5 and 100μM)promoted nuclear translocation ofβ-catenin.By transfection of single-site mutation plasmids,we found that hypochlorous acid induced chlorination of theβ-catenin tyrosine 30 residue,which promoted nuclear translocation.Altogether,our study indicates that maintaining low levels of hypochlorous acid plays a key role in the recovery of neurological function.展开更多

In thi...Haynaldia villosa possesses a lot of important agronomic traits and has been a powerful gene resource for wheat improvement. However, only several wheat-H, villosa translocation lines have been reported so far. In this study, we attempted to develop an efficient method for inducing wheat-H, villosa chromosomal translocations. Triticum durum- Haynaldia villosa amphiploid pollen treated with 1 200 rad ^60Co-y-rays was pollinated to Triticum aestivum cv. 'Chinese Spring'. Ninety-eight intergeneric translocated chromosomes between T. durum and H. villosa were detected by genomic in situ hybridization in 44 of 61 M1 plants, indicating a translocation occurrence frequency of 72.1%; much higher than ever reported. There were 26, 62 and 10 translocated chromosomes involving whole arm translocations, terminal translocations, and intercarlary translocations, respectively. Of the total 108 breakage-fusion events, 79 involved interstitial regions and 29 involved centric regions. The ratio of small segment terminal translocations (W.W-V) was much higher than that of large segment terminal translocations (W-V.V). All of the M1 plants were self-sterile, and their backcross progeny was all obtained with 'Chinese Spring' as pollen donors. Transmission analysis showed that most of the translocations were transmittable. This study provides a new strategy for rapid mass production of wheat-alien chromosomal translocations, especially terminal translocations that will be more significant for wheat improvement.展开更多

Pollen collected from the spikes 1, 2, and 3 days after irradiation were transferred to emasculated spikes of the common wheat cv. ‘Chinese Spring'. Genomic in situ hybridization was used to identify wheat-H, villosa chromosome translocations in the M1 generation. Transmission of the identified translocation chromosomes was analyzed in the BC1, BC2, and BC3 generations. The results indicated that all three irradiation doses were highly efficient for inducing wheat-alien translocations without affecting the viability of the M1 seeds. Within the range of 800-1,600 rad, both the efficiency of translocation induction and the frequency of interstitial chromosome breakage-fusion increased as the irradiation dosage increased. A higher translocation induction frequency was observed using pollen collected from the spikes 1 day after irradiation over that of 2 or 3 days after irradiation. More than 70% of the translocations detected in the M1 generation were transmitted to the BC1 through the female gametes. All translocations recovered in the BC1 generation were recovered in the following BC2, and BC3 generations. The transmission ability of different translocation types in different genetic backgrounds showed an order of ‘whole-arm translocation 〉 small alien segment translocation 〉 large alien segment translocation', through either male or female gametes, In general, the transmission ability through the female gametes was higher than that through the male gametes. By this approach, 14 translocation lines that involved different H. villosa chromosomes have been identified in the BC3 using EST-STS markers, and eight of them were homozygous.展开更多

However simply observing demographic growth of a translocated population is not sufficient to infer species recovery. Adequate genetic representation of the source population(s) and their long-term viability should also be considered. The woylie Bettongia penicillata ogilbyi has been subject to more formal translocations for conservation than any other marsupial that, up until recently, has resulted in one of the most suc- cessful species recoveries in Australia. We used mitochondrial and nuclear DNA markers to assess the genetic outcomes of trans- located woylie populations. These populations have lost genetic variability, differentiated fxom their source population and the supplementation program on two island populations appears to have failed. We discuss the conservation implications that our re- suits have for managing threatened species, outline some general recommendations for the management of present and future translocations and discuss the appropriate sampling design for the establishment of new populations or captive breeding programs that may mitigate the genetic 'erosion' seen in our study species. This research provides some practical outcomes and a pragmatic understanding of translocation biology. The findings are directly applicable to other translocation programs .展开更多
In acidic paddy fields of South China,rice(Oryza sativa L.)faces the dual challenges of cadmium(Cd)toxicity and silicon(Si)deficiency.Although previous studies have highlighted the functions of Si application timing a...In acidic paddy fields of South China,rice(Oryza sativa L.)faces the dual challenges of cadmium(Cd)toxicity and silicon(Si)deficiency.Although previous studies have highlighted the functions of Si application timing and strategies in mitigating Cd-stressed rice,the precise mechanisms underlying the health restoration of Cd-toxic rice and the assurance of grain safety remain elusive.This study explored Cd translocation and detoxification in the shoots of rice regulated by various Si fertilization regimes:Si(T)(all Si added before transplanting),Si(J)(all Si added at jointing),and Si(TJ)(half Si added both before transplanting and at jointing).The findings revealed that the regime of Si(TJ)was more beneficial to rice health and grain safety than Si(T)and Si(J).The osmotic regulators such as proline,soluble sugars,and soluble proteins were significantly boosted by Si(TJ)compared to other Si treatments,and which enhanced membrane integrity,balanced intracellular pH,and increased Cd tolerance of rice.Furthermore,Si(TJ)was more effective than Si(T)and Si(J)on the Cd sequestration in the cell wall,Cd bio-passivation,and the down-regulated expression of the Cd transport genes.The concentrations of Cd in the xylem and phloem treated with Si(TJ)were reduced significantly.Additionally,Si(TJ)facilitated much more Cd bound with the outer layer proteins of grains,and promoted Cd chelation and complexation by phytic acid,phenolics,and flavonoids.Overall,Si(TJ)outperformed Si(T)and Si(J)in harmonizing the phycological processes,inhibiting Cd translocation,and enhancing Cd detoxification in rice plant.Thereby the split Si application strategy offers potential for reducing Cd toxicity in rice grain.展开更多
Global climate change is characterized by asymmetric warming,i.e.,greater temperature increases in winter,spring,and nighttime than in summer,autumn,and daytime.Field experiments were conducted using four wheat cultiv...Global climate change is characterized by asymmetric warming,i.e.,greater temperature increases in winter,spring,and nighttime than in summer,autumn,and daytime.Field experiments were conducted using four wheat cultivars,namely‘Yangmai 18’(YM18),‘Sumai 188’(SM188),‘Yannong 19’(YN19),and‘Annong 0711’(AN0711),in the two growing seasons of 2019-2020 and 2020-2021,with passive night warming during different periods in the early growth stage.The treatments were night warming during the tillering-jointing(NW_(T-J)),jointing-booting(NWJ-B),and booting-anthesis(NWB-A)stages,with ambient temperature(NN)as the control.The effects of night warming during different stages on wheat yield formation were investigated by determining the characteristics of dry matter accumulation and translocation,as well as sucrose and starch accumulation in wheat grains.The wheat yields of all four cultivars were significantly higher in NW_(T-J)than in NN in the 2-year experiment.The yield increases of semi-winter cultivars YN19 and AN0711 were greater than those of spring cultivars YM18 and SM188.Treatment NW_(T-J)increased wheat yield mainly by increasing the 1,000-grain weight and the number of fertile spikelets,and it increased dry matter accumulation in various organs of wheat at the anthesis and maturity stages by increasing the growth rate at the vegetative growth stage.The flag leaf and spike showed the largest increases in dry matter accumulation.NW_(T-J)also increased the grain sucrose and starch contents in the early and middle grain-filling stages,promoting yield formation.Overall,night warming between the tillering and jointing stages increased the pre-anthesis growth rate,and thus,wheat dry matter production,which contributed to an increase in wheat yield.展开更多

