Objective:To evaluate the antioxidant activities and total phenolic contents of brown seaweeds belonging to Turbinaria spp.[Turbinaria conoides(T.conoides)and Turbinaria ornata(T.ornata)collected from Gulf of Mannar of southeastern coast of India in various in vitro systems.Methods:The antioxidant activity was evaluated using different in vitro systems,viz.,I,I-diphenyl-2-picrylhydrazyl(DPPH),2,2'-azino-bis-3 ethylbenzothiozoline-6-sulfonic acid diammonium salt(ABTS),H_20_2/H0 radical scavenging,Fe^(2+)ion chelating ability,and reducing potential.Folin-Ciocalteu method was used to determine the total phenolic content of the extracts,and the results were expressed as mg of gallic acid equivalents(GE)/g of the seaweed extracts.Thiobarbituric acid-reactive substances assay was employed to assess the ability of the seaweed extracts to inhibit lipid oxidation.Results:Ethyl acetate(EtOAc)fraction of T.conoides registered significantly higher phenolic content(105.97 mg GE/g)than that of T.ornata(69.63 mg GE/g).Significantly higher antioxidant potential as determined by DPPH(64.14%)radical scavenging activity was registered in EtOAc fraction of T.ornata.A higher ABTS(?)radical scavenging(IC_(50)3.16μg/mL),Fe^(2+)chelating(IC_(50)0.46 mg/mL),H_2O_2 scavenging(IC_(50)4.25 mg/mL),lipid peroxidation inhibitory(TBARS,IC_(50)0.21 mg/mL),and reducing abilities(IC_(50)52.67 mg/mL)(P<0.05)were realized in EtOAc fraction of T.ornata than other fractions.Conclusions:This study indicated the potential use of T.conoides and T.ornata as candidate species to be used as food supplements/functional foods to increase shelf-life of food items for human consumption,and nutraceuticals to deter deleterious free radical-induced life-threatening diseases. 展开更多

In this study,the biosorption of Malachite green(MG)onto Turbinaria conoides,brown marine algae,was studied with respect to initial pH,temperature,initial dye concentration,and sorbent dosage.The optimum initial pH and temperature values for MG removal were found to be 8.0 and 30℃,respectively.Sorbent dosage was found to strongly influence the removal of MG.Equilibrium studies were carried out to test the validity of the Langmuir(q_(max)=66.6 mg/g and b=0.526 mL mol/L)and the Freundlich(n=1.826 and K=3.751 mg/g)isotherms.The kinetic studies indicated the validity of the pseudo first-order and second-order equation. 展开更多

Safety of Malaysian marine endophytic fungal extract S2 from a brown seaweed Turbinaria conoides

Objective:To evaluate the in vivo acute toxicity and antioxidant activity of the marine endophytic fungus extract S2 isolated from Turbinaria conoides.Methods:Two doses(100 mg/kg and 400 mg/kg)of the S2 extract were administered to rats orally for acute toxicity and antioxidant test.The body weight,relative weight of six organs,haematological,biochemical and antioxidant properties were investigated on Day 14.Results:A single oral dose treatment did not cause any mortality or observable adverse effects in rats.No significant variations in the body and organ weights between the control and the treated groups were observed.Heamatological analysis and clinical blood chemistry also did not reveal any toxic effects of the extract.The total white blood cell count and haemoglobin levels were increased.The levels of total serum cholesterol in males treated with 100 and 400 mg/kg were significantly(P<0.05)decreased(1.28 and 1.34 mmol/L respectively)compared to control(1.55 mmol/L)rats.Pathologically,neither gross abnormalities nor histopathological changes were observed.This study showed strong evidence of the non-toxic effects of S2 extract.Furthermore the extract exhibited significant(P<0.05)antioxidant activity through increased levels of superoxide dismutase and glutathione peroxidase enzy mes in serum,liver and kidney.Conclusions:The research findings from the present study showed the potential of marine natural products particularly in Malaysia as a source of bioactive compounds.Marine endophytic fungi as a potential source of anticancer drugs with great potential as they are potent yet safe,thus deserving further extensive investigation. 展开更多

