Due to seasonal climate alterations,the microstructure and permeability of granite residual soil are easily affected by multiple dry-wet cycles.The X-ray micro computed tomography(micro-CT)acted as a nondestructive to...Due to seasonal climate alterations,the microstructure and permeability of granite residual soil are easily affected by multiple dry-wet cycles.The X-ray micro computed tomography(micro-CT)acted as a nondestructive tool for characterizing the microstructure of soil samples exposed to a range of damage levels induced by dry-wet cycles.Subsequently,the variations of pore distribution and permeability due to drywet cycling effects were revealed based on three-dimensional(3D)pore distribution analysis and seepage simulations.According to the results,granite residual soils could be separated into four different components,namely,pores,clay,quartz,and hematite,from micro-CT images.The reconstructed 3D pore models dynamically demonstrated the expanding and connecting patterns of pore structures during drywet cycles.The values of porosity and connectivity are positively correlated with the number of dry-wet cycles,which were expressed by exponential and linear functions,respectively.The pore volume probability distribution curves of granite residual soil coincide with the χ^(2)distribution curve,which verifies the effectiveness of the assumption of χ^(2)distribution probability.The pore volume distribution curves suggest that the pores in soils were divided into four types based on their volumes,i.e.micropores,mesopores,macropores,and cracks.From a quantitative and visual perspective,considerable small pores are gradually transformed into cracks with a large volume and a high connectivity.Under the action of dry-wet cycles,the number of seepage flow streamlines which contribute to water permeation in seepage simulation increases distinctly,as well as the permeability and hydraulic conductivity.The calculated hydraulic conductivity is comparable with measured ones with an acceptable error margin in general,verifying the accuracy of seepage simulations based on micro-CT results.展开更多

Some int...Melanosomes, isolated by centrifugal separation from culture broth of B16 melanoma cells derived from mouse, were observed by scanning electron microscopy (SEM), and by transmission electron microscopy (TEM). Some interesting structural features were found inside and outside of the melanosomes. By SEM observation, the melanosomes were ellipsoid shape, their surface was not smooth and was covered with rough substructure, 10 to 20 nm particles. By TEM, uneven structure and micro particles were observed in the melanosomes. Furthermore, three-dimensional analysis was tried by using the ultra-high voltage electron microscopy(UHVEM). Micrographs of the melanosomes were taken at various tilted angles by UHVEM, after preparing 500 nm thickness specimens stained with lead citrate. From the micrographs collected, the three-dimensional structures were reconstructed by using i-mode software. Melanin stained by lead and non stained parts was clearly observed in the reconstructed structure. Non stained parts were round, regular size, and distributed widely in the melanosomes.展开更多

The authors are interested in investigating the structure and bonding in lanthanide and actinide metal complexes using 166Er, t55Gd, and 237Np Mtissbauer spectroscopies in connection with single-crystal and/or powder X-ray diffraction, making clear the differences on their structures as well as the differences in the participation of 4f and 5f orbitals in the chemical bonds. In this article, the crystal structures of two novel Gd(Ⅲ) acetylacetonato complexes, Gd(pta)3 · 2H2O (pta = 1,1,1 -trifluoro-5,5-dimethy 1-2,4-hexanedione) and Gd(bfa)3 · 2H2O (bfa = 1, 1, 1 -trifluoro-4-phenyl-2-4-butanedione) were reported. Though both of them were dihydrate and had distorted square antiprismatical structure, Gd(pta)3 · 2H2O crystallizes in the P 2 1/n (#14) monoclinic space group and its lattice parameters are a = 1.4141(6) nm, b = 1.0708(3) nm, c =2.2344(4) nm, β =952.4(2)°, and Gd(bfa)3· 2H2O crystallizes in P 212121 orthorhombic space group and its lattice parameters were a = 1.322 (1) nm, b = 2.295 (1) nm, c = 1. 0786(8) nm. In the meantime, the authors had finished a systematic investigation on the ^155Gd Mossbauer isomer shift (δ) of various Gd(Ⅲ) metal complexes having a different coordination number (C.N.) and different ratios coordinating oxygen to nitrogen. A tendency for the 6 value to decrease with an increase in the C.N, and the number of the nitrogen atom coordinating to Gd was confirmed. This indicated that the Gd-O and/or Gd-N bond in the investigated Gd(Ⅲ) metal complexes had a small covalent contribution, which was possible to be deduced from the O and/or N atoms of the lisands donating electrons to 6s, 5d, and 4f orbitals of Gd.展开更多

