Myricetin alleviates ovalbumin-induced allergic rhinitis in mice by regulating Th1/Th2 balance 被引量:2

Objective:To evaluate the effect of myricetin on ovalbumin(OVA)-induced allergic rhinitis in mice.Methods:Mice were sensitized and challenged using OVA(5%,500 mL)intraperitoneally and intranasally,respectively,on an alternative day for 14 days,followed by administration of myricetin(50,100,and 200 mg/kg)till day 21.Nasal symptoms,biochemical parameters,protein expressions,and histopathology were observed.Results:OVA-induced increased nasal symptoms including rubbing,sneezing,and discharge were significantly reduced by myricetin(100and 200 mg/kg)(P<0.05).Myricetin also protected against histamine challenge and attenuated elevated serum immunoglobulin E(IgE;total and OVA-specific),total IgG1,andβ-hexosaminidase levels,as well as leukotriene C4 and interleukins levels in nasal lavage fluid(P<0.05).Western blot analysis showed that myricetin significantly upregulated the protein expression of T-box expressed in T cells,while downregulating the protein expression of GATA binding protein 3,NF-κB,and IκB-α(P<0.05).Additionally,OVA-induced histopathological abberations in the nasal mucosa was markedly ameliorated by myricetin treatment(P<0.05).Conclusions:Myricetin exerts anti-allergic effects against OVAinduced allergic rhinitis via regulating Th1/Th2 balance. 展开更多

Objective:To evaluate the therapeutic effect of Glycyrrhetinic Acid on cough variant asthma(CVA)mice and to investigate the possible mechanism in reducing lung inflammation.Methods:48 young female Balb/c mice were divided into Control,CVA,Prednisone Acetate,Glycyrrhetinic Acid high-dose,Glycyrrhetinic Acid middle-dose and Glycyrrhetinic Acid lowdose groups randomly,with 8 mice in each group.The CVA mice model was established by ovalbumin(OVA)sensitization and OVA challenge,the animal asthma behavior was observed after drug administration,and the index of the lung of mice were recorded.The level of OVAsIgE in the bronchoalveolar lavage fluid(BALF)was tested by ELISA.The pathological changes of the lung tissue were observed by Hematoxylin and Eosin(H&E)staining.The protein expressions of T-bet,IFN-γ,Gata3,IL-4 and IL-13 in the lung tissue were determined by Western blot.Results:Compared with the CVA group,the index of lung of mice,the OVA-sIgE level in BALF and expression levels of Th2-related factor in the lung tissue of mice in Prednisone Acetate and Glycyrrhetinic Acid groups were significantly decreased(P<0.05 or P<0.01),the infiltration of inflammatory cells in the lung tissue was reduced,while expressions of Th1-related factor in the lung tissue was significantly increased(P<0.05 or P<0.01).Conclusion:Glycyrrhetinic acid has therapeutic effect on CVA mice,the underlying mechanism of Glycyrrhetinic acid alleviating lung impairment and airway inflammation may be associated with mediating the Th1/Th2 imbalance in the lung tissue. 展开更多