Effect of fucoidan from Turbinaria ornata against marine ornamental fish pathogens

Objective:To evaluate the antimicrobial capacity of fucoidan from the brown seaweed Turbinaria ornata against the marine ornamental fish bacterial pathogens.Methods:Fucoidan was isolated by the ethanol extraction method and the functional groups were identified by Fourier Transform Infrared Spectroscopy analysis.Subsequently,structural characterization was done by 1H NMR analysis.In vitro antibacterial activity of fucoidan was performed by the agar plate diffusion method and Minimum Inhibitory Concentration.Results:The characteristic C-0-S bending vibration of sulfate substituent the axial C-4 was observed at 839 cm^(-1).Characterisitic signal of the fucoidan was detected in a different ppm of 1H NMR analysis.The maximum antibacterial activity(16.23±0.11)mm was obtained for Vibrio parahaemolyticus and the minimum activity(5.1±0.24)mm was recorded for Yersinia enterocolitica.The Minimum Inhibitory Concentration value was recorded between 2.5 to 10 mg/mL to the respective pathogens.Conclusions:The present study proved that fucoidan possessed the significant antibacterial activity against the tested fish bacterial pathogens.It could be further used as a natural antibiotic in an aquaculture system to control the bacterial diseases.However,the present study suggested that the further in vivo study is required to get the better understanding of the mode of action of fucoidan. 展开更多

Objective:To study ecological parameters and biochemical composition of brown seaweed,Turbinaria ornata(T.ornata)collected from Hurghada shores,Red Sea coast of Egypt during September,October and November,2015.Methods:T.ornata and its associated seaweeds were collected,identified and their abundances were estimated.Water of collection site was analyzed physicochemically as well as qualitative and quantitative analyses of phytoplankton.T.ornata was analyzed for protein,total carbohydrate,lipids,alginic acid,agar,pigments,minerals and heavy metals.Results:The results showed that macroalgal species recorded along Hurghada shores belong to Phaeophyta,Rhodophyta and Chlorophyta.At collection site,the moderate temperature,slight alkaline pH,low turbidity,high dissolved oxygen and valuable nutrient content of saline water exerted the massive growth of T.ornata with maximum abundance(24%)during October.The phytoplankton community was quite diverse with a maximum numbers of taxa(104.2×10^(8) cell/L)recorded during October.Analysis of T.ornata alga powder showed that high soluble carbohydrate(2.80±0.10 mg/g dry/weight)and chlorophyll c(0.0017±0.0001 mg/g fresh weight)contents were recorded during September;while high contents of protein(37.70±0.60 mg/g dry weight),lipids(3.10±0.06 mg/g dry weight),polysaccharides(agar and alginates),carotenoids(0.0160±0.0004 mg/g fresh weight),minerals and heavy metals were recorded during November.Conclusions:The study revealed that physicochemical analyses of water were varied slightly during the three months and suitable for the growth of T.ornata.It contains high amount of most biochemical constituents during October. 展开更多

