METHODS:Assays based on the serovar-specific DNA sequence of S. enteritidis from GenBank, and a serovar-specific real-time, fluorescence-based quantitative polymerase chain reaction (FQ-PCR) were developed for the detection of S. enteritidis. We used this assay to detect genomic DNA of S. enteritidis in the blood and the internal organs, including heart, liver, spleen, kidney, pancreas, and gallbladder, from mice after oral challenge at different time points respectively.RESULTS:The results showed that the spleen was positive at 12 h post inoculation (PI), and the blood was at 14 h PI. The organism was detected in the liver and heart at 16 h PI, the pancreas was positive at 20 h PI, and the final organs to show positive results were the kidney and gallbladder at 22 h PI. The copy number of S. enteritidis DNA in each tissue reached a peak at 24-36 h PI, with the liver and spleen containing high concentrations of S. enteritidis, whereas the blood, heart, kidney, pancreas, and gallbladder had low concentrations. S. enteritidis populations began to decrease and were not detectable at 3 d PI, but were still present up to 12 d PI in the gallbladder, 2 wk for the liver, and 3 wk for the spleen without causing apparent symptoms. CONCLUSION:The results provided significant data for understanding the life cycle of S. enteritidis in the internal organs, and showed that the liver and spleen may be the primary sites for setting itself up as a commensa over a long time after oral challenge. Interestingly, it may be the first time reported that the gallbladder is a site of carriage for S. enteritidis over a 12 d period. This study will help to understand the mechanisms of action of S. enteritidis infection in vivo.展开更多

In our study, a porcine cDNA microarray representing 3 358 pig ge...Pig (Sus scrofa) fat accumulation can be reduced by feeding with high dosages of clenbuterol, but the molecular mechanism has not yet been explained. In our study, a porcine cDNA microarray representing 3 358 pig genes was successfully developed. This microarray is the first porcine DNA microarray in China and its false positive rate is 0.98%, which means the microarray platform is reliable. The microarray can be used to study gene expression profiles in multiple pig tissues because the present genes percentage of adipose, skeletal muscle, heart, liver, lung, kidney, and spleen were all more than 60%. This microarray was used to identify the genes responding to clenbuterol stimulation in pig internal organs, including heart, liver, lung, spleen, and kidney. Many genes were identified including enzymes involved in lipids metabolism (lipoprotein lipase up-regulated in liver, heart and lung, ATP-citrate lyase and carnitine palmitoyltransferase II precursor up-regulated in liver, succinyl-CoA up-regulated in lung, mitochondrial malate dehydrogenase down-regulated in spleen), and signaling pathway genes (cAMP-protein kinase A signaling pathway was found up-regulated in liver, heart, lung, and kidney as reported previously, while transforming growth factor was found down-regulated in heart and lung). However, no common gene responding to clenbuterol administration was found in all tissues. The expression levels of 14 genes were analyzed using real-time PCR with 82.1% of them induced to express similar magnitudes as in the microarray analyses. This work offers some understanding of how clenbuterol so effectively reduces pig adipose accumulation on the molecular level.展开更多

A total of 360 1-day-old Ross 308 broilers were randomly divided into 4 groups with 3 replicates and 30 broilers per replicate (15 male, 15 female). The treatments were fed with basal diet supplemented with 0%, 0.01%, 0.02% and 0.03% phytase. The results showed that: 1) Phytase supplementation increased the body weight gain and the body weight of Ross 308 broilers (P serum calcium (Ca) consistency was 0.02%. According to the above analysis, the feasible supplementation of phytase (enzyme activity for 5000 u/g) in broiler dietary was 0.02%.展开更多