Objective:Explore the role of Th1/Th2 balance in the pathogenesis of primary dysmenorrhea through study the effect of Th1/Th2 balance drift on serum prostaglandin F_(2α),prostaglandin,6-keto-prostaglandin F_(1α),TXB2,COX-2,AVP and OT in Primary Dysmenorrhea Rats with cold coagulation and blood stasis syndrome.Method:The rat model of primary dysmenorrhea with cold coagulation and blood stasis syndrome was established.On the basis of this model,Th1 shift model and Th2 shift model were established.The rats were divided into blank group,model group,Th1 shifting group and Th2 shifting group.The writhing reaction of rats in each group was compared,and the contents of PGF_(2α),PGE_(2),6-keto-PGF_(1α),TXB2,COX-2,AVP and OT in serum were detected.Results:Writhing latency:There was no significant difference between Th1 and Th2 shifting groups(P>0.05),Th2 shifting group was shorter than Th1 shifting group(P<0.05).Writhing times in 30 minutes:Compared with the model group,the writhing times of Th1 shifting group decreased significantly(P<0.01),while Th2 shifting group increased significantly(P<0.01).Compared with the blank group,the contents of PGF_(2α),TXB2,COX-2,AVP and OT were increased(P<0.05,P<0.01),and the contents of 6-keto-PGF_(1α)and PGE_(2)were decreased(P<0.01).Compared with the model group,the contents of PGF_(2α),TXB2,COX-2 and AVP in Th1 shifting group were decreased(P<0.05,P<0.01),and the contents of 6-keto-PGF_(1α)and PGE_(2)were increased(P<0.01),while the contents of PGF_(2α),TXB2,COX-2,AVP and OT in Th2 shifting group were increased(P<0.01),and the contents of 6-keto-PGF_(1α)and PGE_(2)were decreased(P<0.01).Conclusion:The intervention of Th1/Th2 cell balance on the expression of PGF_(2α),PGE_(2),6-keto-PGF_(1α),TXB2,COX-2,AVP and OT may be one of the pathogenesis of PD with cold coagulation and blood stasis syndrome. 展开更多

The authors aimed to assess Thl (T-helper cell 1)/Th2 (T-helper cell 2) balance, through evaluation of serum IFN-γ (interferon gamma) and IL-4 (interleukin 4), during asthma exacerbation and study the effect of anti inflammatory therapy. A randomized prospective case-control study was designed. The sludy included 30 asthmatic patients, aging 8-14 years. All were diagnosed as partly controlled asthmatics. Twenty, age and sex matched, healthy children were included in the study as control group All participants were subjected to medical history, clinical examination, pulmonary function testing, eosinophilic blood counting, estimation of serum interleukine-4 and interferon gamma. Patients were treated for 6 weeks with 2 different anti inflammatory drugs. All methods were then repeated for follow up. IL-4 serum level was significantly higher in subjects with partly controlled asthma than in control subjects (P = 0.01), and then in asthmatic patients after therapy (P = 0.0000), while IFN-), serum level was significantly lower in subjects with partly controlled asthma than in control subjects (P = 0.01), and than in asthmatic patients after therapy (P = 0.0000). Interferon gamma showed a significant negative correlation with IL-4 among the healthy control group (r = -0.559, P = 0.010). Both LTA (leukotriene antagonist) and ICS (inhaled corticosteroids) therapy lead to significant improvement, but there were no statistically significant differences (P 〉 0.05) between them as regard the pulmonary functions and the laboratory evaluating parameters. Both serum levels of IL-4 and IFN-γ, could be used as a reliable inflammatory biomarker for the evaluation and follow up of asthmatic patients. 展开更多

目的:探讨血塞通注射液联合阿司匹林治疗老年缺血性卒中的疗效及对血清Thl/Th2细胞因子水平的影响。方法:选择老年缺血性卒中患者300例,随机分为对照组和观察组各150例。所有患者均进行改善脑循环及相应合并症对症治疗,对照组给予阿司... 目的:探讨血塞通注射液联合阿司匹林治疗老年缺血性卒中的疗效及对血清Thl/Th2细胞因子水平的影响。方法:选择老年缺血性卒中患者300例,随机分为对照组和观察组各150例。所有患者均进行改善脑循环及相应合并症对症治疗,对照组给予阿司匹林肠溶片治疗,观察组在对照组基础上给予血塞通注射液治疗,比较两组治疗前后FMA、MBI评分、NIHSS评分、IFN-γ、IL-4因子变化及临床疗效和生活质量水平情况。结果:治疗后,观察组中医证候积分比对照组下降更明显(P<0.05);治疗后,两组IL-6及IL-4水平均降低,IFN-γ及IL-2均升高;观察组IL-6及IL-4水平低于对照组,IL-2及IFN-γ高于对照组(P<0.05);两组患者治疗后FMA、MBI评分明显升高,而NIHSS评分明显降低,观察组改善更明显(P<0.05)。治疗后两组NSE、BNP水平明显下降,NGF水平明显上升,观察组更明显(P<0.05)。观察组基本治愈率46.67%,总有效率95.33%均高于对照组24.67%及88.00%(P<0.05)。观察组治疗后QLQ-C30评分显著高于对照组(P<0.05)。结论:血塞通注射液联合阿司匹林可显著改善老年缺血性卒中患者脑神经功能缺损状况,调节机体Thl/Th2细胞因子水平,疗效显著。 展开更多

