Explants collected in early...A protocol for micropropagation using nodal explants from mature Pinus massoniana trees has been developed. Time of explant collection is crucial for the initial success of aseptic culture. Explants collected in early March gave the highest percentage of explant survival (64.5%) and shoot-forming percentage (52.3%). Thidiazuron (TDZ) concentration significantly influenced shoot formation; 4 mu M TDZ was optimum, with 4.8 shoots produced per explant with a mean length of 7.1 cm after 120 days of culture. Regenerated shoots rooted for 60 days in basic medium with 1 mu M NAA were ready for growth in pots. This is the first report on plantlet regeneration in vitro from mature trees of P. massoniana that provides a reliable method for propagating selected elites.展开更多

Our objective was to enhance the frequency of plant regeneration in Himalayan poplar (Populus ciliata wall. ex Royle). The effect of TDZ alone and in combi- nation with adenine and NAA was studied on the regen- eration potential of petiole explants. The explants were excised from Himalayan poplar plants grown in glass- houses. After surface sterilization the explants were cul- tured on shoot induction medium. High percentage shoot regeneration (86 %) was recorded on MS medium sup- plemented with 0.004 mg L-1 TDZ and 79.7 mg L-1 adenine. The regenerated shoots for elongation and multi- plication were transferred to MS + 0.5 mg L-1 BAP + 0.2 mg L-1 IAA + 0.3 mg L-1 GA3. Root re- generation from shoots developed in vitro was observed on MS medium supplemented with 0.10 mg L-1 IBA. Hi- malayan poplar plantlets could be produced within 2 months after acclimatization in a sterile mixture of sand and soil. The effectiveness of thidiazuron(TDZ)on adventitious bud and shoot formation from leaves of Robinia pseudoacacia was tested in combination with naphthaleneacetic acid (NAA)or 2,4-dichiorophenoxyacetic acid (2,4-D...The effectiveness of thidiazuron(TDZ)on adventitious bud and shoot formation from leaves of Robinia pseudoacacia was tested in combination with naphthaleneacetic acid (NAA)or 2,4-dichiorophenoxyacetic acid (2,4-D),With the increasing of TDZ level,the induction rate of adventitious buds appeared an increasing trend and the highest rate was obtained by the combination of 2mg/L TDZ whth 0.1mg/L NAA.The excessive level of TDZ could induce useless cellae and easily produce hyperhydricity.Inhibition of shoot elongation by TDZ was overcome through transferring shoot cultures to a shoot proliferation medium lacking TDZ orcontaining NAA,benzyladenine (BA)and TDZ.Indole-3-butyric acid (IBA)was used to stimulate the rooting in vitro .Based on the number and length of rooting ,the treatment with 0.5mg/L IBA was the best.展开更多
