[Objectives]This study was conducted to study the total polyphenol content and antioxidant capacity of different extraction fractions of Pholidota chinensis.[Methods]Different polar fractions of P.chinensis were obtained by ultrasonic extraction method, and determined for polyphenol contents by UV spectrophotometry, and their antioxidant activity was evaluated by DPPH free radical scavenging and ferric ion reducing antioxidant power(FRAP).SPSS analysis software was used to analyze the correlation between total polyphenol contents and antioxidant activity of different extracts.[Results]The contents of total polyphenols in different polar fractions ranged from 1.52 to 6.41 mg/g,and the order from low to high was:water<petroleum ether<ethyl acetate<95%ethanol.The reduction capacity of the total antioxidant capacity method ranged from 12.54 to 22.45μmol/L,and the order was 95%ethanol>ethyl acetate>petroleum ether>water. The DPPH median inhibition concentration ranged from 0.35 to 2.65 mg/ml,and the order was water>petroleum ether>ethyl acetate>95%ethanol.The total polyphenol content of P.chinensis was best in the 95%ethanol extraction fraction, which showed the strongest total antioxidant capacity,so the total polyphenol content of the 95%ethanol fraction was positively correlated with its antioxidant capacity, while the DPPH free radical-scavenging capacity was better in the water fraction, indicating that the free radical-scavenging capacity of the water fraction was better.[Conclusions]This study provides a theoretical basis for the further study of the chemical components of P.chinensis and the development and utilization of natural antioxidants. 展开更多

The microwave-low-pressure process(MWLP)of total polyphenols from Chaenomeles speciosa fruit was studied, and the advantages of MWLP were further evaluated by comparing with ultra high pressure(UHP)and microwave-assisted extraction(MAE).The influences of liquid/solid ratio, extraction time, pressure, and ethanol concentration on the performance of MWLP were investigated.Thereafter, the interactive variables were further optimized by the stepwise multiple quadratic regression model on the basis of the previous univariate analysis. The results showed that temperature as an intermediate variable in MWLP significantly affected the yields of polyphenols and 3-o-caffeoyl-quinic acid, which was determined by pressure and ethanol concentration. The optimized parameters were proved to be valid because the results predicted by the stepwise multiple quadratic regression model equations fit well with the experimental results.Compared with UHP,the predominance of MWLP was that the extraction time was shortened and the cost of extraction equipment was lowered.MWLP is an effective technology since MWLP was superior to MAE based on extraction yield, solvent loss and reproducibility. 展开更多

This study discusses the composition analysis and comparison of Jaboticaba under different processing conditions for fermented liquid, jam, fruit powder, and fruit vinegar. The differences and comparison of the contents of total polyphenols, anthocyanins, resveratrol, superoxide dismutase, small molecular peptides and ellagic acid in their products were analyzed. After analysis and comparison of results, the content of total polyphenols was found to be the highest 44.82 ± 0.89 (mg/g) after being fermented directly from fresh fruit. The fresh fruit was dried at low temperature and ground into powder, with a maximum ellagic acid content of 12.799 ± 0.12 (mg/g). Fresh fruit was then made into jam, with the highest anthocyanin content of 57.39 ± 1.20 (mg/g). The fruit vinegar was produced by fermentation. Except for the content of small molecule peptides, which was similar to the content of fermentation broth and fruit powder, they were 20.63 ± 1.61, 23.84 ± 2.51 and 20.52 ± 1.21 (mg/g), and the rest of the composition was not as good as other samples. In the fresh fruit fermentation broth samples, resveratrol was produced and detected, and its content was 2.14 (mg/L), while it was not detected in other samples, and a superoxide dismutase (SOD)-like enzyme was detected in the fresh fruit fermentation broth. The highest activity was 49002.5 units/mL. Jaboticaba is prepared using a fermentation broth processing method with fresh fruit, and its total polyphenol content is higher than that of fruit powder, jam, and fruit vinegar. The commercial value of Jaboticaba and the establishment of composition content data are relatively improved, which is more promising for the future. 展开更多