目的:探讨慢性乙型肝炎在拉米呋定治疗过程中IL—12对PBMC产生THI/TH2细胞因子的影响。方法:在拉米呋定治疗前和治疗3mo、6mo、9mo后,20例慢性乙型肝炎的PBMC在PHA(100μg·mL^-1)、HBcAg(1μg·mL^-1)、habeas(1μg·m... 目的:探讨慢性乙型肝炎在拉米呋定治疗过程中IL—12对PBMC产生THI/TH2细胞因子的影响。方法:在拉米呋定治疗前和治疗3mo、6mo、9mo后,20例慢性乙型肝炎的PBMC在PHA(100μg·mL^-1)、HBcAg(1μg·mL^-1)、habeas(1μg·mL^-1)单独或联合IL—12(10ng·mL^-1)刺激下体外培养48h,收集上清液,用ELIS检测:IFN-γ和IL-10的水平。结果:在拉米呋定治疗3mo后,无论是抗原单独诱导还是联合IL-12共同诱导,PBMC产生-IFN-γ的水平同治疗前相比无显著变化;在治疗6mo后,抗原单独诱导产生IFN-γ的水平同治疗前相比无显著性差异,但IL-12诱导产生IFN-γ的增殖效应明显加强;在治疗9mo后,无论是抗原单独诱导还是联合IL-12共同诱导,PBMC产生IFN-γ的水平同治疗前相比均显著增高,同时IL-10的水平也显著降低。在治疗同一阶段,抗原联合IL-12诱导组产生IFN-γ水平明显高于抗原单独诱导组。结论:拉米呋定治疗慢性乙型肝炎过程中,随着HBV DNA的抑制,IL-l2对乙型肝炎患者PBMC产生IFN-γ协同效应逐渐增强,对IL-10的产生有抑制作用,提示拉咪呋定联合IL-12可能提高慢性乙型肝炎的疗效。 展开更多

Scutellarin alleviates complete freund’s adjuvant-induced rheumatoid arthritis in mice by regulating the Keap1/Nrf2/HO-1 pathway

Scutellarin(SCU)is a herbal flavonoid glucuronide with multiple pharmacological activities,including antioxidant,anti-inflammation,vascular relaxation,anti-platelet,and myocardial protection.However,the effect of SCU on complete Freund’s adjuvant(CFA)-induced rheumatoid arthritis(RA)had not been studied.In this study,we investigated the beneficial effects of SCU in the CFA-induced RA mice model and the anti-arthritic activity was evaluated by paw edema.Enzyme-linked immunosorbent assay(ELISA)was carried out to evaluate the plasma levels of immunoglobulin(Ig)G,IgE,tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6,receptor activator of nuclear factor-κB ligand(RANKL),and osteoprotegerin(OPG).Histological slides were prepared from the harvested paws of mice to determine the pathological changes in the joints.The proportions of T helper type 1(Th1)and T helper type 2(Th2)cells of CD4+T lymphocyte subsets were analyzed by flow cytometry.The expression of Kelch-like ECHassociated protein 1(Keap1),nuclear factor erythroid 2-related factor 2(Nrf2),and heme oxygenase-1(HO-1)was analyzed using real-time quantitative PCR(RT-qPCR)and western blotting assays.The present study demonstrated that SCU prevented CFA-induced RA,and inhibited the expression of inflammation factors,IgG,IgE,TNF-α,IL-1β,and IL-6.While SCU also reduced the RANKL level,it increased OPG expression in RA mice.The Th1/Th2 ratio was significantly lower in mice treated with SCU.Additionally,HO-1 expression was reduced while the expression of Keap1 and Nrf2 was elevated following SCU treatment.Results provide preliminary evidence to employ SCU in arthritis treatment which might be related to the regulation of Th1/Th2 balance and the Keap1/Nrf2/HO-1 pathway. 展开更多

