Caseous lymphaden...Caseous lymphadenitis (CL) is a chronic infectious disease caused by Corynebacterium pseudotuberculosis (C. pseudotuberculosis) that is transmitted mainly by contact with contaminated exudate wounds. Caseous lymphadenitis affects different species of animals and is considered an occupational zoonotic disease. CL is responsible for important economic losses in the small ruminant industry, which include decreased production, damaged quality of milk and wool, reproductive disorders, total or partial confiscation of carcasses, and depreciation of the skin. Caseous lymphadenitis disease can present in two clinical pictures: a skin or superficial picture and a visceral or deep picture. The presumptive diagnosis of CL in the skin is based on the exploration of superficial lymph nodes. These have little value in diagnosing visceral CL, where the main sign is emaciation. The prevention and control of CL involve identifying the skin condition and debriding the abscesses. In addition to the treatment of superficial lesions, the animal facilities must be repaired and adapted. To avoid exposure of negative herds to CL, new animals must be subjected to observation to identify any of the clinical manifestations. Another form of control is vaccination, though only some countries have commercial vaccines. General information on the etiological agent and its characteristics can be used to improve the understanding of this disease.展开更多

T...Cysticercus tenuicollis is a metacestode stage of Taenia hydatigena residing in the visceral organs of small ruminants and its effects cause economic losses due to condemnation of visceral organs in slaughterhouses. This study reports on the prevalence of Cysticercus tenuicollis in small ruminants slaughtered in Lira Municipal Abattoir. A total of 70 sheep and 70 goats were sampled and examined after slaughter for the presence of Cysticercus tenuicollis in the visceral organs using standard meat inspection procedures. An abattoir data collection sheet was used to record data, which was then analysed in SPSS version 20. The study revealed 51.4% of goats and 68.6% of sheep to be infested with Cysticercus tenuicollis. Following the district of origin for the small ruminants, Otuke district had the highest prevalence of the parasite (82.6%), followed by Kole (72.4%), Alebtong (66.7%), Apac (59.4%) and Lira (31.4%). Cysticercus tenuicollis was more frequently seen in the intestines (57.8%) of sheep and goats than in any other visceral organs. The liver had a prevalence of 20.7%, the lungs 17.8%, the kidneys 3.5% and the fetal sac;(15.7%) in goats and (12.9%) in sheep. Cysticercus tenuicollis was detected in 68.8% of females and 58.8% of male small ruminants. It is recommended to routinely deworm goats and sheep by farmers most especially in districts with the highest prevalence of Cysticercus tenuicollis infestation. The sale of the condemned parts of sheep and goats and the feeding of dogs on these contaminated materials should be stopped forthwith. It is also being suggested that a national study be carried out to determine the prevalence and economic impact of Cysticercus tenuicollis in small ruminants.展开更多
The development of receptive endometrium(RE) from pre-receptive endometrium(PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epith...The development of receptive endometrium(RE) from pre-receptive endometrium(PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epithelium undergo a series of significant changes, including cell proliferation and apoptosis. However, the molecular mechanisms are not yet fully understood. In this study, a higher circRNA3669 level was observed in PE than in RE of goats. Functional assays revealed that this overexpression promoted the proliferation of goat endometrial epithelial cells(GEECs) by activating the expression of genes related to the PI3K/AKT-mTOR and MAPK pathways,thereby inhibiting apoptosis in vitro. Furthermore, circRNA3669 functioned as a competing endogenous RNA(ceRNA) to upregulate Reticulocalbin-2(RCN2) expression at the post-transcriptional level by interacting with and downregulating miR-26a in GEECs. In addition, RCN2, which is highly expressed in the PE of goats, was found to be regulated by β-estradiol(E2) and progesterone(P4). Our results demonstrated that RCN2 also affected the key proteins PI3K, AKT, mTOR, JNK, and P38 in the PI3K/AKT-mTOR and MAPK pathways, thereby facilitating GEECs proliferation and suppressing their apoptosis in vitro. Collectively, we constructed a new circRNA3669-miR-26aRCN2 regulatory network in GEECs, which further provides strong evidence that circRNA could potentially play a crucial regulatory role in the development of RE in goats.展开更多

In this study, the polymorphisms of insulin-like growth factor 1 (IGF1) gene in 561 goats of ten breeds were detected by polymerase chain reaction (PCR)-single strand conformation polymorphism (SSCP) and their association with litter size and birth weight in three breeds were investigated. The effects of IGF1 polymorphisms on the breeding value for litter size and birth weight were examined using least square methods. Two deletions (CA) were detected in the microsatellite and two mutations (A1637G, T1640C) were found in 5′-flanking regulatory region. No significant association between the polymorphisms in 5′-flanking region of IGF1 and birth weight was found in the three breeds of goats. In Gulin Ma goats, two polymorphisms were found to affect litter size traits. In Chuandong White goats and Guizhou White goats, no significant difference (P0.05) in litter size between goats carrying different genotypes was observed. Further evaluation and confirmation studies in more goat populations with larger sample sizes are necessary.展开更多

