The Main Biological Hazards in Animal Biosafety Level 2 Facilities and Strategies for Control

Concern about the biological hazards involved in microbiological research, especially research involving laboratory animals, has increased in recent years. Working in an animal biosafety level 2 facility （ABSL-2）, commonly used for research on infectious diseases, poses various biological hazards. Here, the regulations and standards related to laboratory biosafety in China are introduced, the potential biological hazards present in ABSL-2 facilities are analyzed, and a series of strategies to control the hazards are presented.

Estimation of water volume transports through the main straits of the East China Sea

The structure of current speed and the variability of volume transports of the Kuroshio in the Tokara-kaikyo and Osumi-kaikyo are discussed on the basis of data of KER in the period from 1977 to 1984. The average geostrophic transport through these two straits is estimated to be 24. 5×106 m3/s and only 1/12 of the transport is through the Osumi-kaiky5. Countercurrents on both sides of the Kuroshio trunk are observed in the Tokara-kaikyo. Calculation indicates that the average geostrophic current speed is less than the GEK current speed, systematically. On the basis of the current measurements, the northward transports through the Taiwan Strait in winter and summer are estimated to be 1. 05×106and 3. 16×106m3/s, respectively. From Chu's data (1976) the average transport of the Kuroshio flowing into the East China Sea passing through the passage east of Taiwan is about 29. 3×106m3/s. From Miita and Ogawa's data (1984) the average transport through the Tsushima-kaikyo is 3. 6×106m3/s. Thus the volume transports through the above four straits are roughly in balance, the total outflowing transport is slightly larger than the total inflowing transport. The possible reasons resulting in the difference of transports are also discussed.

Regional homogeneity of intrinsic brain activity related to the main alexithymia dimensions

Background Alexithymia is a multidimensional personality construct.Objective This study aims to investigate the neuronal correlates of each alexithymia dimension by examining the regional homogeneity(ReHo) of intrinsic brain activity in a resting situation.Methods From university freshmen, students with alexithymia and non-alexlthymia were recruited. Their alexithymic traits were assessed using the Toronto Alexithymia Scale-20. The ReHo was examined using a resting-state functional MRI approach.Results This study suggests significant gro叩 differences in ReHo in multiple brain regions distributed in the frontal lobe, parietal lobe, temporal lobe, occipital lobe and insular cortex. However, only the ReHo in the insula was positively associated with difficulty identifying feelings, a main dimension of alexithymia. The ReHo in the lingual gyrus,precentral gyrus and postcentral gyrus was positively associated with difficulty describing feelings in participants with alexithymia. Lastly, the ReHo in the right dorsomedial prefrontal cortex(DMPFC_R) was negatively related to the externally oriented thinking style of participants with alexithymia.Conclusion In conclusion, these results suggest that the main dimensions of alexithymia are correlated with specific brain regions' function, and the role of the insula,lingual gyrus, precentral gyrus, postcentral gyrus and DMPFC_R in the neuropathology of alexithymia should be further investigated.

Discovery of and Preliminary studies on a Rapid-Leafing Rice Genotype at the Vegetative Growth Stage

The leafing rates of fourteen rice varieties were measured in a sowing-time experiment, and a rapid leafing genotype at the vegetative growth stage was discovered in an indica variety Yanhui 559. The leaf number on the main culm of Yanhui 559 was always 4-5 leaves more than that of Lemont, and the leafing rate of Yanhui 559 was significantly higher than that of Lemont based on similar growth durations from sowing to heading. Furthermore, the difference of the leafing rate was significant at the vegetative growth stage, but not distinctive at the panicle initiation stage. Genetic analysis of the leafing rates in the two backcross populations of Yanhui 559 and Lemont showed that major and quantitative genes controlled the expression of rapid leafing character. Based on results of investigation for some plants with similar growth durations in the backcrossing populations, the rapid leafing genotypes exhibited earlier tillering and more tiller numbers per plant, and its yield components including the number of panicles per plant and number of grains per panicle were superior to those of the slow leafing genotypes. Further research and application feasibility of the rapid leafing genotype in breeding were discussed.

Histidine decarboxylase and urinary methylimidazoleacetic acid in gastric neuroendocrine cells and tumours

AIM: To study histidine decarboxylase(HDC) expression in normal and neoplastic gastric neuroendocrine cells in relationship to the main histamine metabolite. METHODS: Control tissues from fundus(n = 3) and corpus(n = 3) mucosa of six patients undergoing operations for gastric adenocarcinoma, biopsy and/or gastric surgical specimens from 64 patients with primary gastric neuroendocrine tumours(GNETs), as well as metastases from 22 of these patients, were investigated using conventional immunohistochemistry and double immunofluorescence with commercial antibodies vs vesicular monoamine transporter 2(VMAT-2), HDC and ghrelin. The urinary excretion of the main histamine metabolite methylimidazoleacetic acid(U-Me Im AA) was determined using highperformance liquid chromatography in 27 of the 64 patients.RESULTS: In the gastric mucosa of the control tissues, co-localization studies identified neuroendocrine cells that showed immunoreactivity only to VMAT-2 and others with reactivity only to HDC. A third cellpopulation co-expressed both antigens. There was no co-expression of HDC and ghrelin. Similar results were obtained in the foci of neuroendocrine cell hyperplasia associated with chronic atrophic gastritis type A and also in the tumours. The relative incidence of the three aforementioned markers varied in the tumours that were examined using conventional immunohistochemistry. All of these GNETs revealed both VMAT-2 and HDC immunoreactivity, and their metastases showed an immunohistochemical pattern and frequency similar to that of their primary tumours. In four patients, increased U-Me Im AA excretion was detected, but only two of the patients exhibited related endocrine symptoms. CONCLUSION: Human enterochromaffin-like cells appear to partially co-express VMAT-2 and HDC. Coexpression of VMAT-2 and HDC might be required for increased histamine production in patients with GNETs.
