Objective To determine the optimum extraction method and condition of poly-saccharides fromTrollius chinensis (TCP).Methods With the yield of TCP as index and water as solvent,reflux extraction,ultrasonic extraction a...Objective To determine the optimum extraction method and condition of poly-saccharides fromTrollius chinensis (TCP).Methods With the yield of TCP as index and water as solvent,reflux extraction,ultrasonic extraction and supercritical CO_2 extraction were employed to determine the efficient extraction method of TCP.Moreover,the conditions of reflux extraction were optimized by single factor test and orthogonal test.The content of polysaccharide was measured with phenol-sulfuric acid method.Results The data displayed that the crude polysaccharide(CPS)yield of regular reflux extraction was(11.7±0.12)%,superior to ultrasonic extraction and supercritical CO_2 extraction.The optimum conditions of reflux extraction were as follows:extraction temperature of 95℃,extraction time of 3hand the liquid-solid ratio of 20∶1.Under these conditions,the polysaccharide yield was (1.45±0.03)%,which was well in close agreement with the predicted value.Conclusion Regular reflux extraction was regarded as the optimal extraction method of TCP.The optimum conditions of reflux extraction were as follows:extraction temperature of 95℃,extraction time of 3hand the liquid-solid ratio of 20∶1.展开更多
AIM: To study the absorption properties and mechanism of two important components, trolline and veratric acid, from the flowers of Trollius chinensis, in order to better understand the contribution of these two compou...AIM: To study the absorption properties and mechanism of two important components, trolline and veratric acid, from the flowers of Trollius chinensis, in order to better understand the contribution of these two compounds to the effectiveness of these flowers. METHOD: The human Caco-2 cell monolayer model was employed to study the transport of trolline and veratric acid from apical side(AP) to basal side(BL), and from BL to AP by determining the transport rates as the function of time and concentration and calculating apparent permeability coefficients(Papp). RESULTS: Trolline and veratric acid were transported across Caco-2 cell monolayer through different mechanisms in a concentration dependent manner.Trolline was transported at a Papp level of 10-6 cm·s-1 with a Papp AP→BL/Papp BL→AP ratio of more than 1.8 or less than 0.8, while veratric acid was transported at a Papp level of 10-5 cm·s-1 with a Papp AP→BL/Papp BL→AP ratio of close to 1.0. CONCLUSION: Trolline is moderately absorbed through an associative mechanism involving active and passive transport, and veratric acid is well-absorbed mainly through passive diffusion. These factors should be taken into account when chemically assessing the pharmacodynamic material basis of the flowers of T. chinensis.展开更多
Acid hydrolysis mechanisms of orientin-2"-O-galactopyranoside(OGA),orientin and other flavone C-glycosides in the trollflowers[Trollius chinensis Bunge) were studied in this report for the first time.Hydrolysis pa...Acid hydrolysis mechanisms of orientin-2"-O-galactopyranoside(OGA),orientin and other flavone C-glycosides in the trollflowers[Trollius chinensis Bunge) were studied in this report for the first time.Hydrolysis parameters including temperature,acidity,solvent and reaction time were comprehensively investigated.OGA could be hydrolyzed to orientin,followed by an isomerization to isoorientin via a reversible Wessely-Moser rearrangement reaction under stronger acidic conditions.A first-order kinetic model fitted the hydrolysis process of OGA well.Under the optimal hydrolysis conditions of 80 ℃,1.0 mol/L H^+ and 7 h reaction time,about 77%OGA was transformed to orientin with no detectable isoorientin.These results could be helpful for better understanding of the acid hydrolysis kinetics of flavone C-glycosides,as well as the preparation of these valuable components under controlled acid hydrolysis conditions.展开更多