An animal model with fluorosis was constructed, and the fluorescent probe technique of DPH was used to measue the erythrocytic membrane liquidity. At the same time, the contents of MDA and the activity of SOD in serum were measured. The results showed that the erythrocytic membrane liquidity in the control group and flurosis group were 5.6742 ± 0.4417 and 3.7248 ± 0.4521 (P 〈0.01) respectively, the contents of MDA in serum were 2.0408 ± 0.198 and 4.494± 0.438 (P 〈0.01) respectively, the activities of SOD were 175.638 ± 22.201 and 113.714 ± 34.258 (P 〈0.01) respectively. The correlation analysis indicated that the relation between the activity of SOD and the liquidity of erythrocytic membrane was positive correlation ( r= 0.7321, P 〈0.05), whereas the relation between the contents of MDA and the liquidity of erythrocytic membrane was negative correlation (r = -0.6438, P 〈0.01). The erythrocytic membrane liquidity decreased in milk goat with fluorosis, which played a role in the occurrence and development of the fluorosis. There was correlation among the erythrocytic membrane liquidity and the contents of MDA, the activity of SOD.展开更多
Background: To understand the impact of feeding a high-concentrate diet to mid-lactating goats for a long time on liver metabolism and inflammatory response, two dimensional polyacrylamide gel electrophoresis(2-DE)...Background: To understand the impact of feeding a high-concentrate diet to mid-lactating goats for a long time on liver metabolism and inflammatory response, two dimensional polyacrylamide gel electrophoresis(2-DE) and real-time PCR method were employed to detect proteins differentially expressed in liver and their m RNAs expression in goats fed high concentrate diet(HC) or low concentrate diet(LC). Twelve lactating dairy goats were randomly assigned to either a HC diet group(65 % concentrate of dry matter; n = 6) or a LC diet group(35 % concentrate of dry matter; n = 6) for 10 wk.Results: Twenty differentially expressed spots(≥2.0-fold changes) in the hepatic tissues were excised and successfully identified using MALDI TOF/TOF. Of these, 8 proteins were up-regulated, while the rest 12 proteins were down-regulated in HC goats compared to LC. Differential expressed proteins including alpha enolase 1(ENO1), glutamate dehydrogenase 1(GLUD1), glutathione S-transferase A1(GSTA1), ATP synthase subunit 5β(ATP5β), superoxide dismutase [Cu-Zn](SOD1), cytochrom c oxidase subunit Via(COX6A1) and heat shock protein 60(HSP60) were further verified by real-time PCR and/or western blot at m RNA or protein expression level. Consistent with the 2-DE results, a significant decrease of β-actin protein expression and SOD enzyme activity was observed in liver of HC goats(P 〈 0.05), while ENO1 protein expression was significantly up-regulated in HC compared to LC goats(P 〈 0.05). However, western blot analysis did not show a significant difference of hepatic HSP60 protein between HC and LC group, which did not match the decrease of HSP60 content detected by 2-DE analysis. Real-time PCR showed that glutathione S-transferase P1(GSTP1) and SOD1 m RNA expression was significantly decreased in liver of HC goats, while cytochrom c oxidase(COX3) and ATPase 8(ATP8) m RNAs expression were markedly increased compared to LC(P 〈 0.05). Gene Ontology(GO) analysis revealed that HC diet resulted in altered expression of proteins related to catalytic and mitochondrial metabolism in the liver, and may increase the stress response with up-regulating the expression of differentiation 14(CD14) cluster and serum amyloid A(SAA) as well as C-reactive protein(CRP) in the liver.Conclusions: These results suggest that feeding high concentrate diet to lactating goats for 10 wk leads to the activation of the inflammatory response, and decreases the anti-oxidant capacity, and subsequently impairs the mitochondrial function in the liver.展开更多

However, its use as immunostimulant in the prevention of Peste des petits ruminants (PPR) in goats has not been investigated. In this study, the use of Levamisole in enhancing immune response to PPR vaccination including its effects on weight gain was investigated among groups of goats (groups A, B, C and D). Virus neutralization test was used to determine the antibody profile in vaccinated goats while the total and differential leucocyte counts and body mass index (BMI) were determined using standard procedures. Goats in group A (Levamisole primed and PPR vaccinated) seroconverted to more than 3-folds of the initial pre-vaccination geometric mean titre (GMT) of neutralizing antibody beginning from second week post-vaccination and remained high throughout the period of the experiment. Similarly, there was significant increase (p 0.05) in BMI of animals in other groups throughout the period of the experiment. The results of this study further confirm the immunostimulatory effect of Levamisole when used in combination with a vaccine.展开更多
Background Mastitis not only deteriorates the composition or quality of milk,but also damages the health and pro-ductivity of dairy goats.Sulforaphane(SFN)is a phytochemical isothiocyanate compound with various pharma...Background Mastitis not only deteriorates the composition or quality of milk,but also damages the health and pro-ductivity of dairy goats.Sulforaphane(SFN)is a phytochemical isothiocyanate compound with various pharmacologi-cal effects such as anti-oxidant and anti-inflammatory.However,the effect of SFN on mastitis has yet to be elucidated.This study aimed to explore the anti-oxidant and anti-inflammatory effects and potential molecular mechanisms of SFN in lipopolysaccharide(LPS)-induced primary goat mammary epithelial cells(GMECs)and a mouse model of mastitis.Results In vitro,SFN downregulated the mRNA expression of inflammatory factors(tumor necrosis factor-α(TNF-α),interleukin(IL)-1βand IL-6),inhibited the protein expression of inflammatory mediators(cyclooxygenase-2(COX2),and inducible nitric oxide synthase(iNOS))while suppressing nuclear factor kappa-B(NF-κB)activation in LPS-induced GMECs.Additionally,SFN exhibited an antioxidant effect by increasing Nrf2 expression and nuclear translocation,up-regulating antioxidant enzymes expression,and decreasing LPS-induced reactive oxygen species(ROS)produc-tion in GMECs.Furthermore,SFN pretreatment promoted the autophagy pathway,which was dependent on the increased Nrf2 level,and contributed significantly to the improved LPS-induced oxidative stress and inflammatory response.In vivo,SFN effectively alleviated histopathological lesions,suppressed the expression of inflammatory factors,enhanced immunohistochemistry staining of Nrf2,and amplified of LC3 puncta LPS-induced mastitis in mice.Mechanically,the in vitro and in vivo study showed that the anti-inflammatory and anti-oxidative stress effects of SFN were mediated by the Nrf2-mediated autophagy pathway in GMECs and a mouse model of mastitis.Conclusions These results indicate that the natural compound SFN has a preventive effect on LPS-induced inflam-mation through by regulating the Nrf2-mediated autophagy pathway in primary goat mammary epithelial cells and a mouse model of mastitis,which may improve prevention strategies for mastitis in dairy goats.展开更多
Background:SCNT(somatic cell nuclear transfer)is of great significance to biological research and also to the livestock breeding.However,the survival rate of the SCNT cloned animals is relatively low compared to other...Background:SCNT(somatic cell nuclear transfer)is of great significance to biological research and also to the livestock breeding.However,the survival rate of the SCNT cloned animals is relatively low compared to other transgenic methods.This indicates the potential epigenetic variations between them.DNA methylation is a key marker of mammalian epigenetics and its alterations will lead to phenotypic differences.In this study,ASMT(acetylserotonin-Omethyltransferase)ovarian overexpression transgenic goat was produced by using SCNT.To investigate whether there are epigenetic differences between cloned and WT(wild type)goats,WGBS(whole-genome bisulfite sequencing)was used to measure the whole-genome methylation of these animals.Results:It is observed that the different m Cp G sites are mainly present in the intergenic and intronic regions between cloned and WT animals,and their CG-type methylation sites are strongly correlated.DMR(differentially methylated region)lengths are located around 1000 bp,mainly distributed in the exonic,intergenic and intronic functional domains.A total of 56 and 36 DMGs(differentially methylated genes)were identified by GO and KEGG databases,respectively.Functional annotation showed that DMGs were enriched in biological-process,cellularcomponent,molecular-function and other signaling pathways.A total of 10 identical genes related to growth and development were identified in GO and KEGG databases.Conclusion:The differences in methylation genes among the tested animals have been identified.A total of 10 DMGs associated with growth and development were identified between cloned and WT animals.The results indicate that the differential patterns of DNA methylation between the cloned and WT goats are probably caused by the SCNT.These novel observations will help us to further identify the unveiled mechanisms of somatic cell cloning technology,particularly in goats.展开更多

