目的构建稳定表达的tdTomato转基因小鼠并建系,观察其细胞及组织的荧光表达水平以及其干细胞共培养后的荧光表达水平。方法使用Gateway clone技术和DNA显微注射法构建含有tdTomato表达载体的受精卵,移植至假孕母鼠体内,待其自然生产。... 目的构建稳定表达的tdTomato转基因小鼠并建系,观察其细胞及组织的荧光表达水平以及其干细胞共培养后的荧光表达水平。方法使用Gateway clone技术和DNA显微注射法构建含有tdTomato表达载体的受精卵,移植至假孕母鼠体内,待其自然生产。使用双向鉴定法鉴定并挑取饲养阳性小鼠并建系。另提取tdTomato阳性小鼠的神经干细胞分别与eGFP小鼠的骨髓间充质干细胞、原代神经干细胞共培养。结果成功构建了tdTomato转基因小鼠并建系,与野生型小鼠相比,tdTomato转基因小鼠生化指标和生长性状无明显差异,其组织切片及脑源神经干细胞均可观察到红色荧光。示踪实验表明注射部位可观察到明显的荧光成像,共培养实验可以清晰的观察到细胞分化后荧光的稳定表达。结论tdTomato转基因小鼠的组织和细胞能稳定表达红色荧光,可以利用荧光鼠的自发荧光特性,研究干细胞共培养后的分化方向。 展开更多

Cell fusion in the liver, revisited

The liver contains a consider... There is wide agreement that cell fusion is a physiological process in cells in mammalian bone, muscle and placenta. In other organs, such as the cerebellum, cell fusion is controversial. The liver contains a considerable number of polyploid cells: They are commonly believed to originate by genome endoreplication, although the contribution of cell fusion to polyploidization has not been excluded. Here, we address the topic of cell fusion in the liver from a historical point of view. We discuss experimental evidence clearly supporting the hypothesis that cell fusion occurs in the liver, specifically when bone marrow cells were injected into mice and shown to rescue genetic hepatic degenerative defects. Those experiments-carried out in the latter half of the last century-were initially interpreted to show "transdifferentiation", but are now believed to demonstrate fusion between donor macrophages and host hepatocytes, raising the possibility that physiologically polyploid cells, such as hepatocytes, could originate, at least partially, through homotypic cell fusion. In support of the homotypic cell fusion hypothesis, we present new data generated using a chimera-based model, a much simpler model than those previously used. Cell fusion as a road to polyploidization in the liver has not been extensively investigated, and its contribution to a variety of conditions, such as viral infections, carcinogenesis and aging, remains unclear. 展开更多