The accumulation of intracellular lipids and fatty acids in the myocardium usually results in lipotoxicity, which impairs myocardial function. Adipsin may play an important protective role in the pathogenesis of DCM. The aim of this study is to investigate the regulatory effect of Adipsin on DCM lipotoxicity and its molecular mechanism.MethodsA high-fat diet (HFD)-induced type 2 diabetes mellitus model was constructed in mice with adipose tissue-specific overexpression of Adipsin (Adipsin-Tg). Liquid chromatography-tandem mass spectrometry (LC–MS/MS), glutathione-S-transferase (GST) pull-down technique, Co-immunoprecipitation (Co-IP) and immunofluorescence colocalization analyses were used to investigate the molecules which can directly interact with Adipsin. The immunocolloidal gold method was also used to detect the interaction between Adipsin and its downstream modulator.ResultsThe expression of Adipsin was significantly downregulated in the HFD-induced DCM model (P < 0.05). Adipose tissue-specific overexpression of Adipsin significantly improved cardiac function and alleviated cardiac remodeling in DCM (P < 0.05). Adipsin overexpression also alleviated mitochondrial oxidative phosphorylation function in diabetic stress (P < 0.05). LC–MS/MS analysis, GST pull-down technique and Co-IP studies revealed that interleukin-1 receptor-associated kinase-like 2 (Irak2) was a downstream regulator of Adipsin. Immunofluorescence analysis also revealed that Adipsin was co-localized with Irak2 in cardiomyocytes. Immunocolloidal gold electron microscopy and Western blotting analysis indicated that Adipsin inhibited the mitochondrial translocation of Irak2 in DCM, thus dampening the interaction between Irak2 and prohibitin (Phb)-optic atrophy protein 1 (Opa1) on mitochondria and improving the structural integrity and function of mitochondria (P < 0.05). Interestingly, in the presence of Irak2 knockdown, Adipsin overexpression did not further alleviate myocardial mitochondrial destruction and cardiac dysfunction, suggesting a downstream role of Irak2 in Adipsin-induced responses (P < 0.05). Consistent with these findings, overexpression of Adipsin after Irak2 knockdown did not further reduce the accumulation of lipids and their metabolites in the cardiac myocardium, nor did it enhance the oxidation capacity of cardiomyocytes expose to palmitate (PA) (P < 0.05). These results indicated that Irak2 may be a downstream regulator of Adipsin.ConclusionsAdipsin improves fatty acid β-oxidation and alleviates mitochondrial injury in DCM. The mechanism is related to Irak2 interaction and inhibition of Irak2 mitochondrial translocation.展开更多