The major objective of this paper was to elucidate which genes and pathways were involved in the regulation of milk protein synthesis in the dairy goat mammary gland. Total 36 primiparous Guanzhong dairy goats were allotted in 12 groups according to their mammary development stages: days 90 and 150 of virgin, days 30, 90, and 150 of pregnancy, days 1, 10, 35, and 60 of lactation and days 3, 7, and 21 of involution (three animals per group). Mammary tissue RNA was isolated for quantitative real- time RT-PCR of four casein genes alpha-s 1 casein (CSN 1S 1 ), alpha-s2 casein (CSN 1 S2), beta-casein (CSN2) and casein kappa (CSN3), four whey protein genes lactoglobulin (LGB), laetalbumin (LALBA), laetofarrin (LTF), and Whey acidic protein (WAP) and the genes which were potentially to regulate dairy goat milk protein synthesis at the level of transcription or translation [prolactin receptor (PRLR), AKT1, signal transducers and activators of transcription 5 (STAT5), E74-Like Factor 5 (ELF5), eukaryotic translation initiation factor 4E binding protein 1 (EIF4E-BP1), S6kinase (S6K) and caveolin 1]. The results showed that all genes were up-regulated in lactation period. The expressions of PRLR, AKT1, STAT5, ELF5, and S6K were similar to mRNA expressions of milk proteins. Our results indicated that milk protein synthesis in dairy goat mammary gland was possibly regulated by these genes.展开更多

Recently, among adult animals and the resulting offspring, a large number of individuals with low levels of natural resistance have begun to appear, which negatively affects the productivity of adults, and growth and development, as well as the safety of the resulting offspring. In this regard, the aim of the study was to study the effects of full-fledged feeding of females during pregnancy on the level of natural resistance of the received goats. To achieve this goal, a number of modern physiological and biochemical research methods were used. According to the results obtained, it was established that with the organization of full-fledged feeding, the period of suckling of the goat develops normally and the resulting baby goats are born with increased levels of natural resistance and more massive. In the goats of the experimental group, these advantages persist steadily until the age of one year, and to a certain extent ensure the normal preservation of the received goats and their growth and development. This makes it possible for farmers to fatten goats unused in breeding from 8 - 9 months of age and donate them for meat as an additional economic supplement to production.展开更多

In the sequence of a 648 bp fragment representing intron 1, exon 2, and part of intron 2 of the MLPH mammalian pigmentation gene, we identified a novel g.469C> G mutation in intron 2, and genotyped it in 266 Nigerian goats using PCR-RFLP analysis. The C allele had frequencies of 0.9625, 0.9804 and 0.97405 in West African Dwarf (WAD),Sahel(SH) and Red Sokoto (RS) breeds, respectively. The G allele was the highest in WAD (0.0375), followed by RS (0.02595), and then SH (0.0196). Overall low FIS and FST and high Nm values demonstrate little differentiation within and among the goat breeds at this intronic locus. This g.469C> G polymorphism in MLPH gene is the first in any goat breed and also first in Nigerian goats. Our results suggest that this intronic SNP locus is maintained at Hardy-Weinberg equilibrium (P < 0.05) and the lack of association of this SNP with coat color may indicate its neutrality in goats.展开更多

Growth traits include birth weight (BW), one-month weight (1 MW), weaning weight (WW) and 6-month weight (6 MW). Also the milk production traits was monthly milk production on 2, 3, 4, 5th months (quantitative traits) and some milk composition included dry matter (%DM), fat (%F), and protein (%P) percentage on during of production (qualitative traits). There were no significant differences in sex, birth type and genetic group for BW. Also the effect of genetic groups on 1 MW, WW, and 6 MW was significant, while effects of sex and birth type on 1 MW, WW and 6 MW was not significant. Also MW (Mothe's weight) had no significant effect on BW, 1 MW, WW and 6 MW. The average of daily milk records was 0.48 kg for nativel and 1.1 kg for crossed goats. The maximum monthly milk records in two groups were in second month (native: 0.64 -4- 0.05, crossed: 1.44 ~: 0.12) and the minimum were in fifth month (native: 0.36 + 0.03, crossed: 0.77 -4- 0.05). %DM, %F and %P in 5th month were higher than other months in two groups. Effects of genetic groups on milk production and also on average of%F, %DM and %P were significant. This study showed that the crossbreeding has a significant effect on most milk production and growth traits in Iranian native goats.展开更多