Both T helper IL-17-producing cells (Thl7 cells) and regulatory T cells (Tregs) have been found to be increased in malignant pleural effusion (MPE). However, the possible imbalance between Thl7 cells and Tregs, as well as the association of.Thl7/Treg and Thl/Th2 cells in MPE remains to be elucidated. The objective of the present study was to investigate the distribution of Th 17 cells in relation to Tregs, as well as Thl/Th2 balance in MPE. The number ofThl7, Tregs, Thl, and Th2 cells in MPE and peripheral blood was determined by using flow cytometry. The relationship among the number of Thl7, Tregs, Thl, and Th2 cells was explored. It was found that the number of Thl7, Tregs, Thl, and Th2 cells was all increased in MPE as compared with the corresponding peripheral blood. The number of Thl7 cells was correlated negatively with Tregs in MPE, but not in blood. Thl7 cells and Thl7/Treg ratio were positively, and Tregs were negatively, correlated with Thl cells, but not with either Th2 cells or Th1/Th2 ratio in MPE. This study supports earlier data that both Thl7 cells and Treg are present at higher frequencies in MPE than in the autologous blood. For the first time, we show that Thl7/Treg imbalance exists in MPE. 展开更多

目的研究Th1/Th2细胞因子谱在慢性阻塞性肺疾病急性加重(AECOPD)患者中的变化。方法该次研究抽取的以30例慢性阻塞性肺疾病急性加重患者、30例慢性阻塞性肺疾病稳定者和30名健康者作为研究对象,其入院时间为2013年1—12月。用流式微球... 目的研究Th1/Th2细胞因子谱在慢性阻塞性肺疾病急性加重(AECOPD)患者中的变化。方法该次研究抽取的以30例慢性阻塞性肺疾病急性加重患者、30例慢性阻塞性肺疾病稳定者和30名健康者作为研究对象,其入院时间为2013年1—12月。用流式微球阵列分析术(CBA)检测各组Th1/Th2细胞因子包括IL-4、IL-6、IL-10、TNF-α、IFN-γ的表达水平。结果 AECOPD组血清产生IL-4(27.22±1.51)pg/mL、IL-6(47.73±4.72)pg/mL低于慢性阻塞性肺疾病稳定组(34.79±1.37)、(72.38±5.31)pg/mL,(P<0.05)。AECOPD组血清产生IL-10(35.76±2.56)pg/mL低于慢性阻塞性肺疾病稳定组(45.15±2.08)pg/mL,(P<0.05)。AECOPD组血清产生TNF-α(19.11±1.08)pg/mL、IFN-γ(40.33±3.69)pg/mL高于慢性阻塞性肺疾病稳定组(12.93±0.36)、(22.28±1.16)pg/mL,(P<0.05)。而慢性阻塞性肺疾病稳定组其IL4(34.79±1.37)pg/mL、IL-6(72.38±5.31)pg/mL、IL-10(45.15±2.08)pg/mL、TNF-α(12.93±0.36)pg/mL、IFN-γ(22.28±1.16)pg/mL的表达水平与健康对照组(33.72±1.66)、(67.06±6.89)、(43.84±2.45)、(13.81±0.38)、(23.59±1.21)pg/mL相比,均差异无统计学意义(P>0.05)。结论慢性阻塞性肺疾病急性加重患者存在Thl/Th2细胞因子失衡。 展开更多