In one example, vitrification and peeling show the annuli hidden under the chorion. Sectioning and 3-D reconstruction display an intact digestive tract from the inverted introvert to the terminal anus. The inverted introvert forms a rugby cavum. The following digestive tract is rope-like coiling, parallel to the body axis, about 650 μm in length, and uniform in diameter (-80 μm). An exquisitely preserved pipe-like structure is hidden in the middle of the rope-like structure, diameter 20--40 lam, with a length of -120 μm. We interpret this pipe-like structure as the possible epidermis of the gut and its surroundings as the possible residue of musculature, similar to that in Priapulans. The two symmetrical rod-shape structures connecting the body wall and digestive tract are interpreted as the possible retractor muscles. After comparing the well preserved Left-form and Right-form Body of Markuelia, we suggest that they may represent a dimorphism. Counted directly, one sample of Markuelia hunanensis possesses 62 annulations and the other 68.展开更多

Through introducing Zernike phase contrast and heavy metal staining, images taken at 8 keV displayed sufficient contrast for observing nanoscale fine structures, such as the spermatheca cochleate duct and the subapex of the flagellum, which have some implications for the study of the sperm transfer process and genital evolution in insects. This work shows that both the spatial resolution and the contrast characteristic of hard X-ray microscopy are quite promising for insect morphology studies and, particularly, provide an attractive alternative to the destructive techniques used for investigating internal soft tissues.展开更多

Furthermore, the number of pinholes is small. The analysis of the sample surface and interface further verifies this result by using x-ray photoemission spectroscopy. At the same time, PTCDA is found to have the ability of restraining the diffusion of chemical constituents from ITO to the hole transport layer, which is beneficial to the improvement of the performance and the useful lifetime of the organic light emitting diodes (OLEDs).展开更多

Until recentl...The geometric and spatial characteristics of pore structures determine the permeability and water retention of soils, which have important effects on soil functional diversity and ecological restoration. Until recently, there have not been tools and methods to visually and quantitatively describe the characteristics of soil pores. To solve this problem, this research reconstructs the geometry and spatial distribution of soil pores by the marching cubes method, texture mapping method and the ray casting method widely used in literature. The objectives were to explore an optimal method for three-dimensional visualization of soil pore structure by comparing the robustness of the three methods on soil CT images with single pore structure and porosity ranging from low (2–5%) to high (12–18%), and to evaluate the reconstruction performance of the three methods with different geometric features. The results demonstrate that there are aliases (jagged edges) and deficiency at the boundaries of the model reconstructed by the marching cubes method and pore volumes are smaller than the ground truth, whereas the results of the texture mapping method lack the details of pore structures. For all the soil images, the ray casting method is preferable since it better preserves the pore characteristics of the ground truth. Furthermore, the ray casting method produced the best soil pore model with higher rendering speed and lower memory consumption. Therefore, the ray casting method provides a more advanced method for visualization of pore structures and provides an optional technique for the study of the transport of moisture and the exchange of air in soil.展开更多

This clearly does not mee...Detection of sulfur-oxidizing bacteria has largely been dependent on targeted gene sequencing technology or traditional cell cultivation, which usually takes from days to months to carry out. This clearly does not meet the requirements of analysis for time-sensitive samples and/or complicated environmental samples. Since energy-dispersive X-ray spectrometry(EDS) can be used to simultaneously detect multiple elements in a sample, including sulfur, with minimal sample treatment, this technology was applied to detect sulfur-oxidizing bacteria using their high sulfur content within the cell. This article describes the application of scanning electron microscopy imaging coupled with EDS mapping for quick detection of sulfur oxidizers in contaminated environmental water samples, with minimal sample handling. Scanning electron microscopy imaging revealed the existence of dense granules within the bacterial cells, while EDS identified large amounts of sulfur within them. EDS mapping localized the sulfur to these granules. Subsequent 16S rRNA gene sequencing showed that the bacteria detected in our samples belonged to the genus Chromatium, which are sulfur oxidizers. Thus, EDS mapping made it possible to identify sulfur oxidizers in environmental samples based on localized sulfur within their cells, within a short time(within 24 h of sampling). This technique has wide ranging applications for detection of sulfur bacteria in environmental water samples.展开更多

In this study,the permeabilities of Berea and Otway sandstones were measured under different confining pressures,and porosity was investigated through mercury intrusion porosimetry(MIP).The total porosities of the Ber...In this study,the permeabilities of Berea and Otway sandstones were measured under different confining pressures,and porosity was investigated through mercury intrusion porosimetry(MIP).The total porosities of the Berea and Otway sandstones were approximately 17.4%and 25%,respectively.Pore size distributions of each sandstone were almost the same,but the pores in the Otway sandstone were slightly narrower.However,the permeability of the Otway sandstone was smaller than that of the Berea sandstone by one order of magnitude.Three-dimensional(3D)void geometry and geometrical properties of the void spaces relevant to flow were compared to obtain the relation between the permeability differences and porosities of the two sandstones.The 3D geometrical analysis using microfocus X-ray computed tomography(CT)was performed,and the pore geometries of both sandstones were compared using the 3D medial axis(3DMA)method.Pore and throat radii,pore coordination number,tortuosity,number of connecting paths,connecting path volume,and other factors were determined using 3DMA.The Otway sandstone was characterized by a small effective throat/pore radius ratio.Based on the fluid flow mechanism,the lower effective throat/pore radius ratio results in a lower permeability induced by the fluid energy loss,which means that the 3D geometrical shape of void spaces affects the permeability value.展开更多