This paper analyzes the influence of key weather variables (temperature and rainfall) and seasonal variations in daylength on daytime activity budgets. This study showed that the percentage of daytime spent in feeding by feral goats on Rum Island decreased with average monthly temperature, while the percentage of daytime spent in lying increased and rainfall had no significant effects on the percentage daytime spent in feeding or lying. The daylength varied greatly from January to December with the longest daylength being in June and July, and the shortest one in December and January. The amount of daytime spent feeding decreased from summer to winter, although the percentage of daytime spent feeding increased greatly from summer to winter. The seasonal variations in daylength seem to be an important factor in constraining feral goats' activity, especially in the cold and wet winters when forage biomass and quality are both at their lowest level. We discuss the influence of such constraining effects on the forage intake of these goats and their winter survival rate, as well as its implication for population regulation of these goats.展开更多
[Objective] The aim was to study the effect of the muscle histological characteristics of Haimen goats on meat quality.[Method] Twelve-month-old Haimen goats and Bore × Haimen F1(♂)were used as the materials t...[Objective] The aim was to study the effect of the muscle histological characteristics of Haimen goats on meat quality.[Method] Twelve-month-old Haimen goats and Bore × Haimen F1(♂)were used as the materials to systematically research the muscle histological properties of different sexes and muscles.[Result] The results revealed that the muscle fiber area and myofibril diameter,sarcomere length,light band length,dark band length,hall band length of quadriceps femoris of Haimen female goats were extremely significantly higher than male goats(P〈0.01);However,that of longissimus dorsi muscle was extremely significantly lower than male goats(P〈0.01).There was significant difference(P〈0.05)or extremely significant difference(P〈0.01)between muscle microstructure and ultrastructure among the same sex and different muscles.There were a large number of mitochondria,glycogen granules and fat droplet within muscle fiber from Haimen male goats,which indicated that oxidative metabolism rate was high,water retention was good and the meat was tender,succulent and distinct.[Conclusion] The study provided theoretical basis for breeding,development and utilization of Haimen goats and variety evaluation of local goats in China.展开更多
[Objective] This study aimed to investigate the genetic diversity of Macheng black goat and its correlation with heterosis.[Method] Ten microsatellite markers were selected for polymorphism investigation and statistic...[Objective] This study aimed to investigate the genetic diversity of Macheng black goat and its correlation with heterosis.[Method] Ten microsatellite markers were selected for polymorphism investigation and statistical analysis of Boer goat and Macheng black goat populations.[Result] The results showed that totally 175 alleles were found in 10 microsatellite loci; to be specific,the maximum number of detected alleles was 23,and the minimum number was 10; the effective number of alleles (Ne) was 6.4-18.1,with absolute difference value of 1.6-8.1 from the observed number of alleles.The highest gene frequency was 0.239 1 and the lowest was 0.002 7.The polymorphic information contents of all the ten microsatellite markers were above 0.95.The observed heterozygosity (Ho) ranged from 0.616 7 to 0.984 4 and the expected heterozygosity (He) ranged from 0.844 1 to 0.944 6.The average expected heterozygosity of Boer goat and Macheng black goat was respectively 0.894 0 and 0.906 7.Various body weight and body size indices of Boer goatxMacheng black goat hybrids were improved in varying degrees compared with Macheng black goat (with an increase range of 0.32%-30.06%).The average heterosis rates of body height and chest girth were relatively high,while average heterosis rate of body weight was relatively low.[Conclusion] The genetic distance between Boer goat and Macheng black goat was 0.379 5,which is consistent with the geographical distribution of Boer goat and Macheng black goat populations and is fully relevant to the heterosis of Boer goat × Macheng black goat hybrids,indicating that investigating polymorphism via microsatellite loci is one of the feasible means to predict and analyze heterosis between varieties.展开更多
[Objective] The aim of this study was to reveal the relationship between inihibin (INH) α precursor gene and seasonal reproduction of goats, and investigate the evolutionary conservation of INHα precursor gene. [ ...[Objective] The aim of this study was to reveal the relationship between inihibin (INH) α precursor gene and seasonal reproduction of goats, and investigate the evolutionary conservation of INHα precursor gene. [ Method] Cloning and sequence analysis of 5' flanking region and exon of inihibinα (INHE) precursor gene in twenty ewes between non-seasonal estrous breed (Haimen goats) and seasonal estrous breed (Anhui white goats) was analyzed in this study. [ Result] Compared with Anhui white goats, INHα precursor gene in Haimen goats had three SNP but no amino acid change, while its nucleotide homology was 99.7% and amino acid homology was 100%. The nucleotide homology of INHα precursor gene in goat, cattle, pig, person, chicken, horse, rat and dog ranged from 12.7% to 96.5%. [ Conclusion] INHα precursor gene tends to be highly conserved in species, and any change of nucleotide and amino acid maybe directly influence the function of the whole gene coding and regulation.展开更多

