Polysaccharides are the important biologically active components found in the peel of Dioscorea opposita Thunb.(DTTP).The influences of 4 extraction methods,namely hot water extraction(W),acidic extraction(HA),hot-com...Polysaccharides are the important biologically active components found in the peel of Dioscorea opposita Thunb.(DTTP).The influences of 4 extraction methods,namely hot water extraction(W),acidic extraction(HA),hot-compressed water extraction(HCW)and enzyme-assisted extraction(EAE),on the yields,physicochemical properties,hypoglycemic and antioxidant activities of polysaccharides from DTTP were studied and compared.Among these DTTP polysaccharides,DTTP-HA was outstanding in several respects.DTTP-HA was the most water soluble;it had the highest total carbohydrate content(85.08%),the highest uronic acid(13.20%)and the highest thermal stability.DTTP-HA and DTTP-W possessed a triple-helix structure.These 4 kinds of polysaccharides have the same types of monosaccharides,but in different molar percentages.Extraction method had a significant impact on the microstructures of the extracted polysaccharides.DTT-HA exhibited irregular structure with many holes.Among the 4 extracted methods,the DTTP-HA and DTTP-W initially exhibited higher hypoglycemic and antioxidant activities.The better bioactivities of DTTP-HA may be related to the above factors.The findings indicated that acid extraction is an effective method to extract polysaccharides with high biological activities from DTTP.展开更多
BACKGROUND Gastric cancer is one of the most common and deadly malignancies worldwide.Despite recent medical progress, the 5-year survival rate of gastric cancer is still unsatisfactory. 5-fluorouracil(5-Fu) is one of...BACKGROUND Gastric cancer is one of the most common and deadly malignancies worldwide.Despite recent medical progress, the 5-year survival rate of gastric cancer is still unsatisfactory. 5-fluorouracil(5-Fu) is one of the first-line antineoplastic treatments for gastric cancer, as it can effectively induce cancer cell apoptosis.However, the effect of 5-Fu is limited due to drug resistance of the malignant tumor. Previous studies have reported that Sotetsuflavone from Cycas revoluta Thunb. can markedly suppress lung cancer cell proliferation by apoptosis,though its effect on gastric cancer remains unknown.AIM To investigate the inhibitory effect of Cycas revoluta Thunb. and to determine whether it can overcome gastric cancer cell drug resistance to 5-Fu.METHODS Cell viability was examined to determine whether the natural extract of Cycas revoluta Thunb. induced gastric cancer cell death. The half-maximal effective concentration and the half-maximal lethal concentration were calculatede.Wound-healing and transwell assays were performed to examine gastric cancer cell motility. Clonogenic assays were performed to investigate the synergistic effects of Cycas revoluta Thunb. with 5-Fu, and apoptotic bodies were detected by Hoechst staining. Western blotting was performed to examine the expression of related proteins and to investigate the molecular mechanism of Cycas revoluta Thunb.-induced cancer cell apoptosis. The expressions of proteins, including mammalian target of rapamycin(mTOR) and p-AKT, were detected in different combinations of treatments for 48 h, then analyzed by ECL detection.RESULTS Gastric cancer cells were more sensitive to the natural extract of Cycas revoluta Thunb. compared to normal gastric epithelial cells, and the extract effectively inhibited gastric cancer cell migration and invasion. The extract improved the anti-cancer effect of 5-Fu by enhancing the chemosensitization of gastric cancer cells. Extract plus 5-Fu further reduced the expression of the drug-resistancerelated proteins p-AKT and mTOR after 48 h compared to 5-Fu alone. Compared to 5-Fu treatment alone, mTOR and p-AKT expression was significantly reduced by about 50% and 75%, respectively. We also found that the natural extract of Cycas revoluta Thunb. further increased 5-Fu-induced gastric cancer cell apoptosis. Expression of apoptosis-related protein X-linked inhibitor of apoptosis protein and apoptosis inducing factor were significantly reduced and increased,respectively, in the 5-Fu-resistant gastric cancer line SGC-7901/R treated with extract plus 5-Fu, while the expression of survivin did not change.CONCLUSION The natural extract of Cycas revoluta Thunb. effectively inhibited gastric cancer cell growth and enhanced the anti-cancer effect of 5-Fu through the AKT-mTOR pathway.展开更多
