The action mechanism of matrix metalloproteinases-2 (MMP-2) and tissue inhibitor of metalloproteinases-2 (TIMP-2) in the genesis, development and degeneration of haemangioma was investigated by detecting their expression in the tissue of haemangioma in different phases by using the immunohistochemistry. Fifty paraffin-embedded specimens of skin capillary haemangioma were collected, which were documented in the Department of Pathology, Renmin Hospital of Wuhan University from 2000 to 2006. All samples were stained by regular HE method, and proliferative cell nuclear antigen (PCNA) was tested by immunohistochemical S-P method. The samples were classified according to the Mulliken criteria and the expression pattern of PCNA. Immunohistochemical S-P method was ap- plied to detect the expression of MMP-2 and TIMP-2 in proliferative and degenerative phases of cutaneous capillary haemangioma, and in normal skin tissues. In combination with the detection of the expression of factor Ⅷ-related antigen, it was verified that in haemangioma tissues, the cells expressing MMP-2 and TIMP-2 were vascular endothelial cells. The MMP-2 and TIMP-2 expression was quantitatively analyzed by image analysis system (HPIAS-1000), and one-way ANOVA(107) and SNK(q) test were done to analyze average absorbance (A) and positive area rate of immunohistochemically positive particles by using SPSS11.5. The results showed: (1) Among 50 samples of haemangioma, there were 26 proliferative haemangiomas, and 24 degenerative haemangiomas, respectively; (2) The expression of MMP-2 was weak in normal vascular endothelial cells, cytoplasm of connective tissues and extracellular matrix around blood vessels. The expression of MMP-2 in proliferative group was significantly higher than in degenerative group and control group (normal skin) (P〈0.05), but there was no statistically significant difference between the latter two groups; (3) TIMP-2 was highly expressed in normal tissues, degenerative vascular endothelial cells, cytoplasm of connective tissues and extracellular matrix around blood vessels. The expression level of TIMP-2 in proliferative phase was significantly lower than in degenerative phase (P〈0.05), and the expression of TIMP-2 in proliferative phase was significantly different from that in degenerative phase and normal tissues (P〈0.05). It was concluded that in proliferative phase of haemangioma, MMP-2 may promote over-proliferation of endothelial cells of haemangioma, and in degenerative phase, TIMP-2 can inhibit the proliferation of endothelial cells of haemangioma. The two substances play important roles in the genesis, development and degeneration of haemangiomas. 展开更多

目的分析尿液胰岛素样生长因子结合蛋白-7(IGFBP-7)和金属蛋白酶组织抑制剂-2(TIMP-2)在脓毒症急性肾损伤(AKI)中的早期预测价值。方法选取2020年1月—2021年12月山西省人民医院急诊科监护室收治的脓毒症患者58例,并根据KDIGO的诊断标... 目的分析尿液胰岛素样生长因子结合蛋白-7(IGFBP-7)和金属蛋白酶组织抑制剂-2(TIMP-2)在脓毒症急性肾损伤(AKI)中的早期预测价值。方法选取2020年1月—2021年12月山西省人民医院急诊科监护室收治的脓毒症患者58例,并根据KDIGO的诊断标准分成脓毒症AKI组22例和非AKI组36例。分别在入组后0 h、24 h、48 h、72 h采集患者尿液及血液标本,并检测血肌酐(SCr)与尿IGFBP-7、TIMP-2水平。分析SCr、尿IGFBP-7、TIMP-2与脓毒症AKI的相关性。受试者工作特征曲线(ROC)分析SCr、尿IGFBP-7及TIMP-2在脓毒症早期急性肾损伤中的诊断价值。结果与非AKI组比较,AKI组各时点SCr、尿IGFBP-7、尿TIMP-2水平均升高,差异具有统计学意义(F/P=147.848/<0.001、330.849/<0.001、136.777/<0.001)。SCr与尿IGFBP-7、尿TIMP-2均呈正相关(r/P=0.680/<0.001,0.392/0.002),尿IGFBP-7与尿TIMP-2呈正相关(r/P=0.618/<0.001)。ROC曲线分析显示,SCr、尿IGFBP-7、尿TIMP-2及尿IGFBP-7联合TIMP-2预测脓毒症AKI的曲线下面积(AUC)分别为0.586、0.907、0.738、0.914,尿IGFBP-7、尿IGFBP-7联合TIMP-2优于SCr(Z/P=4.117/<0.001,4.196/<0.001);尿IGFBP-7、尿IGFBP-7联合TIMP-2优于尿TIMP-2(Z/P=2.663/0.008,3.378/0.008)。结论尿IGFBP-7、尿TIMP-2与尿IGFBP-7联合TIMP-2均可以作为早期预测脓毒症急性肾损伤发生的指标,且尿IGFBP-7、尿IGFBP-7联合TIMP-2的预测价值优于尿TIMP-2。 展开更多