文摘[Objective] The paper aims to study the relationship between genetic polymorphisms of three gene loci and growth character of Haimen Goats to provide reference to promote Haimen Goat growth character and cultivate Haimen Goat new strain of fast-growth.[Method] We processed analysis and related statistical analysis on three gene loci of Haimen Goats with PCR-RFLP and PCR-SSCP.[Result] The results suggested that there were two genotypes in MSTN gene IntronⅡ,GH gene exonⅠ and GH gene exonⅡ in Haimen Goats.Different genotypes of these gene loci had different effects on relative growth character of Haimen Goats.[Conclusion] This study had a significant meaning about promoting Haimen Goat avoirdupois by marker assistant option.
文摘Objective:To investigate the effect of Moringa oleifera leaf extract on semen quality parameters in Teressa goat buck.Methods:A total of 25 semen samples from six bucks were selected for the study.Spermatozoa of 150×10^(6) were incubated in 0,300,500 and 700μg of leaf extract as groupⅠ,Ⅱ,Ⅲ and Ⅳ,respectively.Liquid stored semen samples were analysed for motility,viability,total sperm abnormalities,plasma membrane,acrosomal and nuclear integrities,seminal plasma intracellular enzymes[aspartate aminotransferase(AST),alanine aminotransferase(ALT)and lactate dehydrogenase(LDH)]and total antioxidant capacity(TAC)and spermatozoa malondialdehyde(MDA)and cholesterol efflux in comparison with the control group(groupⅠ)for up to 72 h.Results:Moringa oleifera leaf extract(500μg/150×10^(6) spermatozoa)-treated semen had significantly higher motility,viability,plasma membrane,acrosomal and nuclear integrities,and TAC(P<0.05),and had significantly lower total sperm abnormalities,AST,ALT,LDH,MDA production and cholesterol efflux compared to those in other Moringa oleifera leaf extract treated(300 or 700μg/150×10^(6) spermatozoa)and control groups at different hours of liquid storage(P<0.05).Semen quality parameters and TAC showed an increasing trend and total sperm abnormalities,MDA production,leakage of intra-cellular enzymes and cholesterol efflux showed a decreasing trend from group Ⅰto group Ⅲand then an opposite trend from group Ⅲ to group Ⅳ at different hours of liquid storage.Conclusions:Moringa oleifera leaf extract 500μg/150×10^(6) spermatozoa can be used for liquid semen preservation in Teressa goat at Andaman and Nicobar Islands.
文摘Caseous lymphadenitis (CL) is a chronic infectious disease caused by Corynebacterium pseudotuberculosis (C. pseudotuberculosis) that is transmitted mainly by contact with contaminated exudate wounds. Caseous lymphadenitis affects different species of animals and is considered an occupational zoonotic disease. CL is responsible for important economic losses in the small ruminant industry, which include decreased production, damaged quality of milk and wool, reproductive disorders, total or partial confiscation of carcasses, and depreciation of the skin. Caseous lymphadenitis disease can present in two clinical pictures: a skin or superficial picture and a visceral or deep picture. The presumptive diagnosis of CL in the skin is based on the exploration of superficial lymph nodes. These have little value in diagnosing visceral CL, where the main sign is emaciation. The prevention and control of CL involve identifying the skin condition and debriding the abscesses. In addition to the treatment of superficial lesions, the animal facilities must be repaired and adapted. To avoid exposure of negative herds to CL, new animals must be subjected to observation to identify any of the clinical manifestations. Another form of control is vaccination, though only some countries have commercial vaccines. General information on the etiological agent and its characteristics can be used to improve the understanding of this disease.
文摘Cysticercus tenuicollis is a metacestode stage of Taenia hydatigena residing in the visceral organs of small ruminants and its effects cause economic losses due to condemnation of visceral organs in slaughterhouses. This study reports on the prevalence of Cysticercus tenuicollis in small ruminants slaughtered in Lira Municipal Abattoir. A total of 70 sheep and 70 goats were sampled and examined after slaughter for the presence of Cysticercus tenuicollis in the visceral organs using standard meat inspection procedures. An abattoir data collection sheet was used to record data, which was then analysed in SPSS version 20. The study revealed 51.4% of goats and 68.6% of sheep to be infested with Cysticercus tenuicollis. Following the district of origin for the small ruminants, Otuke district had the highest prevalence of the parasite (82.6%), followed by Kole (72.4%), Alebtong (66.7%), Apac (59.4%) and Lira (31.4%). Cysticercus tenuicollis was more frequently seen in the intestines (57.8%) of sheep and goats than in any other visceral organs. The liver had a prevalence of 20.7%, the lungs 17.8%, the kidneys 3.5% and the fetal sac;(15.7%) in goats and (12.9%) in sheep. Cysticercus tenuicollis was detected in 68.8% of females and 58.8% of male small ruminants. It is recommended to routinely deworm goats and sheep by farmers most especially in districts with the highest prevalence of Cysticercus tenuicollis infestation. The sale of the condemned parts of sheep and goats and the feeding of dogs on these contaminated materials should be stopped forthwith. It is also being suggested that a national study be carried out to determine the prevalence and economic impact of Cysticercus tenuicollis in small ruminants.

文摘The development of receptive endometrium(RE) from pre-receptive endometrium(PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epithelium undergo a series of significant changes, including cell proliferation and apoptosis. However, the molecular mechanisms are not yet fully understood. In this study, a higher circRNA3669 level was observed in PE than in RE of goats. Functional assays revealed that this overexpression promoted the proliferation of goat endometrial epithelial cells(GEECs) by activating the expression of genes related to the PI3K/AKT-mTOR and MAPK pathways,thereby inhibiting apoptosis in vitro. Furthermore, circRNA3669 functioned as a competing endogenous RNA(ceRNA) to upregulate Reticulocalbin-2(RCN2) expression at the post-transcriptional level by interacting with and downregulating miR-26a in GEECs. In addition, RCN2, which is highly expressed in the PE of goats, was found to be regulated by β-estradiol(E2) and progesterone(P4). Our results demonstrated that RCN2 also affected the key proteins PI3K, AKT, mTOR, JNK, and P38 in the PI3K/AKT-mTOR and MAPK pathways, thereby facilitating GEECs proliferation and suppressing their apoptosis in vitro. Collectively, we constructed a new circRNA3669-miR-26aRCN2 regulatory network in GEECs, which further provides strong evidence that circRNA could potentially play a crucial regulatory role in the development of RE in goats.