Objective:To investigate the potential antibacterial activities of ethanol extracts of Turbinaria ornata(T.ornata),Oleic acid(OA)and palmitic acid(PA)extracted from T.ornata as well as mixtures of OA and PA(1:1)against some bacterial species.Methods:Brown seaweed T.ornata was collected from Hurghada shores,Red Sea coast of Egypt.OA and PA were extracted from T.ornata.Ethanol extracts of T.ornata,OA,PA and mixtures of these two fatty acids(1:1)were tested for their antibacterial activities against Escherichia coli,Bacillus cereus,Bacillus subtilis,Klebsiella pneumonia and Staphylococcus aureus by the disc diffusion method.Results:Ethanol extracts of T.ornata,OA,PA and mixtures of OA and PA(1:1)showed antibacterial activities that increased significantly(least significant difference at 0.05 level)with increasing concentrations against all tested bacteria.Different concentrations of ethanol extracts of T.ornata and extracted OA showed its highest activity against Bacillus subtilis,while PA and mixtures of PA and OA(1:1)showed its highest activity against Bacillus cereus.The maximum inhibition activities were shown for mixtures of OA and PA(1:1).Scanning electron microscope showed that mixtures of OA and PA(1:1)caused plasmolysis and reduction in cell size of Escherichia coli.Conclusions:Different concentrations of T.ornata and its fatty acids showed activities against all tested bacteria.Therefore,it is a potential source of natural antimicrobial compounds. 展开更多

本研究通过120d的室内培养实验研究了盾形陀螺珊瑚(Turbinaria peltata)、根枝藻(Rhizoclonium sp.)和点斑篮子鱼(Siganus guttatus)之间的相互作用关系.结果显示,高质量浓度的NO3-(5.00~10.00mg/dm3)刺激了丝状绿藻根... 本研究通过120d的室内培养实验研究了盾形陀螺珊瑚(Turbinaria peltata)、根枝藻(Rhizoclonium sp.)和点斑篮子鱼(Siganus guttatus)之间的相互作用关系.结果显示,高质量浓度的NO3-(5.00~10.00mg/dm3)刺激了丝状绿藻根枝藻在缸内的爆发式生长,在实验27d时根枝藻在缸壁的面积覆盖率达到90.5%,藻丝长度为10~15cm.在海藻的胁迫作用下,盾形陀螺珊瑚的PSⅡ最大光化学量子产量(Fv/Fm)均值逐渐从0.712降至0.626,降幅为10.7%~13.5%.放入4尾4~6cm长的点斑篮子鱼后,实验缸壁根枝藻的面积覆盖率在2d内从90.5%下降至10.0%.但解除根枝藻胁迫后盾形陀螺珊瑚的Fv/Fm值并没有明显变化,与对照组的差异也不显著;随着海藻胁迫因子的消除,盾形陀螺珊瑚的Fv/Fm值逐渐升高,并在第120天时恢复至初始状态.本研究结果从珊瑚光合作用效率的角度验证了啃食者的存在确能缓解富营养化对造礁珊瑚的不利影响,为“下行控制假说”提供了新的证据. 展开更多

Objective:To evaluate the antimicrobial activity of Turbinaria conoides(T.conoides).Padina gymnospora(P.gymnospora) and Sargassum tenerrimum against human bacterial and fungal pathogens.Methods:The antimicrobial activities of the extracts against various organisms were tested by using disc diffusion method.Results:The methanol extract showed the better result than the other extracts.Whereas,the strong antibacterial inhibition was noted in methanol extracts of P.gymnospora against Bacillus subtilus(26.33±1.86) and the mild inhibition of ethanol extracts from T.conoides against Klebsiella pneumoniae(2.33±0.51).Acetone extraction of P. gymnospora had strong antifungal inhibition against Cryptococcus neoformans(23.00±1.78), and acetone extract of T.conoides had mild inhibition against Aspergillus niger(3.00±0.89). Conclusions:The seven different solvent extracts of seaweeds used in the present study have shown significant bacterial action.Further,a detailed study on the principle compound in the seaweeds which is responsible for antimicrobial activity is still needed and it can be achieved by using advanced separation techniques. 展开更多

