BACKGROUND: Previous studies have demonstrated that reduced estrogen levels may accelerate the formation of advanced glycation endproducts (AGE) in brain tissue, raise the concentration of lipid peroxidation products in vivo, and speed up deterioration of learning and memory. A tonifying kidney recipe is hypothesized to improve the ability of learning and memory in ovariectomized rats by downregulating AGE and lipid peroxidation products. OBJECTIVE: To simulate a postmenopausal state, bilateral ovariectomy (OVX) was performed in rats, and the effects of tonifying kidney recipe (TKR) on AGE and lipid peroxidation in the rat cerebral cortex, hippocampus, and blood serum levels was measured. In addition, the effects on learning and memory were evaluated, and the effect of AGE -specific inhibitor aminoguanidine (AG) was compared with TKR. DESIGN, TIME AND SETTING: A randomized, in vivo, control experiment was performed at the scientific research center (Provincial Key Laboratory) in the Fourth Hospital of Hebei Medical University (Shijiazhuang, Hebei Province, China) from May 2005 to January 2007. MATERIALS: Forty healthy, adult, female, Sprague Dawley rats were used for this study. TKR was composed of prepared rehmannia rhizome, epimedium herb, desert-living cistanche, and Szechwan lovage rhizome, which were provided by Shijiazhuang Medical Materials Company (China). A TKR extraction was prepared for further use. AG was provided by Sigma (USA). Forty rats were randomly divided into four groups: sham, OVX, AG, and TKR, with 10 rats in each group. METHODS: The rat ovaries were resected in the OVX, AG, and TKR groups, whereas the same volume of fat was resected in the sham group. At four weeks after OVX, the AG group received 1% AG water solution by lavage; the TKR group was administrated by lavage once per day at a dose of 6.3 g (crude drug)/kg; OVX and sham groups received equal volumes of tap water. MAIN OUTCOME MEASURES: Learning and memory behavior of rats was tested in a Y-electric maze 16 weeks after the OVX procedure. The contents of advanced glycation endproducts in the rat cerebral cortex, hippocampus, serum, and urine were detected by competitive ELISA and spectrofluorophotometer. The contents of lipid peroxidation in rat serum, cerebral cortex, and hippocampus were assayed using a biochemical method. RESULTS: Compared with the sham group, serum content of advanced glycation endproducts in the OVX group was significantly increased, and lipid peroxidation content increased in cerebral cortex, hippocampus, and serum (t = 3.04-4.22, P 〈 0.05 0.01). Both AG and TKR decreased the amount of AGE in cerebral cortex and serum (t = 2.53, 3.64, P 〈 0.05, 0.01), increased AGE urine content (t = 3.25 4.87, P 〈 0.01), and decreased lipid peroxidation content in cerebral cortex, hippocampus, and serum (t = 2.80 3.70, P 〈 0.05 0.01). In comparison to the OVX and sham groups, the correct escape rate in the Y-electric maze was significantly increased (t = 3.46, 3.28, P 〈 0.01), and escape latency was significantly decreased (t=3.12, 2.48 P 〈 0.05) in the AG and TKR groups, which indicated that both AG and TKR improved learning and memory The OVX group had a significantly lower correct escape compared with the sham group (t = 4.21, P 〈 0.01 ).CONCLUSION: The tonifying kidney recipe decreased deposition of advanced glycation endproducts and lipid peroxidation in ovariectomized rats, and concomitantly improved learning and memory. The effect of TKR was equal to that of AG. 展开更多

Tonifying Shen strategy works as an important alternative and complementary method that is widely used to treat osteoporosis in Traditional Chinese Medicine(TCM) based on the proper identification of Zheng. Shen deficiency Zheng is one of the main types of osteoporosis while Shen-Yin and-Yang deficiencies represent two basic principles/types of Shen deficiency. Currently, Tonifying Shen strategy, in particular, the Tonifying Shen-Yin and-Yang principles, has been demonstrated to exert osteoprotective effects. However, the mechanisms by which Tonifying Shen strategy and/or Tonifying Shen-Yin and-Yang principles function in treating osteoporosis are still not clearly understood. Here we first briefly explore current understanding of Tonifying Shen strategy, such as Tonifying Shen prescriptions, herbs, and effective components, in treating osteoporosis. Furthermore, the mechanism of Tonifying Shen-Yang principle in treating osteoporosis is reviewed from the clinic experience, animal, and mechanistic investigation. Moreover, the mechanisms by which Tonifying Shen-Yin principle in treating osteoporosis are established. Finally, we compare the similarity and difference between Tonifying Shen-Yang and-Yin principles in treating osteoporosis. Our findings indicate that both principles exert bone-protective effects in treating osteoporosis by simultaneously stimulating the differentiation of mesenchymal stem cells(MSCs) into osteoblast and the differentiation of hematopoietic stem cells(HSCs) into osteoclast. However,Tonifying Shen-Yin principle is in favor of increasing bone mineral density(BMD) while Tonifying Shen-Yang principle is in favor of osteoporosis-related syndromes. Our findings indicate that Tonifying Shen-Yin and-Yang principles not only share some similarity but also obtain some difference in treating osteoporosis. 展开更多