Results indicated that there was no difference in the total area and volume of mitochondria between 5-day-old drosophila and 20-day-old drosophila in all sections, but the ratio of mitochondrial total areas to axon total areas, as well as mitochondrial density of 20-day-old drosophila, was lower than that of 5-day-old drosophila. The number of mitochondria, whose volume was less than 1 000 000 IJm3, and between 1 000 000 pm3 and 10 000 000 pm3, was higher in 20-day-old drosophila than that in 5-day-old drosophila. The number of mitochondria with a volume between 1 000 000 pm3 and 100 000 000 IJm3 was apparently higher than those with a volume less than 1 000 000 t.lm3 or larger than 100 000 000 IJm3. In addition, the number of mitochondria with a volume more than 100 000 000 tJm3 was small; however, the volume was nearly 70% of the total volume in both 5-day-old and 20-day-old drosophila. In contrast, the number of mitochondria with a volume between 1 000 000 t.Jm3 and 10 000 000 IJm3 was large, but the volume was less than 30% of the total volume. These experimental findings suggest that changes in mitochondrial morphology and number in motor neurons from the dorsal longitudinal muscle of drosophila are present during different ages.展开更多

These t...The aim of this study was to apply the existing techniques that enable examination ofmacadamia kernels to provide a better understanding of physico-chemical properties of kernels during postharvest processing. These techniques, such as X-ray tomography, could be applied for quality monitoring in the macadamia industry. The objectives of this study were to investigate the browning centre symptoms that usually occur in macadamia nuts-in-shell. The applied techniques included confocal microscopy, X-ray tomography and magnetic resonance imaging (MRI). Five different varieties of macadamia nuts (A38, 246, 816, 842 and Daddow) were selected to include distinct characteristics, such as drop pattern and growing location. Analysis of the microstructure of kernels by confocal microscopy showed the distribution of possible brown pigment compounds as well as the distribution of lipids, carbohydrates and proteins inside macadamia cells. Physical properties data, including shell density and seed to volume ratio, were obtained by X-ray tomography. Magnetic resonance diffusion tensor imaging used in this study showed marked differences in microstructure which indicate that different varieties exhibit different microstructures expressed as fraction ofanisotropy and apparent diffusion coefficient that appear to be related to the occurrence of the brown centre. Hence, the findings of this study have potential to improve the existing postharvest techniques used in the macadamia processing industry. They will be of benefit to the industry in terms of improved quality control and cost reduction.展开更多