Dietary interventions with naturalproducts have shown promise in preventing and treating NAFLD. Sulforaphane (SFN) is a phytocompoundwith antioxidant and anti-inflammatory properties, and previous research has demonstrated that SFN canameliorate hepatic lipid accumulation and inflammation. However, the molecular mechanisms underlying thesebeneficial effects remain unclear. In this study, we confirmed the protective effects of SFN on excessive lipidaccumulation and inflammatory injury in a high-fat, high-fructose diet-induced non-alcoholic steatohepatitis(NASH) mouse model. We found that SFN attenuates the inflammatory injury in a macrophage cell line andthe liver of NASH mice, owing to the promotion of M1-type macrophage polarization toward the M2-type andthe regulation of inflammatory mediators. Further analysis demonstrated that this SFN-induced macrophageM2-type polarization occurs in a Krüppel-like factor 4 (KLF4)-dependent manner. In summary, we uncovereda new mechanism of action underlying SFN activity and provide evidence that dietary intervention with SFNmight be protective against NASH.展开更多
BACKGROUND Helicobacter pylori(H.pylori)eradication treatment for primary gastric mucosaassociated lymphoid tissue(MALT)lymphoma has already been established.However,t(11;18)(q21;q21)/API2-MALT1 translocation-positive...BACKGROUND Helicobacter pylori(H.pylori)eradication treatment for primary gastric mucosaassociated lymphoid tissue(MALT)lymphoma has already been established.However,t(11;18)(q21;q21)/API2-MALT1 translocation-positive lesions are a type of primary gastric MALT lymphoma in which a response to eradication treatment is difficult to achieve.In addition,trisomy 18 may be associated with diffuse large B-cell lymphoma(DLBCL)transformation of gastric MALT lymphoma.CASE SUMMARY A 66-year-old man was diagnosed with MALT lymphoma in the ascending colon by colonoscopy and biopsy.Two years later,esophagogastroduodenoscopy revealed chronic atrophic gastritis that was positive for H.pylori,and eradication treatment was administered.Two years and nine months later(at the age of 70),a new ulcerative lesion suggestive of MALT lymphoma appeared in the gastric body,and six months later,a similar lesion was also found in the fundus.One year later(4 years and 3 months after H.pylori eradication),at the age of 72,the lesion in the gastric body had become deeper and had propagated.A biopsy revealed a pathological diagnosis of DLBCL.Both MALT lymphoma lesions in the ascending colon and DLBCL lesions in the stomach were positive for the t(11;18)(q21;q21)/API2-MALT1 translocation,and trisomy 18q21 was also detected.After 6 courses of R-CHOP(rituximab,cyclophosphamide,doxorubicin,vincristine and prednisone)chemotherapy,all of the above lesions disappeared[complete remission(CR)],and CR has been maintained for more than 3 years.In addition,both the colonic and gastric lesions were proven to have the same clonality.CONCLUSION Because the patient had a MALT1 translocation with trisomy 18q21,it was thought that this gastric MALT lymphoma developed independently of H.pylori infection and progressed.展开更多

Non-alcoholic fatty liver disease(NAFLD)is emerging as a common cause of chronic liver disease in children and adults.NAFLD can progress to steatohepa-titis and potentially even hepatocellular carcinoma.Early identifi...Non-alcoholic fatty liver disease(NAFLD)is emerging as a common cause of chronic liver disease in children and adults.NAFLD can progress to steatohepa-titis and potentially even hepatocellular carcinoma.Early identification of pati-ents at risk for progressive disease is crucial for managing NAFLD.Recent studies have identified long noncoding RNAs(lncRNAs),circular RNAs,and microRNAs as playing important roles in the pathogenesis of NAFLD.These noncoding RNAs are involved in modulating several metabolic pathways such as hepatic glucose and lipid metabolism,oxidative stress,and even carcinogenesis.Elevated levels of lncARSR and lncRNA nuclear-enriched abundant transcript 1 have been found in patients with NAFLD.In addition,lncRNAs such as PRYP4-3 and RP11-128N14.5 can distinguish patients with NAFLD from healthy indi-viduals.Increased MEG3 expression has been observed in both NAFLD and non-alcoholic steatohepatitis,suggesting that it may help predict patients at risk for disease progression.With advances in transcriptomics,we may discover additional targets to help in the identification and prognostication of NAFLD.展开更多