三亚鹿回头海域盾形陀螺珊瑚和精巧扁脑珊瑚反射率光谱分析

为了进一步深入研究不同形状和不同颜色珊瑚的光谱特征,选择三亚湾鹿回头海域两种常见造礁石珊瑚(褐色片状珊瑚:盾形陀螺珊瑚(Turbinaria peltata)和蓝灰色块状珊瑚:精巧扁脑珊瑚(Platygyra daeda))为样本进行测量和分析。于2015年7月2... 为了进一步深入研究不同形状和不同颜色珊瑚的光谱特征,选择三亚湾鹿回头海域两种常见造礁石珊瑚(褐色片状珊瑚:盾形陀螺珊瑚(Turbinaria peltata)和蓝灰色块状珊瑚:精巧扁脑珊瑚(Platygyra daeda))为样本进行测量和分析。于2015年7月22日上午采集两种珊瑚样品各7组。样品块大小~6 cm,并将其暂养于中国科学院海南热带海洋生物重点实验站岸基实验室珊瑚养殖缸,养殖缸内水温控制在~26℃。待样品块暂养≥4小时后用光纤光谱仪测量其反射率,光谱采集条件为无云遮挡的晴天。所用光纤光谱仪(海洋光学USB2000+),波段为200~850 nm,光谱分辨率1.34 nm,步长0.6 nm,视场角为25°。珊瑚样品置于缸内的平台上,过滤后恒温~26℃的海水持续注入以保证缸内水温恒定;多余的海水自动从养殖缸上壁溢出以排除因光线折射入水体后引起的"汇聚现象";养殖缸内壁采用黑色尼龙布贴壁,以避免玻璃缸壁光线反射对测量结果的影响。光纤光谱仪的探头与样品间距保持在5 cm,每个样品重复测量10次取平均值以代表该样品的光谱反射率。测量光源为太阳光,每次测量前校正一次光谱仪,选用可见光波段的反射率光谱进行数据分析。反射率光谱导数分析可以放大光谱间的差异,四阶导数光谱法在提高检测灵敏度、改善分辨率和加强抗干扰力等方面具有独特的优点,故此对所测珊瑚光谱反射率数据进行反射率光谱数据一阶导数、二阶导数和四阶导数分析,根据盾形陀螺珊瑚和精巧扁脑珊瑚反射率光谱导数之间的差异确定两种珊瑚光谱的敏感可区分波段。分析结果发现,可见光范围内两种珊瑚反射率差异明显;后者反射率光谱明显高于前者,仅~700 nm出现类似较高反射率。盾形陀螺珊瑚反射率介于4%~15%之间,波峰和波谷明显。400~450 nm反射率相对较低约为4%~5%; 480 nm后急升至~10%, 502, 578, 604和652 nm附近为明显波峰;随后激增至700 nm的~36%。精巧扁脑珊瑚反射率介于6%~16%之间; 400~420 nm波长附近反射率值相对较低,为~6%; 420~470 nm急剧升高至~15%, 486 nm附近出现宽大波峰,为该珊瑚的特征峰; 486, 577, 607和650 nm处也存在四个明显波峰;随后剧增至700 nm的~37%。光谱反射率导数分析结果表明盾形陀螺珊瑚和精巧扁脑珊瑚可区分波段为:一阶导数483.7~492.6, 496.2~500和533.5~540.5 nm。二阶导数414~422.7, 499.4~504, 520.2~523.3, 534.2~536.6, 557.5~561和671.8~675 nm。四阶导数414~417.6, 427.4~430.3, 433.4~436.5, 452.3~455.5和657.1~659.1 nm。 展开更多