Ptychography is a diffraction-based X-ray microscopy technique in which an extended sample is scanned by a coherent beam with overlapped illuminated areas and complex transmission function of the sample is obtained by...Ptychography is a diffraction-based X-ray microscopy technique in which an extended sample is scanned by a coherent beam with overlapped illuminated areas and complex transmission function of the sample is obtained by applying iterative phase retrieval algorithms to the diffraction patterns recorded at each scanned position.It permits quantitatively imaging of non-crystalline specimens at a resolution limited only by the X-ray wavelength and the maximal scattering angle detected.In this paper,the development of soft X-ray ptychography method at the BL08U1 A beamline of Shanghai Synchrotron Radiation Facility is presented.The experimental setup,experimental parameters selection criteria,and post-experimental data analyzing procedures are presented in detail with a prospect of high-resolution image reconstruction in real time.The performance of this newly implemented method is demonstrated through the measurements of a resolution test pattern and two real samples:Pt-Co alloy nanoparticles and a breast cancer cell.The results indicate that strong scattering specimens can be reconstructed to sub-20 nm resolution,while a sub-25 nm resolution for biological specimens can be achieved.展开更多
Heavy ion beams with high linear energy transfer exhibit more beneifcial physical and biological performance than conventional X-rays, thus improving the potential of this type of radiotherapy in the treatment of canc...Heavy ion beams with high linear energy transfer exhibit more beneifcial physical and biological performance than conventional X-rays, thus improving the potential of this type of radiotherapy in the treatment of cancer. However, these two radiotherapy modalities both cause inevitable brain injury. The objective of this study was to evaluate the effects of heavy ion and X-ray irra-diation on the cytoskeleton and cytomechanical properties of rat cortical neurons, as well as to determine the potential mechanism of neuronal injury after irradiation. Cortical neurons from 30 new-born mice were irradiated with heavy ion beams at a single dose of 2 Gy and X-rays at a single dose of 4 Gy;subsequent evaluation of their effects were carried out at 24 hours after irradiation. An immunolfuorescence assay showed that after irradiation with both the heavy ion beam and X-rays, the number of primary neurons was signiifcantly decreased, and there was ev-idence of apoptosis. Radiation-induced neuronal injury was more apparent after X-irradiation. Under atomic force microscopy, the neuronal membrane appeared rough and neuronal rigidity had increased. These cell changes were more apparent following exposure to X-rays. Our ifnd-ings indicated that damage caused by heavy ion and X-ray irradiation resulted in the structural distortion and rearrangement of the cytoskeleton, and affected the cytomechanical properties of the cortical neurons. Moreover, this radiation injury to normal neurons was much severer after irradiation with X-rays than after heavy ion beam irradiation.展开更多
In this article, we report the results of our detailed investigations of the growth kinetics of zero-dimensional nanocrystals as well as one-dimensional nanorods by the combined use of small angel X-ray scattering (S...In this article, we report the results of our detailed investigations of the growth kinetics of zero-dimensional nanocrystals as well as one-dimensional nanorods by the combined use of small angel X-ray scattering (SAXS), transmission electron microscopy (TEM) along with other physical techniques. The study includes growth kinetics of gold nanocrystals formed by the reduction of HAuCl4 by tetrakis(hydroxymethyl) phosphonium chloride in aqueous solution, of CdSe nanocrystals formed by the reaction of cadmium stearate and selenium under solvothermal conditions, and of ZnO nanorods formed by the reaction of zinc acetate with sodium hydroxide under solvothermal conditions in the absence and presence of capping agents. The growth of gold nanocrystals does not follow the diffusion-limited Ostwald ripening, and instead follows a Sigmoidal rate curve. The heat change associated with the growth determined by isothermal titration calorimetry is about 10 kcal·mol^-1 per I nm increase in the diameter of the nanocrystals. In the case of CdSe nanocrystals also, the growth mechanism deviates from diffusion-limited growth and follows a combined model containing both diffusion and surface reaction terms. Our study of the growth kinetics of uncapped and poly(vinyl pyrollidone) (PVP)-capped ZnO nanorods has yielded interesting insights. We observe small nanocrystals next to the ZnO nanorods after a lapse of time in addition to periodic focusing and defocusing of the width of the length distribution. These observations lend support to the diffusion-limited growth model for the growth of uncapped ZnO nanorods. Accordingly, the time dependence on the length of uncapped nanorods follows the L3 law as required for diffusion-limited Ostwald ripening. The PVP-capped nanorods, however, show a time dependence, which is best described by a combination of diffusion (L^3) and surface reaction (L^2) terms.展开更多

Recently, the development of advanced analytical transmission electron microscopy(TEM) techniques, especially quantitative high-angle annular darkfield(HAADF) imaging and high-energy resolution spectroscopy analysis in scanning transmission electron microscopy(STEM) at the atomic scale, strengthens the power of(S)TEM in analyzing the structural/chemical information of heterogeneous catalysts. Three-dimensional reconstruction from two-dimensional projected images and the real-time recording of structural evolution during catalytic reactions using in-situ(S)TEM methods further broaden the scope of(S)TEM observation. The atomic-scale structural information obtained from high-resolution(S)TEM has proven to be of significance for better understanding and designing of new catalysts with enhanced performance.展开更多

文摘Due to seasonal climate alterations,the microstructure and permeability of granite residual soil are easily affected by multiple dry-wet cycles.The X-ray micro computed tomography(micro-CT)acted as a nondestructive tool for characterizing the microstructure of soil samples exposed to a range of damage levels induced by dry-wet cycles.Subsequently,the variations of pore distribution and permeability due to drywet cycling effects were revealed based on three-dimensional(3D)pore distribution analysis and seepage simulations.According to the results,granite residual soils could be separated into four different components,namely,pores,clay,quartz,and hematite,from micro-CT images.The reconstructed 3D pore models dynamically demonstrated the expanding and connecting patterns of pore structures during drywet cycles.The values of porosity and connectivity are positively correlated with the number of dry-wet cycles,which were expressed by exponential and linear functions,respectively.The pore volume probability distribution curves of granite residual soil coincide with the χ^(2)distribution curve,which verifies the effectiveness of the assumption of χ^(2)distribution probability.The pore volume distribution curves suggest that the pores in soils were divided into four types based on their volumes,i.e.micropores,mesopores,macropores,and cracks.From a quantitative and visual perspective,considerable small pores are gradually transformed into cracks with a large volume and a high connectivity.Under the action of dry-wet cycles,the number of seepage flow streamlines which contribute to water permeation in seepage simulation increases distinctly,as well as the permeability and hydraulic conductivity.The calculated hydraulic conductivity is comparable with measured ones with an acceptable error margin in general,verifying the accuracy of seepage simulations based on micro-CT results.
文摘Melanosomes, isolated by centrifugal separation from culture broth of B16 melanoma cells derived from mouse, were observed by scanning electron microscopy (SEM), and by transmission electron microscopy (TEM). Some interesting structural features were found inside and outside of the melanosomes. By SEM observation, the melanosomes were ellipsoid shape, their surface was not smooth and was covered with rough substructure, 10 to 20 nm particles. By TEM, uneven structure and micro particles were observed in the melanosomes. Furthermore, three-dimensional analysis was tried by using the ultra-high voltage electron microscopy(UHVEM). Micrographs of the melanosomes were taken at various tilted angles by UHVEM, after preparing 500 nm thickness specimens stained with lead citrate. From the micrographs collected, the three-dimensional structures were reconstructed by using i-mode software. Melanin stained by lead and non stained parts was clearly observed in the reconstructed structure. Non stained parts were round, regular size, and distributed widely in the melanosomes.

