OBJECTIVE: To explore the effects of Yigan Tiaozhi Decoction on serum nitric oxide(NO), endotoxin and retinol binding protein 4(RBP4) in patients with nonalcoholic fatty liver disease(NAFLD), and to provide evidence f...OBJECTIVE: To explore the effects of Yigan Tiaozhi Decoction on serum nitric oxide(NO), endotoxin and retinol binding protein 4(RBP4) in patients with nonalcoholic fatty liver disease(NAFLD), and to provide evidence for clinical treatment of such patients. METHODS: A total of 92 patients with NAFLD admitted to Army General Hospital 263 Clinic from March 2013 to March 2017 were selected, and all patients were divided into the control group(n = 46) and the observation group(n = 46) according to the random number table method. The patients in the control group were given conventional treatment, and the patients in the observation group were treated with Yigan Tiaozhi Decoction on the basis of the control group. Liver function, blood lipid levels, serum NO, endotoxin, RBP4 levels and clinical efficacy were observed and compared between the 2 groups before and after treatment. RESULTS: Before treatment, there was no significant difference in serum alanine aminotransferase, glutamyltranspeptidase, aspartate aminotransferase, total cholesterol and triglyceride levels between the 2 groups(P > 0.05); after treatment, the serum levels of alanine aminotransferase, glutamyltranspeptidase, aspartate aminotransferase, total cholesterol and triglyceride in the observation group were lower than those in the control group, and the difference was statistically significant(P < 0.05); before treatment, there was no significant difference in serum NO, endotoxin and RBP4 levels between the 2 groups(P > 0.05); after treatment, the serum NO, endotoxin and RBP4 levels in the observation group were lower than those in the control group, and the difference was statistically significant(P < 0.05); the total effective rate of treatment in the observation group(89.13%) was significantly higher than that in the control group(76.09%), and the difference was statistically significant(P < 0.05). CONCLUSION: Yigan Tiaozhi Decoction is effective in NAFLD, which can reduce serum NO, endotoxin and RBP4 levels, improve liver function and reduce blood lipid levels.展开更多

Methods: One hundred and thirty-eight patients were enrolled and random...Objective: To observe the clinical efficacy and safety of Tiaozhi Yanggan Decoction (调脂养肝汤,TZYGD) in treating non-alcoholic fatty liver. Methods: One hundred and thirty-eight patients were enrolled and randomized into two groups according to the random number table in a ratio of 3:1, with 8 cases eventually dropping out. The symptoms, signs, liver function markers, blood lipids, iconographic indices and clinical comprehensive efficacy after a 12-week treatment course were assessed in 101 patients treated with TZYGD in the treated group and 29 patients treated with Thiola in the control group. Results: The total effective rate in the treated group and the control group was 81.19% and 68.97%, respectively, showing a significant difference between the two groups with the former being significantly higher than the latter (P〈0.05). Moreover, the improvements in the symptoms, signs, liver function, blood lipids and iconographic indices in the treated group were favorable with no serious adverse reactions. Conclusion: TZYGD is effective and highly safe in treating non-alcoholic fatty liver.展开更多

Methods Chromatography was performed on a Dionex Acclaim C 18 column (250 mm × 4.6 mm, 5.0 μm) at 30 ℃ .The mobile phase was composed of acetonitrile-potassium dihydrogen phosphate solution (0.015 mol/L, 40:60, including 1.7 g/L sodium dodecyl sulfate and phosphoric acid used to regulate pH value to 3.0), with a flow rate of 1.0 mL/min. The detection wavelength was 270 nm. Results The calibration curves of magnoflorine, columbamine, jatrorrhizine, epiberberine, coptisine, palmatine, and berberine were linear in the range of 1.07- 10.65, 0.78-7.55, 0.75-7.50, 1.60-15.95, 2.69-26.85, 2.31-23.10, and 6.04-60.40 mg/mL. The average recoveries of magnoflorine, columbamine, jatrorrhizine, epiberberine, coptisine, palmatine, and berberine were 101.0%, 101.2%, 100.1%, 100.0%, 100.1%, 101.1%, and 99.7%, respectively. Conclusion The method could be used for the quantitative determination of the preparation.展开更多