The problem of constructing global view of heterogeneous information sources for information sharing is becoming more and more important due to the availability of multiple information sources within the virtual organ...The problem of constructing global view of heterogeneous information sources for information sharing is becoming more and more important due to the availability of multiple information sources within the virtual organization. Global view is defined to provide a unified representation of the information in the different sources by analyzing concept schemas associated with them and resolving possible semantic heterogeneity. An ontology-based method for global view construction is proposed. In the method, ( 1 ) Based on the formal ontologies, the concept of semantic affinity is introduced to assess the level of semantic relationship between information classes from different information sources; (2) Information classes are classified by semantic affinity levels using clustering procedures so that their different representations can be analyzed for unification; (3) Global view is constructed starting from selected clusters by unifying representation of their elements. The application example of using the method in the joint-aerial defense organization is illustrated and the result shows that the proposed method is feasible.展开更多
Waste heat recovery is one of the possible solutions to improve the efficiency of internal combustion engines.Instead of wasting the exhaust stream of an energy conversion system into the environment,its residual ener...Waste heat recovery is one of the possible solutions to improve the efficiency of internal combustion engines.Instead of wasting the exhaust stream of an energy conversion system into the environment,its residual energy content can be usefully recovered,for example in Organic Rankine Cycles(ORC).This technology has been largely consolidated in stationary power plants but not yet for mobile applications,such as road transport,due to the limitations in the layout and to the constraints on the size and weight of the ORC system.An ORC system installed on the exhaust line of a bus powered by a natural gas spark ignition engine has been investigated.The thermal power available at engine exhaust has been evaluated by measuring gas temperature and mass flow rate during real driving operation.The waste thermal power has been considered as heat input for the ORC plant simulation.A detailed heat exchanger model has been developed because it is a crucial component for the ORC performance.The exergy analysis of the ORC was performed comparing different working fluids:R601,R1233zd(E)and two zeotropic blends of the two organic pure fluids.The model allowed the evaluation of the ORC produced energy over the driving cycle and the potential benefit on the engine efficiency.展开更多
The Coronavirus Disease-2019 (COVID-19) outbreak has become a global health emergency owing to its magnitude, attributed deaths, and its propensity to spread across the world. In-fact, owing to its quick spread across...The Coronavirus Disease-2019 (COVID-19) outbreak has become a global health emergency owing to its magnitude, attributed deaths, and its propensity to spread across the world. In-fact, owing to its quick spread across international boundaries and the resulting caseload, the disease has been declared as a Public Health Emergency of International Concern on 30 January 2020. It is worth noting that out of the 395 cases detected in other nations, 165 (41.8%) have a positive history of travel to China. As of now, the World Health Organization has not recommended for any restrictions on the travel or trade aspects, but has clearly specified that implementation of International Health Regulations should be strictly done at the airports and seaports. In conclusion, the COVID-19 outbreak has created an alarm across the globe as the causative virus is novel in nature. However, strengthening of standard infection control practices and adoption of preventive measures for travelers can significantly minimize the threat of further transmission of the disease.展开更多

A new function that reflects both the technical and economic performances was put forward and used as the objective function. An organic Rankine cycle (ORC) was analyzed through the energetic and exergetic analyses, and the reasons for low efficiency were pinpointed. Results indicate that geothermal water directly transferring heat to the working fluid reduces energy dissipation and increases cycle efficiencies. The net power output with an internal heat exchanger (IHE) is averagely 5.3% higher than that without an IHE. R601a and R601 can be used to replace R123 for geothermal water below 110℃. We also review efforts made by major international and regional organizations in gov...In this paper, we discuss the environmental damages caused by the use and carriage of Heavy Fuel Oil(HFO) by ships in Arctic waters. We also review efforts made by major international and regional organizations in governing the use and carriage of HFO by ships in Arctic waters after implementation of the Polar Code, and analyze the obstacles and difficulties that lie ahead. By illustrating the features of the Arctic Council and the International Maritime Organization(IMO), which govern Arctic waters, we examine measures taken by these two organizations in tackling HFO issues. These include: assessing indigenous and local community’s reliance on HFO transportation in the Arctic, developing risk prevention measures for ships operating in Arctic waters, seeking economic alternatives to HFO that contribute to a greener economy, developing a package of HFO governance measures, strengthening cooperation between relevant international and regional organizations that govern HFO to provide suggestions for follow-up discussions on HFOs, and optimize governance by relevant organizations and determine a better global solution to governing the use and carriage of HFO by ships. In addition, we discuss the feasibility of an IMO-imposed HFO ban from the perspective of the Arctic governance to clearly grasp the path of its future development.展开更多

The first group consists of professionals and companies in eco-sustainable businesses, such as law firms, insurance companies, investment firms, banking, technological innovation, mass media, medical research and pharmaceutical research. The second group will consist of persons engaged in organic/eco-sustainable agriculture whose crops and animal husbandry practices can be transferred successfully to Climate Change Haven regions. The present research focuses on the social and economic variables that must be taken into account to insure that each new Climate Change Haven Community becomes successfully integrated with the local population and forms a cohesive, harmonious social structure. Examples are given from the United States, France, Spain, Portugal and Italy.

文摘In this paper, we discuss the environmental damages caused by the use and carriage of Heavy Fuel Oil(HFO) by ships in Arctic waters. We also review efforts made by major international and regional organizations in governing the use and carriage of HFO by ships in Arctic waters after implementation of the Polar Code, and analyze the obstacles and difficulties that lie ahead. By illustrating the features of the Arctic Council and the International Maritime Organization(IMO), which govern Arctic waters, we examine measures taken by these two organizations in tackling HFO issues. These include: assessing indigenous and local community’s reliance on HFO transportation in the Arctic, developing risk prevention measures for ships operating in Arctic waters, seeking economic alternatives to HFO that contribute to a greener economy, developing a package of HFO governance measures, strengthening cooperation between relevant international and regional organizations that govern HFO to provide suggestions for follow-up discussions on HFOs, and optimize governance by relevant organizations and determine a better global solution to governing the use and carriage of HFO by ships. In addition, we discuss the feasibility of an IMO-imposed HFO ban from the perspective of the Arctic governance to clearly grasp the path of its future development.