文摘Some functional lanthanide metal complexes, such as acetylacetonato complexes, ethylenediaminetetraacetato complexes, were successfully applied for diagnostic technique. The authors are interested in investigating the structure and bonding in lanthanide and actinide metal complexes using 166Er, t55Gd, and 237Np Mtissbauer spectroscopies in connection with single-crystal and/or powder X-ray diffraction, making clear the differences on their structures as well as the differences in the participation of 4f and 5f orbitals in the chemical bonds. In this article, the crystal structures of two novel Gd(Ⅲ) acetylacetonato complexes, Gd(pta)3 · 2H2O (pta = 1,1,1 -trifluoro-5,5-dimethy 1-2,4-hexanedione) and Gd(bfa)3 · 2H2O (bfa = 1, 1, 1 -trifluoro-4-phenyl-2-4-butanedione) were reported. Though both of them were dihydrate and had distorted square antiprismatical structure, Gd(pta)3 · 2H2O crystallizes in the P 2 1/n (#14) monoclinic space group and its lattice parameters are a = 1.4141(6) nm, b = 1.0708(3) nm, c =2.2344(4) nm, β =952.4(2)°, and Gd(bfa)3· 2H2O crystallizes in P 212121 orthorhombic space group and its lattice parameters were a = 1.322 (1) nm, b = 2.295 (1) nm, c = 1. 0786(8) nm. In the meantime, the authors had finished a systematic investigation on the ^155Gd Mossbauer isomer shift (δ) of various Gd(Ⅲ) metal complexes having a different coordination number (C.N.) and different ratios coordinating oxygen to nitrogen. A tendency for the 6 value to decrease with an increase in the C.N, and the number of the nitrogen atom coordinating to Gd was confirmed. This indicated that the Gd-O and/or Gd-N bond in the investigated Gd(Ⅲ) metal complexes had a small covalent contribution, which was possible to be deduced from the O and/or N atoms of the lisands donating electrons to 6s, 5d, and 4f orbitals of Gd.

文摘In the light of Synchrotron Radiation X-ray Tomographic Microscopy (SRXTM), the internal structure of Markuelia hunanensis is revealed. In one example, vitrification and peeling show the annuli hidden under the chorion. Sectioning and 3-D reconstruction display an intact digestive tract from the inverted introvert to the terminal anus. The inverted introvert forms a rugby cavum. The following digestive tract is rope-like coiling, parallel to the body axis, about 650 μm in length, and uniform in diameter (-80 μm). An exquisitely preserved pipe-like structure is hidden in the middle of the rope-like structure, diameter 20--40 lam, with a length of -120 μm. We interpret this pipe-like structure as the possible epidermis of the gut and its surroundings as the possible residue of musculature, similar to that in Priapulans. The two symmetrical rod-shape structures connecting the body wall and digestive tract are interpreted as the possible retractor muscles. After comparing the well preserved Left-form and Right-form Body of Markuelia, we suggest that they may represent a dimorphism. Counted directly, one sample of Markuelia hunanensis possesses 62 annulations and the other 68.

文摘A penetrating view of the three-dimensional nanostructure of female spermatheca and male flagellum in the species Aleochara verna is obtained with 100-nm resolution using a hard X-ray microscope, which provides a fast noninvasive imaging technology for insect morphology. Through introducing Zernike phase contrast and heavy metal staining, images taken at 8 keV displayed sufficient contrast for observing nanoscale fine structures, such as the spermatheca cochleate duct and the subapex of the flagellum, which have some implications for the study of the sperm transfer process and genital evolution in insects. This work shows that both the spatial resolution and the contrast characteristic of hard X-ray microscopy are quite promising for insect morphology studies and, particularly, provide an attractive alternative to the destructive techniques used for investigating internal soft tissues.

