越冬对池塘专养模式下中华鳖(Trionyx sinensis)机体形态、血清生化、脏器相关功能酶活力及肌肉和裙边物性的影响 被引量:4

水生动物往往会通过调整自身生理生化特征而形成各自固有的越冬生存适应对策,因此,深入探究越冬对具有冬眠习性的目标水产养殖动物机体生理状况的影响过程与机制,对指导其安全养殖具重要参考价值。以池塘专养模式下2^(+)龄中华鳖(Triony... 水生动物往往会通过调整自身生理生化特征而形成各自固有的越冬生存适应对策,因此,深入探究越冬对具有冬眠习性的目标水产养殖动物机体生理状况的影响过程与机制,对指导其安全养殖具重要参考价值。以池塘专养模式下2^(+)龄中华鳖(Trionyx sinensis)同生群个体为研究对象,从机体形态、血清生化、脏器相关功能酶活力和肌肉与裙边物性等四个维度开展了中华鳖越冬前后生理生化特征的差异研究,以进一步了解越冬对中华鳖生理生化的影响特征。结果表明:(1)越冬期间实验中华鳖总体上呈现较明显的生长态势,所测30项体尺性状和12项质量性状测定值间的差异相似性分别为46.7%和8.3%;(2)所测16项血清生化指标中,越冬后显著降低的为TG等5项(P<0.05),显著增加的为TP等4项(P<0.05),无显著差异的为GLB等7项(P>0.05);(3)所测8项脏器酶学指标中,越冬后酶活力显著下降的为肝脏CAT、POD和脂肪酶(P<0.05),显著升高的为胃蛋白酶等相关酶指标(P<0.05);(4)所测7项物性指标中,越冬后裙边弹性和咀嚼性均显著增加(P<0.05),其余为下降或无显著差异(P>0.05)。表明越冬为引起肝脏功能发生正常切换的重要时期,但不会引起心肝功能产生实质性改变。研究结果可为中华鳖越冬生理生态学研究和池养中华鳖越冬管理技术体系构建提供基础资料。 展开更多

Study on Preventive Effect of Cecropin and Lysozyme on Skin Fester Disease in Trionyx sinensis 被引量:1

[ Objective ] This study was to investigate the preventive effect of cecropin and lysozyme on skin fester disease in Trionyx sinensis. [ Method ] By employing intramuscular injection method, the preventive effect of mixtures of cecropin (10 mg/kg) and lysozyme( 10 mg/kg) and of cecropin(6 mg/kg) and lysozyme( 5 mg/kg) were challenged on Aeromonas encheleia-caused skin fester disease in T. sinensis. The mortality and relative percent survival were used evaluate the preventive effect of different drugs. The serum agglutination titer, cytophagocytosis and bactericidal activity were detected to compare the effect of drugs on the immunity and disease resistance of T. sinensis. [ Result] The residue concentrations of gentamicin and lysozyme in serum were compared to evaluate the side effect of drugs used challenged. The results showed that combined use of cecropin and lysozyme gave the best preventive effect and immunity in T. sinensis, with an extremely significant difference from other treatments ( P 〈 0.01 ). Analysis of drug residue in serum indicated that lysozyme was completely metabolized 6 days after medication, while gentamicin residue was still observed 12 days later. [ Conclusion] The combined application of cecropin and lysozyme showed better preventive effect than antibiotics in fester disease of T. sinensis. Moreover, no drug residue and side effect were observed in the combined use treatment. This method avoids the side effect of antibiotics, worthy of extension at commercial scale. 展开更多

Purification Effect of Trionyx sinensis Breeding Wastewater by a Synthetic Biological Pond

A synthetic biological pond composed of aquatic plants, a microorganism and aquatic animals was applied in the purification of the wastewater discharged from a Tnonyx sinensis breeding greenhouse, and the removal effects of TN, NH4+ -N, TP and COD in the wastewater were analyzed. The results show that after the breeding wastewater was purified by the synthetic biological pond for about five months, TN, NH~ -N, TP and COD concentration in the breeding wastewater were reduced by 78%, 92%, 81% and 60% respectively, while DO content in the breeding wastewater increased by about four times, and there was no obvious change in water pH; previous yellow or brown and turbid sewage became yel- lowish and slightly turbid, and it had no smell. It is obviously seen that the purification effect of T. sinensis breeding wastewater by the synthetic biological pond composed of aquatic plants, a microorganism and aquatic animals is obvious, and the synthetic biological pond has good promotion and application prospects. 展开更多

Trionyx sinensis Hemorrhagic Syndrome Virus(TSHSV)is an arterivirus newly discovered in Chinese softshell turtles.Little is known about the effect of antibodies against the virus or the distribution of the virus in different organs of infected turtles.In this study,a partial protein of TSHSV-HP4 was produced using a prokaryotic expression system,and its polyclonal antibody was generated.The polyclonal antibody was confirmed by western blot and dot enzyme-linked immunosorbent assay(dot-ELISA).The distribution of TSHSV in different organs of T.sinensis was examined by immunohistochemistry(IHC)and the expression of immune-related genes was analyzed using quantitative real-time polymerase chain reaction(qRT-PCR).The results indicated that the recombinant TSHSV-HP4 protein was successfully expressed,and the generated polyclonal antibody showed specific binding to viral particles in the lung tissues of infected turtles.The IHC assay indicated that the virus was highly localized in various cells,including intestinal lymphocytes,enterocytes,kidney epithelial cells,spleen cells,lung macrophages,and cardiomyocytes.The qRT-PCR analysis revealed that TSHSV was detected in all organs tested,including the lungs,liver,kidneys,spleen,and heart.The numbers of viral mRNA copies in lung and heart tissues were significantly higher in the virus-antibody group than in the virus group.The interferonstimulated genes(ISGs),myxovirus resistance protein 2(MX2)and radical S-adenosyl methionine domain containing 2(RSAD2)were highly upregulated in all groups of infected turtles.Antibody-dependent enhancement(ADE)seemed to occur after stimulation by the polyclonal antibody,because significantly greater expression of the two genes was detected in the virus-antibody group than in the virus group.Overall,these results are important in understanding the cell localization of TSHSV and the immune response of infected turtles. 展开更多