Antibacterial Effects of Extracts of Two Types of Red Sea Algae

Introduction: Intestinal bacteria are exposed many external influences, including drugs, causing the emergence of strains resistant to the effects of antibiotics. Consequently, the discovery of new antibiotics that affect resistant strains is required. Marine algae offer a source of renewable natural compounds with antimicrobial effects. Therefore, the aim of this study was to detect some of these compounds and examine their impact on enteric bacteria. Methodology: Escherichia coli, Salmonella typhi, Shigella dysenteriae, Klebsiella pneumoniae, and Enterobacter aerogenes were tested with extracts of Turbinaria triquetra and Halimeda opuntia extracted with methanol, ethanol, petroleum ether, or dimethyl formamide solvents. We measured bacterial growth inhibition, the minimal inhibitory concentrations (MICs), and potassium leakage, and analyzed the bacterial cells with scanning electron microscopy and energy-dispersive X-ray spectroscopy. Results: The T. triquetra extract produced with methanol strongly affected the bacteria tested. When the results for T. triquetra and H. opuntia were compared with those of omacillin, the T. triquetra and H. opuntia extracts in most solvents were more effective than the antibiotic. Differences in the bacterial growth inhibition and MICs depended on the type of alga and the solvent used. At the end of the incubation period, potassium leakage had increased by 62.98% for E. coli, 61.24% for S. typhi, 61.32% for S. dysenteriae, 64.02% for K. pneumoniae, and 63.10% for E. aerogenes when treated T. triquetra. Conclusion: Turbinaria triquetra extracted with methanol strongly affected the growth of the bacteria tested. Therefore, it is a potential source of natural antibacterial compounds. 展开更多

【目的】探究不同环境条件对盾形陀螺珊瑚(Turbinaria peltata)摄食及共生藻光合生理的影响。【方法】测量不同养殖温度(24、28、32℃)、pH(7.6、7.8、8.0、8.2)、无机盐浓度(0.1、10μmol/L)条件下盾形陀螺珊瑚摄食率、共生虫黄藻密度... 【目的】探究不同环境条件对盾形陀螺珊瑚(Turbinaria peltata)摄食及共生藻光合生理的影响。【方法】测量不同养殖温度(24、28、32℃)、pH(7.6、7.8、8.0、8.2)、无机盐浓度(0.1、10μmol/L)条件下盾形陀螺珊瑚摄食率、共生虫黄藻密度、净光合速率、呼吸速率等指标变化。【结果】与对照组(24℃)相比,温度升高(28、32℃)条件下盾形陀螺珊瑚摄食率提高、共生虫黄藻密度降低,其净光合速率提高、呼吸速率下降(P<0.05);低pH条件(7.6、7.8)可降低盾形陀螺珊瑚的摄食率、共生虫黄藻密度,降低净光合速率和暗呼吸速率(P<0.05);高无机盐条件(10μmol/L)显著提升珊瑚的摄食率、共生虫黄藻密度,同时提高净光合速率、降低呼吸速率(P<0.05)。【结论】环境因子的变化对盾形陀螺珊瑚的摄食速率、共生虫黄藻密度、光合呼吸比均有显著影响,在人工育苗的过程中需针对不同珊瑚的特点进行相应的养殖条件设置。 展开更多

Objective:To investigate the antimalarial and larvicidal activities of seaweeds against Plasmodium falciparum strain andAnopheles stephensi mosquito larva. Methods: The three different seaweeds,Jania rubens,Turbinaria ornata (T. ornata) and Ulva fasciata were collected from Southeast India. The seaweeds were extracted using hexane and ethyl acetate solvents and evaluated for their mosquito-larvicidal and antiplasmodial property. The bioactive compounds of the seaweed extract were studied by Fourier transform infrared spectroscopy and gas chromatography-mass spectrometry. Results:The hexane extract ofT. ornata (HTO) has shown significant inhibitory effects at the IC50 value 22.98 μg/mL, when compared to the positive control, chloroquine (12.32 μg/mL). The larvicidal activity of HTO on the fourth instar larvae of Anopheles stephensi was found to be effective with the LC50 and LC90 values (μg/mL) of 1.38 and 3.34. The Fourier transform infrared spectroscopy analysis ofHTO revealed the presence of alkenes, aromatics, amides and amines groups, whereas in gas chromatography-mass spectrometry, 11 different bioactive components were recorded. The bioactive component, hentriacontane in the HTO might be responsible for the recorded antiplasmodial and larvicidal activities. Conclusions:The finding of the present study reveals that the constituents of T. ornata have new sources of antimalarial and larvicidal agents in future. 展开更多