目的:检测尿液中基质金属蛋白酶-2(MMP-2)及基质金属蛋白酶组织抑制剂-2(TIMP-2),探讨MMP-2及TIMP-2在诊断梗阻性膀胱顺应性改变中的应用价值。方法:2016年11月—2020年1月期间前列腺增生致膀胱顺应性改变患者资料。研究组患者尿动力检... 目的:检测尿液中基质金属蛋白酶-2(MMP-2)及基质金属蛋白酶组织抑制剂-2(TIMP-2),探讨MMP-2及TIMP-2在诊断梗阻性膀胱顺应性改变中的应用价值。方法:2016年11月—2020年1月期间前列腺增生致膀胱顺应性改变患者资料。研究组患者尿动力检查存在膀胱出口梗阻(BOO),且膀胱顺应性(BC)20≤BC<40 mL/cmH2O;对照组为随机抽取前列腺增生引起BOO者,且BC≥40 mL/cmH2O。留取患者尿液并行血肌酐及胱抑素C检测,同时收集尿液于―80℃冻存待检测。尿样解冻后采用ELISA试剂盒测定MMP-2及TIMP-2尿液中含量。结果:37例患者纳入研究组,两组间年龄、IPSS评分、体重指数(BMI)、膀胱过度活动症评分(OABSS)差异无统计学意义(P>0.05);除BC外两组间尿动力学参数(残余尿量、最大逼尿肌力、膀胱初感觉容量等)差异无统计学意义(P>0.05)。研究组尿液MMP-2及TIMP-2含量分别为(0.75±0.26)ng/mL、(359.8±123.7)pg/mL,高于对照组(0.52±0.21)ng/mL、(289.8±101.9)pg/mL,差异具有统计学意义(P<0.05)。结论:尿液中MMP-2与TIMP-2表达情况与梗阻因素导致的BC改变密切相关。 展开更多

The relationship between angiogenesis and eukaryotic translation initiation factor 4E (EIF4E) expression level in non Hodgkin lymphoma (NHL) was studied. Mean microvessel density (MVD) and EIF4E were detected in 52 lymph node samples paraffin sections of patients with newly diagnosed NHL by the way of immunohistochemistry. Antisense EIF4E cDNA was cloned into plasmid pcDNA3.1 (+) and transfected into Raji cells. A series of angiogenesis related factors,including vascular endothelial growth factor (VEGF), matrix metalloproteinases 9 (MMP-9) and tissue inhibitor of metalloproteinases-2 (TIMP-2) proteins were detected by Western blot. The results showed that: (1) The Expression of EIF4E and MVD was higher in aggressive lymphomas than in indolent lymphomas(P〈0.05)and the expression of EIF4E was positively correlated with MVD in lymph node of NHL(r=0. 695, P〈0.01). (2) Antisense EIF4E eukaryocytic expression vector (pcDNA3. 1-EIF4Eas) was constructed successfully. (3) EIF4E, VEGF and MMP-9 were expressed at high levels in Raji cells as compared to normal human peripheral blood monocular cells (NHPMC), and blockage of EIF4E expression brought down the expression of VEGF and MMP-9. However, TIMP-2 was undetectable in Rail cells, although a moderate level of TIMP-2 was detected in NHPMC. It was concluded that the increased EIF4E expression was associated with aggressive property of NHL. 展开更多