Samp...This study aims to investigate the level of soil pollution and the grade of accumulation of metals and heavy metals by wheat plants from the soil in different parts of the crop: root, stem, leaf, spike and grain. Sampling campaigns took place in February, April and July when wheat plants were at different growth stages. A number of eight soil samples and eight wheat plant samples were collected. The sampled wheat plant was taken at the same time and from the same place as the soil. Concentrations of Al (aluminium), Cr (chromium), Mn (manganese), Fe (iron), Ni (nickel), Co (cobalt), Cu (copper), Zn (zinc), Sr (strontium), Cd (cadmium) and Pb (lead) were determined by inductively coupled plasma mass spectrometry. Dynamics of dry- or fresh-weight of fruit, peel photosynthetic rate and chlorophyll content, and the characteristics of translocation and distribution of radiolabelled assimilates from leaf or fruit were examined in d...Dynamics of dry- or fresh-weight of fruit, peel photosynthetic rate and chlorophyll content, and the characteristics of translocation and distribution of radiolabelled assimilates from leaf or fruit were examined in developing satsuma mandarin (Citrus unshiu Marc. cv. Miyagawa wase) fruit from primary stage of fruit enlargement up to fruit full ripe. Change in fruit photosynthetic rate was some what related to the change in the chlorophyll content of peel. Fruit photosynthetic rate markedly declined as chlorophyll degradation occurred in the peel. Before full ripe stage of the fruit, photosynthates produced by a 14C-fed leaf were mainly distributed to juice sacs even during periods when dry matter accumulation in peel was more rapid than that in juice sacs. At the full ripe stage, peel photosynthetic rate approached zero and peel became the major sink of leaf photosynthates. Most of the peel assimilates, however, remained in situ for up to 48 h after feeding 14CO 2 to the fruit, only a small portion being transported to other parts of fruit. The percentage of fruit photosynthates exported decreased with fruit development and ripening, but the peak rate of export to juice sacs amount to as high as 12%. The sugar content and dry weights of peel and juice sacs in shaded fruit were lower than that in the control fruit. These results show that peel assimilate was mainly consumed in peel respiration and growth and thus the dependence on leaf photosynthates decreased. Part of this assimiate was used in sugar accumulation in juice sacs of fruit.展开更多
文摘With regard to wildlife translocations and the assessment of potential risk of disease transmission, several advances have been made in conservative projects. However, other factors like the large number of species received at screening centers from different locations, rescued after being hit by vehicles, taken by the public or confiscated from illegal trade by the authorities, have increased the risk of spreading, emergence or reemergence of zoonosis. Besides the notorious importance of the procedure improvement for managing wildlife, the access to as much as possible information about the occurrence of potential infections on each particular species can be a tool of great value for mitigating the disease risk. In the present paper, it was showed the evolution of processes for wildlife translocations mostly related to mammals, we also discussed some aspects related to sylvatic animals as reservoir host of zoonosis and finally were presented several tables recording numerous mammals hosts and their respective parasitic protozoa.
文摘We analysed ejaculated spermatozoa from five infertile men with different balanced reciprocal translocations to contribute to the study of meiotic segregation of chromosomes 18, X and Y and also to evaluate sperm morphology by transmission electron microscopy (TEM) analysis. Conventional lymphocyte karyotype analyses highlighted dif- ferent reciprocal balanced translocations: t(12; 13), t(4;9), t(X;8), t(8; 10) and t(3; 16). Semen analysis was performed by light and TEM. Fluorescence in situ hybridization was performed directly on sperm nuclei using centromeric probes for chromosomes 18, X and Y. The carriers of the balanced reciprocal translocations considered in the present study showed a very similar pattern of sperm pathologies: diffused presence of apoptosis and immaturity. All patients showed meiotic segregation derangements, highlighted by the presence of sperm diploidies and sex chromosome disomies particularly related to the failure of the first meiotic division. However, an increased incidence of chromosome 18 aneuploidy was detected in spermatozoa from t(X;8) and t(8;10) carriers. We have also reported values from sex chromosomes such as t(X;8), although the X chromosome was involved in translocation. Since patients with reciprocal translocations and spermatogenetic impairment are candidates for intracytoplasmic sperm injection cycles, the study of sperm parameters, and particularly of the level of aneuploidy rates, would provide better information for couples at risk and would contribute to the data in the literature for a better understanding of the effects of chromosomal rearrangement on the whole meiotic process and, in particular, on chromosomes not involved in translocation.

Chinese Spring and Chinese Spring-Th. bessarabicum amphiploid. Theselines included Tj01 and Tj02 (2n=44) containing a pair of wheat-Th. bessarabicumtranslocation chromosomes besides a pair of added Th. bessarabicum chromosomes, Tj03(2n=44) with a pair of added interspecific translocation chromosomes, Tj04 (2n=44)containing a pair of interspecific translocation chromosomes besides an added pair ofTh. bessarabicum chromosome arms and Tj05 (2n=46) containing a pair of interspecifictranslocation chromosomes besides two pairs of added intact alien chromosomes. Thebreakpoints of all the translocations were found to be not around centromere. Meanwhile,all the lines showed normal plant growth, development and fertility, while the translocationchromosomes transmitted regularly. The obtained translocations might be of use fortransferring elite genes from Th. bessarabicum into wheat.

文摘Multinanoparticles interacting with the phospholipid membranes in solution were studied by dissipative particle dynamics simulation.The selected nanoparticles have spherical or cylindrical shapes,and they have various initial velocities in the dynamical processes.Several translocation modes are defined according to their characteristics in the dynamical processes,in which the phase diagrams are constructed based on the interaction strengths between the particles and membranes and the initial velocities of particles.Furthermore,several parameters,such as the system energy and radius of gyration,are investigated in the dynamical processes for the various translocation modes.Results elucidate the effects of multiparticles interacting with the membranes in the biological processes.
文摘Acute lymphoblastic leukemia (ALL) is the most common malignancy in children, with the majority of cases being of precursor B-cell phenoltype. Conventional cytogenetic analysis plays an important role in the diagnosis of B-cell ALL, identifying characteristic chromosomal abnormalities associated with a given prognosis therein facilitating optimized treatment. The more recent introduction of microarray technology to the analysis of B-cell ALL has afforded both higher resolution for the detection of known abnormalities and an ability to identify novel copy number abnormalities (CNAs) with potential clinical relevance. In the current study, microarray analysis was performed on 20 cytogenetically abnormal B-cell ALL cases (10 pediatric and 10 adult), while a novel microarray-based balanced-translocation detection methodology (translocation CGH or tCGH) was applied to that subset of cases with a known or suspected recurrent balanced translocation. Standard microarray analysis identified that CNAs was not detected by previous conventional cytogenetics in 75% (15/20) cases. tCGH identified 9/9 (100%) balanced translocations defining BCR/ABL1 (x4), ETV6/RUNX1 (x3), and MLL/AFF1 (x2) breakpoints with high resolution. The results illustrate the improved molecular detail afforded by these technologies and a comparison of translocation breakpoints, CNAs and patient age offers new insights into tumor biology with potential prognostic significance.