Objective:To examine the acanthamoebicidal effects of ethyl acetate,aqueous and butanol fractions of dried flower buds of Lonicera japonica(L.japonica) Thunb.(Flos Lonicerae) in vitro.Methods:Acanthamoeba triangularis...Objective:To examine the acanthamoebicidal effects of ethyl acetate,aqueous and butanol fractions of dried flower buds of Lonicera japonica(L.japonica) Thunb.(Flos Lonicerae) in vitro.Methods:Acanthamoeba triangularis isolates were obtained from environmental water samples and identified by PCR.They were exposed to ethyl acetate,water and butanol fractions of L.japonica Thunb.at concentrations ranging from 0.5 mg/m L to 1.5 mg/m L.The extracts were evaluated for growth inhibition at 24,48 and 72 h,respectively.Chlorogenic acid at a concentration of 1 mg/mL was examined for inhibition of encystment.Results:Ethyl acetate fraction at a concentration of 1.5 mg/mL evoked a significant reduction of trophozoite viability by 48.9% after 24 h,49.2% after 48 h and 33.7% after 72 h Chlorogenic acid,the major active constituent of L.japonica Thunb.at the concentration of 1 mg/mL reduced the cysts/trophozoite ratio by 100% after 24 h,84.0% after 48 h and 72.3% after 72 h.This phenolic compound at concentration of 1 mg/mL concurrent with 0.6% hydrogen peroxide inhibited hydrogen peroxide-induced encystment by 92.8% at 72 h.Conclusions:Results obtained from this study show that ethyl acetate fraction at 1.5 mg/m L is the most potent fraction of L.japonica Thunb.and its major constituent chlorogenic acid showed the remarkable inhibition of encystment at a concentration of 1 mg/mL.展开更多

[Methods] Samples of 35 different sources of H. cordata were collected from Guizhou,and polyphenols,total flavonoids,total chlorogenic acid and 7 kinds of flavonoids( xanthol-3-O-glucoside,hyperoside,quercitrin,rutin,quercetin,kaempferol and isorhamnetin),and DPPH free radical scavenging rate of the extracts were determined. Relationship between the contents of phenolics and the DPPH free radical scavenging rate of the extracts were analyzed by SPSS software. [Results] There were some significant differences in polyphenols,total flavonoids,total chlorogenic acid and the seven flavonoids between samples of different habitats,and the contents of polyphenols,total flavonoids and total chlorogenic acid increased linearly with altitude. There was a significant positive correlation between polyphenols,total flavonoids and total chlorogenic acid content and DPPH free radical scavenging rate of extracts. The contents of kaempferol-3-O-glucoside,hyperoside,quercitrin,rutin,quercetin kaempferol and isorhamnetin were positively correlated with the DPPH free radical scavenging rates of the extracts,except for the significant correlation with quercitrin. [Conclusions]The experimental results suggest that the antioxidant capacity of H. cordata is derived from the total contribution of the phenolic components,indicating that these components have overall equivalence of antioxidant capacities. This provides a new research idea for solving the key problems of quality control of traditional Chinese medicine.展开更多
Microwave-assisted extraction(MAE)was used for extraction of effective components of sarcandra glabra(Thunb.),and then chromatographic fingerprint of sarcandra glabra(Thunb.)was studied by high performance liquid chro...Microwave-assisted extraction(MAE)was used for extraction of effective components of sarcandra glabra(Thunb.),and then chromatographic fingerprint of sarcandra glabra(Thunb.)was studied by high performance liquid chromatography/diode array detector(HPLC/DAD).The conditions of MAE were optimized by an orthogonal experiment,and then the authentication and validation of the chromatographic fingerprint were conducted.Nine peaks were identified as common peaks in the fingerprint chromatograms,and isofraxidin was considered as a reference compound and quantified.Relative standard deviations of retention time and peak area of each component were less than 3% and 8%,respectively.Similarity and difference analysis were conducted by use of PCA and relation coefficient.Twenty batches of sarcandra glabra(Thunb.)samples from two different producing areas could be classified into two different groups in the PCA model.The results showed that MAE-HPLC/DAD method was simple,efficient and stable for the study of complex chromatographic fingerprint of sarcandra glabra(Thunb.),which could provide more reliable and precise information for quality evaluation.展开更多