The fruit of Eucalyptus globulus Labill.,also known as“Yikouzhong(YKZ)”in China, belongs to the myrtaceae family. The aim of this study was to characterize the chemical composition of YKZ extract by GC-MS combined with LC-MS/MS.A total of 34 compounds were initially identified by GC-MS and 54 compounds were identified by LC-MS/MS positive and negative total ion flow maps. The total phenols and flavonoid-like substances were quantified using an enzyme standardization method to determine the presence of phenolic substances in them.At the same time,a network pharmacology approach based on LC-MS/MS was used to construct a“drug-component-target”network for myocardial protection, and 696 potential anti-myocardial fibrosis targets were obtained. The PPI analysis identified 72 core targets, and the enrichment analysis revealed that the core targets may exert anti-myocardial fibrosis effects by regulating Lipid and atherosclerosis and Proteoglycans signaling pathways.Taken together, these results suggest that YKZ anti-cardiac fibrosis may act through multiple components and targets, and this study provides a scientific basis for the functional development and application of YKZ against myocardial fibrosis. 展开更多

Milk thistle(Silybum marianum)is a crucial medicinal plant containing a large amount of oil.In the study, the changes in storage oil during seed germination and seedling transition from heterotrophic phases were investigated. The results showed that seed oil decreased from 19.53%to 0.88%on the 7th day of seedling development.Oil hydrolysis continued until the 4th day of germination with a low slope, but then increased the use of oils in seed germination end seedling growth metabolism. The results indicated that the quantitative changes in fatty acids, presented at lower amount, were relatively higher than dominant fatty acids. There were decreasing phenolic content in the developing seedlings, but overall, lowest level of total phenolic content can be attributed to the control(30.52 mg⋅100 g⋅Oil^(-1)).In contrast, the maximum peroxide value(2.58 meq⋅kg Oil^(-1))in the developing seedling was observed on the last day of the experiment. The results showed that there was a significant correlation between saturated fatty acid, unsaturated fatty acid, and lipase activity. However, the correlation between lipase activity and polyunsaturated fatty acids was significantly higher than between lipase activity and monounsaturated fatty acids(R^(2)=90%and R^(2)=77%,respectively).Therefore,the lipolysis process acts selectively in milk thistle oils.According to the results,C12:0 exhibits a greater impact on the early seedling growth rather than on the germination process and is one of the determining factors in the transition from heterotroph to autotroph. Also,it can be a marker for TAGs breakdown. 展开更多

This study was carried out to characterize total residual catechins and their fractions, polyphenolic contents and antioxidant activities of black teas enriched with high levels of theaflavin-3,3’-digallate. The made teas were processed from eleven selected cultivars. A comparative study was carried out between the processed teas and those from commercially grown Kenyan cultivars in relation to the above chemical parameters. A correlation matrix analysis was also conducted to find out whether a relationship existed between the antioxidant activities and the said chemical parameters. The total residual catechins were found to range between 3.10% and 8.08%. The total polyphenol levels varied between 19.00% and 28.90%, while the antioxidant activities of the teas ranged from 82.70% to 91.70%. There was a significant p < 0.001 correlation between the antioxidant activity and total polyphenols (r = 0.8948). There was also a high correlation p < 0.001 between the antioxidant activity and total catechins (r = 0.8878). Out of the four catechin fractions, the antioxidant activity correlated most with EGCG (r = 0.8774). The total polyphenolic contents and antioxidant activities for most of the cultivars were comparable to those of the green tea reference standard. From the figures obtained, it can be concluded that the most of the newly developed black teas of the selected cultivars have higher quality and enhanced antioxidant activities and that they can be recommended for commercial production. 展开更多