文摘It has been shown clinically that continuous removal of ischemia/reperfusion-induced reactive oxygen species is not conducive to the recovery of late stroke.Indeed,previous studies have shown that excessive increases in hypochlorous acid after stroke can cause severe damage to brain tissue.Our previous studies have found that a small amount of hypochlorous acid still exists in the later stage of stroke,but its specific role and mechanism are currently unclear.To simulate stroke in vivo,a middle cerebral artery occlusion rat model was established,with an oxygen-glucose deprivation/reoxygenation model established in vitro to mimic stroke.We found that in the early stage(within 24 hours)of ischemic stroke,neutrophils produced a large amount of hypochlorous acid,while in the recovery phase(10 days after stroke),microglia were activated and produced a small amount of hypochlorous acid.Further,in acute stroke in rats,hypochlorous acid production was prevented using a hypochlorous acid scavenger,taurine,or myeloperoxidase inhibitor,4-aminobenzoic acid hydrazide.Our results showed that high levels of hypochlorous acid(200μM)induced neuronal apoptosis after oxygen/glucose deprivation/reoxygenation.However,in the recovery phase of the middle cerebral artery occlusion model,a moderate level of hypochlorous acid promoted the proliferation and differentiation of neural stem cells into neurons and astrocytes.This suggests that hypochlorous acid plays different roles at different phases of cerebral ischemia/reperfusion injury.Lower levels of hypochlorous acid(5 and 100μM)promoted nuclear translocation ofβ-catenin.By transfection of single-site mutation plasmids,we found that hypochlorous acid induced chlorination of theβ-catenin tyrosine 30 residue,which promoted nuclear translocation.Altogether,our study indicates that maintaining low levels of hypochlorous acid plays a key role in the recovery of neurological function.

文摘Chromosomal aberrations have been associated with cancer development since their discovery more than a hundred years ago.Chromosomal translocations,a type of particular structural changes involving heterologous chromosomes,have made a critical impact on diagnosis,prognosis and treatment of cancers.For example,the discovery of translocation between chromosomes 9 and 22 and the subsequent success of targeting the fusion product BCR-ABL transformed the therapy for chronic myelogenous leukemia.In the past few decades,tremendous progress has been achieved towards elucidating the mechanism causing chromosomal translocations.This review focuses on the basic mechanisms underlying the generation of chromosomal translocations.In particular,the contribution of frequency of DNA double strand breaks and spatial proximity of translocating loci is discussed.

文摘Haynaldia villosa possesses a lot of important agronomic traits and has been a powerful gene resource for wheat improvement. However, only several wheat-H, villosa translocation lines have been reported so far. In this study, we attempted to develop an efficient method for inducing wheat-H, villosa chromosomal translocations. Triticum durum- Haynaldia villosa amphiploid pollen treated with 1 200 rad ^60Co-y-rays was pollinated to Triticum aestivum cv. 'Chinese Spring'. Ninety-eight intergeneric translocated chromosomes between T. durum and H. villosa were detected by genomic in situ hybridization in 44 of 61 M1 plants, indicating a translocation occurrence frequency of 72.1%; much higher than ever reported. There were 26, 62 and 10 translocated chromosomes involving whole arm translocations, terminal translocations, and intercarlary translocations, respectively. Of the total 108 breakage-fusion events, 79 involved interstitial regions and 29 involved centric regions. The ratio of small segment terminal translocations (W.W-V) was much higher than that of large segment terminal translocations (W-V.V). All of the M1 plants were self-sterile, and their backcross progeny was all obtained with 'Chinese Spring' as pollen donors. Transmission analysis showed that most of the translocations were transmittable. This study provides a new strategy for rapid mass production of wheat-alien chromosomal translocations, especially terminal translocations that will be more significant for wheat improvement.