目的 :研究乙型肝炎病毒 (HBV)母婴垂直传播与T辅助细胞 1、2型细胞因子的相关性。方法 :将研究对象分为两组 :研究组为 78例乙型肝炎病毒表面抗原 (HBsAg)阳性孕妇 ;对照组为 40例正常孕妇。采用双抗夹心酶联免疫吸附法 (DAS-ELISA)检... 目的 :研究乙型肝炎病毒 (HBV)母婴垂直传播与T辅助细胞 1、2型细胞因子的相关性。方法 :将研究对象分为两组 :研究组为 78例乙型肝炎病毒表面抗原 (HBsAg)阳性孕妇 ;对照组为 40例正常孕妇。采用双抗夹心酶联免疫吸附法 (DAS-ELISA)检测孕妇外周静脉血及其新生儿脐静脉血血清中乙型肝炎五项指标 (HBVM)及细胞因子干扰素γ(IFN γ)、白细胞介素 12 (IL 12 )、白细胞介素 6 (IL 6 )水平。结果 :研究组孕妇分娩的新生儿有 10例宫内感染 ,宫内感染率为 12 82 %。胎儿宫内感染组孕妇血清中IFN -γ、IL - 12水平显著低于宫内未感染组及对照组孕妇 ,IL - 6水平则显著高于宫内未感染组及对照组孕妇 (P <0 0 5 )。宫内未感染组与对照组相比 ,上述三种细胞因子水平差异均无显著性(P >0 0 5 )。上述各组孕妇血清中IFN -γ与IL - 12水平均呈显著正相关 (P <0 0 1,P <0 0 5 ,P <0 0 5 ) ;IL - 12与IL -6呈显著负相关 (P <0 0 1,P <0 0 5 ,P <0 0 5 ) ;IFN -γ与IL - 6亦呈显著负相关 (P <0 0 1,P <0 0 5 ,P <0 0 5 )。各组新生儿脐血清中IFN -γ、IL - 12、IL - 6水平差异无显著性 (P >0 0 5 )。结论 :孕妇细胞免疫功能紊乱导致Th1型细胞因子IFN -γ、IL - 12水平下降 ,而Th 2型细胞因子IL - 展开更多

目的:探讨细胞因子、Th1/Th2细胞及调节性T细胞(Treg)、转录因子Foxp3在佐剂性关节炎(AA)大鼠肺功能损害中的作用机制。方法:将24只Wistar大鼠随机分为正常对照(NC)组和模型(Model)组,每组12只,向Model组大鼠右后足跖皮内注射弗氏完全佐... 目的:探讨细胞因子、Th1/Th2细胞及调节性T细胞(Treg)、转录因子Foxp3在佐剂性关节炎(AA)大鼠肺功能损害中的作用机制。方法:将24只Wistar大鼠随机分为正常对照(NC)组和模型(Model)组,每组12只,向Model组大鼠右后足跖皮内注射弗氏完全佐剂0.1mL致炎,复制成AA模型。致炎48d后,观察两组大鼠足跖肿胀度(E)及关节炎指数(AI),HE染色观察肺组织病理学改变,计算两组大鼠肺系数(LI)、肺泡炎积分,免疫组化染色法检测Foxp3、TGF-β1蛋白表达情况。通过小动物肺功能仪检测肺功能,ELISA法测定细胞因子的变化,流式细胞术(FCM)测定Treg的表达。结果:与NC组相比,Model组大鼠E、AI、LI、1s内平均呼气流量(FEV1/FVC%)、肺泡炎积分、血清TNF-α、Th1/Th2、肺组织TGF-β1、CD4+ CD25- T细胞的表达水平明显升高,且两者差异有统计学意义(P<0.05或P<0.01);用力肺活量(FVC)、25%肺活量的最大呼气流量(FEF25)、50%肺活量的最大呼气流量(FEF50)、75%肺活量的最大呼气流量(FEF75)、最大呼气中期流量(MMF)、用力最大呼气流量(PEF)、肺动态顺应性(Cldyn)、血清IL-10、CD4+ Treg、CD4+ CD25- Treg、肺组织Foxp3蛋白表达水平显著降低(P<0.01)。Spearman相关分析结果显示,AA大鼠肺功能参数分别与E、AI、TNF-α、IL-10、Th1/Th2及CD4+ Treg、CD4+CD25+Treg、CD4+ CD25- T细胞、Foxp3、TGF-β1表达呈相关性,且相关有统计学意义(P<0.05或P<0.01)。结论:大鼠在致炎后对抗原刺激呈高敏反应状态,Th1/Th2状态失衡、CD4+ CD25- T细胞转化成CD4+ CD25+ Treg受阻,免疫调节功能紊乱,释放大量细胞因子和炎症介质,导致局部关节的病变和肺组织损害,从而发生AA肺功能降低。 展开更多

