The CP2 gene of TSV and N gene of YHDV in Gen Bank was anal...The aim is to develop a liquid chip technique to detect Taura syndrome virus( TSV) and yellow head disease virus( YHDV) on Penaeus orientalis simultaneously. The CP2 gene of TSV and N gene of YHDV in Gen Bank was analysed by using the software DNAStar 7. 0 to design the TSV-and YHDV-specific primers. The primers were labeled with biotin and subjected to amination modification. They were then coupled with fluorescence-coded microspheres and then used for hybridization with RT- PCR products of TSV and YHDV. The liquid chip detection technique for detection of TSV and YHDV was established by using BD FACSArray to detect fluorescence signal in the reaction system. This assay system had a high sensitivity to TSV and YHDV,with the detection of limit of 100 pg. Moreover,the assay was specific for the detection of TSV,YHDV and was not susceptible to cross with other viruses,including white spot syndrome virus( WSSV),spring viremia of carp virus( SVCV),infectious haematopoietic necrosis virus( IHNV). In conclusion,the liquid chip assay technique established in this study is highly sensitive and specific to TSV and YHDV detection. Moreover,it provides a novel,convenient and rapid approach for the detection of TSV and YHDV.展开更多
The Infectious hypodermal and hematopoietic necrosis virus(IHHNV) and Taura syndrome virus(TSV) are two important shrimp viruses in cultured shrimp in America.These two viruses were transmitted to China at the beginni...The Infectious hypodermal and hematopoietic necrosis virus(IHHNV) and Taura syndrome virus(TSV) are two important shrimp viruses in cultured shrimp in America.These two viruses were transmitted to China at the beginning of the 21st century.In this study,214 shrimp samples of Penaeus vannamei were collected from seven different areas of China and tested by PCR for IHHNV and TSV infection.The results showed that there were a high prevalence of IHHNV(65.42%) and low prevalence of TSV(3.27%) in the tested samples.Several samples were found to be co-infected with these two viruses.A 3 kb fragment of 7 positive IHHNV samples and a structure protein region(ORF2) of three TSV positive samples were amplified and sequenced.The sequence comparison indicated that both IHHNV and TSV sequenced in China have a low genetic variations compared with the prototype IHHNV and TSV from Hawaii.Phylogenetic analysis showed that TSV isolates were clustered into two groups,Asia and America group,which was genetically correlated to geographic distribution.展开更多

Taura syndrome virus(TSV) is one of the important pathogenic agents of Penaeus vannami,which causes prawn infection with serious disease in China in recent years.245 samples collected from 26 farms of Zhangzhou,Xiamen...Taura syndrome virus(TSV) is one of the important pathogenic agents of Penaeus vannami,which causes prawn infection with serious disease in China in recent years.245 samples collected from 26 farms of Zhangzhou,Xiamen,Shenzhen,Yangjiang,Ningbo and Guangzhou.were detected for TSV with RT-PCR,the positive rates were 100%,33.3%,40.7%,50% and 40%,respectively.Southern blotting and nucleotide sequencing were performed to confirm the specificity of the amplified RT-PCR products.The homology of 231bp nucleotide sequences reached 98.3%-100% when analysed and compared with the GenBank using the BLAST program.Phylogenetic analyses clustered the TSV isolates into two groups: one contained USA and Chinese Taiwan isolates;the other contained Vietnam isolate(YN1) and 21 isolates of China's Mainland.The 21 China isolates clustered into one branch,while the YN1 distributed in another branch.5 strains from Guangzhou and 2 strains from Shenzhen constituted one subcluster of the China branch.展开更多