(Scrophulariaceae) is a highly sought after medicinal plant with therapeutic properties as cognition enhancer as well as for other brain and body functions. Research was conducted to optimize a thin cell layer explant based micropropagation system to assist mass propagation. Thin cell layers (TCL) derived from leaf and internode segments were used as explants. Murashige and Skoog medium was used to formulate shoot induction, elongation, and rooting media. Shoot induction media were prepared by supplementing three concentrations (0.1, 1.0, and 10.0 μM) of four cytokinins 6-benzylaminopurine, 2-isopentenyl-adenine, 6-3-Hydroxybenzylaminopurine, and thidiazuron to study adventitious shoot bud induction response. An optimum shoot bud induction response was observed on MS medium supplemented with 10.0 μM 6-benzylaminopurine for both leaf and stem transverse thin cell layer (tTCL) explants. The average number of shoot buds from leaf tTCL explants was 59, whereas, on an average, 33 shoot buds were regenerated from internode tTCL explants. Elongation of adventitious shoot buds was achieved best in a liquid medium using Liquid Lab Rocker® system. Elongated shoots recorded 100% rooting in MS medium supplemented with 5 μM indole butyric acid. Bacopa micropropagation employing tTCL explants for initial shoot bud induction and using LLR® boxes in subsequent elongation step can achieve cost effective way to regenerate high volume of plantlets and biomass required for herbal industry. Leaf and stem tTCL explants both were suitable for Agrobacterium tumefaciens (EHA105) mediated genetic transformation. Successful transformation was scored within three days of co-cultivation with Agrobacterium suspension on the basis of Enhanced Green Fluorescent Protein (EGFP) expression as an early and non-destructible screening device. Transformation frequencies of 83% and 76% were accomplished for leaf and stem tTCL explants, respectively. Greenhouse grown Bacopa plants were analyzed as fresh and dry methanolic extracts for total polyphenol content (811.93 ± 7.98 and 814 ± 17.64 GAE mg g-1) and the Trolox Equivalent Antioxidant Capacity values were 1918.25 ± 173.12 and 3163.14 ± 403.25 μmol/g, respectively. 展开更多

More and more Parkia biglobosa plants’ stem barks, and roots are collected for traditional medicine uses. This work aimed to quantify Parkia biglobosa plant parts total polyphenols, total flavonoids contents and their antioxidant activities. The hypothesis was that the plant parts would have different phytochemical contents. So, leaves, stem barks, root barks and mature fruit pulp were collected. Alongside the leaves, the stem and root barks were sliced, air-dried at room temperature for 30 days. Thereafter, the samples were crushed and sieved. Following this, the powders were extracted with distilled water by maceration and decoction. As a result, the decoction was more efficient than the maceration. Leaves had the highest total polyphenol content (14.68), followed by stem barks (11.69), and root barks (9.09 ± 0.43 mg GAE/g) (0.0001 ≤ p ≤ 0.0002). However, for total flavonoid contents, the stem barks were better than the other parts. Indeed, stem barks delivered 0.74 and were followed by leaves for 0.6, and root bark for 0.49 ± 0.02 mg EQ/g (0.0001 ≤ p ≤ 0.0169). Finally, the antioxidant activities were 3.07 and 2.86 ± 0.1 μmol T.E/g, respectively for stem bark, and leaves (p = 0.0532). In conclusion, there is no need to debark the stems, because, in decoction, leaves and stem barks would have the same efficacy. 展开更多

Numerous plants are used to cure or prevent diseases. These plants are promising candidates for functional foods products. The most frequent form in which they are used is an aqueous infusion or decoction. In this study, we compared the antioxidant properties of ten Peruvian plants infusions and investigated their relation to the phenolic content. The studied plants were: Uncaria tomentosa (cat’s claw), Lepidium meyenii (maca), Berberis vulgaris L. (barberry, agracejo), Phyllantus niruri (chanca piedra), Annona muricata L. (graviola, soursop), Gentianella alborosea (hercampure), Geranium dielsianum (pasuchaca), Tabebuia ochracea (tahuari), Notholaena nivea (“cuti cuti”) and Tiquilia paronychioides (“flor de arena”). Infusions of all studied plants have shown antioxidant activity, though there was a large diversity between the results. The antioxidant properties, determined with DPPH and ABTS scavenging assays as well as FRAP test, were strongly correlated with total phenolic content, while there was no correlation with the carotenoid content. 展开更多
