康艾注射液治疗子宫颈癌术后疗效及对IGF-2、SCC、CYFRA21-1水平变化的影响 被引量:8

目的:分析康艾注射液对子宫颈癌术后患者的疗效及对IGF-2、SCC、CYFRA21-1水平变化的影响。方法:子宫颈癌患者91例,根据随机数字法分为对照组和观察组。所有患者术前均采用新辅助化疗后行全麻下开腹根治性宫颈切除手术治疗,对照组患... 目的:分析康艾注射液对子宫颈癌术后患者的疗效及对IGF-2、SCC、CYFRA21-1水平变化的影响。方法:子宫颈癌患者91例,根据随机数字法分为对照组和观察组。所有患者术前均采用新辅助化疗后行全麻下开腹根治性宫颈切除手术治疗,对照组患者术后采用常规对症治疗,观察组患者在对照组治疗基础上加用康艾注射液治疗。观察两组患者治疗后疗效,健康状况、Th1/Th2细胞相关因子、IGF-2、SCC、CYFRA21-1水平变化情况。结果:观察组患者有效率为71.74%,对照组为48.89%,观察组患者有效率高于对照组(P〈0.05)。两组患者治疗后IGF-2、SCC、CYFRA21-1、IL-6和IL-10水平均较治疗前下降,INF-γ和IL-2水平均较治疗前上升(P〈0.05)。观察组患者治疗后IGF-2、SCC、CYFRA21-1、血清Th1/Th2细胞相关因子水平均优于对照组(P〈0.05)。观察组患者治疗后健康状况优于对照组(P〈0.05)。结论:子宫颈癌术后采用康艾注射液治疗可降低患者血清IGF-2、SCC、CYFRA21-1水平,调节患者血清Th1/Th2细胞相关因子水平,提高近期疗效,改善机体健康状况。 展开更多

Both T helper IL-17-producing cells (Thl7 cells) and regulatory T cells (Tregs) have been found to be increased in malignant pleural effusion (MPE). However, the possible imbalance between Thl7 cells and Tregs, as well as the association of.Thl7/Treg and Thl/Th2 cells in MPE remains to be elucidated. The objective of the present study was to investigate the distribution of Th 17 cells in relation to Tregs, as well as Thl/Th2 balance in MPE. The number ofThl7, Tregs, Thl, and Th2 cells in MPE and peripheral blood was determined by using flow cytometry. The relationship among the number of Thl7, Tregs, Thl, and Th2 cells was explored. It was found that the number of Thl7, Tregs, Thl, and Th2 cells was all increased in MPE as compared with the corresponding peripheral blood. The number of Thl7 cells was correlated negatively with Tregs in MPE, but not in blood. Thl7 cells and Thl7/Treg ratio were positively, and Tregs were negatively, correlated with Thl cells, but not with either Th2 cells or Th1/Th2 ratio in MPE. This study supports earlier data that both Thl7 cells and Treg are present at higher frequencies in MPE than in the autologous blood. For the first time, we show that Thl7/Treg imbalance exists in MPE. 展开更多

T-bet, GATA-3 and Foxm1 expression in chronic sinusitis mucosa and their correlation with inflammatory molecule expression

Objective: To study the T-bet, GATA-3 and Foxm1 expression in chronic sinusitis mucosa and their correlation with inflammatory molecule expression. Methods: Patients with sinusitis and deflection of nasal septum who received nasal endoscopic surgery in Shengli hospital between June 2015 and February 2017 were selected and enrolled in CRS group and control group respectively. The nasal mucosa tissue was collected during operation to determine the mRNA expression of T-bet, GATA-3 and Foxm1 as well as the protein expression of inflammatory molecules. Results: T-bet, GATA-3 and Foxm1 mRNA expression as well as IL-1β, IFN-γ, TNF-α, IL-4, IL-5, IL-19, IL-20R1, IL-20R2, MMP2, MMP9, PI3K, Akt, GSK-3β and p38MAPK protein expression in nasal mucosa tissue of CRS group were significantly higher than those of control group;T-bet, GATA-3 and Foxm1 mRNA expression in nasal mucosa tissue of patients with CRS were positively correlated with IL-1β, IFN-γ, TNF-α, IL-4, IL-5, IL-19, IL-20R1, IL-20R2, MMP2, MMP9, PI3K, Akt, GSK-3β and p38MAPK protein expression. Conclusions: The high expression of T-bet, GATA-3 and Foxm1 in chronic sinusitis mucosa tissue can activate Th1 and Th2 immune response and cause mucosal inflammatory response. 展开更多

We investigated the relative abundance and activit... Background Recent recognition is that Th2 response is insufficient to fully explain the aetiology of asthma. Other CD4^+ T cells subsets might play a role in asthma. We investigated the relative abundance and activities of Thl, Th2, Th17 and CD4^+CD25^+ Treg cells in patients with allergic asthma. Methods Twenty-two patients with mild asthma, 17 patients with moderate to severe asthma and 20 healthy donors were enrolled. All patients were allergic to house dust mites. Plasma total IgE, pulmonary function and Asthma Control Questionnaire were assessed. The proportions of peripheral blood Thl, Th2, Th17 and CD4^+CD25^+ Treg cells were determined by flow cytometry. The expression of cytokines in plasma and Jn the culture supernatant of peripheral blood mononuclear cells was determined by enzyme linked, immunosorbent assay. Results The frequency of blood Th2 cells and IL-4 levels in plasma and culture supernatant of peripheral blood mononuclear cells were increased in all patients with allergic asthma. The frequency of Th17 cells and the plasma and culture supernatant levels of IL-17 were increased, whereas the frequency of CD4^+CD25^+ Treg cells and plasma IL-10 levels were decreased in patients with moderate to severe asthma. Dermatophagoides pteronyssinus specific IgE levels were positively correlated with the percentage of blood Th2 cells and plasma IL-4 levels. Forced expiratory volume in the first second was negatively correlated with the frequency of Th17 cells and plasma IL-17 levels, and positively correlated with the frequency of Treg cells. However, mean Asthma Control Questionnaire scores were positively correlated with the frequency of Th17 cells and plasma IL-17 levels, and negatively correlated with the frequency of Treg cells. Conclusions Imbalances in Thl/Th2 and Th17/Treg were found in patients with allergic asthma. Furthermore, elevated Th17 cell responses, the absence of Tregs and an imbalance in Th17/Treg levels were associated with moderate to severe asthma. 展开更多

Previous studies have shown that tumor cells predominantly express Th2 type cytokines and transcription factors. GATA-3, as a Th2-specific transcription factor, plays a central role in positive-regulating Th2 development. So whether the expression of GATA-3 in tumor cells has any effect on tumor development is a question of interest. In the present study, we inhibited the expression of GATA-3 in tumor cells through antisense RNA blockade technique, and observed its effects on tumor in vitro and in vivo. Our results showed that antisense GATA-3 treatment could inhibit the expression of TNF-α and Th2 cytokines in tumor cells, and antisense-induced blockade of GATA-3 could also depress tumor growth in tumor-bearing mice. We suggest that the ratio of T-bet/GATA-3 can be evaluated as a more important marker of the status of Thl/Th2 type.