文摘Through the investigation of the sample surface and interface of 3, 4, 9, 10-perylenetetracarboxylic dianhydride (PTCDA)/indium-tin-oxide (ITO) thin films using atomic force microscopy, it has been found that the surface is complanate, the growth is uniform and the defects cover basically the surface of ITO. Furthermore, the number of pinholes is small. The analysis of the sample surface and interface further verifies this result by using x-ray photoemission spectroscopy. At the same time, PTCDA is found to have the ability of restraining the diffusion of chemical constituents from ITO to the hole transport layer, which is beneficial to the improvement of the performance and the useful lifetime of the organic light emitting diodes (OLEDs).

文摘The geometric and spatial characteristics of pore structures determine the permeability and water retention of soils, which have important effects on soil functional diversity and ecological restoration. Until recently, there have not been tools and methods to visually and quantitatively describe the characteristics of soil pores. To solve this problem, this research reconstructs the geometry and spatial distribution of soil pores by the marching cubes method, texture mapping method and the ray casting method widely used in literature. The objectives were to explore an optimal method for three-dimensional visualization of soil pore structure by comparing the robustness of the three methods on soil CT images with single pore structure and porosity ranging from low (2–5%) to high (12–18%), and to evaluate the reconstruction performance of the three methods with different geometric features. The results demonstrate that there are aliases (jagged edges) and deficiency at the boundaries of the model reconstructed by the marching cubes method and pore volumes are smaller than the ground truth, whereas the results of the texture mapping method lack the details of pore structures. For all the soil images, the ray casting method is preferable since it better preserves the pore characteristics of the ground truth. Furthermore, the ray casting method produced the best soil pore model with higher rendering speed and lower memory consumption. Therefore, the ray casting method provides a more advanced method for visualization of pore structures and provides an optional technique for the study of the transport of moisture and the exchange of air in soil.

文摘The three-dimensional(3D)model is of great significance to analyze the performance of nonwovens.However,the existing modelling methods could not reconstruct the 3D structure of nonwovens at low cost.A new method based on deep learning was proposed to reconstruct 3D models of nonwovens from multi-focus images.A convolutional neural network was trained to extract clear fibers from sequence images.Image processing algorithms were used to obtain the radius,the central axis,and depth information of fibers from the extraction results.Based on this information,3D models were built in 3D space.Furthermore,self-developed algorithms optimized the central axis and depth of fibers,which made fibers more realistic and continuous.The method with lower cost could reconstruct 3D models of nonwovens conveniently.

文摘Detection of sulfur-oxidizing bacteria has largely been dependent on targeted gene sequencing technology or traditional cell cultivation, which usually takes from days to months to carry out. This clearly does not meet the requirements of analysis for time-sensitive samples and/or complicated environmental samples. Since energy-dispersive X-ray spectrometry(EDS) can be used to simultaneously detect multiple elements in a sample, including sulfur, with minimal sample treatment, this technology was applied to detect sulfur-oxidizing bacteria using their high sulfur content within the cell. This article describes the application of scanning electron microscopy imaging coupled with EDS mapping for quick detection of sulfur oxidizers in contaminated environmental water samples, with minimal sample handling. Scanning electron microscopy imaging revealed the existence of dense granules within the bacterial cells, while EDS identified large amounts of sulfur within them. EDS mapping localized the sulfur to these granules. Subsequent 16S rRNA gene sequencing showed that the bacteria detected in our samples belonged to the genus Chromatium, which are sulfur oxidizers. Thus, EDS mapping made it possible to identify sulfur oxidizers in environmental samples based on localized sulfur within their cells, within a short time(within 24 h of sampling). This technique has wide ranging applications for detection of sulfur bacteria in environmental water samples.

Crystal data: H36Cl2K4W12Na2O56, Mr = 3411.77, monoclinic, space group P21/c, a = 11.5734(16), b = 14.3136(19), c = 15.459(2)A, β = 105.601(2)°, V = 2466.5(6)A^3, Z = 2, Dc = 4.594 g/cm^3, F(000) = 3008, μ = 28.447 mm^-1, R = 0.0574 and wR = 0.1239 (Ⅰ〉 2σ(Ⅰ)). Single-crystal X-ray diffraction analysis results reveal that a three-dimensional architecture in the title compound is constructed from the H2W12O42^10- units linked by potassium.