文摘In order to develop more wheat-Haynaldia villosa translocations involving different chromosomes and chromosome segments of H. villosa, T. durum-H, villosa amphiploid was irradiated with ^60Co γ-rays at doses of 800, 1,200, and 1,600 rad. Pollen collected from the spikes 1, 2, and 3 days after irradiation were transferred to emasculated spikes of the common wheat cv. ‘Chinese Spring'. Genomic in situ hybridization was used to identify wheat-H, villosa chromosome translocations in the M1 generation. Transmission of the identified translocation chromosomes was analyzed in the BC1, BC2, and BC3 generations. The results indicated that all three irradiation doses were highly efficient for inducing wheat-alien translocations without affecting the viability of the M1 seeds. Within the range of 800-1,600 rad, both the efficiency of translocation induction and the frequency of interstitial chromosome breakage-fusion increased as the irradiation dosage increased. A higher translocation induction frequency was observed using pollen collected from the spikes 1 day after irradiation over that of 2 or 3 days after irradiation. More than 70% of the translocations detected in the M1 generation were transmitted to the BC1 through the female gametes. All translocations recovered in the BC1 generation were recovered in the following BC2, and BC3 generations. The transmission ability of different translocation types in different genetic backgrounds showed an order of ‘whole-arm translocation 〉 small alien segment translocation 〉 large alien segment translocation', through either male or female gametes, In general, the transmission ability through the female gametes was higher than that through the male gametes. By this approach, 14 translocation lines that involved different H. villosa chromosomes have been identified in the BC3 using EST-STS markers, and eight of them were homozygous.
文摘Translocations are an important conservation strategy for many species. However simply observing demographic growth of a translocated population is not sufficient to infer species recovery. Adequate genetic representation of the source population(s) and their long-term viability should also be considered. The woylie Bettongia penicillata ogilbyi has been subject to more formal translocations for conservation than any other marsupial that, up until recently, has resulted in one of the most suc- cessful species recoveries in Australia. We used mitochondrial and nuclear DNA markers to assess the genetic outcomes of trans- located woylie populations. These populations have lost genetic variability, differentiated fxom their source population and the supplementation program on two island populations appears to have failed. We discuss the conservation implications that our re- suits have for managing threatened species, outline some general recommendations for the management of present and future translocations and discuss the appropriate sampling design for the establishment of new populations or captive breeding programs that may mitigate the genetic 'erosion' seen in our study species. This research provides some practical outcomes and a pragmatic understanding of translocation biology. The findings are directly applicable to other translocation programs .

文摘In acidic paddy fields of South China,rice(Oryza sativa L.)faces the dual challenges of cadmium(Cd)toxicity and silicon(Si)deficiency.Although previous studies have highlighted the functions of Si application timing and strategies in mitigating Cd-stressed rice,the precise mechanisms underlying the health restoration of Cd-toxic rice and the assurance of grain safety remain elusive.This study explored Cd translocation and detoxification in the shoots of rice regulated by various Si fertilization regimes:Si(T)(all Si added before transplanting),Si(J)(all Si added at jointing),and Si(TJ)(half Si added both before transplanting and at jointing).The findings revealed that the regime of Si(TJ)was more beneficial to rice health and grain safety than Si(T)and Si(J).The osmotic regulators such as proline,soluble sugars,and soluble proteins were significantly boosted by Si(TJ)compared to other Si treatments,and which enhanced membrane integrity,balanced intracellular pH,and increased Cd tolerance of rice.Furthermore,Si(TJ)was more effective than Si(T)and Si(J)on the Cd sequestration in the cell wall,Cd bio-passivation,and the down-regulated expression of the Cd transport genes.The concentrations of Cd in the xylem and phloem treated with Si(TJ)were reduced significantly.Additionally,Si(TJ)facilitated much more Cd bound with the outer layer proteins of grains,and promoted Cd chelation and complexation by phytic acid,phenolics,and flavonoids.Overall,Si(TJ)outperformed Si(T)and Si(J)in harmonizing the phycological processes,inhibiting Cd translocation,and enhancing Cd detoxification in rice plant.Thereby the split Si application strategy offers potential for reducing Cd toxicity in rice grain.

文摘Global climate change is characterized by asymmetric warming,i.e.,greater temperature increases in winter,spring,and nighttime than in summer,autumn,and daytime.Field experiments were conducted using four wheat cultivars,namely‘Yangmai 18’(YM18),‘Sumai 188’(SM188),‘Yannong 19’(YN19),and‘Annong 0711’(AN0711),in the two growing seasons of 2019-2020 and 2020-2021,with passive night warming during different periods in the early growth stage.The treatments were night warming during the tillering-jointing(NW_(T-J)),jointing-booting(NWJ-B),and booting-anthesis(NWB-A)stages,with ambient temperature(NN)as the control.The effects of night warming during different stages on wheat yield formation were investigated by determining the characteristics of dry matter accumulation and translocation,as well as sucrose and starch accumulation in wheat grains.The wheat yields of all four cultivars were significantly higher in NW_(T-J)than in NN in the 2-year experiment.The yield increases of semi-winter cultivars YN19 and AN0711 were greater than those of spring cultivars YM18 and SM188.Treatment NW_(T-J)increased wheat yield mainly by increasing the 1,000-grain weight and the number of fertile spikelets,and it increased dry matter accumulation in various organs of wheat at the anthesis and maturity stages by increasing the growth rate at the vegetative growth stage.The flag leaf and spike showed the largest increases in dry matter accumulation.NW_(T-J)also increased the grain sucrose and starch contents in the early and middle grain-filling stages,promoting yield formation.Overall,night warming between the tillering and jointing stages increased the pre-anthesis growth rate,and thus,wheat dry matter production,which contributed to an increase in wheat yield.