文摘Research on the identity of genes and their relationship with traits of economic importance in farm animals could assist in the selection of livestock. In this study, the polymorphisms of insulin-like growth factor 1 (IGF1) gene in 561 goats of ten breeds were detected by polymerase chain reaction (PCR)-single strand conformation polymorphism (SSCP) and their association with litter size and birth weight in three breeds were investigated. The effects of IGF1 polymorphisms on the breeding value for litter size and birth weight were examined using least square methods. Two deletions (CA) were detected in the microsatellite and two mutations (A1637G, T1640C) were found in 5′-flanking regulatory region. No significant association between the polymorphisms in 5′-flanking region of IGF1 and birth weight was found in the three breeds of goats. In Gulin Ma goats, two polymorphisms were found to affect litter size traits. In Chuandong White goats and Guizhou White goats, no significant difference (P0.05) in litter size between goats carrying different genotypes was observed. Further evaluation and confirmation studies in more goat populations with larger sample sizes are necessary.
文摘To research the relation among the erythrocytic membrane.liquidity and the contents of MDA, the activity of SOD in serum of the milk goat with fluorosis. An animal model with fluorosis was constructed, and the fluorescent probe technique of DPH was used to measue the erythrocytic membrane liquidity. At the same time, the contents of MDA and the activity of SOD in serum were measured. The results showed that the erythrocytic membrane liquidity in the control group and flurosis group were 5.6742 ± 0.4417 and 3.7248 ± 0.4521 (P 〈0.01) respectively, the contents of MDA in serum were 2.0408 ± 0.198 and 4.494± 0.438 (P 〈0.01) respectively, the activities of SOD were 175.638 ± 22.201 and 113.714 ± 34.258 (P 〈0.01) respectively. The correlation analysis indicated that the relation between the activity of SOD and the liquidity of erythrocytic membrane was positive correlation ( r= 0.7321, P 〈0.05), whereas the relation between the contents of MDA and the liquidity of erythrocytic membrane was negative correlation (r = -0.6438, P 〈0.01). The erythrocytic membrane liquidity decreased in milk goat with fluorosis, which played a role in the occurrence and development of the fluorosis. There was correlation among the erythrocytic membrane liquidity and the contents of MDA, the activity of SOD.

文摘Background: To understand the impact of feeding a high-concentrate diet to mid-lactating goats for a long time on liver metabolism and inflammatory response, two dimensional polyacrylamide gel electrophoresis(2-DE) and real-time PCR method were employed to detect proteins differentially expressed in liver and their m RNAs expression in goats fed high concentrate diet(HC) or low concentrate diet(LC). Twelve lactating dairy goats were randomly assigned to either a HC diet group(65 % concentrate of dry matter; n = 6) or a LC diet group(35 % concentrate of dry matter; n = 6) for 10 wk.Results: Twenty differentially expressed spots(≥2.0-fold changes) in the hepatic tissues were excised and successfully identified using MALDI TOF/TOF. Of these, 8 proteins were up-regulated, while the rest 12 proteins were down-regulated in HC goats compared to LC. Differential expressed proteins including alpha enolase 1(ENO1), glutamate dehydrogenase 1(GLUD1), glutathione S-transferase A1(GSTA1), ATP synthase subunit 5β(ATP5β), superoxide dismutase [Cu-Zn](SOD1), cytochrom c oxidase subunit Via(COX6A1) and heat shock protein 60(HSP60) were further verified by real-time PCR and/or western blot at m RNA or protein expression level. Consistent with the 2-DE results, a significant decrease of β-actin protein expression and SOD enzyme activity was observed in liver of HC goats(P 〈 0.05), while ENO1 protein expression was significantly up-regulated in HC compared to LC goats(P 〈 0.05). However, western blot analysis did not show a significant difference of hepatic HSP60 protein between HC and LC group, which did not match the decrease of HSP60 content detected by 2-DE analysis. Real-time PCR showed that glutathione S-transferase P1(GSTP1) and SOD1 m RNA expression was significantly decreased in liver of HC goats, while cytochrom c oxidase(COX3) and ATPase 8(ATP8) m RNAs expression were markedly increased compared to LC(P 〈 0.05). Gene Ontology(GO) analysis revealed that HC diet resulted in altered expression of proteins related to catalytic and mitochondrial metabolism in the liver, and may increase the stress response with up-regulating the expression of differentiation 14(CD14) cluster and serum amyloid A(SAA) as well as C-reactive protein(CRP) in the liver.Conclusions: These results suggest that feeding high concentrate diet to lactating goats for 10 wk leads to the activation of the inflammatory response, and decreases the anti-oxidant capacity, and subsequently impairs the mitochondrial function in the liver.
文摘The use of Levamisole hydrochloride as an immunostimulant has been successful in the control of certain diseases such as Derzsy’s disease in chicken as well as in increasing lymphocytes and weight gain in pigs. However, its use as immunostimulant in the prevention of Peste des petits ruminants (PPR) in goats has not been investigated. In this study, the use of Levamisole in enhancing immune response to PPR vaccination including its effects on weight gain was investigated among groups of goats (groups A, B, C and D). Virus neutralization test was used to determine the antibody profile in vaccinated goats while the total and differential leucocyte counts and body mass index (BMI) were determined using standard procedures. Goats in group A (Levamisole primed and PPR vaccinated) seroconverted to more than 3-folds of the initial pre-vaccination geometric mean titre (GMT) of neutralizing antibody beginning from second week post-vaccination and remained high throughout the period of the experiment. Similarly, there was significant increase (p 0.05) in BMI of animals in other groups throughout the period of the experiment. The results of this study further confirm the immunostimulatory effect of Levamisole when used in combination with a vaccine.

文摘Background Mastitis not only deteriorates the composition or quality of milk,but also damages the health and pro-ductivity of dairy goats.Sulforaphane(SFN)is a phytochemical isothiocyanate compound with various pharmacologi-cal effects such as anti-oxidant and anti-inflammatory.However,the effect of SFN on mastitis has yet to be elucidated.This study aimed to explore the anti-oxidant and anti-inflammatory effects and potential molecular mechanisms of SFN in lipopolysaccharide(LPS)-induced primary goat mammary epithelial cells(GMECs)and a mouse model of mastitis.Results In vitro,SFN downregulated the mRNA expression of inflammatory factors(tumor necrosis factor-α(TNF-α),interleukin(IL)-1βand IL-6),inhibited the protein expression of inflammatory mediators(cyclooxygenase-2(COX2),and inducible nitric oxide synthase(iNOS))while suppressing nuclear factor kappa-B(NF-κB)activation in LPS-induced GMECs.Additionally,SFN exhibited an antioxidant effect by increasing Nrf2 expression and nuclear translocation,up-regulating antioxidant enzymes expression,and decreasing LPS-induced reactive oxygen species(ROS)produc-tion in GMECs.Furthermore,SFN pretreatment promoted the autophagy pathway,which was dependent on the increased Nrf2 level,and contributed significantly to the improved LPS-induced oxidative stress and inflammatory response.In vivo,SFN effectively alleviated histopathological lesions,suppressed the expression of inflammatory factors,enhanced immunohistochemistry staining of Nrf2,and amplified of LC3 puncta LPS-induced mastitis in mice.Mechanically,the in vitro and in vivo study showed that the anti-inflammatory and anti-oxidative stress effects of SFN were mediated by the Nrf2-mediated autophagy pathway in GMECs and a mouse model of mastitis.Conclusions These results indicate that the natural compound SFN has a preventive effect on LPS-induced inflam-mation through by regulating the Nrf2-mediated autophagy pathway in primary goat mammary epithelial cells and a mouse model of mastitis,which may improve prevention strategies for mastitis in dairy goats.