[Methods]The extraction ra...[Objectives]To optimize extraction process of total flavonoids from Akebia trifoliata( Thunb.) Koidz.,so as to provide references for development and use of Akebia trifoliata( Thunb.) Koidz. [Methods]The extraction rate of total flavonoids of Akebia trifoliata( Thunb.)Koidz. was taken as observation indicator. On the basis of single factor experiment,central composite design( CCD) was used to evaluate the effects of the extraction temperature,extraction time,and ethanol concentration on the extraction process. Multiple linear regression and binomial fitting were used,and response surface methodology( RSM) was used to select the optimum extraction process. [Results] The optimum extraction process conditions for total flavonoids of Akebia trifoliata( Thunb.) Koidz. was extraction temperature: 83. 92 ℃; extraction time:96. 47 min; ethanol concentration: 63. 92%; extraction times: two times; solid to liquid ratio: 1 ∶ 20; extraction rate of total flavonoids:4. 55%. [Conclusions] The central composite design-response surface methodology( CCD-RSM) is simple,convenient,and feasible for extraction of total flavonoids from Akebia trifoliata( Thunb.) 文摘Microwave-assisted extraction(MAE)was used for extraction of effective components of sarcandra glabra(Thunb.),and then chromatographic fingerprint of sarcandra glabra(Thunb.)was studied by high performance liquid chromatography/diode array detector(HPLC/DAD).The conditions of MAE were optimized by an orthogonal experiment,and then the authentication and validation of the chromatographic fingerprint were conducted.Nine peaks were identified as common peaks in the fingerprint chromatograms,and isofraxidin was considered as a reference compound and quantified.Relative standard deviations of retention time and peak area of each component were less than 3% and 8%,respectively.Similarity and difference analysis were conducted by use of PCA and relation coefficient.Twenty batches of sarcandra glabra(Thunb.)samples from two different producing areas could be classified into two different groups in the PCA model.The results showed that MAE-HPLC/DAD method was simple,efficient and stable for the study of complex chromatographic fingerprint of sarcandra glabra(Thunb.),which could provide more reliable and precise information for quality evaluation.
文摘A new kaempferol glycoside, named kaempferol 3-O-[6'-O-β-D-glucopyranosyl-6''-O-α-L-rhamnopy ranosyl-2''-O-α-L-rharnnopyranosyl]-β-D-glucopyranoside, was isolated from Hydrangea macrophylla (Thunb.) Seringe. Its structure was established by spectroscopic techniques including MS, IR, UV, and 2D NMR.

文摘[Objectives]To optimize extraction process of total flavonoids from Akebia trifoliata( Thunb.) Koidz.,so as to provide references for development and use of Akebia trifoliata( Thunb.) Koidz. [Methods]The extraction rate of total flavonoids of Akebia trifoliata( Thunb.)Koidz. was taken as observation indicator. On the basis of single factor experiment,central composite design( CCD) was used to evaluate the effects of the extraction temperature,extraction time,and ethanol concentration on the extraction process. Multiple linear regression and binomial fitting were used,and response surface methodology( RSM) was used to select the optimum extraction process. [Results] The optimum extraction process conditions for total flavonoids of Akebia trifoliata( Thunb.) Koidz. was extraction temperature: 83. 92 ℃; extraction time:96. 47 min; ethanol concentration: 63. 92%; extraction times: two times; solid to liquid ratio: 1 ∶ 20; extraction rate of total flavonoids:4. 55%. [Conclusions] The central composite design-response surface methodology( CCD-RSM) is simple,convenient,and feasible for extraction of total flavonoids from Akebia trifoliata( Thunb.) Koidz.,and the prediction results are reliable.