Herbal Medicines used to tonify Kidney and ameltorate the marrow microenvironmentwere applied in the treatment of 50 patients of aplastic anemia. Stanazol was also used to some cases.Forty-one cases (82. 0 % )responded well. It is significantly better than the 56. 5 % response among thosetreated with stanazol alone. Experimentally, the herbal medicines appear to stimulate hemopoietic pro-genitor cell proliferation, improve the marrow microenyironment and repair marrow damage induced bycyclophosphamide in vivo. The hemopoietic promoting effect might be partly due to the desialation of themarrow stroma cells. The herbal medicines also exert a positive effect on immune system by increasingleukocyte phagocytic activity and elevating the interleukin-2 production. Moreover , the decrease of super-oxide dismutase resulting from cyclophosphamide injection could be reversed by the use of the herbalmedicines. In conclusion, to tonify the Kidney and ameliorate marrow microenvironment medicines pro-duce broad biological results that are beneficial to the body. 展开更多

Objective: To observe the acting pattern on the immune system by Kidney tonifying (KT) drugs. Methods: Three composite recipes were used on the 7- and 14-day corticosterone rat model to study the effect of recipes on the different patterns of immuno-modulatory and neuro- endocrinological systems. Results:The immune system of Spleen Invigorating(SI) group was well protected in either 7- or 14-day experiments, with the SI recipe showing no effect on the neuroendocrine system. It suggested that SI recipe might have direct action on immune system. On the other hand, KT group with no effect on neuroendocrine-immune (NEI) system in 7-day experiment, but showed a remarkable protective effect of the whole NEI system in 14-day experiment. Conclusion: The results indicated that KT recipe acts on the neuroendocrine system first, and then influence the immune system. It means that the action took place through down pathway of NEI network. 展开更多

Objective Mesenchymal stem cells(MSCs) represent a promising population for supporting new clinical concepts in cellular therapy, and they can be reprogrammed into induced pluripotent stem cells(iPSCs) by defined factors. Methods This method opened up a new era of stem cell research, because the transplantation rejection of iPSCs is the bottleneck of its clinical application, so seeking alternative compounds and animal origin diagnostic reagents to achieve full chemical iPSCs is to be done to solve this problem. Results The application of these iPSCs has largely been associated with well known undesirable effects such as the development of cancers in certain experimental models. This has called for the search and use of reprogramming factors that are safe. Chinese materia medica(CMM) with tonifying kidney function(TKF) offers an alternative source. On the other hand, human umbilical cord mesenchymal stem cells(hUMSCs) are known to be a "young" source of MSCs, hUMSCs transplantation is an attractive approach for acute kidney injury repair. Therefore, In this study, we investigated whether the treatment of CMM with TKF on hUMSCs could enhance the repair in mice model of acute kidney injury after transplantation. Conclusion Our results showed that the treatment of hUMSCs with kidney tonifying CMM increased their multipotency, improved the renal function of mice and enhanced subsequent homing to the injured kidney in an acute kidney injury mice model. 展开更多

Cerebral ischemia-reperfusion injury(CIRI) refers to a pathological phenomenon that aggravates the injury after the restoration of blood perfusion and oxygen supply to the cerebral ischemia-induced tissues and organs,... Cerebral ischemia-reperfusion injury(CIRI) refers to a pathological phenomenon that aggravates the injury after the restoration of blood perfusion and oxygen supply to the cerebral ischemia-induced tissues and organs, with a relatively high incidence. The traditional Chinese medicine(TCM) believes that Qi deficiency and blood stasis are the cause of CIRI. Therefore, Chinese medicine for tonifying Qi and activating blood is regarded as an important choice for the treatment of CIRI. In recent years, it has been found that many Chinese herbal medical ingredients and compound Chinese medicine(CCM) have significant anti-CIRI effects, and their mechanisms of action mainly include improving brain blood supply, neuroprotection, regulating signal pathways such as TLR4/HO-1/Bcl-2, protecting mitochondrial function, regulating related protein levels, and regulating oxidative molecule levels. This article summarizes and introduces the pharmacological mechanisms of Tonifying-Qi and activating-blood Chinese medicine and CCM which have the function of anti-CIRI. Our goal is to provide effective reference for further researches on the cerebral protection of related TCMs or compounds and their clinical application. 展开更多

Objective: To observe the effect of tonifying Kidney herbs (TKH) on pituitary gland, ovary, and adrenal gland of androgen sterilized rats (ASR).Methods: ASR model was established by injecting testosterone propionate subcutaneously to Sprague-Dawley (SD) female rats of 9 days age. Morphological and endocrinological change of pituitary gland, ovary and adrenal gland in rats of 100–107 days old before and after feeding TKH extract were observed by light and electron microscope, cell cultures, immunohistochemical studies, and radioimmunoassay. Results: In the ASR groups, there were intracytoplasmic lipid drops, crinophagy, vacuole, and autophagy in pituitary gland, anovulation in ovary, and hyperplasia of reticular zone in adrenal gland. At the same time, serum follicular stimulating hormone and luteinizing hormone decreased (P < 0.05-0.005) and serum testosterone and dehydroepiandrosterone levels increased significantly (P < 0.01-0.001). These morphologiacl and endocrinological changes were all returned to normal range after the TKH treatment. 展开更多