文摘Background:SCNT(somatic cell nuclear transfer)is of great significance to biological research and also to the livestock breeding.However,the survival rate of the SCNT cloned animals is relatively low compared to other transgenic methods.This indicates the potential epigenetic variations between them.DNA methylation is a key marker of mammalian epigenetics and its alterations will lead to phenotypic differences.In this study,ASMT(acetylserotonin-Omethyltransferase)ovarian overexpression transgenic goat was produced by using SCNT.To investigate whether there are epigenetic differences between cloned and WT(wild type)goats,WGBS(whole-genome bisulfite sequencing)was used to measure the whole-genome methylation of these animals.Results:It is observed that the different m Cp G sites are mainly present in the intergenic and intronic regions between cloned and WT animals,and their CG-type methylation sites are strongly correlated.DMR(differentially methylated region)lengths are located around 1000 bp,mainly distributed in the exonic,intergenic and intronic functional domains.A total of 56 and 36 DMGs(differentially methylated genes)were identified by GO and KEGG databases,respectively.Functional annotation showed that DMGs were enriched in biological-process,cellularcomponent,molecular-function and other signaling pathways.A total of 10 identical genes related to growth and development were identified in GO and KEGG databases.Conclusion:The differences in methylation genes among the tested animals have been identified.A total of 10 DMGs associated with growth and development were identified between cloned and WT animals.The results indicate that the differential patterns of DNA methylation between the cloned and WT goats are probably caused by the SCNT.These novel observations will help us to further identify the unveiled mechanisms of somatic cell cloning technology,particularly in goats.

文摘The lactating mammary gland is a prodigious protein-producing factory, but the milk protein synthesis mechanisms are not well understood. The major objective of this paper was to elucidate which genes and pathways were involved in the regulation of milk protein synthesis in the dairy goat mammary gland. Total 36 primiparous Guanzhong dairy goats were allotted in 12 groups according to their mammary development stages: days 90 and 150 of virgin, days 30, 90, and 150 of pregnancy, days 1, 10, 35, and 60 of lactation and days 3, 7, and 21 of involution (three animals per group). Mammary tissue RNA was isolated for quantitative real- time RT-PCR of four casein genes alpha-s 1 casein (CSN 1S 1 ), alpha-s2 casein (CSN 1 S2), beta-casein (CSN2) and casein kappa (CSN3), four whey protein genes lactoglobulin (LGB), laetalbumin (LALBA), laetofarrin (LTF), and Whey acidic protein (WAP) and the genes which were potentially to regulate dairy goat milk protein synthesis at the level of transcription or translation [prolactin receptor (PRLR), AKT1, signal transducers and activators of transcription 5 (STAT5), E74-Like Factor 5 (ELF5), eukaryotic translation initiation factor 4E binding protein 1 (EIF4E-BP1), S6kinase (S6K) and caveolin 1]. The results showed that all genes were up-regulated in lactation period. The expressions of PRLR, AKT1, STAT5, ELF5, and S6K were similar to mRNA expressions of milk proteins. Our results indicated that milk protein synthesis in dairy goat mammary gland was possibly regulated by these genes.
文摘The traditional way of breeding pastured animals is year-round pasture keeping without any additional fertilizing. Recently, among adult animals and the resulting offspring, a large number of individuals with low levels of natural resistance have begun to appear, which negatively affects the productivity of adults, and growth and development, as well as the safety of the resulting offspring. In this regard, the aim of the study was to study the effects of full-fledged feeding of females during pregnancy on the level of natural resistance of the received goats. To achieve this goal, a number of modern physiological and biochemical research methods were used. According to the results obtained, it was established that with the organization of full-fledged feeding, the period of suckling of the goat develops normally and the resulting baby goats are born with increased levels of natural resistance and more massive. In the goats of the experimental group, these advantages persist steadily until the age of one year, and to a certain extent ensure the normal preservation of the received goats and their growth and development. This makes it possible for farmers to fatten goats unused in breeding from 8 - 9 months of age and donate them for meat as an additional economic supplement to production.
文摘Pigmentation plays important adaptation and physiological efficiency roles in animals. In the sequence of a 648 bp fragment representing intron 1, exon 2, and part of intron 2 of the MLPH mammalian pigmentation gene, we identified a novel g.469C> G mutation in intron 2, and genotyped it in 266 Nigerian goats using PCR-RFLP analysis. The C allele had frequencies of 0.9625, 0.9804 and 0.97405 in West African Dwarf (WAD),Sahel(SH) and Red Sokoto (RS) breeds, respectively. The G allele was the highest in WAD (0.0375), followed by RS (0.02595), and then SH (0.0196). Overall low FIS and FST and high Nm values demonstrate little differentiation within and among the goat breeds at this intronic locus. This g.469C> G polymorphism in MLPH gene is the first in any goat breed and also first in Nigerian goats. Our results suggest that this intronic SNP locus is maintained at Hardy-Weinberg equilibrium (P < 0.05) and the lack of association of this SNP with coat color may indicate its neutrality in goats.
文摘The performance of some growth and milk traits of selected Qomi native (Lori black) goats and hybrids of saanen with native goats during 2008-2010 was compared. Growth traits include birth weight (BW), one-month weight (1 MW), weaning weight (WW) and 6-month weight (6 MW). Also the milk production traits was monthly milk production on 2, 3, 4, 5th months (quantitative traits) and some milk composition included dry matter (%DM), fat (%F), and protein (%P) percentage on during of production (qualitative traits). There were no significant differences in sex, birth type and genetic group for BW. Also the effect of genetic groups on 1 MW, WW, and 6 MW was significant, while effects of sex and birth type on 1 MW, WW and 6 MW was not significant. Also MW (Mothe's weight) had no significant effect on BW, 1 MW, WW and 6 MW. The average of daily milk records was 0.48 kg for nativel and 1.1 kg for crossed goats. The maximum monthly milk records in two groups were in second month (native: 0.64 -4- 0.05, crossed: 1.44 ~: 0.12) and the minimum were in fifth month (native: 0.36 + 0.03, crossed: 0.77 -4- 0.05). %DM, %F and %P in 5th month were higher than other months in two groups. Effects of genetic groups on milk production and also on average of%F, %DM and %P were significant. This study showed that the crossbreeding has a significant effect on most milk production and growth traits in Iranian native goats.