文摘In this study,the permeabilities of Berea and Otway sandstones were measured under different confining pressures,and porosity was investigated through mercury intrusion porosimetry(MIP).The total porosities of the Berea and Otway sandstones were approximately 17.4%and 25%,respectively.Pore size distributions of each sandstone were almost the same,but the pores in the Otway sandstone were slightly narrower.However,the permeability of the Otway sandstone was smaller than that of the Berea sandstone by one order of magnitude.Three-dimensional(3D)void geometry and geometrical properties of the void spaces relevant to flow were compared to obtain the relation between the permeability differences and porosities of the two sandstones.The 3D geometrical analysis using microfocus X-ray computed tomography(CT)was performed,and the pore geometries of both sandstones were compared using the 3D medial axis(3DMA)method.Pore and throat radii,pore coordination number,tortuosity,number of connecting paths,connecting path volume,and other factors were determined using 3DMA.The Otway sandstone was characterized by a small effective throat/pore radius ratio.Based on the fluid flow mechanism,the lower effective throat/pore radius ratio results in a lower permeability induced by the fluid energy loss,which means that the 3D geometrical shape of void spaces affects the permeability value.
文摘This study aimed to reconstruct a three-dimensional map of axonal mitochondria using Fiji and Neurolucida software, and to observe directly the morphology and distribution of mitochondria in axons of motor neurons in dorsal longitudinal flight muscles of drosophila aged 5 days and 20 days, using electron microscopy. Results indicated that there was no difference in the total area and volume of mitochondria between 5-day-old drosophila and 20-day-old drosophila in all sections, but the ratio of mitochondrial total areas to axon total areas, as well as mitochondrial density of 20-day-old drosophila, was lower than that of 5-day-old drosophila. The number of mitochondria, whose volume was less than 1 000 000 IJm3, and between 1 000 000 pm3 and 10 000 000 pm3, was higher in 20-day-old drosophila than that in 5-day-old drosophila. The number of mitochondria with a volume between 1 000 000 pm3 and 100 000 000 IJm3 was apparently higher than those with a volume less than 1 000 000 t.lm3 or larger than 100 000 000 IJm3. In addition, the number of mitochondria with a volume more than 100 000 000 tJm3 was small; however, the volume was nearly 70% of the total volume in both 5-day-old and 20-day-old drosophila. In contrast, the number of mitochondria with a volume between 1 000 000 t.Jm3 and 10 000 000 IJm3 was large, but the volume was less than 30% of the total volume. These experimental findings suggest that changes in mitochondrial morphology and number in motor neurons from the dorsal longitudinal muscle of drosophila are present during different ages.
文摘The aim of this study was to apply the existing techniques that enable examination ofmacadamia kernels to provide a better understanding of physico-chemical properties of kernels during postharvest processing. These techniques, such as X-ray tomography, could be applied for quality monitoring in the macadamia industry. The objectives of this study were to investigate the browning centre symptoms that usually occur in macadamia nuts-in-shell. The applied techniques included confocal microscopy, X-ray tomography and magnetic resonance imaging (MRI). Five different varieties of macadamia nuts (A38, 246, 816, 842 and Daddow) were selected to include distinct characteristics, such as drop pattern and growing location. Analysis of the microstructure of kernels by confocal microscopy showed the distribution of possible brown pigment compounds as well as the distribution of lipids, carbohydrates and proteins inside macadamia cells. Physical properties data, including shell density and seed to volume ratio, were obtained by X-ray tomography. Magnetic resonance diffusion tensor imaging used in this study showed marked differences in microstructure which indicate that different varieties exhibit different microstructures expressed as fraction ofanisotropy and apparent diffusion coefficient that appear to be related to the occurrence of the brown centre. Hence, the findings of this study have potential to improve the existing postharvest techniques used in the macadamia processing industry. They will be of benefit to the industry in terms of improved quality control and cost reduction.
文摘Plasma X-ray sources for biological microscopy have been produced by focusing single shots from Nd:glass laser onto carbon rod targets at irradiances between 1 × 1013 W⋅cm−2 and 3 × 1013 W⋅cm−2 to expose test objects. The optimum parameters needed for obtaining high accurate information on the samples under test namely: the minimum energies and irradiances at a range of angles between the incoming laser beam and the normal to the resist, the depth of exposure of the photoresist as a function of incident laser energy (and irradiance) were concluded in this work.

文摘To explore ways to improve the accuracy of quantitative analysis of samples in the micrometer to nanometer range of magnitudes,we adopted analytical transmission electron microscopy(AEM/EDS)for qualitative and quantitative analysis of pyrite materials.Additionally,the k factor of pyrite is calculated experimentally.To develop an appropriate non-standard quantitative analysis model for pyrite materials,the experimentally calculated k factor is compared with that estimated from the non-standard quantitative analytical model of the instrument software.The experimental findings demonstrate that the EDS attached to a TEM can be employed for precise quantitative analysis of micro-and nanoscale regions of pyrite materials.Furthermore,it serves as a reference for improving the results of the EDS quantitative analysis of other sulfides.