目的:探讨大负荷体能训练对大学生篮球运动员细胞免疫功能的影响。方法:14名男子大学生篮球运动员进行为期16周的集训,在训练期间分次对运动员外周血淋巴细胞亚群(CD3+、CD4+/CD8+淋巴细胞比值、NK细胞比例)和Th1/Th2细胞因子 mRNA表达... 目的:探讨大负荷体能训练对大学生篮球运动员细胞免疫功能的影响。方法:14名男子大学生篮球运动员进行为期16周的集训,在训练期间分次对运动员外周血淋巴细胞亚群(CD3+、CD4+/CD8+淋巴细胞比值、NK细胞比例)和Th1/Th2细胞因子 mRNA表达进行检测。结果:在两次分别持续了6周的训练期后,运动员外周血CD3+淋巴细胞数于增加后出现显著下降,从第7周的峰值(59.74±7.18)下降至第16周的最低值(52.02±7.92)(P<0.001);CD8+细胞数第16周(31.15±6.25)与第3周(37.98±7.05)相比显著下降(P<0.05);NK细胞数从第7周的最高值(19.62±5.21)下降至第14周的最低值(14.41±7.93)(P<0.05);IL-4 mRNA表达训练后与基础值(6.56±0.71)相比显著增加,从第7周的7.04±0.35和第14周的7.30±0.25,直至第16周达到最大值7.36±0.45(P<0.05),其他指标的变化不具有统计学意义。结果提示,大负荷训练使运动员细胞免疫功能削弱,Th1/Th2平衡向Th2方向漂移。 展开更多

目的 观察酪氨酸蛋白激酶(TPK)、蛋白激酶C(PKC)和磷酯酰肌醇-3-激酶(P13-K)特异性抑制剂(分别为tyrphostin-25、D-sphingosine和wortmannin)对日本血吸虫感染小鼠虫卵肉芽肿病变的影响,并探讨其作用机制。 方法 于小鼠感染日本血吸虫后... 目的 观察酪氨酸蛋白激酶(TPK)、蛋白激酶C(PKC)和磷酯酰肌醇-3-激酶(P13-K)特异性抑制剂(分别为tyrphostin-25、D-sphingosine和wortmannin)对日本血吸虫感染小鼠虫卵肉芽肿病变的影响,并探讨其作用机制。 方法 于小鼠感染日本血吸虫后第35天起,经尾静脉分别注射3种信号转导分子抑制剂,连续5 d。在小鼠感染后6和8wk,观察小鼠肝肉芽肿病变,并用ELISA夹心法和硝酸还原酶法分别测定小鼠血清IFN-γ、IL-4和一氧化氮(NO)水平。结果 感染小鼠应用TPK和PKC抑制剂后均可显著抑制肝肉芽肿病变,PKC抑制剂可使肉芽肿减少率达56.2%~63.4%(P<0.01)。PKC抑制剂主要抑制Th2细胞因子IL-4的表达,其抑制率为34.1%和65.6%(P<0.01),而对NO水平的检测结果进一步证明了PKC抑制剂对IL-4表达的抑制作用。 结论 在日本血吸虫感染早期应用PKC抑制剂干预T淋巴细胞信号转导可显著抑制小鼠肝肉芽肿病变,其机制可能是由于抑制了Th2优势应答并介导Th2向Th1免疫反应偏移。 展开更多