文摘[Objective] The aim of this study was to explore the genetic law of litter size of Inner Mongolia Cashmere Goat.[Method] A total of 22 721 litter records from 3 044 mothers were statistically analyzed.[Result] As shown by the least square variance analysis,the sex,parity and birth type had significant effect on lamb birth weight.As indicated by restricted maximum likelihood method,the parity,population and maternal effect influenced litter size significantly.The birth weight heritability of single lamb,twins and total was 0.16,0.40 and 0.17,respectively.[Conclusion] The study provided a certain basis for the establishment of Inner Mongolia Cashmere Goat line with superior reproductive performance.
文摘Daytime activity budgets of feral goats ( Capra hircus ) were studied in 1981, 1982 and 2000 on the Isle of Rum, northwest Scotland. This paper analyzes the influence of key weather variables (temperature and rainfall) and seasonal variations in daylength on daytime activity budgets. This study showed that the percentage of daytime spent in feeding by feral goats on Rum Island decreased with average monthly temperature, while the percentage of daytime spent in lying increased and rainfall had no significant effects on the percentage daytime spent in feeding or lying. The daylength varied greatly from January to December with the longest daylength being in June and July, and the shortest one in December and January. The amount of daytime spent feeding decreased from summer to winter, although the percentage of daytime spent feeding increased greatly from summer to winter. The seasonal variations in daylength seem to be an important factor in constraining feral goats' activity, especially in the cold and wet winters when forage biomass and quality are both at their lowest level. We discuss the influence of such constraining effects on the forage intake of these goats and their winter survival rate, as well as its implication for population regulation of these goats.

文摘[Objective] The aim was to study the effect of the muscle histological characteristics of Haimen goats on meat quality.[Method] Twelve-month-old Haimen goats and Bore × Haimen F1(♂)were used as the materials to systematically research the muscle histological properties of different sexes and muscles.[Result] The results revealed that the muscle fiber area and myofibril diameter,sarcomere length,light band length,dark band length,hall band length of quadriceps femoris of Haimen female goats were extremely significantly higher than male goats(P〈0.01);However,that of longissimus dorsi muscle was extremely significantly lower than male goats(P〈0.01).There was significant difference(P〈0.05)or extremely significant difference(P〈0.01)between muscle microstructure and ultrastructure among the same sex and different muscles.There were a large number of mitochondria,glycogen granules and fat droplet within muscle fiber from Haimen male goats,which indicated that oxidative metabolism rate was high,water retention was good and the meat was tender,succulent and distinct.[Conclusion] The study provided theoretical basis for breeding,development and utilization of Haimen goats and variety evaluation of local goats in China.
文摘[Objective] This study aimed to investigate the genetic diversity of Macheng black goat and its correlation with heterosis.[Method] Ten microsatellite markers were selected for polymorphism investigation and statistical analysis of Boer goat and Macheng black goat populations.[Result] The results showed that totally 175 alleles were found in 10 microsatellite loci; to be specific,the maximum number of detected alleles was 23,and the minimum number was 10; the effective number of alleles (Ne) was 6.4-18.1,with absolute difference value of 1.6-8.1 from the observed number of alleles.The highest gene frequency was 0.239 1 and the lowest was 0.002 7.The polymorphic information contents of all the ten microsatellite markers were above 0.95.The observed heterozygosity (Ho) ranged from 0.616 7 to 0.984 4 and the expected heterozygosity (He) ranged from 0.844 1 to 0.944 6.The average expected heterozygosity of Boer goat and Macheng black goat was respectively 0.894 0 and 0.906 7.Various body weight and body size indices of Boer goatxMacheng black goat hybrids were improved in varying degrees compared with Macheng black goat (with an increase range of 0.32%-30.06%).The average heterosis rates of body height and chest girth were relatively high,while average heterosis rate of body weight was relatively low.[Conclusion] The genetic distance between Boer goat and Macheng black goat was 0.379 5,which is consistent with the geographical distribution of Boer goat and Macheng black goat populations and is fully relevant to the heterosis of Boer goat × Macheng black goat hybrids,indicating that investigating polymorphism via microsatellite loci is one of the feasible means to predict and analyze heterosis between varieties.

文摘[Objective] The aim of this study was to reveal the relationship between inihibin (INH) α precursor gene and seasonal reproduction of goats, and investigate the evolutionary conservation of INHα precursor gene. [ Method] Cloning and sequence analysis of 5' flanking region and exon of inihibinα (INHE) precursor gene in twenty ewes between non-seasonal estrous breed (Haimen goats) and seasonal estrous breed (Anhui white goats) was analyzed in this study. [ Result] Compared with Anhui white goats, INHα precursor gene in Haimen goats had three SNP but no amino acid change, while its nucleotide homology was 99.7% and amino acid homology was 100%. The nucleotide homology of INHα precursor gene in goat, cattle, pig, person, chicken, horse, rat and dog ranged from 12.7% to 96.5%. [ Conclusion] INHα precursor gene tends to be highly conserved in species, and any change of nucleotide and amino acid maybe directly influence the function of the whole gene coding and regulation.