文摘Ptychography is a diffraction-based X-ray microscopy technique in which an extended sample is scanned by a coherent beam with overlapped illuminated areas and complex transmission function of the sample is obtained by applying iterative phase retrieval algorithms to the diffraction patterns recorded at each scanned position.It permits quantitatively imaging of non-crystalline specimens at a resolution limited only by the X-ray wavelength and the maximal scattering angle detected.In this paper,the development of soft X-ray ptychography method at the BL08U1 A beamline of Shanghai Synchrotron Radiation Facility is presented.The experimental setup,experimental parameters selection criteria,and post-experimental data analyzing procedures are presented in detail with a prospect of high-resolution image reconstruction in real time.The performance of this newly implemented method is demonstrated through the measurements of a resolution test pattern and two real samples:Pt-Co alloy nanoparticles and a breast cancer cell.The results indicate that strong scattering specimens can be reconstructed to sub-20 nm resolution,while a sub-25 nm resolution for biological specimens can be achieved.
文摘Heavy ion beams with high linear energy transfer exhibit more beneifcial physical and biological performance than conventional X-rays, thus improving the potential of this type of radiotherapy in the treatment of cancer. However, these two radiotherapy modalities both cause inevitable brain injury. The objective of this study was to evaluate the effects of heavy ion and X-ray irra-diation on the cytoskeleton and cytomechanical properties of rat cortical neurons, as well as to determine the potential mechanism of neuronal injury after irradiation. Cortical neurons from 30 new-born mice were irradiated with heavy ion beams at a single dose of 2 Gy and X-rays at a single dose of 4 Gy;subsequent evaluation of their effects were carried out at 24 hours after irradiation. An immunolfuorescence assay showed that after irradiation with both the heavy ion beam and X-rays, the number of primary neurons was signiifcantly decreased, and there was ev-idence of apoptosis. Radiation-induced neuronal injury was more apparent after X-irradiation. Under atomic force microscopy, the neuronal membrane appeared rough and neuronal rigidity had increased. These cell changes were more apparent following exposure to X-rays. Our ifnd-ings indicated that damage caused by heavy ion and X-ray irradiation resulted in the structural distortion and rearrangement of the cytoskeleton, and affected the cytomechanical properties of the cortical neurons. Moreover, this radiation injury to normal neurons was much severer after irradiation with X-rays than after heavy ion beam irradiation.
文摘In this article, we report the results of our detailed investigations of the growth kinetics of zero-dimensional nanocrystals as well as one-dimensional nanorods by the combined use of small angel X-ray scattering (SAXS), transmission electron microscopy (TEM) along with other physical techniques. The study includes growth kinetics of gold nanocrystals formed by the reduction of HAuCl4 by tetrakis(hydroxymethyl) phosphonium chloride in aqueous solution, of CdSe nanocrystals formed by the reaction of cadmium stearate and selenium under solvothermal conditions, and of ZnO nanorods formed by the reaction of zinc acetate with sodium hydroxide under solvothermal conditions in the absence and presence of capping agents. The growth of gold nanocrystals does not follow the diffusion-limited Ostwald ripening, and instead follows a Sigmoidal rate curve. The heat change associated with the growth determined by isothermal titration calorimetry is about 10 kcal·mol^-1 per I nm increase in the diameter of the nanocrystals. In the case of CdSe nanocrystals also, the growth mechanism deviates from diffusion-limited growth and follows a combined model containing both diffusion and surface reaction terms. Our study of the growth kinetics of uncapped and poly(vinyl pyrollidone) (PVP)-capped ZnO nanorods has yielded interesting insights. We observe small nanocrystals next to the ZnO nanorods after a lapse of time in addition to periodic focusing and defocusing of the width of the length distribution. These observations lend support to the diffusion-limited growth model for the growth of uncapped ZnO nanorods. Accordingly, the time dependence on the length of uncapped nanorods follows the L3 law as required for diffusion-limited Ostwald ripening. The PVP-capped nanorods, however, show a time dependence, which is best described by a combination of diffusion (L^3) and surface reaction (L^2) terms.

文摘Heterogeneous catalysts are the most important catalysts in industrial reactions. Nanocatalysts, with size ranging from hundreds of nanometers to the atomic scale, possess activities that are closely connected to their structural characteristics such as particle size, surface morphology, and three-dimensional topography. Recently, the development of advanced analytical transmission electron microscopy(TEM) techniques, especially quantitative high-angle annular darkfield(HAADF) imaging and high-energy resolution spectroscopy analysis in scanning transmission electron microscopy(STEM) at the atomic scale, strengthens the power of(S)TEM in analyzing the structural/chemical information of heterogeneous catalysts. Three-dimensional reconstruction from two-dimensional projected images and the real-time recording of structural evolution during catalytic reactions using in-situ(S)TEM methods further broaden the scope of(S)TEM observation. The atomic-scale structural information obtained from high-resolution(S)TEM has proven to be of significance for better understanding and designing of new catalysts with enhanced performance.