文摘Background Diabetic cardiomyopathy (DCM) causes the myocardium to rely on fatty acid β-oxidation for energy. The accumulation of intracellular lipids and fatty acids in the myocardium usually results in lipotoxicity, which impairs myocardial function. Adipsin may play an important protective role in the pathogenesis of DCM. The aim of this study is to investigate the regulatory effect of Adipsin on DCM lipotoxicity and its molecular mechanism.MethodsA high-fat diet (HFD)-induced type 2 diabetes mellitus model was constructed in mice with adipose tissue-specific overexpression of Adipsin (Adipsin-Tg). Liquid chromatography-tandem mass spectrometry (LC–MS/MS), glutathione-S-transferase (GST) pull-down technique, Co-immunoprecipitation (Co-IP) and immunofluorescence colocalization analyses were used to investigate the molecules which can directly interact with Adipsin. The immunocolloidal gold method was also used to detect the interaction between Adipsin and its downstream modulator.ResultsThe expression of Adipsin was significantly downregulated in the HFD-induced DCM model (P < 0.05). Adipose tissue-specific overexpression of Adipsin significantly improved cardiac function and alleviated cardiac remodeling in DCM (P < 0.05). Adipsin overexpression also alleviated mitochondrial oxidative phosphorylation function in diabetic stress (P < 0.05). LC–MS/MS analysis, GST pull-down technique and Co-IP studies revealed that interleukin-1 receptor-associated kinase-like 2 (Irak2) was a downstream regulator of Adipsin. Immunofluorescence analysis also revealed that Adipsin was co-localized with Irak2 in cardiomyocytes. Immunocolloidal gold electron microscopy and Western blotting analysis indicated that Adipsin inhibited the mitochondrial translocation of Irak2 in DCM, thus dampening the interaction between Irak2 and prohibitin (Phb)-optic atrophy protein 1 (Opa1) on mitochondria and improving the structural integrity and function of mitochondria (P < 0.05). Interestingly, in the presence of Irak2 knockdown, Adipsin overexpression did not further alleviate myocardial mitochondrial destruction and cardiac dysfunction, suggesting a downstream role of Irak2 in Adipsin-induced responses (P < 0.05). Consistent with these findings, overexpression of Adipsin after Irak2 knockdown did not further reduce the accumulation of lipids and their metabolites in the cardiac myocardium, nor did it enhance the oxidation capacity of cardiomyocytes expose to palmitate (PA) (P < 0.05). These results indicated that Irak2 may be a downstream regulator of Adipsin.ConclusionsAdipsin improves fatty acid β-oxidation and alleviates mitochondrial injury in DCM. The mechanism is related to Irak2 interaction and inhibition of Irak2 mitochondrial translocation.

文摘The aim of this study was to investigate whether punicalagin(PU)could prevent obesity-related cardiac dysfunction by promoting DNA demethy lation,and to explore its possible mechanism.C57BL/6J mice were fed with standard diet,high-fat diet(HFD),HFD supplemented with resveratrol,low-dose PU(LPU)and high-dose PU(HPU)for 8 weeks.Compared with HFD group,body weight was significantly lower in PU treatment groups,number of cardionwocytes and the protein level of myosin heavy chain 7B were significantly higher in PU treatment groups.Levels of 5-hydroxymethylcytosine and 5-formylcytosine were significantly lower in HFD group than in other groups.Compared with the HFD group,the protein level of ten-eleven translocation enzyme(TET)2 was significantly higher in PU treatment groups,p-AMP-activated protein kinase(AMPK)was significantly higher in LPU group.Levels of total antioxidant capacity and the protein levels of complexesⅡ/Ⅲ/Ⅴ,oxoglutarate dehydrogenase,succinate dehydrogenase B and fumarate hdrolase were significantly lower in HFD group than PU treatment group.The ratio of(succinic acid+fumaric acid)/a-ketoglutarate was significantly higher in HFD group than other groups.In conclusion,PU up-regulated TETs enzyme activities and TET2 protein stability through alleviating mitochondrial dysfunction and activating AMPK,so as to promote DNA demethylation,thus preventing obesity-related cardiac dysfunction.