Forage with cholesterol and lipid plus 1.59 g/kg of YSTZT was fed to Group A every day;for Group B, 22.54 mg/kg gypenoside tablet was added to forage with cholesterol and lipid; for Group C, hyperlipid forage was given and for Group D, only ordinary forage was given. Biochemical parameters were measured and pathomorphological examinations were carried out 6 weeks later.Results: (1) YSTZT obviously lowered the levels of serum total cholesterol(TC), triglyceride(TG), atherosclerotic index(AI), apoprotein(ApoB), lipoprotein [Lp(a)], oxygen low density lipoprotein cholesterol(ox LDL), hydroxyproline(HYP), plasma Ca 2+ , thromboxane B 2(TXB 2), and increased the levels of serum high density lipoprotein cholesterol(HDL C), apoprotein A 1(Apo A 1), ApoA 1 /ApoB, plasma 6 keto PGF 1α ( P <0.01). (2) Pathomorphological examination showed that in Group A aortic intimal atherosclerotic plaque area and arterial intima thickness were obviously reduced, smooth muscle cell hyperplasia and elastic fibers were not seen.Conclusion: YSTZT can inhibit experimental hyperlipemia and atherogenesis. It is an ideal and effective medicine in preventing and treating hyperlipemia and atherosclerosis.展开更多

Methods: Using the aging model induced by hypodermic injection of 5%D-glalactose in mice for forty days. The content of malondialdehyde(MDA) and activity of superoxide dismutase(SOD), glutathione peroxidase (GSH-Px) in serum, activity of monoamine oxidase B (MAO-B), Na+-K+-ATPase in brain were investigated after treatment. Results: Compared with the aging model group, it showed that YSTZT could obviously inhibit the increase of level of MDA in serum and the activities of MAO-B in brain; and enhance the activities of SOD,GSH-Px in serum, and the activities of Na+-K+-ATPase in brain. Furthermore, YSTZT could obviously elevate the tolerance to fatigue and hypoxia in mice. Conclusion: YSTZT could effectively improve the free radical metabolism of senile body, it is an ideal and effective medicine in preventing aging, which is also one of the experimental basis in proving the theory of 'Spleen and Kidney Deficiency complicated with Stasis and Phlegm causes aging'.展开更多
OBJECTIVE:To assess the effect of Jianpi Huazhuo Tiaozhi granules(JHTG)on oxidative stress damage to macrophages and explore the relationship between the levels of this damage and the nicotinamide adenine dinucleotide...OBJECTIVE:To assess the effect of Jianpi Huazhuo Tiaozhi granules(JHTG)on oxidative stress damage to macrophages and explore the relationship between the levels of this damage and the nicotinamide adenine dinucleotide phosphate oxidase(NOX)/reactive oxygen species(ROS)-nuclear transcription factor kappa B(NF-κB)signaling pathway.METHODS:Macrophages cultured in vitro were divided into seven groups:control,model control,inhibitor,positive control,2.5%JHTG,5%JHTG,and10%JHTG.An oxidative stress injury model was established by stimulating macrophages with oxidized low-density lipoprotein.Cell survival and apoptosis were detected by 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide and flow cytometry assays,respectively.Malondialdehyde and superoxide dismutase levels were detected by enzyme-linked immunosorbent assays,while ROS levels were detected using a fluorescence probe.Proteins and m RNAs associated with the NOX/ROS-NF-κB pathway,including NOX4,p22 phox,inhibitor of NF-κB kinase-α(IKK-α),inhibitor of NF-κB kinase-β(IKK-β),and NF-κB were detected by Western blot and PCR,respectively.RESULTS:After JHTG treatment,there were fewer damaged and apoptotic macrophages,while superoxide dismutase levels were elevated.The JHTG-treated groups also showed reduced ROS levels.The molecular changes following JHTG treatment included decreased expression of NOX4 and p22 phox at the protein level and decreased IKK-α,IKK-β,and NF-κB expression at the m RNA level.All of these effects were correlated with the JHTG concentration.CONCLUSION:These results demonstrated that the molecular mechanism underlying the antioxidant activity of JHTG in macrophages is through inhibiting the NOX/ROS-NF-κB pathway.展开更多