文摘Non-alcoholic fatty liver disease (NAFLD) has become a global issue and a severe threat to public health.However, to date, no approved therapeutic drugs have been developed. Dietary interventions with naturalproducts have shown promise in preventing and treating NAFLD. Sulforaphane (SFN) is a phytocompoundwith antioxidant and anti-inflammatory properties, and previous research has demonstrated that SFN canameliorate hepatic lipid accumulation and inflammation. However, the molecular mechanisms underlying thesebeneficial effects remain unclear. In this study, we confirmed the protective effects of SFN on excessive lipidaccumulation and inflammatory injury in a high-fat, high-fructose diet-induced non-alcoholic steatohepatitis(NASH) mouse model. We found that SFN attenuates the inflammatory injury in a macrophage cell line andthe liver of NASH mice, owing to the promotion of M1-type macrophage polarization toward the M2-type andthe regulation of inflammatory mediators. Further analysis demonstrated that this SFN-induced macrophageM2-type polarization occurs in a Krüppel-like factor 4 (KLF4)-dependent manner. In summary, we uncovereda new mechanism of action underlying SFN activity and provide evidence that dietary intervention with SFNmight be protective against NASH.
文摘BACKGROUND Helicobacter pylori(H.pylori)eradication treatment for primary gastric mucosaassociated lymphoid tissue(MALT)lymphoma has already been established.However,t(11;18)(q21;q21)/API2-MALT1 translocation-positive lesions are a type of primary gastric MALT lymphoma in which a response to eradication treatment is difficult to achieve.In addition,trisomy 18 may be associated with diffuse large B-cell lymphoma(DLBCL)transformation of gastric MALT lymphoma.CASE SUMMARY A 66-year-old man was diagnosed with MALT lymphoma in the ascending colon by colonoscopy and biopsy.Two years later,esophagogastroduodenoscopy revealed chronic atrophic gastritis that was positive for H.pylori,and eradication treatment was administered.Two years and nine months later(at the age of 70),a new ulcerative lesion suggestive of MALT lymphoma appeared in the gastric body,and six months later,a similar lesion was also found in the fundus.One year later(4 years and 3 months after H.pylori eradication),at the age of 72,the lesion in the gastric body had become deeper and had propagated.A biopsy revealed a pathological diagnosis of DLBCL.Both MALT lymphoma lesions in the ascending colon and DLBCL lesions in the stomach were positive for the t(11;18)(q21;q21)/API2-MALT1 translocation,and trisomy 18q21 was also detected.After 6 courses of R-CHOP(rituximab,cyclophosphamide,doxorubicin,vincristine and prednisone)chemotherapy,all of the above lesions disappeared[complete remission(CR)],and CR has been maintained for more than 3 years.In addition,both the colonic and gastric lesions were proven to have the same clonality.CONCLUSION Because the patient had a MALT1 translocation with trisomy 18q21,it was thought that this gastric MALT lymphoma developed independently of H.pylori infection and progressed.
文摘[Objectives]This study was conducted to investigate the effects of slow-release nitrogen fertilizer on dry matter accumulation and translocation of summer maize.[Methods]With Zhoudan 9 as the test variety,six different treatment were set up:blank control(CK1),slow-release urea 75 kg/hm^(2)(C1),slow-release urea 150 kg/hm^(2)(C2),slow-release urea 225 kg/hm^(2)(C3),slow-release urea 300 kg/hm^(2)(C4)and ordinary urea 300 kg/hm^(2)(CK2),to study the change law of dry matter accumulation and translocation in summer maize.[Results]Treatment slow-release urea 225 kg/hm^(2)(C4)showed summer maize yield,dry matter translocation between organs,grain contribution rate and proportion of grain dry matter in the full ripe stage higher than other treatments.Considering the weight loss and cost factors,slow-release urea 225 kg/hm^(2)(C3)could be recommended as the fertilizing amount for summer maize.[Conclusions]This study provides theoretical reference for rational selection of fertilizers for reducing fertilizer application and increasing fertilizer efficiency,and for production of summer maize in Shajiang black soil region.
文摘Non-alcoholic fatty liver disease(NAFLD)is emerging as a common cause of chronic liver disease in children and adults.NAFLD can progress to steatohepa-titis and potentially even hepatocellular carcinoma.Early identification of pati-ents at risk for progressive disease is crucial for managing NAFLD.Recent studies have identified long noncoding RNAs(lncRNAs),circular RNAs,and microRNAs as playing important roles in the pathogenesis of NAFLD.These noncoding RNAs are involved in modulating several metabolic pathways such as hepatic glucose and lipid metabolism,oxidative stress,and even carcinogenesis.Elevated levels of lncARSR and lncRNA nuclear-enriched abundant transcript 1 have been found in patients with NAFLD.In addition,lncRNAs such as PRYP4-3 and RP11-128N14.5 can distinguish patients with NAFLD from healthy indi-viduals.Increased MEG3 expression has been observed in both NAFLD and non-alcoholic steatohepatitis,suggesting that it may help predict patients at risk for disease progression.With advances in transcriptomics,we may discover additional targets to help in the identification and prognostication of NAFLD.
文摘This study aims to investigate the level of soil pollution and the grade of accumulation of metals and heavy metals by wheat plants from the soil in different parts of the crop: root, stem, leaf, spike and grain. Sampling campaigns took place in February, April and July when wheat plants were at different growth stages. A number of eight soil samples and eight wheat plant samples were collected. The sampled wheat plant was taken at the same time and from the same place as the soil. Concentrations of Al (aluminium), Cr (chromium), Mn (manganese), Fe (iron), Ni (nickel), Co (cobalt), Cu (copper), Zn (zinc), Sr (strontium), Cd (cadmium) and Pb (lead) were determined by inductively coupled plasma mass spectrometry. Bioconcentration and translocation factors were calculated for the samples analysed.
文摘Dynamics of dry- or fresh-weight of fruit, peel photosynthetic rate and chlorophyll content, and the characteristics of translocation and distribution of radiolabelled assimilates from leaf or fruit were examined in developing satsuma mandarin (Citrus unshiu Marc. cv. Miyagawa wase) fruit from primary stage of fruit enlargement up to fruit full ripe. Change in fruit photosynthetic rate was some what related to the change in the chlorophyll content of peel. Fruit photosynthetic rate markedly declined as chlorophyll degradation occurred in the peel. Before full ripe stage of the fruit, photosynthates produced by a 14C-fed leaf were mainly distributed to juice sacs even during periods when dry matter accumulation in peel was more rapid than that in juice sacs. At the full ripe stage, peel photosynthetic rate approached zero and peel became the major sink of leaf photosynthates. Most of the peel assimilates, however, remained in situ for up to 48 h after feeding 14CO 2 to the fruit, only a small portion being transported to other parts of fruit. The percentage of fruit photosynthates exported decreased with fruit development and ripening, but the peak rate of export to juice sacs amount to as high as 12%. The sugar content and dry weights of peel and juice sacs in shaded fruit were lower than that in the control fruit. These results show that peel assimilate was mainly consumed in peel respiration and growth and thus the dependence on leaf photosynthates decreased. Part of this assimiate was used in sugar accumulation in juice sacs of fruit.
